March2 Alleviates Aortic Aneurysm/Dissection by Regulating PKM2 Polymerization.

BACKGROUND: Aortic aneurysm/dissection (AAD) is a life-threatening disease lacking effective pharmacological treatment. Protein ubiquitination plays a pivotal role in cardiovascular diseases. However, the possible contribution of the E3 ubiquitin ligase March2 (membrane-associated RING [really interesting new gene] finger protein 2) to the cause of AAD remains elusive. METHODS: Integrated single-cell RNA sequencing analysis was conducted in human AAD tissues. Based on the screening results, we generated a mouse line of smooth muscle cell-specific March2 knockout. β-Aminopropionitrile monofuma

Pressure-Driven Solid-State Radical Polymerization toward Carbon Nanothread.

Mechanochemical radical polymerization has unique advantages in the synthesis of polymers due to its reduced solvent consumption and adaptability of insoluble monomers. However, it suffers from the uncontrollable degradation of the formed polymers during reaction, and a new synthetic strategy with precise controllability needs to be developed. Here, by employing high static pressure up to 30 GPa, we found 1,3,5-trifluorobenzene undergoes radical polymerization by breaking the conjugated π-bonds and forms a carbon nanothread with high selectivity (Polymer-I polymorph). Based on the crystal stru

Membrane composition and curvature in SNX9-mediated actin polymerization.

Sorting nexin 9 (SNX9) is a membrane-binding scaffold protein that contributes to viral uptake and inflammation and is associated with worse outcomes in several cancers. It is involved in endocytosis of epidermal growth factor receptors, β1-integrin and membrane type 1 matrix metalloprotease, and formation of mitochondrial-derived vesicles. The SNX9 Bin-Amphiphysin-Rvs (BAR)-Phox homology (PX) domains bind phosphoinositide lipids and the Src homology 3 (SH3) domain interacts with dynamin and Neural-Wiskott Aldrich syndrome protein (N-WASP) to stimulate Arp2/3 complex-mediated actin polymerizat

Polymerization mechanism of the Candida albicans virulence factor candidalysin.

Candida albicans is a commensal fungus that can cause epithelial infections and life-threatening invasive candidiasis. The fungus secretes candidalysin (CL), a peptide that causes cell damage and immune activation by permeation of epithelial membranes. The mechanism of CL action involves strong peptide assembly into polymers in solution. The free ends of linear CL polymers can join, forming loops that become pores upon binding to membranes. CL polymers constitute a therapeutic target for candidiasis, but little is known about CL self-assembly in solution. Here, we examine the assembly mechanis

A Coformer Approach for Supramolecular Polymerization at High Concentrations.

Insolubility of functional molecules caused by polymorphism sometimes poses limitations for their solution-based processing. Such a situation can also occur in the preparation processes of supramolecular polymers formed in a solution. An effective strategy to address this issue is to prepare amorphous solid states by introducing a "coformer" molecule capable of inhibiting the formation of an insoluble polymorph through co-aggregation. Herein, inspired by the coformer approach, we demonstrated a solubility enhancement of a barbiturate π-conjugated compound that can supramolecularly polymerize t

Titanium-Catalyzed Polymerization of a Lewis Base-Stabilized Phosphinoborane.

The reaction of the Lewis base-stabilized phosphinoborane monomer tBuHPBH2 NMe3 (2 a) with catalytic amounts of bis(η5 :η1 -adamantylidenepentafulvene)titanium (1) provides a convenient new route to the polyphosphinoborane [tBuPH-BH2 ]n (3 a). This method offers access to high molar mass materials under mild conditions and with short reaction times (20 °C, 1 h in toluene). It represents an unprecedented example of a transition metal-mediated polymerization of a Lewis base-stabilized Group 13/15 compound. Preliminary studies of the substrate scope and a potential mechanism are reported.

PTEN Regulates Dendritic Arborization by Decreasing Microtubule Polymerization Rate.

