We previously reported that plasmalogens, a class of phospholipids, were decreased in a setting of dilated cardiomyopathy (DCM). Plasmalogen levels can be modulated via a dietary supplement called alkylglycerols (AG) which has demonstrated benefits in some disease settings. However, its therapeutic potential in DCM remained unknown. To determine whether an optimized AG supplement could restore plasmalogen levels and attenuate cardiac dysfunction/pathology, we placed a cardiac-specific transgenic DCM mouse model of both sexes on chow +/-1.5 % AG supplementation at ∼10 weeks of age for 16 weeks.

Glucokinase-maturity onset diabetes of the young (GCK-MODY) represents a rare genetic disorder due to mutation in the glucokinase (GCK) gene. The low incidence of vascular complications in GCK-MODY makes it a natural paradigm for interrogating molecular mechanisms promoting vascular health under prolonged hyperglycemia. Clinical rate of misdiagnosis has remained high, and a reliable serum lipid biomarker that precedes genetic screening can facilitate correct diagnosis and treatment. Herein, we comprehensively quantitated 565 serum lipids from 25 classes in 105 subjects (42 nondiabetic controls

Rhizomelic chondrodysplasia punctata (RCDP) is a rare genetic disorder caused by mutations in peroxisomal genes essential for plasmalogen biosynthesis. Plasmalogens are a class of membrane glycerophospholipids containing a vinyl-ether-linked fatty alcohol at the sn-1 position that affect functions including vesicular transport, membrane protein function and free radical scavenging. A logical rationale for the treatment of RCDP is therefore the therapeutic augmentation of plasmalogens. The objective of this work was to provide a preliminary characterization of a novel vinyl-ether synthetic plas

Ethanolamine plasmalogen (PlsEtn), a major phospholipid in neuronal membranes [60-90 mol% of ethanolamine glycerophospholipid (EtnGpl)], is specifically decreased in brains from patients with Alzheimer's disease (AD). The present study investigated how PlsEtn administration affects cognitive deficits and lipid composition in an animal model of AD. AD model rats were infused with amyloid-β (Aβ) into the cerebral ventricle and divided into three groups. Control, Egg, and Ascidian groups were then orally administered vehicle, egg yolk EtnGpl (260 μmol as EtnGpl/kg BW/day; 10 μmol as PlsEtn/kg BW/

OBJECTIVE: Oxygen toxicity causes chronic bronchopulmonary dysplasia (BPD) in extremely preterm infants. Polyunsaturated fatty acids (PUFA) and plasmalogens are the two main substrates for lipid peroxidation in the pulmonary surfactant. In the present study, we tested whether low concentrations of both were associated with development of BPD and whether both were further reduced during mechanical ventilation with oxygen.
DESIGN: Prospective, noninterventional, descriptive study.
SETTING: Level III neonatal intensive care unit in a university hospital.
PATIENTS: In 25 extremely low birth wei

The gold standard therapy for Parkinson's disease (PD), L-3,4-dihydroxyphenylalanine (L-DOPA), induces dyskinesias in the majority of patients after years of treatment. Ethanolamine plasmalogens (PlsEtn) play critical roles in membrane structure mediated functions and as a storage depot of polyunsaturated fatty acids such as docosahexaenoic acid. We previously showed that a PlsEtn precursor PPI-1011 reduced already established L-DOPA-induced dyskinesias (LID) in 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) lesioned monkeys as a PD model. We hypothesize that development of LID can be pre

Fluorescently labelled ether phospholipid (1-O-alkyl/alkenyl-2-acyl- glycerophosphocholine) readily internalizes at low temperatures (2 degrees C) after insertion into the plasma membrane of cultured fibroblasts. This fate differs markedly from that of its diacyl phospholipid analogue, which remains associated with the plasma membrane under similar conditions. Analysis by thin-layer chromatography reveals that the translocation involves transfer of the intact ether phosphatidylcholine molecules. Relative to control cells, a 2-fold increase of ether phosphatidylcholine uptake was noted when pla

Zellweger syndrome, neonatal adrenoleukodystrophy, and infantile Refsum's disease are genetic disorders characterized by the virtual absence of catalase-positive peroxisomes and a general impairment of peroxisomal functions. Recent studies in these three disorders have provided morphologic evidence of peroxisomal "ghosts" of density 1.10 g/cm3 that contain membrane proteins but lack a majority of the matrix enzyme activities. We report here the biochemical studies in a female infant with clinical features of infantile Refsum's disease whose liver and fibroblasts contained cytosolic catalase bu

Chronic inflammation in white adipose tissue (WAT) contributes to obesity-associated insulin resistance. Plasmalogens, ether phospholipids synthesized in peroxisomes, have been implicated in immune regulation. GNPAT (glyceronephosphate O-acyltransferase) is a peroxisomal enzyme that catalyzes the first and rate-limiting step of plasmalogen biosynthesis. Here, we show that dietary supplementation with alkyl glycerol (AG), a plasmalogen precursor, improves insulin sensitivity and reduces adipose inflammation in high-fat diet (HFD)-fed mice, independent of changes in body weight or invariant natu

