Palytoxin congeners.

Palytoxin, isolated from a zoanthid of the genus Palythoa, is the most potent marine toxin known. Intoxication by palytoxin leads to vasoconstriction, hemorrhage, ataxia, muscle weakness, ventricular fibrillation, pulmonary hypertension, ischemia and death. Palytoxin and its numerous derivatives (congeners) may enter the food chain and accumulate mainly in fishes and crabs, causing severe human intoxication and death following ingestion of contaminated products. Furthermore, toxic effects in individuals exposed via inhalation or skin contact to marine aerosol in coincidence with Ostreopsis blo

Improved detection of multiple environmental antibiotics through an optimized sample extraction strategy in liquid chromatography-mass spectrometry analysis.

A solid-phase extraction/liquid chromatography/electrospray ionization/multi-stage mass spectrometry (SPE-LC-ESI-MS/MS) method was optimized in this study for sensitive and simultaneous detection of multiple antibiotics in urban surface waters and soils. Among the seven classes of tested antibiotics, extraction efficiencies of macrolides, lincosamide, chloramphenicol, and polyether antibiotics were significantly improved under optimized sample extraction pH. Instead of only using acidic extraction in many existing studies, the results indicated that antibiotics with low pK a values (<7) were e

Integrated GC-MS and UHPLC-HRMS/MS profiling of bioactive compounds from Streptomyces sp. BPA-6 isolated from bees-collected pollen.

Antimicrobial resistance is one of the most current global health challenges, due to the increasing ineffectiveness of antibiotics and the increasing difficulty in treating infections. Approximately two-thirds of microbial antibiotics are produced by actinomycetes, of which approximately 74% come from the genus Streptomyces. In this study, a novel actinomycete strain, Streptomyces sp. BPA-6, isolated from pollen collected by Algerian bees, was investigated for its antimicrobial potential and chemical composition by GC-MS and HPLC-MS analyses. The strain showed strong antimicrobial activity aga

Quantification of human and veterinary antibiotics in water and sediment using SPE/LC/MS/MS.

An analytical method was developed and tested for four different groups of veterinary antibiotics in both river water and sediment matrices. Solid phase extraction (SPE) was used to enrich and to clean up the aqueous sample. Also, Mcllvaine and ammonium hydroxide buffer solutions were used to extract the compounds from the sediment matrix. High performance liquid chromatography (HPLC) equipped with tandem mass spectrometry (MS/MS) was used to separate and quantify the samples. The range of recoveries (in percent) for tetracyclines (TCs), sulfonamides (SAs), macrolides (MLs), and ionophore poly

Aminal Protection of Epoxide Monomer Permits the Introduction of Multiple Secondary Amine Moieties at Poly(ethylene glycol).

In contrast to acetal groups, aminal moieties are almost unknown in polymer chemistry. The aminal-protected isopropyl-hexahydro-pyrimidine glycidyl amine (PyGA) for the anionic ring-opening polymerization (AROP) is introduced. The monomer is prepared in a two-step synthesis and can be polymerized in a well-controlled manner under AROP conditions. Several poly(ethylene glycol) block and triblock copolymers are synthesized in a molecular weight range from 2 700 to 11 400 g mol-1 with up to 11 mol% PyGA. The molecular weight distributions are monomodal with low dispersity (Đ = Mw /Mn ) below 1.2.

Methods to improve antibacterial properties of PEEK: A review.

As a thermoplastic and bioinert polymer, polyether ether ketone (PEEK) serves as spine implants, femoral stems, cranial implants, and joint arthroplasty implants due to its mechanical properties resembling the cortical bone, chemical stability, and radiolucency. Although there are standards and antibiotic treatments for infection control during and after surgery, the infection risk is lowered but can not be eliminated. The antibacterial properties of PEEK implants should be improved to provide better infection control. This review includes the strategies for enhancing the antibacterial propert

Physicochemical properties of antibiotics: A review with an emphasis on detection in the aquatic environment.

Antibiotics have extensively been applied to rescue a great number of lives through prevention and treatment of contagious diseases and infections. They are either natural or human-made substances, which are broadly employed for promoting the health condition of human, plant, and animal. However, antibiotics are known to exert detrimental impacts on useful and nontarget microbiota of the biological system due to the overuse, continuous discharge into the environment, and subsequently aggregation in various environmental matrices. Physical and chemical properties help to evaluate whether a subs

In vivo subchronic effects of ciguatoxin-related compounds, reevaluation of their toxicity.

Ciguatoxins are marine compounds that share a ladder-shaped polyether structure produced by dinoflagellates of the genus Gambierdiscus and Fukuyoa, and include maitotoxins (MTX1 and MTX3), ciguatoxins (CTX3C) and analogues (gambierone), components of one of the most frequent human foodborne illness diseases known as ciguatera fish poisoning. This disease was previously found primarily in tropical and subtropical areas but nowadays, the dinoflagellates producers of ciguatoxins had spread to European coasts. One decade ago, the European Food Safety Authority has raised the need to complete the t

Standardized Protocol to Evaluate the Effect of Surface Treatments, Luting Cements, And Thermocycling On PEEK-Composite Bond Strength.