Phosphatase and tensin homolog (PTEN) is a major negative regulator of the phosphatidylinositol-3-kinase (PI3K)/Akt/mechanistic target of rapamycin (mTOR) pathway. Loss-of-function mutations in PTEN have been found in a subset of patients with macrocephaly and autism spectrum disorder (ASD). PTEN loss in neurons leads to somal hypertrophy, aberrant migration, dendritic overgrowth, increased spine density, and hyperactivity of neuronal circuits. These neuronal overgrowth phenotypes are present on Pten knock-out (KO) and reconstitution with autism-associated point mutations. The mechanism underl

Polymerization Reactions and Modifications of Polymers by Ionizing Radiation.

Ionizing radiation has become the most effective way to modify natural and synthetic polymers through crosslinking, degradation, and graft polymerization. This review will include an in-depth analysis of radiation chemistry mechanisms and the kinetics of the radiation-induced C-centered free radical, anion, and cation polymerization, and grafting. It also presents sections on radiation modifications of synthetic and natural polymers. For decades, low linear energy transfer (LLET) ionizing radiation, such as gamma rays, X-rays, and up to 10 MeV electron beams, has been the primary tool to produ

Acetaldehyde polymerization on Co(0001): the role of CO.

The adsorption and polymerization of acetaldehyde (CH3CHO) have been investigated on clean and CO pre-covered Co(0001) surfaces using the temperature programmed desorption (TPD) method. On the clean Co(0001) surface, CH3CHO molecules can polymerize to produce paraldehyde with very low efficiency. With pre-dosed CO molecules on Co(0001), the decomposition of CH3CHO is greatly inhibited. When the coverage of pre-dosed CO is <0.33 ML, no enhancement of CH3CHO polymerization is observed. However, when the pre-dosed CO coverage is >0.33 ML, the polymerization of CH3CHO is significantly enhanced dur

Inhibition of actin polymerization by marine toxin pectenotoxin-2.

Pectenotoxin-2 (PCTX-2) is one of the polyether macrolide toxins isolated from scallops involved in diarrheic shellfish poisoning via actin depolymerization. In the present study, we examined the bioactive mechanism of PCTX-2 in smooth muscle cells and clarify mode of action of the PCTX-2-induced actin depolymerization using purified skeletal actin. PCTX-2 (300 nM-3 µM) non-selectively inhibited vascular smooth muscle contractions elicited by high K+ or phenylephrine in a dose-dependent manner. However, elevated cytosolic Ca2+ and myosin light chain phosphorylation stimulated by high K+ were o

L-tyrosine polymerization-based ultrasensitive multi-analyte enzymatic biosensor.

Novel amperometric bi-enzyme biosensor based on tyrosinase (Tyr)-catalyzed L-tyrosine polymerization to effectively immobilize Tyr and glucose oxidase (GOx) simultaneously has been developed and is demonstrated to be efficient in monitoring multi-analyte (bisphenol A (BPA), phenol, Cr(III), glucose, and Cr(VI)). For the first time, the polymer from L-tyrosine oxidation (PLT) was utilized to in-situ immobilize biomacromolecules and was demonstrated to be effective in entrapping high-load and high-activity enzymes. The prepared bi-enzyme biosensor (PLT-Tyr-GOx/Au electrode) works well in the bio

Nitrogen oxides under pressure: stability, ionization, polymerization, and superconductivity.

Nitrogen oxides are textbook class of molecular compounds, with extensive industrial applications. Nitrogen and oxygen are also among the most abundant elements in the universe. We explore the N-O system at 0 K and up to 500 GPa though ab initio evolutionary simulations. Results show that two phase transformations of stable molecular NO2 occur at 7 and 64 GPa, and followed by decomposition of NO2 at 91 GPa. All of the NO(+)NO3(-) structures are found to be metastable at T = 0 K, so experimentally reported ionic NO(+)NO3(-) is either metastable or stabilized by temperature. N2O5 becomes stable

Poly(isobutylene) nanoparticles via cationic polymerization in nonaqueous emulsions.

