Incidence, risk factors and the effect of polyomavirus infection in hematopoietic stem cell transplant recipients.

Objective The effect of polyomavirus infection in HSCT recipients is poorly understood. Methods We evaluated 38 HSCT recipients. Polyomavirus was detected by nested qualitative polymerase chain reaction (PCR) assays of urine. The risk factors for BK virus and JC virus were analysed. The kidney and liver functions of infected and uninfected patients were compared. Results BK virus, JC virus, and simian virus 40 were detected in 21%, 42%, and 0% of HSCT recipients respectively. HCMV infection was found to be an independent risk factor for JC virus infection (odds ratio (OR): 8.528), while transp

Antibodies reacting with JCPyV_VP2 _167-15mer as a novel serological marker for JC polyomavirus infection.

BACKGROUND: JC polyomavirus (JCPyV) is a widespread human polyomavirus that usually resides latently in its host, but can be reactivated under immune-compromised conditions potentially causing Progressive Multifocal Leukoencephalopathy (PML). Detection of antibodies against the major capsid protein VP1 currently is the main marker for assessment of infection with JCPyV. METHODS: Based on a peptide microarray, peptide JCPyV_VP2_167-15mer was selected and a peptide ELISA was developed for detection of antibodies directed against this peptide. Epitope mapping and computational modelling was perf

Reversible ureteral obstruction due to polyomavirus infection after percutaneous nephrostomy catheter placement.

BK virus (BKV) is a human polyomavirus that remains latent in the urinary tract epithelium in most individuals. However, in immunocompromised states, including after hematopoietic stem cell transplantation (HSCT), BKV may reactivate and cause infection predominantly affecting the bladder, commonly manifested as hemorrhagic cystitis. Renal insufficiency, occasionally requiring hemodialysis, is not uncommon and was previously attributed to medications or the development of tubulointestitial nephritis. We report a series of 6 HSCT recipients who developed obstructive uropathy of the upper urinary

Polyomavirus Infection in Two Young Parrots: Assessment of Viral Distribution in Organs and Affected Tissues.

Avian polyomavirus (APyV) is responsible for diseases in birds belonging to several avian orders, including Psittaciformes, Anseriformes, and other susceptible taxa. APyVs appear to be able to replicate in various organs of susceptible bird species. This study aimed to investigate (i) the distribution of APyV in the organs of two parrot chicks belonging to different species, Moluccan eclectus (Eclectus roratus) and blue-winged macaw (Primolius maracana), which died due to APyV infection and (ii) the lesions associated with the infection. By negative-staining electron microscopy, well-defined v

Co-infection with human polyomavirus BK enhances gene expression and replication of human adenovirus.

Immunocompromised patients are susceptible to multiple viral infections. Relevant interactions between co-infecting viruses might result from viral regulatory genes which trans-activate or repress the expression of host cell genes as well as the genes of any co-infecting virus. The aim of the current study was to show that the replication of human adenovirus 5 is enhanced by co-infection with BK polyomavirus and is associated with increased expression of proteins including early region 4 open reading frame 1 and both the large tumor antigen and small tumor antigen. Clinical samples of whole bl

JC polyomavirus infection is strongly controlled by human leucocyte antigen class II variants.

JC polyomavirus (JCV) carriers with a compromised immune system, such as in HIV, or subjects on immune-modulating therapies, such as anti VLA-4 therapy may develop progressive multifocal leukoencephalopathy (PML) which is a lytic infection of oligodendrocytes in the brain. Serum antibodies to JCV mark infection occur only in 50-60% of infected individuals, and high JCV-antibody titers seem to increase the risk of developing PML. We here investigated the role of human leukocyte antigen (HLA), instrumental in immune defense in JCV antibody response. Anti-JCV antibody status, as a surrogate for J

Frequent infection of cerebellar granule cell neurons by polyomavirus JC in progressive multifocal leukoencephalopathy.