Polyetheretherketone (PEEK) has gained increasing attention as a biomaterial in prosthodontics; however, achieving durable adhesion to veneering composite materials remains a major challenge due to its chemically inert structure. This study presents and validates a standardized, reproducible protocol for evaluating shear bond strength (SBS) between PEEK and an indirect composite resin under different surface treatments, luting cements, and thermocycling conditions. A total of 240 PEEK discs were divided into four groups according to surface treatment: no surface treatment, sandblasting, sulfur

Increased Trypanocidal Activity of the Salinomycin Derivative Ironomycin Is Due to ROS Production and Iron Uptake Impairment.

Salinomycin and its derivatives display promising anti-proliferating activity against bloodstream forms of Trypanosoma brucei. The mechanism of trypanocidal action of these compounds is due to their ionophoretic activity inducing an influx of sodium cations followed by osmotic water uptake, leading to massive swelling of bloodstream-form trypanosomes. Generally, higher trypanocidal activities of salinomycin derivatives are associated with higher cell swelling activities. Although ironomycin (C20-propargylamine derivative of salinomycin) and salinomycin showed identical cell swelling activities

Chemical Diversity, Origin, and Analysis of Phycotoxins.

Microalgae, particularly those from the lineage Dinoflagellata, are very well-known for their ability to produce phycotoxins that may accumulate in the marine food chain and eventually cause poisoning in humans. This includes toxins accumulating in shellfish, such as saxitoxin, okadaic acid, yessotoxins, azaspiracids, brevetoxins, and pinnatoxins. Other toxins, such as ciguatoxins and maitotoxins, accumulate in fish, where, as is the case for the latter compounds, they can be metabolized to even more toxic metabolites. On the other hand, much less is known about the chemical nature of compound

POx-Lipids as an Alternative to PEG-Lipids? Multimethod Assessment of Chemistry and Structure.

The characterization of polymer-lipid conjugates is essential for their successful application in pharmaceutical formulations, particularly in lipid nanoparticles (LNPs). While poly(ethylene glycol) (PEG) lipids are widely used in LNPs, growing concerns over PEG immunogenicity have prompted the search for alternatives. Here, we investigate a series of poly(2-ethyl-2-oxazoline) (PEtOx) lipids as promising PEG substitutes featuring biocompatibility, tunable hydrophilicity, and stealth-like properties. We apply a comprehensive, multimethod approach utilizing liquid chromatography (LC) and matrix-

Polyethylene Glycol (PEG)-Based Wet-Adhesive Absorbable Bone Wax for Osseous Hemostasis and Repair.

The non-absorbability and biological inertia of traditional bone wax often result in foreign body retention, inhibit bone healing, and increase the risk of infection. Herein, a novel absorbable bone wax was developed by mixing polyethylene glycol (PEG), tannic acid (TA), sodium carboxymethyl cellulose (CMC), and low-molecular-weight poly-L-lactic acid (PLLA) via one-pot method. Among them, PEG functions as the matrix, endowing it with plasticity and hydrophilicity, thereby facilitating its facile, close fit to the surface of bone defects. TA, with its profusion of phenolic hydroxyl groups, bes

Multifunctional Thiolated Hyaluronic Acid-graft-PEDOT Dressing With Antifouling, On-Demand Drug Delivery and Electrical Stimulation for Wound Healing.

Recent advances in electro-responsive wound dressings have enabled on-demand drug delivery or electrical stimulation (ES) to accelerate healing; however, integrating both functions within a single platform remains challenging. Here, we report a multifunctional dressing based on a conductive, porous substrate comprising a thiolated hyaluronic acid (THA)-grafted PEDOT copolymer, P(EDOT-co-EDOT-g-THA). The material is fabricated by copolymerizing 3,4-ethylenedioxythiophene (EDOT) and S-((2,3-dihydrothieno[3,4-b][1,4]dioxin-2-yl)methyl)ethanethioate (EDOT-SAc) on flexible Au-coated electrospun PCL

Rapid Access to Ironomycin Derivatives by Click Chemistry.

Salinomycin, a natural carboxylic polyether ionophore, shows a very interesting spectrum of biological activities, including selective toxicity toward cancer stem cells (CSCs). Recently, we have developed a C20-propargylamine derivative of salinomycin (ironomycin) that exhibits more potent activity in vivo and greater selectivity against breast CSCs compared to the parent natural product. Since ironomycin contains a terminal alkyne motif, it stands out as being an ideal candidate for further functionalization. Using copper-catalyzed azide-alkyne cycloaddition (CuAAC), we synthesized a series o
