ATP-sensitive potassium channels counteract anoxia in neurones of the substantia nigra.

The ATP-sensitive potassium channel (KATP channel) is a unique ionophore in that it appears to reflect cell metabolism. In the brain, the highest density of binding sites for the KATP channel is the substantia nigra. To evaluate the role of the KATP channel in this key brain area for motor control, we used exposure to cyanide to lower intracellular ATP and thereby mimic anoxia and ischemia. Treatment with cyanide caused the activation of a potassium current in a sub-population of nigral neurones with distinct pharmacological and electrophysiological properties. The response to cyanide was abol

Nitrile-synthesizing enzyme: Screening, purification and characterization.

Cyanide is known as a toxic compound for almost all living organisms. We have searched for cyanide-resistant bacteria from the soil and stock culture collection of our laboratory, and have found the existence of a lot of microorganisms grown on culture media containing 10 mM potassium cyanide. Almost all of these cyanide-resistant bacteria were found to show β-cyano-L-alanine (β-CNAla) synthetic activity. β-CNAla synthase is known to catalyze nitrile synthesis: the formation of β-CNAla from potassium cyanide and O-acetyl-L-serine or L-cysteine. We found that some microorganisms were able to de

Surviving cyanide poisoning: A case report highlighting the role of early antidote use.

Survival after high-dose oral cyanide ingestion is rare, and when untreated cases often result in death within an hour. Immediate treatment however, can be lifesaving. We describe a patient who fully recovered after prompt intervention. He arrived at the emergency department unconscious, with red skin and a Glasgow Coma Scale of 3 and was suspected of an intentional intoxication with an unknown white crystalline powder. He rapidly suffered a cardiac arrest. Blood gas analysis showed severe metabolic acidosis, high lactate, and slightly elevated methemoglobin. In suspect of cyanide poisoning, h

[Structural Optimization in Nitrile Synthesis via Desulfonylative Smiles Rearrangement].

Nitriles are important organic compounds that are frequently found in pharmaceuticals, natural products, agrichemicals, and functional materials. In addition, nitriles serve as versatile building blocks in synthetic chemistry, as the nitrile moiety can be readily transformed into a variety of valuable functional groups, such as carbonyl derivatives, amines, and heterocycles. However, conventional methods for nitrile synthesis often rely on highly toxic metal cyanides, such as copper(I) cyanide (CuCN) and potassium cyanide (KCN). In this review, I introduce our recent works on a cyanide- and tr

Potential for error when assessing blood cyanide concentrations in fire victims.

The present study explores toxicologic significance of blood cyanide concentrations in fire victims. Headspace gas chromatography was used for cyanide detection. Analysis of blood samples from ten fire victims (postmortem interval = 8 h to 3 to 5 d) detected zero to 11.9 mg/L of cyanide and a large difference in cyanide concentrations among victims. Carboxyhemoglobin (COHb) saturation was in the range of 24.9 to 84.2%. To examine the effects of methemoglobinemia and postmortem interval on blood cyanide concentrations in fire victims, an experiment was carried out using rabbits as the animal mo

The effect of storage upon cyanide in blood samples.

CDPA blood of human origin was 'spiked' with a solution of potassium cyanide at four different concentrations. When such blood was left in contact with the atmosphere for up to 10 min an appreciable amount of the cyanide was lost. Stoppered tubes containing the 'spiked' blood were stored at -20 degrees C and 4 degrees C for up to 6 months. Most samples, especially those 'spiked' at the highest concentrations, showed a loss of cyanide.

Ecotoxicity of cyanide complexes in industrially contaminated soils.

This study deals with acute and chronic ecotoxicity of leachates from industrially contaminated soils. Analyses focused on cyanides (complex and free forms) to study their possible involvement in leachates toxicity. No acute toxicity on the Microtox and 48 h-Daphnia magna tests was found in leachates collected over 18 months, but a high chronic toxicity was recorded on the reproduction of Ceriodaphnia dubia (EC50-7d=0.31±0.07%) and on the algal growth of Pseudokirchneriella subcapitata (EC50-72 h=0.27±0.09%). Ceriodaphnids were as sensitive to free cyanide as to complex forms (EC50-7d as CN(-)

The effects of cyanide on the growth and respiration of Enterobacter aerogenes in continuous culture.

