Barnyardgrass EcPIP2;4 and EcPIP2;5 are associated with altered quinclorac redistribution in Arabidopsis.

Barnyardgrass (Echinochloa crus-galli var. zelayensis) is a major weed in rice fields, and reduced sensitivity to the auxin-type herbicide quinclorac has been increasingly reported. Our previous work found two plasma membrane intrinsic protein genes, EcPIP2;4 and EcPIP2;5, that showed differential quinclorac-induced expression between susceptible and resistant barnyardgrass populations, suggesting their possible involvement in quinclorac translocation. Here, quinclorac-related phenotypes in Arabidopsis lines carrying EcPIP2;4 or population-derived EcPIP2;5 constructs were evaluated. Full-lengt

Transcription factor TaPIL1-mediated iron accumulation in wheat.

Iron (Fe) is an essential micronutrient that helps prevent iron deficiency anemia in humans. To address this problem, iron biofortification represents a promising strategy for developing iron-rich wheat cultivars. In this study, we identified the bHLH transcription factor TaPIL1 as a critical regulator of the wheat response to iron deficiency. Under Fe-deficient conditions, the expression level of TaPIL1 was increased. TaPIL1-5D overexpression (OE) lines accumulated higher iron contents in roots, shoots, and white flour derived from grain than wild type (WT), whereas the tapil1 CRISPR mutants

Expression of Trichoderma spp. endochitinase gene improves red rot disease resistance in transgenic sugarcane.

Sugarcane (Saccharum spp.)is an economically useful crop grown globally for sugar, ethanol and biofuel production. The crop is vulnerable to fungus Colletotrichum falcatum known to cause red rot disease. The pathogen hydrolyses stalk parenchyma cells where sucrose is accumulated resulting in upto 75% losses in sugar recovery. In this study, transgenic sugarcane having resistance against red rot was developed by introducing Trichoderma spp. endochitinase following Agrobacterium mediated transformation. The transgene introduction and expression in genetically modified plants were verified throug

Overexpression of HvVDE gene improved light protection in transgenic tobacco (Nicotiana tabacum).

Hosta is commonly acknowledged as a popular and preferred plant for landscaping and gardening. The 'sunburn' caused by prolonged exposure to strong sunlight is reducing the ornamental values of Hosta plants. However, there is a scarcity of research focusing on the genetic components linked to light-induced harm in Hosta . Here, the violaxanthin de-epoxidase (VDE) homolog from Hosta ventricosa was isolated and functionally identified through conducting HvVDE -overexpression tobacco (Nicotiana tabacum ) lines. The results showed that HvVDE encodes a putative protein comprising 481 amino acids wi

Catalytic and functional aspects of different isozymes of glycolate oxidase in rice.

BACKGROUND: Glycolate oxidase (GLO) is a key enzyme for photorespiration in plants. There are four GLO genes encoding and forming different isozymes in rice, but their functional differences are not well understood. In this study, enzymatic and physiological characteristics of the GLO isozymes were comparatively analyzed. RESULTS: When expressed heterologously in yeast, GLO1, GLO4 and GLO1 + 4 showed the highest activities and lowest K m for glycolate as substrate, whereas GLO3 displayed high activities and affinities for both glycolate and L-lactate, and GLO5 was catalytically inactive with

Root-specific reduction of cytokinin causes enhanced root growth, drought tolerance, and leaf mineral enrichment in Arabidopsis and tobacco.

Optimizing root system architecture can overcome yield limitations in crop plants caused by water or nutrient shortages. Classic breeding approaches are difficult because the trait is governed by many genes and is difficult to score. We generated transgenic Arabidopsis thaliana and tobacco (Nicotiana tabacum) plants with enhanced root-specific degradation of the hormone cytokinin, a negative regulator of root growth. These transgenic plants form a larger root system, whereas growth and development of the shoot are similar. Elongation of the primary root, root branching, and root biomass format

Elucidating the role of rice thioredoxin-m in mitigating copper toxicity.

The pervasive accumulation of elevated heavy metal concentrations in the environment exerts significant selective pressures, driving the progressive evolution of a comprehensive suite of adaptive strategies. These include both physiological tolerance and biochemical detoxification mechanisms. Our research identified a novel role of a rice-specific m-type thioredoxin (OsTrx-m) in conferring tolerance specifically to copper-induced heavy metal stress. Upon exposure to CuSO4, recombinant OsTrx-m underwent a rapid functional transformation from a disulfide reductase to a holdase chaperone, accompa

On-Site Visualization and Sensing Platform for Detecting Genetically Modified Papaya "55-1" in Papaya-Derived Products.

Genetically modified (GM) papaya "55-1" is the world's first papaya cultivar resistant to Papaya ringspot virus (PRSV). To facilitate government regulation and protect consumers' rights to informed choices, scientists are expected to develop a rapid and straightforward method for detecting "55-1". In this study, we established an on-site visualization and sensing platform based on recombinase polymerase amplification (RPA) to detect "55-1" in papaya-derived products. Amplified signals were generated exclusively in samples containing "55-1", with a detection limit of 20 copies, demonstrating hi

miR156-SPLs module regulates flowering and controls plant height by modulating gibberellin biosynthesis in citrus.

Plant height and optimal flowering time are key determinants of crop yield and economic value. However, the regulatory mechanisms governing these traits, particularly in woody plants, remain unclear. In this study, overexpression of a citrus microRNA156 (miR156) family member, Ci-miR156c, resulted in significant phenotypic changes in citrus, including reduced plant height and delayed flowering. miR156-mediated repression of SQUAMOSA PROMOTER-BINDING PROTEIN-LIKE (SPL) genes is a highly conserved regulatory mechanism in plants. Overexpression of OsPGIP2 confers Sclerotinia sclerotiorum resistance in Brassica napus through increased activation of defense mechanisms.

