Role of polyamines in intestinal adaptation in the rat.

The cellular mechanisms controlling adaptive intestinal mucosal hypo- and hyperplasia are poorly understood but changes in tissue polyamine levels and in the activity of the key enzymes controlling their synthesis (ornithine decarboxylase: ODC) and degradation (diamine oxidase: DAO) have been implicated. Therefore, in two models of adaptive mucosal hyperplasia (pancreatico-biliary diversion, PBD, achieved by surgically transposing the jejunum to lie between pylorus and ampulla of Vater, and 90% small bowel resection, SBR-both studied 8 weeks after surgery) and in one model of hypoplasia (8 day

Structure-Function Studies and Mechanism of Action of Snake Venom L-Amino Acid Oxidases.

Snake venom L-amino acid oxidases (SV-LAAOs) are the least studied venom enzymes. These enzymes catalyze the stereospecific oxidation of an L-amino acid to their corresponding α-keto acid with the liberation of hydrogen peroxide (H2O2) and ammonia (NH3). They display various pathological and physiological activities including induction of apoptosis, edema, platelet aggregation/inhibition, hemorrhagic, and anticoagulant activities. They also show antibacterial, antiviral and leishmanicidal activity and have been used as therapeutic agents in some disease conditions like cancer and anti-HIV drug

Arginase 2 deletion leads to enhanced M1 macrophage activation and upregulated polyamine metabolism in response to Helicobacter pylori infection.

We reported that arginase 2 (ARG2) deletion results in increased gastritis and decreased bacterial burden during Helicobacter pylori infection in mice. Our studies implicated a potential role for inducible nitric oxide (NO) synthase (NOS2), as Arg2 (-/-) mice exhibited increased NOS2 levels in gastric macrophages, and NO can kill H. pylori. We now bred Arg2 (-/-) to Nos2 (-/-) mice, and infected them with H. pylori. Compared to wild-type mice, both Arg2 (-/-) and Arg2 (-/-) ;Nos2 (-/-) mice exhibited increased gastritis and decreased colonization, the latter indicating that the effect of ARG2

Alterations in polyamine metabolism induced by the pathogen Helicobacter pylori: Implications for gastric inflammation and carcinogenesis.

Helicobacter pylori is a pathogen of the human stomach that infects half of the world's population. The chronicity of the infection is a major risk factor for the development of gastric cancer. Multiple parameters, including the heterogenicity of the virulence factors, the environmental parameters, or the intensity of the host responses, regulate the evolution of the disease. Thus, we have reported that the dysregulation of the polyamine pathway in the infected gastric mucosa represents a major component of the inflammation and the risk for neoplastic transformation. Using different animal mod

Garlic-released NO and H2S modulate Arabidopsis metabolism by reshaping redox balance and redox-dependent post-translational modifications.

Nitric oxide (NO) and hydrogen sulfide (H2S) are key gasotransmitters acting as signaling molecules in plants and animals. Garlic, known for its sulfur-rich compounds, had not previously been identified as a source of NO or H2S emission. Using specific sensors, we show that garlic cloves simultaneously release both gases. To assess their biological impact, 30-day-old Arabidopsis thaliana plants were exposed for 24 h to a garlic-released NO/H2S-enriched atmosphere. Biochemical analyses revealed significant changes in components of reactive oxygen species (ROS), NO, and H2S metabolism, including

Chronic sub-lethal oxidative stress by spermine oxidase overactivity induces continuous DNA repair and hypersensitivity to radiation exposure.

In the aging process and in most degenerative diseases, the oxidant by-products of cellular metabolism lead to oxidative stress. Oxidative stress plays an important role in switching from cell proliferation to its opposite outcome, cell death. The metabolic pathways in charge of the interconversion and degradation of the polyamines are responsible for oxidant by-products. In the past few years, spermine metabolism has been found closely related to DNA oxidation and apoptosis. Moreover, that the ectopical expression of murine spermine oxidase induced DNA damage in the neuroblastoma cell line, a

[Mechanism of thymol inhibiting Botrytis cinerea: PAO-H2O2 system].

Gray mold disease caused by Botrytis cinerea infection is one of the major crop diseases. The application of environmental-friendly fungicides to control gray mold disease has been drawing great attention. Thymol, a natural compound, showed strong antifungal activity against Botrytis cinerea. We investigated the role of polyamine oxidase (PAO)-dependent hydrogen peroxide (H2O2) production in thymol-inhibited B. cinerea growth by using physiological and biochemical approaches. The results showed that: 1) Thymol significantly inhibited the growth of B. cinerea, with remarkable increases in H2O2

Acrolein produced during acute kidney injury promotes tubular cell death.

Acute kidney injury is an important global health concern as it is associated with high morbidity and mortality. Polyamines, essential for cell growth and proliferation, are known to inhibit cardiovascular disease. However, under conditions of cellular damage, toxic acrolein is produced from polyamines by the enzyme spermine oxidase (SMOX). We used a mouse renal ischemia-reperfusion model and human proximal tubule cells (HK-2) to investigate whether acrolein exacerbates acute kidney injury by renal tubular cell death. Acrolein visualized by acroleinRED was increased in ischemia-reperfusion kid

Gene dosage of the spermidine/spermine N(1)-acetyltransferase ( SSAT) gene with putrescine accumulation in a patient with a Xp21.1p22.12 duplication and keratosis follicularis spinulosa decalvans (KFSD).