The effect of cyanide on the physiology of lactate- and oxygen-limited Enterobacter aerogenes NCTC 10336 was studied in chemostat culture (D = 0.1 h-1). In the absence of cyanide, the molar growth yield from oxygen (YO2) under oxygen limitation was 60% of the carbon-limited value. A similar decrease in yield was observed in a lactate-limited culture (excess oxygen) which was continuously fed low concentrations of potassium cyanide. The cultures with the lower growth yields possessed respiratory systems less sensitive to inhibition by cyanide. This was particularly marked in cultures grown in t

Application of cyanide hydrolase from Klebsiella sp. in a biosensor system for the detection of low-level cyanide.

A partially purified preparation of cyanide hydrolase (cyanidase) from a bacterium, Klebsiella sp., was applied as a biocatalyst in a biosensor system for low-level cyanide detection. In the biosensor system cyanide hydrolase converts cyanide into formate and ammonia. The formate produced in the cyanide degradation was detected with a formate biosensor, in which formate dehydrogenase (FDH; E.C. 1.2.1.2) was co-immobilized with salicylate hydroxylase (SHL; E.C. 1.14.13.1) on a Clark electrode. The principle of the formate sensor is that FDH converts formate into carbon dioxide using beta-nicoti

Ferrocyanide ingestion may cause false positives in cyanide determination.

The authors present a patient who ingested a cyanide containing solution and arrived at the hospital without any clinical evidence of intoxication but an elevated blood cyanide level. The authors explain this discrepancy with the following hypotheses: 1) the patient ingested cyanide as an iron-chelated complex; and 2) the sulfuric acid used in the standard microdiffusion technique released cyanide from its iron-bound state to result in the observed elevated blood cyanide. Through a series of in vitro analyses, the authors demonstrate the following: 1) the ingested solution tested positive for

Colorimetric determination of cyanide liberated from apricot kernels.

A simple colorimetric method is described for determining the quantity of hydrogen cyanide produced by the spontaneous decomposition of amygdalin in apricot kernels. The evolved cyanide is collected in sodium hydroxide solution and assayed colorimetrically by reaction with picric acid. Results for duplicate assays, 3.02 and 3.06 mg CN-/g, compare well with those obtained by AOAC method 26.115 which specifies steam distillation and silver nitrate titration; results for triplicate assays were 3.02, 3.03, and 3.08 mg CN-/g by the latter. Recovery of cyanide from potassium cyanide at a level equiv

[Cyanide, in And Then There Were None, Sparkling Cyanide].

And Then There Were None and Sparkling Cyanide, two of Agatha Christie's famous novels describe potassium cyanide-induced deaths. Cyanide, a tasteless, odorless, strongly alkaline poison is a powerful gastrointestinal irritant, following oral ingestion. It reacts with hydrochloric acid in the gastric juice to produce hydrogen cyanide gas, which is absorbed and inhibits the mitochondrial electron transfer system and consequently suppresses adenosine triphosphate (ATP) production. Therefore, the central nervous system, which consumes a large amount of ATP, is first affected and symptoms of poiso

Cyanide induced changes in dynamic pulmonary mechanics in rats.

Effect of subcutaneously (s.c.) administered potassium cyanide (0.5 and 1.0 LD50) and inhalation of hydrogen cyanide (55 ppm or 60.6 mg/m3) for 30 minutes was studied on various physiological parameters related to dynamic pulmonary mechanics in anaesthetized rats. Total pulmonary phospholipid with its fractions were also estimated. Both s.c. (1.0 LD50) and inhalation exposures increased air flow, transthoracic pressure and tidal volume accompanied by significant decrease in pulmonary phospholipids. Inhalation of hydrogen cyanide also exhibited direct effect on the pulmonary cells as evidenced

Cyanide-induced parkinsonism: clinical, MRI, and 6-fluorodopa PET studies.

A 46-year-old man ingested 1,500 mg of potassium cyanide in a suicide attempt. He survived, but later developed a severe parkinsonian syndrome. MRI revealed multiple areas of low-signal intensity in the globus pallidus and posterior putamen. A 6-fluorodopa PET study revealed bilateral decreased uptake in the basal ganglia. This evidence of functional impairment of dopaminergic nigrostriatal neurons is related either to direct toxicity of cyanide or to the effects of cerebral hypoxia secondary to cyanide intoxication.