Progressive multifocal leukoencephalopathy (PML) occurs most often in immunosuppressed individuals. The lesions of PML result from astrocyte and oligodendrocyte infection by the polyomavirus JC (JCV); JCV has also been shown to infect and destroy cerebellar granule cell neurons (GCNs) in 2 human immunodeficiency virus (HIV)-positive patients. To determine the prevalence and pattern of JCV infection in GCNs, we immunostained formalin-fixed paraffin-embedded cerebellar samples from 40 HIV-positive and 3 HIV-negative PML patients for JCV, and glial and neuronal markers. The JCV infection was dete

Expression of p53 in virally infected tubular cells in renal transplant patients with polyomavirus nephropathy.

Polyomavirus (PV) infection is associated with ureteral stenosis, hemorrhagic cystitis, and interstitial nephritis in renal transplant patients. The 3 PVs detected in human beings-BK virus, JC virus, and simian virus 40-each encode highly homologous forms of a large T antigen, a transcriptional and replicational regulatory protein. We describe immunohistochemical findings in 5 renal transplant patients who developed PV nephropathy (PVN) and a sixth patient with both PVN and PV infection of the bladder mucosa. Polyomavirus infection was confirmed by immunohistochemical detection of T antigen in

Human polyomavirus JC control region variants in persistently infected CNS and kidney tissue.

The question of a possible role for JC virus (JCV) genomic rearrangements in the pathogenesis of progressive multifocal leukoencephalopathy (PML) was addressed by analysis of the genomic complexity and the transcriptional control region (TCR) of the JCV DNA population in persistently infected CNS and kidney tissue. After cloning of full-length viral DNA, no extensive changes were detected in the coding regions of the JCV genome by restriction analysis suggesting an intact JCV DNA population. For further analysis of the distribution of JCV subtypes, the non-coding region was amplified by PCR. M

Cellular immune response during BK polyomavirus infection in living donor kidney transplant recipients.

BACKGROUND: BK polyomavirus (BKPyV) is a significant cause of graft impairment and loss following kidney transplantation. Early prediction of which patients are at risk of BKPyV replication and BKPyV-associated nephropathy (BKPyVAN) would be beneficial, preferably prior to transplantation. From an immunological perspective, determinants of susceptibility to BKPyV reactivation remain incompletely understood. The aim of this study was to evaluate the prognostic value of lymphocyte subset counts, CD4/8 T cell ratio, and changes in CD4 + T cell counts over time. Additionally, the concept of Immune

An Elusive Target: Inhibitors of JC Polyomavirus Infection and Their Development as Therapeutics for the Treatment of Progressive Multifocal Leukoencephalopathy.

Progressive multifocal leukoencephalopathy (PML) is a rare demyelinating disease caused by infection with JC Polyomavirus (JCPyV). Despite the identification of the disease and isolation of the causative pathogen over fifty years ago, no antiviral treatments or prophylactic vaccines exist. Disease onset is usually associated with immunosuppression, and current treatment guidelines are limited to restoring immune function. This review summarizes the drugs and small molecules that have been shown to inhibit JCPyV infection and spread. Paying attention to historical developments in the field, we

Chk1 and the Host Cell DNA Damage Response as a Potential Antiviral Target in BK Polyomavirus Infection.

The human BK polyomavirus (BKPyV) is latent in the kidneys of most adults, but can be reactivated in immunosuppressed states, such as following renal transplantation. If left unchecked, BK polyomavirus nephropathy (PyVAN) and possible graft loss may result from viral destruction of tubular epithelial cells and interstitial fibrosis. When coupled with regular post-transplant screening, immunosuppression reduction has been effective in limiting BKPyV viremia and the development of PyVAN. Antiviral drugs that are safe and effective in combating BKPyV have not been identified but would be a benefi

Rat polyomavirus 2 infection in a colony of X-linked severe combined immunodeficiency rats in Japan.

Polyomaviruses (PyVs) infect a wide range of animals and provoke wasting diseases, particularly in immunosuppressed hosts. Recently, a novel Rattus norvegicus polyomavirus 2 (RatPyV2) has been identified in a colony of X-linked severe combined immunodeficiency (X-SCID) rats in the United States. Here, we describe the first report of the RatPyV2 infection in an X-SCID rat colony in Japan. The affected rats exhibited adult-onset wasting. Histologically, we observed large basophilic intranuclear inclusion bodies within the hyperplastic or dysplastic epithelial cells in the salivary glands, Harder

BK polyomavirus-infected nephrogenic adenoma of the urinary bladder in a renal transplant recipient: A case report.