Sclerotinia stem rot (SSR), caused by Sclerotinia sclerotiorum, is the most serious disease affecting the yield of the agriculturally and economically important crop Brassica napus (rapeseed). In this study, Oryza sativa polygalacturonase-inhibiting protein 2 (OsPGIP2) was found to effectively enhanced rapeseed immunity against S. sclerotiorum infection. Leaf extracts of B. napus plants overexpressing OsPGIP2 showed enhanced S. sclerotiorum resistance by delaying pathogen infection. The constitutive expression of OsPGIP2 in rapeseed plants provided a rapid and effective defense response, which

Composite Cucurbitaceae Plants as a Tool to Study Physiological and Molecular Mechanisms of Root Development.

Composite plants are a convenient model for obtaining a whole plant with undisturbed regulatory links between the shoot and root system, while the roots carry the properties specified by the researcher. This approach allows expressing transgenes only in roots, which in some cases may be preferable over the expression of transgenes in all organs of a non-chimeric transgenic plant. Composite plants have been widely used to study the nitrogen fixation process, mycorrhizal symbioses, and the interaction of plant roots with various pathogens. The current protocol for Agrobacterium rhizogenes-mediat

Research progress on the regulatory mechanisms of the PSY promoter.

Carotenoids are essential pigments in the plant photosynthetic apparatus, functioning in light harvesting, photoprotection, and signal transduction, and serving as precursors of vital nutrients such as vitamin A. Phytoene synthase (PSY) is the first rate-limiting enzyme in the plant carotenoid biosynthetic pathway, and its transcriptional regulation primarily depends on cis-acting promoter elements, associated transcription factors, and epigenetic status. The PSY promoter region contains core cis-elements as well as multiple light-, hormone-, and stress-responsive elements, which collectively

Novel epidermal-cortical pattern and root apical silencing of VcCPC promoter activity in Arabidopsis might suggest a joint contribution to hairless roots in blueberry.

BACKGROUND: Root hairs are specialized tubular extensions that play a pivotal role in nutrient absorption and plant-soil interactions. Typically, they originate from epidermal cells, which differentiate into either hair cells or non-hair cells. In blueberry (Vaccinium spp.), the roots are devoid of root hairs. To examine the mechanisms responsible for the hairless root phenotype, this study conducted anatomical and molecular biological investigations on the hairless roots of blueberry. RESULTS: The epidermal cells of blueberry roots exhibit uniformity in size and cross-sectional dimensions, w

Planthopper salivary sheath protein LsSP1 contributes to manipulation of rice plant defenses.

Salivary elicitors secreted by herbivorous insects can be perceived by host plants to trigger plant immunity. However, how insects secrete other salivary components to subsequently attenuate the elicitor-induced plant immunity remains poorly understood. Here, we study the small brown planthopper, Laodelphax striatellus salivary sheath protein LsSP1. Using Y2H, BiFC and LUC assays, we show that LsSP1 is secreted into host plants and binds to salivary sheath via mucin-like protein (LsMLP). Rice plants pre-infested with dsLsSP1-treated L. striatellus are less attractive to L. striatellus nymphs t

Pesticide Detox by Design.

Detoxification (detox) plays a major role in pesticide action and resistance. The mechanisms involved are sometimes part of the discovery and development process in seeking new biochemical targets and metabolic pathways. Genetically modified and chemical-safener-modified crops are a marked exception and often involve herbicide detox by design to achieve the required crop tolerance. This perspective evaluates the role of detox by design or chance and target-site-based selectivity in insecticide, herbicide, and fungicide action and human health and environmental effects.

DcH3.3 and DcNAC1 Regulate the Expression of UGT73A93 Involved in the Changes in Flower Colour and Fungal Resistance in Carnation.

Carnation (Dianthus caryophyllus) contains abundant flavonoid glycosides (FGs), which are important natural functional and colour components. However, there are few reports on the modification of UDP-glycosyltransferases (UGTs) in relation to flavonoids in carnation. In this study, we cloned and characterised a flavonoid 3'-O-glucosyltransferase (UGT73A93) in carnation in vitro. Overexpression of UGT73A93 in carnation and tobacco increased flavonoid glycoside accumulation, particularly kaempferol glycosides, while decreasing anthocyanin content and lightening flower colour. UGT73A93 also enhan

A visible and quantitative pEAQ-RUBY system for analysis of transcription factor-promoter regulation in plants.

Understanding how transcription factors (TFs) regulate target promoters is central to molecular biology. However, widely used reporter-based assays typically require specialized instrumentation, costly substrates, and labor-intensive procedures. The recently developed RUBY reporter gene, which enables visible betalain accumulation, offers a simple and equipment-free alternative for monitoring gene expression. Here, we established a pEAQ-RUBY reporter system for efficient analysis of TF-mediated promoter regulation. We first optimized a quantitative method for betalain extraction in tobacco and

Engineering for disease resistance: persistent obstacles clouding tangible opportunities.

The accelerating pace of gene discovery, coupled with novel plant breeding technologies, provides tangible opportunities with which to engineer disease resistance into agricultural and horticultural crops. This is especially the case for potato, wheat, apple and banana, which are afflicted with fungal and bacterial diseases that impact significantly on each crop's economic viability. Yet public scepticism and burdensome regulatory systems remain the two primary obstacles preventing the translation of research discoveries into cultivars of agronomic value. In this perspective review, the potent
