Structure-function relationships of antifungal monohydroxy unsaturated fatty acids (HUFA) of plant and bacterial origin.

This study investigated the relationships between the structures of hydroxy unsaturated fatty acids (HUFA) and their antifungal activities. Structurally diverse HUFA, including four monohydroxy-18:1 isomers, two monohydroxy 18:2 isomers and two monohydroxy 18:2 isomers were extracted from seeds of plants (Coriaria nepalensis, Thymus vulgaris, Mallotus philippensis and Dimorphotheca sinuata) for which information was available on PlantFAdb database, and from culture supernatants of lactobacilli. They were purified by high-speed counter current chromatography (HSCCC) and identified by LC-MS/MS.

Industrial wastewater treatment plant enriches antibiotic resistance genes and alters the structure of microbial communities.

Antibiotic resistance is an emerging global health crisis, driven largely by overuse and misuse of antibiotics. However, there are examples in which the production of these antimicrobial agents has polluted the environment with active antibiotic residues, selecting for antibiotic resistant bacteria and the genes they carry. In this work, we have used shotgun metagenomics to investigate the taxonomic structure and resistance gene composition of sludge communities in a treatment plant in Croatia receiving wastewater from production of the macrolide antibiotic azithromycin. We found that the tota

The structurally unique photosynthetic Chlorella variabilis NC64A hydrogenase does not interact with plant-type ferredoxins.

Hydrogenases from green algae are linked to the photosynthetic electron transfer chain via the plant-type ferredoxin PetF. In this work the [FeFe]-hydrogenase from the Trebouxiophycean alga Chlorella variabilis NC64A (CvHydA1), which in contrast to other green algal hydrogenases contains additional FeS-cluster binding domains, was purified and specific enzyme activities for both hydrogen (H2) production and H2 oxidation were determined. Interestingly, although C. variabilis NC64A, like many Chlorophycean algal strains, exhibited light-dependent H2 production activity upon sulfur deprivation, C

Effects of plant-induced hypervitaminosis D on cutaneous structure, cell differentiation and cell proliferation in cattle.

Solanum glaucophyllum (Sg) (synonym S. malacoxylon) is a plant toxic to cattle due to its high levels of 1,25-dihydroxyvitamin D3 as glycoside derivatives. Sg causes a disease characterized by wasting and calcification of soft tissues. The effects of vitamin D are not only important in calcium homeostasis, but also in immune regulation, cell growth and cell differentiation. Skin samples in Sg-intoxicated and control heifers were studied histologically. Cellular differentiation and proliferation were analysed by immunohistochemical expression of cytokeratins, involucrin and proliferating cell n

Evolution, structural diversity and functional plasticity of plant derived multifunctional proteases and α-amylases inhibitors.

Plant derived multifunctional inhibitors (MFIs) are an important class of plant defense proteins that disrupt insect digestion by simultaneously impairing protein and carbohydrate catabolism. With the increasing knowledge of inhibitors of plant enzymes, MFIs have become a separate functional group. In this review we propose an extended definition that encompasses the classical inhibitors acting on structurally unrelated enzymes and also inhibitors acting on enzymes of the same substrate pathway, e.g. α-amylase/α-glucosidase and trypsin/chymotrypsin systems. Most of the MFIs consist of single p

SPINet: structure-enhanced point cloud instance segmentation network for multi-plant rice phenotyping.

Accurate 3D phenotyping of rice in multi-plant scenarios is important for evaluating yield-related traits and supporting high-throughput breeding. In particular, grain-level instance segmentation from point clouds provides a direct basis for quantifying panicle structure and grain number. However, this task remains challenging because densely grown rice plants often exhibit severe inter-plant occlusion, organ adhesion, and highly similar local structures, making it difficult to simultaneously capture global plant organization and fine-grained grain geometry. To address these challenges, we pro

Linking Structural and Mechanical Properties to Multimodal Sensory Perception of Fibrousness in Plant-Based Meat Analogs.

