Hydraulic, Gas-Exchange and Leaf-Trait Variation Across Hydrologically Contrasting Sites and Tree Sizes in Populus euphratica.

Plant water status, hydraulic transport and carbon exchange can respond differently across natural hydrologic contrasts. We measured hydraulic, gas-exchange, carbon, morphological and anatomical traits in 33 Populus euphratica trees distributed across four fixed sites (D1-D4) and three tree-size classes. Site and size associations were evaluated with additive models, heteroskedasticity-consistent inference, false-discovery-rate control and a balanced D1-D3 sensitivity analysis. Midday leaf water potential became progressively more negative from D1 to D4, whereas branch xylem hydraulic conducti

Genus-Wide Pan-Genome Analysis of Populus bZIP Transcription Factors with Reanalysis of Public Salt-Stress Transcriptomes.

Basic leucine zipper (bZIP) transcription factors regulate plant development and stress responses, but their genus-wide diversity in Populus remains unclear. We analyzed 19 Populus genomes and retained 1764 bZIP proteins, including 21 independent new loci and four annotation corrections. Of these, 1762 were assigned to 79 orthologous gene groups (OGGs), comprising 43 core, 20 soft-core, 15 shell and one cloud OGG, of which 59 showed copy-number variation. Phylogenetic analysis assigned 74 representative pangenes to 13 subfamilies, with five remaining unclassified and motif patterns differing a

The sex determination gene of Populus deltoides, PdFERR, interacts with ERF96 to promote the development of female flower organs.

The female-specifically expressed response regulator (PdFERR) gene in Populus deltoides, a sex determination gene (an orthologous gene of ARR17 in Populus tremula), was found to promote femaleness in heterologous expression lines of Arabidopsis. None of the genes in the Arabidopsis genome seem to be orthologous to PdFERR. Although originating from two evolutionarily distant plants, the dioecious poplar FERR might promote femaleness in the hermaphroditic Arabidopsis through an evolutionary consistent regulatory pathway. However, there is no molecular evidence to support this viewpoint. In this

Effects of two iron sources on iron and cadmium allocation in poplar (Populus alba) plants exposed to cadmium.

Effects of 10 microM cadmium (supplied as Cd nitrate) on the utilization and allocation of iron (Fe) were investigated in poplar (Populus alba L.) plants grown in nutrient solution with Fe(III)-EDTA or Fe(III)-citrate as the Fe source. The effects of Cd were also compared with those of Fe deprivation. The accumulation of Fe in roots was 10-fold higher in plants grown with Fe-citrate than with Fe-EDTA. Cadmium decreased leaf chlorophyll concentrations and photosynthetic rates, and these decreases were more marked in plants grown with Fe-citrate than with Fe-EDTA. In both Fe treatments, addition

Seasonal shifts in water-source partitioning and fine-root overlap in a Populus simonii-maize shelterbelt system.

INTRODUCTION: Farmland shelterbelts are critical for agricultural production in arid and semiarid regions, but they may compete with crops for limited soil water. This study aimed to quantify seasonal water-source use, soil moisture dynamics, and fine-root distribution in a Populus simonii-maize shelterbelt system. METHODS: The study was conducted in Zhangbei County, northwestern Hebei Province, China. Three land-use conditions were compared: a pure P. simonii shelterbelt (P), a P. simonii-maize intercropping system (PC), and a maize monoculture reference (C). Synchronous sampling was conduct

Gibberellin-mediated PtrLBD39-PtrHDA6-PtrHCT epigenetic path regulates lignin biosynthesis during tension wood formation in Populus trichocarpa.

Tension wood (TW) is a specialized xylem tissue formed in angiosperm trees to counteract mechanical stresses. Increased phytohormones, particularly gibberellins (GAs), and decreased lignin deposition are two well-established features of TW, yet the molecular mechanisms linking GA signaling to lignin repression remain largely unknown. Here we revealed that TW formation in Populus trichocarpa elevates GA accumulation, which activates the expression of PtrLBD39, the most strongly induced transcription factor and a repressor of lignin biosynthesis in TW development. Inhibition of GA biosynthesis a

Drought-induced embolism shapes sex-specific responses by regulating hydraulic conductivity in Populus cathayana and Salix babylonica.