This report describes the morphological and immunohistochemical features of the first case of a nephrogenic adenoma (NA) of the urinary bladder infected with polyomavirus. The findings occurred in a female patient, 6 months post cadaveric renal transplant. Voided urine cytology was positive for decoy cells. Renal transplant biopsy showed no evidence of BKV allograft nephropathy. Cystoscopy demonstrated multiple erythematous patchy and nodular areas. These were found in the left and right posterior wall, and the right lateral wall toward the bladder neck, and were suspicious for either chronic

Persistent infection with human polyomavirus revealed by urinary cytology in a patient with heart transplantation. A case report.

BACKGROUND: Management after heart transplantation must deal with the twin risks of rejection and infection. Early infection with viral infection, particularly cytomegalovirus (CMV), is becoming more prevalent two to three months after transplantation. To our knowledge, there has been no previous report of human polyomavirus (HPOV) infection after heart transplantation. CASE: A 20-year-old male with a history of Kawasaki disease and who had suffered from severe congestive heart failure after a coronary artery bypass graft, underwent heart transplantation. Urinary cytology demonstrated HPOV in

Molecular epidemiology and characterization of goose polyomavirus in China: insights into its impact on hatchability and susceptibility to co-infections.

INTRODUCTION: Goose hemorrhagic polyomavirus (GHPV) is the primary etiological agent of goose hemorrhagic nephritis and enteritis (HGNE), a disease causing substantial economic losses to the global waterfowl industry. Despite documented outbreaks in Europe and Asia, integrated molecular epidemiological evidence linking clinical outbreaks, co-infections, and viral phylogeny remains limited. METHODS: We conducted nationwide epidemiological surveillance across eight provinces in China from March 2023 to April 2025, covering 83 farms and 1,902 samples. An outbreak of reduced hatchability was inve

Lamin subtype expression in Merkel-cell carcinoma: its relation to polyomavirus infection and tumor metastasis.

Merkel-cell carcinoma (MCC) is known for its high metastatic potential. Studies in several tumor types have proposed a role for up- or downregulation of nuclear A- and B-type lamins in facilitating the process of invasion and metastasis, but such studies are lacking for MCC. Therefore, lamin expression levels were examined in 38 tissue samples from 28 patients with MCC using immunohistochemistry. The series comprised primary tumors and different metastases and both Merkel cell polyomavirus (MCPyV) positive and MCPyV-negative cases. This study revealed that primary MCC tumors express variable,

Merkel Cell Polyomavirus Co-Infection in HIV/AIDS Individuals: Clinical Diagnosis, Consequences and Treatments.

Merkel cell polyomavirus (MCV) was named for its role as the causative agent of Merkel cell carcinoma (MCC), which is MCV positive in approximately 80% of cases. MCV is classified as a Group 2A carcinogen, which promotes carcinogenesis by integrating T-antigen into the cell genome. The prevalence of anti-MCV antibodies in the general population can be as high as 90%. MCV typically promotes cancer by integrating T-antigen genes into the host cell genome, and 80% of MCC cases are attributed to MCV activation. In immunocompetent individuals, MCV usually remains latent after infection. However, th

BK polyomavirus infection: more than 50 years and still a threat to kidney transplant recipients.

BK polyomavirus (BKPyV) is a ubiquitous human polyomavirus and a major infection after kidney transplantation, primarily due to immunosuppression. BKPyV reactivation can manifest as viruria in 30%-40%, viremia in 10%-20%, and BK polyomavirus-associated nephropathy (BKPyVAN) in 1%-10% of recipients. BKPyVAN is an important cause of kidney graft failure. Although the first case of BKPyV was identified in 1971, progress in its management has been limited. Specifically, there is no safe and effective antiviral agent or vaccine to treat or prevent the infection. Even in the current era, the mainsta