Texture, particularly fibrousness, is a critical driver of consumer acceptance of plant-based whole-cut meat analogues (PBMAs), as it underpins the characteristic bite and tearing behavior associated with meat. Despite advances in producing and instrumentally characterizing fibrous structures in PBMAs, it remains unclear how macrostructural fiber organization and mechanical anisotropy translate into the human perception of fibrousness during eating, and how visual cues influence this perception. To address this, five PBMA samples with systematically varied fibrous structures were evaluated usi

Plant and fungal acetohydroxyacid synthases (AHAS): Methods for heterologous expression, kinetic characterization, and structural analysis.

Acetohydroxyacid synthase (AHAS; EC 2.2.1.6), also known as acetolactate synthase, catalyzes the first common step in the biosynthesis of the branched-chain amino acids valine, leucine, and isoleucine. AHAS is present in plants and microorganisms but absent in animals. For this reason, it has become an important target for the development of herbicides and antimicrobial agents. AHAS is the target of five major families of commercial herbicides, which collectively represent one of the most successful groups of herbicides used in modern agriculture. Biochemical, kinetic, and structural studies h

Neuroprotective effects of healthful plant-based diets on retinal structure: insights from a large cohort.

BACKGROUND: Retinal sublayer thickness is associated with both ophthalmic and neurodegenerative diseases, serving as a valuable biomarker. Despite the established role of diet in chronic disease prevention, the relationship between healthy dietary patterns and retinal sublayer thickness remains underexplored. OBJECTIVE: This study aims to investigate the associations between four commonly used diet assessment scores and retinal sublayer thickness in a large-scale population. METHODS: This study utilized data from the UK Biobank, including 13,993 participants with complete optical coherence t

Nitrogen cycling microbiomes are structured by plant mycorrhizal associations with consequences for nitrogen oxide fluxes in forests.

Volatile nitrogen oxides (N2 O, NO, NO2 , HONO, …) can negatively impact climate, air quality, and human health. Using soils collected from temperate forests across the eastern United States, we show microbial communities involved in nitrogen (N) cycling are structured, in large part, by the composition of overstory trees, leading to predictable N-cycling syndromes, with consequences for emissions of volatile nitrogen oxides to air. Trees associating with arbuscular mycorrhizal (AM) fungi promote soil microbial communities with higher N-cycle potential and activity, relative to microbial commu

Plant phenological asynchrony and community structure of gall-inducing insects associated with a tropical tree species.

The dynamics of occurrence of target organs in plant populations produces windows of opportunity that directly and indirectly affect the structure of herbivore communities. However, mechanisms that drive herbivore specialization between resource patches are still poorly known. In this study, we tested three hypotheses related to variation in host plant phenology and community structure (i.e., composition, richness, and abundance) of gall-forming species: (a) plants with early leaf-flushing in the season will have greater vegetative growth and high contents of secondary chemical compounds; (b)

Relationship of soybean aphid (Hemiptera: Aphididae) to soybean plant nutrients, landscape structure, and natural enemies.

In the north central United States, populations of the exotic soybean aphid, Aphis glycines Matsumura (Hemiptera: Aphididae), are highly variable across space, complicating effective aphid management. In this study we examined relationships of plant nutrients, landscape structure, and natural enemies with soybean aphid abundance across Iowa, Michigan, Minnesota, and Wisconsin, representing the range of conditions where soybean aphid outbreaks have occurred since its introduction. We sampled soybean aphid and its natural enemies, quantified vegetation land cover and measured soybean nutrients (

Structural properties of plant and mammalian lipoxygenases. Temperature-dependent conformational alterations and membrane binding ability.

Lipoxygenases form a heterogeneous family of lipid peroxidizing enzymes, which have been implicated in the synthesis of inflammatory mediators, in cell development and in the pathogenesis of various diseases with major health and political relevance (atherosclerosis, osteoporosis). The crystal structures of various lipoxygenase-isoforms have been reported, and X-ray coordinates for enzyme-ligand complexes are also available. Although the 3D-structures of plant and animal lipoxygenase-isoforms are very similar, recent small-angle X-ray scattering data suggested a higher degree of motional flexi

Molecular structure, polymorphism, and toxicity of lantadene A, the pentacyclic triterpenoid from the hepatotoxic plant Lantana camara.

