In incompatible plant-pathogen interactions, disease resistance is generated by rapid activation of a multitude of plant defence reactions. Little is known about the dependency of these resistance responses on external factors. The plasticity of plant defence mechanisms in terms of light conditions is studied here. Interaction of Arabidopsis thaliana (L.) Heynh. with an avirulent strain of Pseudomonas syringae pv. maculicola in the dark resulted in increased apoplastic bacterial growth and therefore reduced local resistance as compared to an infection process in the presence of light. Several

Transgenic tobacco (Nicotiana tabacum) with altered levels of mitochondrial alternative oxidase (AOX) were used to examine the potential role of this electron transport chain protein in resistance to tobacco mosaic virus. We examined the effect of AOX expression on the salicylic acid-induced resistance in susceptible plants and the resistance responses of plants harboring the N-gene. A lack of AOX did not compromise the ability of salicylic acid treatment to heighten the resistance of susceptible plants. In plants with the N-gene, a lack of AOX did not compromise the ability of the hypersensit

Members of the MILDEW RESISTANCE LOCUS O (MLO) gene family confer susceptibility to powdery mildews in different plant species, and their existence therefore seems to be disadvantageous for the plant. We recognized that expression of the Arabidopsis MLO2 gene is induced after inoculation with the bacterial pathogen Pseudomonas syringae, promoted by salicylic acid (SA) signaling, and systemically enhanced in the foliage of plants exhibiting systemic acquired resistance (SAR). Importantly, distinct mlo2 mutant lines were unable to systemically increase resistance to bacterial infection after ino

This review highlights the pivotal role of Plant Growth Promoting Rhizobacteria (PGPR) in association with nanotechnology for enhancing plant growth and health. It narratively discusses diverse PGPR groups and genera, outlining their direct mechanisms such as phosphate solubilization, nitrogen fixation, ACC deaminase activity, hydrogen cyanide production, and siderophore production for iron chelation as well as indirect mechanisms through induced systemic resistance (ISR) and systemic acquired resistance (SAR). The application of PGPR as single inoculants and in consortia, together with omics-

Immune responses in eukaryotes involve rapid and profound transcriptional reprogramming. Although mechanisms regulating the amplitude of defense gene expression have been extensively characterized, those controlling the speed of defense gene induction are not well understood. Here, we show that the Arabidopsis Elongator subunit 2 (AtELP2) regulates the kinetics of defense gene induction. AtELP2 is required for rapid defense gene induction and the establishment of full basal and effector-triggered immunity (ETI). Surprisingly, biological or chemical induction of systemic acquired resistance (SA

After a hypersensitive response to invading pathogens, plants show elevated accumulation of salicylic acid (SA), induced expression of plant defense genes, and systemic acquired resistance (SAR) to further infection by a broad range of pathogens. There is compelling evidence that SA plays a crucial role in triggering SAR. We have transformed tobacco with two bacterial genes coding for enzymes that convert chorismate into SA by a two-step process. When the two enzymes were targeted to the chloroplasts, the transgenic (CSA, constitutive SA biosynthesis) plants showed a 500- to 1,000-fold increas

Salicylic acid (SA) plays an important role for plant immunity, especially resistance against biotrophic pathogens. SA quickly accumulates after pathogen attack to activate downstream immunity events and is normally associated with a tradeoff in plant growth. Therefore, the SA level in plants has to be strictly controlled when pathogens are absent, but how this occurs is not well understood. Previously we found that in Arabidopsis (Arabidopsis thaliana), HISTONE DEACETYLASE 6 (HDA6), a negative regulator of gene expression, plays an essential role in plant immunity since its mutation allele sh

Trichoderma atroviride is a root-colonizing fungus that confers multiple benefits to plants. In plants, small RNA (sRNA)-mediated gene silencing (sRNA-MGS) plays pivotal roles in growth, development, and pathogen attack. Here, we explored the role of core components of Arabidopsis thaliana sRNA-MGS pathways during its interaction with Trichoderma. Upon interaction with Trichoderma, sRNA-MGS-related genes paralleled the expression of Arabidopsis defense-related genes, linked to salicylic acid (SA) and jasmonic acid (JA) pathways. SA- and JA-related genes were primed by Trichoderma in leaves aft

Nitric oxide (NO), a small yet highly reactive gaseous molecule, has emerged as a crucial signaling component in plant biology, mainly in mediating its defense responses against biotic stress. Apart from mammalian role of NO, it also actively involved in the broad physiological, biochemical, and immunological processes in plants. NO is synthesized through a set of multiple enzymatic and non-enzymatic pathways, whereupon it rapidly accumulates upon pathogen recognition. NO is implicated in plant immunity by working in synergy with reactive oxygen species (ROS) in triggering the hypersensitive r