Drought is a key constraint on plant growth and development. Poplars and willows are dioecious plants, and sex-specific responses to drought differ across species. However, little is known about how xylem embolism drives sex-specific hydraulic failure. In this study, male and female Populus cathayana and Salix babylonica were used to investigate the role of embolism in sex-specific drought responses. Drought significantly reduced plant growth and photosynthetic capacity, increased membrane damage and reactive oxygen species accumulation, altered vessel structure and aggravated embolism. P. cat

Multi-Omics Analysis Provides Insights Into the Molecular Mechanism of Melatonin-Mediated Cadmium Tolerance in Populus tomentosa.

Cadmium (Cd) contamination, a major constraint on plant growth, causes a serious threat to ecological safety. Populus tomentosa is a promising woody species for Cd phytoremediation, but the mechanism of Cd phytoremediation by melatonin (MT) in this species remains unclear. To elucidate how MT confers Cd tolerance in P. tomentosa, four treatments (control, MT: 100 μmol·L-1 melatonin, Cd: 100 μmol·L-1 CdCl2, and MT + Cd, CM: 100 μmol·L-1 CdCl2 + 100 μmol·L-1 melatonin) on seedlings were conducted, and integrated analyses of physiological traits, noninvasive micro-test (NMT) data, transcriptomes,

Elevated CO2 Modifies Within-Species Differences in Relative Drought Resistance With Implications for Recovery in Populus balsamifera.

Forecasting forest responses to climate change requires mechanistic insight into how rising atmospheric CO2 concentration ([CO2]) affects within-species drought tolerances. We tested whether elevated [CO2] alters drought resistance and recovery using cuttings of two ecotypes of Populus balsamifera (Peace River, PR; Hay River, HR) grown under factorial combinations of [CO2] (400 vs. 1000 μmol mol-1) and water regime (well-watered vs. drought followed by re-watering). Under drought, elevated [CO2] compensated for the water deficit in net photosynthesis (Pn) only in PR, narrowing the ecotypic dif

Neutrophil Immunomodulatory Activity of Nerolidol, a Major Component of Essential Oils from Populus balsamifera Buds and Propolis.

Propolis is a resinous mixture of substances collected and processed from various botanical sources by honeybees. Black poplar (Populus balsamifera L.) buds are one of the primary sources of propolis. Despite their reported therapeutic properties, little is known about the innate immunomodulatory activity of essential oils from P. balsamifera and propolis. In the present studies, essential oils were isolated from the buds of P. balsamifera and propolis collected in Montana. The main components of the essential oil from P. balsamifera were E-nerolidol (64.0%), 1,8-cineole (10.8%), benzyl benzoa

Physiological and biochemical responses and microscopic structure changes of Populus tomentosa Carr seedlings to 4-BDE exposure.

Populus species are very effective in remediation of contaminants. Polybrominated diphenyl ethers (PBDEs) are commonly used as flame retardants and are known to be persistent environmental pollutants. Numerous studies have shown that PBDEs are rising in human tissues and biota. 4-Monobrominated diphenyl ether (4-BDE), one of the less brominated PBDEs, was served as a model compound for biodegradation of lower brominated congeners. The present study was designed to clarify the effects of 4-BDE stress on morphological, physiological, and biochemical impacts of Populus tomentosa Carr in a tissue

Branch architectural features of Populus euphratica in the lower reaches of the Tarim River using terrestrial laser scanning.

Branch architecture is the spatial structure formed by branching within the canopy, which directly affects tree growth, function, and ecological adaptation. To reveal the morphological adaptation mechanisms of Populus euphratica to extremely arid environments, we used terrestrial laser scanning to scan 51 individual P. euphratica trees in the lower reaches of the Tarim River. We reconstructed the branch structure using the quantitative structural model TreeQSM, extracted the geometric parameters of first- to third-order branches, and analyzed the allometric relationships among different branch

Genotype-environment interaction and stability of fiber properties and growth traits in triploid hybrid clones of Populus tomentosa.