Lantadene A (22 beta-angeloyloxy-3-oxo-olean-12-en-28-oic acid), a pentacyclic triterpenoid compound from lantana (Lantana camara) leaves has been obtained in two polymorphic forms I and II. Form I had white, fluffy, and rod-shaped uniform crystals. Form II particles were irregular, shining, and polyhedral. The two forms differed in melting behavior. The powder x-ray diffraction of form I showed sharp peaks whereas from II did not contain distinct peaks. From single-crystal three-dimensional x-ray structure determination, the molecular structure of form I has been established. A/B and B/C ring

Recent gene duplication and structural remodeling drive rapid lineage-specific gene family evolution in plants.

Gene duplication promotes the generation of novel gene functions and trait diversity across species. Here, we present DupHIST, a computational pipeline that reconstructs the hierarchical timing of gene duplications by integrating maximum likelihood (ML)-based phylogeny with substitution-derived timing via statistical smoothing. Applied to over 4.5 million genes from 114 plant genomes, we successfully inferred duplication histories across nearly 130,000 orthogroups. This large-scale analysis showed that 53.0% of genes arose from recent, lineage-specific duplications, with high concentrations in

Seasonal Effect of a Small Hydropower Plant on Benthic Biofilm Microbial Community Structure and Predicted Functional Potential.

Benthic biofilm microbial communities play a critical role in riverine primary production, nutrient cycling, and organic matter decomposition, yet their responses to hydrological alterations due to small hydropower plants (SHPs) remain poorly understood, particularly with respect to seasonal dynamics. This study conducted a one-year monitoring survey across four river sections (upstream, reservoir, dewatered reach, and downstream) in the Oujiang River Basin, Zhejiang Province, China, covering all four seasons. The results showed that the reservoir section exhibited distinct bacterial compositi

Breaking it down: Unveiling the roles of chemical structure, concentrations and technology on micropollutant biotransformation in wastewater treatment plants.

Complex mixtures of anthropogenic micropollutants (MPs) pose risks to ecosystems and health by entering the environment through multiple pathways. Microbial biotransformation is a major removal process, yet the key drivers remain poorly understood. Activated sludge (AS) treatment in wastewater treatment plants (WWTPs) acts as a partial barrier and reduces MP emissions into aquatic ecosystems, yet removal efficiencies vary widely across MPs. Here, we studied the biotransformation of 189 MPs in AS from six Swiss WWTPs operated at three treatment conditions: carbon elimination (Celim), nitrificat

MYB transcription factors in plants: A comprehensive review of their discovery, structure, classification, functional diversity and regulatory mechanism.

The MYB transcription factor (TF) family is one of the largest families in plants and performs highly diverse regulatory functions, particularly in relation to pathogen/pest resistance, nutrient/noxious substance absorption, drought/salt resistance, trichome growth, stamen development, leaf senescence, and flavonoid/terpenoid biosynthesis. Owing to their vital role in various biological regulatory processes, the mechanisms of MYB TFs have been extensively studied. Notably, MYB TFs not only directly regulate targets, such as phytohormones, reactive oxygen species signaling and secondary cell wa

Legacy Effects of Plant Community Structure Are Manifested in Microbial Biofilm Development With Consequences for Ecosystem CO2 Emissions.

To better understand linkages between hydrology and ecosystem carbon flux in northern aquatic ecosystems, we evaluated the relationship between plant communities, biofilm development, and carbon dioxide (CO2) exchange following long-term changes in hydrology in an Alaskan fen. We quantified seasonal variation in biofilm composition and CO2 exchange in response to lowered and raised water table position (relative to a control) during years with varying levels of background dissolved organic carbon (DOC). We then used nutrient-diffusing substrates (NDS) to evaluate cause-effect relationships bet

Prioritised identification of structural classes of natural products from higher plants in the expedition of antimalarial drug discovery.

The emergence and spread of drug-recalcitrant Plasmodium falciparum parasites threaten to reverse the gains made in the fight against malaria. Urgent measures need to be taken to curb this impending challenge. The higher plant-derived sesquiterpene, quinoline alkaloids, and naphthoquinone natural product classes of compounds have previously served as phenomenal chemical scaffolds from which integral antimalarial drugs were developed. Historical successes serve as an inspiration for the continued investigation of plant-derived natural products compounds in search of novel molecular templates fr