The preparation of poly(isobutylene) (PIB) nanoparticles via cationic emulsion polymerization is presented. As a requirement, an oil-in-perfluoroalkane nonaqueous emulsion is developed, which is inert under the carbocationic polymerization conditions. To stabilize the dichloromethane/hexane droplets in the fluorinated, continuous phase, an amphiphilic block copolymer emulsifier is prepared containing PIB and 1H,1H-perfluoroalkylated poly(pentafluorostyrene) blocks. This system allows for the polymerization of isobutylene with number-average molecular weights (Mn) up to 27,000 g mol(-1). The pa

Hypothiocyanous acid oxidation of tubulin cysteines inhibits microtubule polymerization.

Thiol oxidation is a probable outcome of cellular oxidative stress and is linked to degenerative disease progression. In addition, protein thiol redox reactions are increasingly identified as a mechanism to regulate protein structure and function. We assessed the effect of hypothiocyanous acid on the cytoskeletal protein tubulin. Total cysteine oxidation by hypothiocyanous and hypochlorous acids was monitored by labeling tubulin with 5-iodoacetamidofluorescein and by detecting higher molecular weight inter-chain tubulin disulfides by Western blot under nonreducing conditions. Ring-opening metathesis polymerization-based recyclable magnetic acylation reagents.

An operationally simple method for the acylation of amines utilizing carbon-coated metal nanoparticles as recyclable supports is reported. Highly magnetic carbon-coated cobalt (Co/C) and iron (Fe/C) nanobeads were functionalized with a norbornene tag (Nb-tag) through a "click" reaction followed by surface activation employing Grubbs-II catalyst and subsequent grafting of acylated N-hydroxysuccinimide ROMPgels (ROMP=ring-opening metathesis polymerization). The high loading (up to 2.6 mmolg(-1) ) hybrid material was applied in the acylation of various primary and secondary amines. The products w

Inhibition of tubulin polymerization by hypochlorous acid and chloramines.

Protein thiol oxidation and modification by nitric oxide and glutathione are emerging as common mechanisms to regulate protein function and to modify protein structure. We assessed the effect of the oxidants hypochlorous acid and chloramines on the cytoskeletal protein tubulin. Fully computational lead-development of fluoro-olefin polymerization catalysts.

Coordinative polymerization of fluorinated olefins is still a challenging task. We analyzed the catalytic properties of diimido chromium VI compounds computationally by density functional methods. It was found that reactivity predictions depend strongly on the density functional chosen for the computations. Therefore, DFT calculations were calibrated to high level wave function theory calculations. Then geometrical parameters, which can be tuned by a ligand, were scanned to elucidate their influence on the catalytic activity. Having arrived at an optimal parameter set, we randomly assembled ch

Graft polymerization of native chicken feathers for thermoplastic applications.

Inexpensive and biodegradable thermoplastics were developed through graft polymerization of native chicken feather with methyl acrylate as a potential substitute for petroleum products. Poultry feathers are available in large quantities at a low price. However, natural chicken feathers have poor thermoplasticity, cannot be used to develop thermoplastic products, have very limited industrial applications, and are often considered as solid wastes. In this research, the effects of graft polymerization conditions, such as molar ratio of NaHSO(3) to K(2)S(2)O(8), initiator and monomer concentration

A tubulin polymerization microassay used to compare ligand efficacy.

We developed tubulin purification strategies that allowed sufficient material to be produced for compound-screening projects. Tubulins were polymerized in the presence of compounds using either turbidometric or fluorescence polymerization assays. IC50 and EC50 values were calculated and used to determine ratios between host and target tubulin (TT) (e.g., IC50-neuronal tubulin/IC50-TT). This ratio can be compared between compounds to identify the ones which are most selective for a particular TT. We found ratios for different compounds ranged from 0.16 to 4.0 between neuronal and cancer cell tu