BACKGROUND: Clones provide a sensitive method for evaluating genotypic stability and detecting genotype-environment (G × E) interactions because of non-additive genetic effects among clones and there being no genetic effect among ramets of an ortet. With this study, we aimed to confirm and expand earlier findings, estimate stability parameters, and provide accurate estimates of clonal repeatabilities and genetic gains for a triploid breeding program of P. tomentosa Carr. RESULTS: Six 5-year-old clonal trials established in Northern China were used to determine the clonal variation, clone × si

Effect of nitrate and ammonium fertilization on Zn, Pb, and Cd phytostabilization by Populus euramericana Dorskamp in contaminated technosol.

This study aimed at assessing the effect of nitrogen addition under two forms, nitrate and ammonium, on the stabilization of Zn, Pb, and Cd by Populus euramericana Dorskamp grown in contaminated soils for 35 days under controlled conditions. Temporal changes in the soil pore water (SPW) were monitored for pH, dissolved organic carbon (DOC), and total dissolved concentrations of metals in the soils rhizosphere. Rhizospheric SPW pH decreased gradually with NH4(+) addition and increased with NO3(-) addition up to one unit, while it slightly decreased initially then increased for the untreated con

A combination of Populus tremula, Solidago virgaurea and Fraxinus excelsior as an anti-inflammatory and antirheumatic drug. A short review.

Anti-inflammatory properties of Populus tremula are mainly deduced from its components, the well investigated salicylates. Solidago extracts have spasmolytic, antihypertensive and diuretic effects. Fraxinus excelsior itself has hardly been investigated, but its coumarin components proved to have a variety of pharmacological properties, i.e. inhibition of T-cell activation and of the arachidonic acid cascade. Phytodolor N (abbreviated as PD) is a combination of the above mentioned plants. Various in vitro and especially in vivo studies proved its anti-inflammatory and antirheumatic properties,

An Efficient Agrobacterium-Mediated Transformation Method for Hybrid Poplar 84K (Populus alba × P. glandulosa) Using Calli as Explants.

A highly efficient Agrobacterium-mediated transformation method is needed for the molecular study of model tree species such as hybrid poplar 84K (Populus alba × P. glandulosa cv. '84K'). In this study, we report a callus-based transformation method that exhibits high efficiency and reproducibility. The optimized callus induction medium (CIM1) induced the development of calli from leaves with high efficiency, and multiple shoots were induced from calli growing on the optimized shoot induction medium (SIM1). Factors affecting the transformation frequency of calli were optimized as follows: Agro

Metabolism and mineralization of hexahydro-1,3,5-trinitro-1,3,5-triazine inside poplar tissues (Populus deltoides x nigra DN-34).

Poplar tissue cultures and leaf crude extracts (Populus deltoides x nigra DN-34) were exposed to [U-14C]hexahydro-1,3,5-trinitro-1,3,5-triazine (RDX) and incubated under light and in the dark. Poplar tissue cultures were able to partially reduce RDX to hexahydro-1-nitroso-3,5-dinitro-1,3,5-triazine (MNX) and hexahydro-1,3-dinitroso-5-nitro-1,3,5-triazine (DNX), regardless of the presence or absence of light. However, further transformation of RDX, MNX, and DNX required exposure to light and resulted in the formation of formaldehyde (CH2O), methanol (CH3OH), and carbon dioxide (CO2). Similarly,

Benchmarking Assembly-Free K-mer Methods for Species Identification in Complex Plant Groups: A Case Study in Populus.

Species identification in taxonomically complex plant groups is frequently limited by the inadequacy of organellar markers, whose phylogenetic signal is disrupted by cytonuclear discordance and chloroplast capture. Using the taxonomically complex genus Populus as a model, we evaluated an assembly-free k-mer workflow against a curated SNP reference benchmark. Whole-genome resequencing data from 235 Populus individuals were curated to a 202-individual, 34-species reference dataset in which all retained species are strictly monophyletic in a genome-wide SNP analysis. Molecular regulatory mechanisms of Populus deltoides under combined high temperature and short-photoperiod stress associated with DNA methylation.

Populus deltoides is a key species for industrial timber and ecological construction in temperate regions, where increasingly frequent and persistent heat waves pose serious challenges to its survival. However, the epigenetic mechanisms by which DNA methylation regulates environmental responses remain poorly understood. Here, whole-genome bisulfite sequencing and RNA-seq were performed on five P. deltoides genotypes grown in temperate and tropical regions. Results revealed that CG/CHG methylation stability is closely correlated with environmental sensitivity. Significant CG/CHG methylation var