CONTEXT: Plasmalogens are ether phospholipids implicated in neuroendocrine regulation, including reproductive function. Recent studies have suggested that circulating plasmalogen concentrations are associated with reproductive performance in dairy cows; however, practical strategies to increase these concentrations remain limited.
AIMS: We hypothesised that supplementation with Bifidobacterium longum increases circulating choline plasmalogen concentrations and that this response depends on physiological state.
METHODS: Commercial probiotic products were screened using liquid chromatography-m

It has been reported in recent years that blood levels of plasmalogens (Pls) are decreased in various diseases. None of those reports, however, conducted any clinical trials to examine the effect of Pls on those diseases. This article describes our recent report on a therapeutic efficacy of orally administered Pls in mild cognitive impairment (MCI), mild to severe Alzheimer's disease (AD), and Parkinson's disease (PD). A 24-week, multicenter, randomized, double-blind, placebo-controlled trial was performed in patients with MCI (n = 178) and mild AD (n = 98). The study design for moderate AD (n

Background: Plasmalogens (PLS) are a distinctive class of glycerophospholipids with reported neuroprotective properties; however, the mechanisms underlying their potential benefits for age-related cognitive decline remain incompletely understood. Methods: A D-galactose-induced mouse model of aging was used to evaluate the effects of PLS on cognitive performance. Hippocampal synaptic remodeling was assessed by examining the expression of synaptic proteins and receptors. An in vitro BBB model was used to evaluate the transport of PE-PLS 18:0/20:4. Molecular interaction analyses were conducted to

STUDY QUESTION: Does the metabolomic profile of the follicular fluid (FF) of patients with a diminished ovarian reserve (DOR) differ from that of patients with a normal ovarian reserve (NOR)?
SUMMARY ANSWER: The metabolomic signature of the FF reveals a significant decrease in polyunsaturated choline plasmalogens and methyl arginine transferase activity in DOR patients compared to NOR patients.
WHAT IS KNOWN ALREADY: The composition of the FF reflects the exchanges between the oocyte and its microenvironment during its acquisition of gametic competence. Studies of the FF have allowed identif

BACKGROUND: Plasmalogens are essential membrane components predominantly generated by de novo synthesis beginning in peroxisomes. Hence, patients with peroxisome biogenesis defects exhibit markedly reduced plasmalogens. Moreover, a reduction in plasmalogens has been associated with several degenerative and metabolic disorders, as well as aging. Here, we characterized the distribution and changes over time of the most abundant ethanolamine plasmalogen (PlsEtn) species in packed red blood cells (RBCs) from healthy individuals, and established age-specific reference intervals (RIs).
METHODS: Eig

Plasmalogens (Pls) as the hydrophobic bioactive compound have shown potential in enhancing neurological disorders. However, the bioavailability of Pls is limited because of their poor water solubility during digestion. Herein, the hollow dextran sulfate/chitosan - coated zein nanoparticles (NPs) loaded with Pls was prepared. Subsequently, a novel in situ monitoring method utilizing rapid evaporative ionization mass spectrometry (REIMS) coupled with electric soldering iron ionization (ESII) was proposed to assess the lipidomic fingerprint alteration of Pls-loaded zein NPs during in vitro multip

We have developed a two-step selection protocol to generate a population of Chinese hamster ovary (CHO) cell variants that are plasmalogen-deficient, but contain intact, functional peroxisomes (plasmalogen-/peroxisome+). This involved sequential exposures of a mutagenized cell population to photodynamic damage by using two different pyrene-labelled sensors, 9-(1'-pyrene)nonanol and 12-(1'-pyrene)dodecanoic acid. By this procedure we generated several isolates, all except one of which displayed a severe decrease in plasmalogen biosynthesis. Further characterization of one of the plasmalogen-def

Vitamin D3 hypovitaminosis is associated with several neurological diseases such as Alzheimer's disease, Parkinson's disease or multiple sclerosis but also with other diseases such as cancer, diabetes or diseases linked to inflammatory processes. Importantly, in all of these diseases lipids have at least a disease modifying effect. Besides its well-known property to modulate gene-expression via the VDR-receptor, less is known if vitamin D hypovitaminosis influences lipid homeostasis and if these potential changes contribute to the pathology of the diseases themselves. Therefore, we analyzed mo

One of the ways to reduce age-related changes can be a diet correction by adding biologically active substances. We studied the effect of a diet including alkyl glycerol ethers (AGs) and n-3 polyunsaturated fatty acid (PUFA) concentrate isolated from the hepatopancreas of Berrytheuthis magister squid on hematological parameters and plasmalogens level in the liver of elderly rats. The senile animals showed decrease in hemoglobin, a three-fold decrease in leukocytes, a three-fold increase in platelet count, and a double decrease of blood coagulation time in the peripheral blood. Age-related chan