Keratosis follicularis spinulosa decalvans (KFSD) or Siemens-1 syndrome is a rare X-linked disease of unknown etiology affecting the skin and the eye. Although most affected families are compatible with X-linked inheritance, KFSD appears to be clinically and genetically heterogeneous. So far, the gene has been mapped to Xp22.13p22.2 in two extended KFSD families. Analysis of additional recombination events in the first Dutch pedigree located the gene to an interval covering approximately 1 Mb between markers DXS7163 and DXS7593/DXS7105, whereas haplotype reconstruction in the second German fam

Glucocorticoid regulation of amino acid and polyamine metabolism in the small intestine.

Several factors (including diets, changes in intestinal fluora, and hormones) regulate postnatal intestinal growth and development. Based on the early studies involving modification of the adrenal gland, pituitary gland or hypothalamus, exogenous glucocorticoids and glucocorticoid receptor antagonists are now used to study glucocorticoid-mediated metabolism of amino acids in the small intestine. Findings from these studies indicate that physiological levels of glucocorticoids stimulate the catabolism of glutamine and proline for the synthesis of citrulline and arginine in enterocytes during we

Metabolic adjustments during compatible interaction between barley genotypes and stripe rust pathogen.

Stripe rust is a fungal disease that has devastated the barley production for a long time. The present study focused on the role of β-glucan, PR proteins, diamine oxidase (DAO), polyamine oxidase (PAO), key enzymes and metabolites of phenol and proline metabolism in the stripe rust resistance of barley. RD2901 with resistant behavior against stripe rust showed increased levels of PR proteins, phenylalanine ammonia lyase (PAL), tyrosine ammonia lyase (TAL) along with the accumulation of β-glucan and lignin which strengthen the plant cell wall during plant-pathogen interaction. It also depicted

Elucidation of tropane alkaloid biosynthesis in Erythroxylum coca using a microbial pathway discovery platform.

Tropane alkaloids (TAs) are heterocyclic nitrogenous metabolites found across seven orders of angiosperms, including Malpighiales (Erythroxylaceae) and Solanales (Solanaceae). Despite the well-established euphorigenic properties of Erythroxylaceae TAs like cocaine, their biosynthetic pathway remains incomplete. Using yeast as a screening platform, we identified and characterized the missing steps of TA biosynthesis in Erythroxylum coca. We first characterize putative E. coca polyamine synthase- and amine oxidase-like enzymes in vitro, in yeast, and in planta to show that the first tropane ring

2-Hydroxymelatonin mitigates cadmium stress in cucumis sativus seedlings: Modulation of antioxidant enzymes and polyamines.

Cadmium level is continuously increasing in agricultural soils mainly due to anthropogenic activities. Cadmium is one of the most phytotoxic metals in the soils. The present study investigates the possible role of 2-hydroxymelatonin (2-OHMT) in assuagement of Cd-toxicity in cucumber (Cucumis sativus L.) plants. 2-OHMT is an important metabolite produced through interaction of melatonin with oxygenated compounds. Cadmium stress decreased the activity of antioxidant enzymes and polyamines. However, exogenously applied 2-OHMT enhanced plant growth attributes including photosynthetic rate, interce

Serum spermidine oxidase activity in patients with insulin-dependent diabetes mellitus and microvascular complications.

Metabolism of polyamines (spermidine and spermine) is known to be strictly related to the growth processes of eukaryotic cells. Since cell replication processes appear altered in insulin-dependent diabetes mellitus (IDDM), especially when associated with its microvascular complications, the aim of this study was measuring serum spermidine oxidase activity (SOA), a key enzyme in the metabolic pathway of polyamines, in 47 patients with IDDM as compared with 63 healthy control subjects matched for age and sex. Mean SOA levels +/- SD were significantly lower in IDDM patients (177.4 +/- 57.2 mu kat

Arginine and polyamines in Helicobacter pylori-induced immune dysregulation and gastric carcinogenesis.

L-arginine (L-Arg) is metabolized by nitric oxide synthase and arginase enzymes. The gastric pathogen Helicobacter pylori causes peptic ulcer disease and gastric cancer. We have shown that alterations in L-Arg availability and metabolism into polyamines contribute significantly to the dysregulation of the host immune response to this infection. Nitric oxide (NO) derived from inducible NO synthase (iNOS) can kill H. pylori. There are multiple mechanisms leading to failure of this process, including competition for L-Arg substrate by H. pylori arginase, and induction of host macrophage arginase

Biocontrol efficacy and mechanism of Bacillus subtilis PL5 against root rot of sweet potato: Insights into antifungal activity and metabolite function.

Fusarium solani causes sweet potato root rot disease under optimal temperature conditions. Biocontrol using antagonistic microorganisms offers a sustainable and eco-friendly management strategy through dual mechanisms: direct pathogen suppression and induction of systemic plant resistance. Bacillus subtilis PL5 culture broth significantly reduced the plaque diameter of F. solani, impaired spore germination, and curtailed germ tube length. Furthermore, it triggered reactive oxygen species (ROS) accumulation, initiating apoptosis and membrane integrity disruption. In vivo experiments demonstrate