Effect of inhibitors on sugar and amino acid transport in two teleosts.

After the addition of ouabain or potassium cyanide (10(-4)M) to the filling solution, inhibition of the transport of sugars and amino acids was noted in the intestine of both the fish, Ophiocephalus punctatus (carnivorous) and Heteropneustes fossilis (omnivorous). Regional as well as nutrient variations occurred in the transport process. There was a positive but slower transport of nutrients in the beginning but later a negative transport of sugars and amino acids was noted in different portions of the intestine. The negative transport of amino acids was comparatively higher than that of sugar

Organ-distribution of the metabolite 2-aminothiazoline-4-carboxylic acid in a rat model following cyanide exposure.

The reaction of cyanide (CN(-)) with cystine to produce 2-aminothiazoline-4-carboxylic acid (ATCA) is one of the independent detoxification pathways of cyanide in biological systems. In this report, in vivo production of ATCA and its distributions in plasma and organs were studied after a subcutaneous sublethal dose of 4 mg/kg body weight potassium cyanide (KCN) administration to rats. At this sublethal dose of KCN, ATCA concentration was not significantly increased in the plasma samples, however, it was found significantly increased in liver samples. These results suggested that ATCA might no

Fluorometric determination of cyanide in biological fluids and pyridoxal.

Investigations of the physiological disposition of cyanide during thiosulfate therapy has necessitated the development of a convenient and sensitive method for cyanide analysis. A fluorometric method involving the catalytic conversion of pyridoxal to 4-pyridoxylactone was adapted for use with biological fluids by employing microdiffusion analysis. The presence of the cyanide antagonist, sodium thiosulfate, interferes with the formation of the fluorophore. In order to circumvent this interference, the pH of the diffusion media was altered to selectively diffuse cyanide. After testing various ac

The effects of sodium nitroprusside and cyanide on haemoglobin function.

Incubation of human blood with 0.3 mM sodium nitroprusside (SNP) or 1 mM potassium cyanide (KCN) for 180 min produced a 600-fold increase in red cell cyanide concentration, a 4% decrease in oxygen capacity and a 6% increase in oxygen affinity. These effects were not reproduced in patients receiving SNP by infusion probably because in the clinical situation much smaller amounts of the drug were used and red cell cyanide levels were lower. The in vitro observations could be explained by cyanide either combining directly with the haemoglobin or encouraging its spontaneous oxidation to methaemoglo

Dose and time-dependent effects of cyanide on thiosulfate sulfurtransferase, 3-mercaptopyruvate sulfurtransferase, and cystathionine λ-lyase activities.

We assessed the dose-dependent effect of potassium cyanide (KCN) on thiosulfate sulfurtransferase (TST), 3-mercaptopyruvate sulfurtransferase (3-MPST), and cystathionine λ-lyase (CST) activities in mice. The time-dependent effect of 0.5 LD50 KCN on cyanide level and cytochrome c oxidase (CCO), TST, 3-MPST, and CST activities was also examined. Furthermore, TST, 3-MPST, and CST activities were measured in stored mice cadavers. Hepatic and renal TST activity increased by 0.5 LD50 KCN but diminished by ≥2.0 LD50. After 0.5 LD50 KCN, the elevated hepatic cyanide level was accompanied by increased

Ferrate(VI) oxidation of cyanide in water.

Experiments were conducted to test removal of cyanide (free cyanide and several cyanide complexes) in water, under alkaline medium (pH > or = 11), by a new potassium ferrate salt. The removal rate of free cyanide by oxidation with Fe(VI) was greater at pH 11.0 than at pH 12.0. A complete oxidation was obtained with a 2.67 Fe(VI)/CN ratio at pH 11.0. In these conditions, the rate of cyanide oxidation by Fe(VI) was slow, with a reaction rate constant estimated at 0.95 +/- 0.10 s(-1) l mol(-1) at pH 11.0 and 19.6 degrees C in this study. This study revealed that Fe(VI) did not decompose all cyani