One grand challenge for studying plant biotic and abiotic stress responses is to optimize plant growth and plasticity under variable environmental constraints, which in the long run benefits agricultural production. However, efforts in promoting plant immunity are often accompanied by compromised morphological "syndromes" such as growth retardation, sterility, and reduced yield. Such a trade-off is dictated by complex signaling driven by secondary messengers and phytohormones. Salicylic acid (SA) is a well-known phytohormone essential for basal immunity and systemic acquired resistance. Intere

Culture filtrate of Lasiodiplodia theobromae increased respiration rate, phenylalanine ammonia lyase activity, and levels of hydrogen peroxide, lipid peroxides and salicylic acid in B. nigra plants. Salicylic acid (SA) level increased for 1 hr of interaction and reduced later. Development of systemic acquired resistance (SAR) was found restricted in plants infected with L. theobromae due to deficiency of SA, which is a major signal for development of SAR. Exogenously supplied SA did develop resistance and plant death was delayed. It was hypothesized that deficiency of SA could be due to jasmon

BACKGROUND: Addressing global food security under rapid population growth and climate change requires sustainable strategies to protect crops from pathogens while reducing reliance on agrochemicals. Salicylic acid (SA), a key phytohormone in plant defense, is a promising natural elicitor for crop protection. However, its rapid metabolism and phytotoxicity restrict practical use. To overcome these limitations, we developed SA-amino acid conjugates designed to improve protective efficacy of SA while minimizing its toxicity.
RESULTS: Six SA-amino acid conjugates were synthesized through optimize

Plant defence responses to various biotic stresses via systemic acquired resistance (SAR) are induced by avirulent pathogens and chemical compounds, including certain plant hormones in volatile form, such as methyl salicylate and methyl jasmonate. SAR refers to the observation that, when a local part of a plant is exposed to elicitors, the entire plant exhibits a resistance response. In the natural environment, plants are continuously exposed to avirulent pathogens that induce SAR and volatile emissions affecting neighbouring plants as well as the plant itself. However, the underlying mechanis

Detecting plant-derived signal molecules using fluorescent probes is a key topic and a huge challenge for scientists. Salicylic acid (SA), a vital plant-derived defense hormone, can activate global transcriptional reprogramming to systemically express a network of prominent pathogenesis-related proteins against invasive microorganisms. This strategy is called systemic acquired resistance (SAR). Therefore, monitoring the dynamic fluctuations of SA in subcellular microenvironments can advance our understanding of different physiological and pathological functions during the SA-induced SAR mechan

Phytohormones play essential roles in plant-nematode interactions through complex crosstalk. Although these hormones often accumulate in nematode-resistant plants, the roles of abscisic acid (ABA) and ethylene (ET) in rice (Oryza sativa) resistance to the root-knot nematode (RKN) Meloidogyne graminicola (Mg) remain unclear, particularly regarding concentration dependency and underlying mechanisms. Using exogenous hormone gradient treatments, we show that only high concentrations of ABA (200 µM) and ET-releasing compound ethephon (Eth, 500 µM) induce systemic nematode resistance. High-concentra

Plant surface-localized pathogen recognition receptors (PRRs) perceive conserved microbial features, termed pathogen-associated molecular patterns (PAMPs), resulting in disease resistance. PAMP perception leads to calcium influx, MAPK activation, a burst of reactive oxygen species (ROS) mediated by RbohD, accumulation of the defense hormone salicylic acid (SA), and callose deposition. Lectin receptor-like kinases (LecRKs) belong to a specific PRR family and are important players in plant innate immunity. Here, we report that LecRK-IX.2 is a positive regulator of PRR-triggered immunity. Pathoge

Epigenetic modifications have emerged as an important mechanism underlying plant defence against pathogens. We examined the role of JMJ14, a Jumonji (JMJ) domain-containing H3K4 demethylase, in local and systemic plant immune responses in Arabidopsis. The function of JMJ14 in local or systemic defence response was investigated by pathogen growth assays and by analysing expression and H3K4me3 enrichments of key defence genes using qPCR and ChIP-qPCR. Salicylic acid (SA) and pipecolic acid (Pip) levels were quantified and function of JMJ14 in SA- and Pip-mediated defences was analysed in Col-0 a

Pathogenesis-related (PR) proteins are critical defense signaling molecules induced by phytopathogens. They play a vital role in plant's defense signaling pathways and innate immunity, particularly in systemic acquired resistance (SAR) and serve as key molecular markers of plant defense. Overexpressing PR genes, such as chitinase, thaumatin, glucanase, thionin and defensin, either individually or in combination, have significantly boosted plants' defense responses against various pathogens. However, signaling pathways regulating the expression of these versatile proteins remain only partially

The Agrobacterium rhizogenes rolD gene, coding for an ornithine cyclodeaminase involved in the biosynthesis of proline from ornithine, has been inserted in Lycopersicon esculentum cv Tondino with the aim of studying its effects on plant morphological characters including pathogen defense response. The analysis of plants transgenic for rolD did not show major morphological modifications. First generation transgenic plants however were found to flower earlier, and showed an increased number of inflorescences and higher fruit yield. Transformed plants were also analysed for parameters linked to p
