Glycoside hydrolase-mediated utilization of Poria cocos polysaccharide enriches Lactobacillus gasseri and activates the AhR-IL-22 axis to attenuate DSS-induced colitis.

Polysaccharides derived from edible fungi can alleviate ulcerative colitis (UC) by enriching beneficial probiotics. However, the mechanisms underlying their interaction with probiotics remain unclear. Here, The current study investigated how Poria cocos polysaccharide (PCP) enriches L. gasseri and the effects of L. gasseri on dextran sulfate sodium (DSS)-induced UC. Genomic analyses predicted multiple polysaccharide utilization loci in L. gasseri, and growth kinetics together with gene expression assays confirmed that PCP promoted L. gasseri growth and upregulated glycoside hydrolases (GH1 and

Poria cocos polysaccharide-honeycomb manganese oxide nanoparticles adjuvant enhanced immune responses to PCV2 vaccine via TLR4-mediated NF-κB activation enhanced STING pathway.

Adjuvants, as indispensable ingredients in vaccines, possess the capability to enhance the magnitude, breadth, and durability of immune responses. Traditional alum adjuvants principally elicit a potent humoral immune response, yet demonstrate an inability to effectively stimulate a cellular immune response. We previously successfully prepared the Poria cocos polysaccharide-honeycomb manganese oxide nanoparticles (PMn) system, and the PMn has the capability to facilitate the activation of dendritic cells (DCs) and enhance immune responses. Herein, we further investigated the underlying mechanis

Screening of selected wood-damaging fungi for the HIV-1 reverse transcriptase inhibitors.

Extracts obtained using methanol and dichloromethane from 57 species of wood damaging fungi were investigated for their ability to inhibit HIV-1 reverse transcriptase activity in vitro using a non-radioactive assay. Sixty-three samples were tested all together; some species were represented more than one isolate. Thirteen methanolic extracts exhibited more than 40% inhibition and two among them inhibited the enzyme by more than 80%. All extracts obtained with dichloromethane were inferior to methanolic extracts in their inhibitory activity. The most active fungal species discovered in the firs

Macrophage-mimicking nanotherapy for attenuation of acute pancreatitis.

Acute pancreatitis (AP) is a highly fatal pancreatic inflammation. In recent years, synthetic nanoparticles have been extensively developed as drug carriers to address the challenges of systemic adverse reactions and lack of specificity in drug delivery. However, systemically administered nanoparticle therapy is rapidly cleared from circulation by the mononuclear phagocyte system (MPS), leading to suboptimal drug concentrations in inflamed tissues and suboptimal pharmacokinetics. To address this challenge, we herein demonstrate a surface masking strategy that involves coating the surface of se

Multi-omics-guided discovery of CYP5035 enables heterologous production and engineering of pachymic acid in yeast.

Pachymic acid is a key bioactive triterpenoid of Poria cocos, a medicinal fungus widely used in traditional Chinese medicine, and is responsible for its anti-tumor and anti-inflammatory activities. However, its low natural abundance and reliance on pine wood-based cultivation limit large-scale production and therapeutic application. Methyl jasmonate (MeJA) is known to stimulate triterpenoid biosynthesis in plants and fungi. In this study, we treated liquid-cultured Poria cocos mycelia with MeJA and integrated transcriptomic and targeted metabolomic analyses to identify a cytochrome P450 gene,

Removal of copper, chromium and arsenic from preservative-treated wood by chemical extraction-fungal bioleaching.

Large volumes of preservative-treated wood containing toxic Cr, Cu and As salts are decommissioned worldwide. This study investigated the effectiveness of solid-state fermentation with copper-tolerant brown-rot fungi for the remediation of wood treated with chromated copper arsenate (CCA) and acid copper chromate (ACC) formulations. Treatment of CCA- and ACC-wood with the most effective strain, Antrodia vaillantii FRLP-14G, attained extensive leaching of As and/or Cr, but Cu elimination was poor (<18%). Additional research showed that a variety of organic acids, including citrate, are effectiv
