The authors studied the susceptibility of five species of rodent to monkeypox virus inoculated by various routes and the course of the infection. Reactions varied from complete resistance to lethal generalized infection with rash. Rabbits and white mice appeared to be the most susceptible species and young animals were more susceptible than adults. Monkeypox virus was found to infect young animals by natural routes, i.e., per os and intranasally. Transmission by contact occurred among 10-day-old rabbits. Since antibodies to monkeypox virus may persist for over a year in the sera of convalescen

Monkeypox virus is a double-stranded DNA virus species that causes disease in humans and mammals. It is a zoonotic virus belongs the genus Orthopoxviral, the family of Poxviridae, associated with the smallpox virus in many aspects. The first human case of monkeypox was reported throughout the Democratic Republic of Congo in 1970. In April 2022, several cases were recorded in widespread regions of Africa, the Northern and western hemispheres. The current review spotlights taxonomic classification, clinical presentations during infection, and the pathogenicity of the monkeypox virus in humans. F

Poxviruses target innate immunity mediators such as tumor necrosis factors, interleukins, interferons, complement, and chemokines. It also targets adaptive immunity such as CD4+ T cells, CD4+ T cells, and B cells. Emerging of the recent epidemic of monkeypox virus (MPXV), a zoonotic disease native to Central and Western Africa, besides the lack of permitted treatments for poxviruses infections, encouraged researchers to identify effective inhibitors to help in preventing and treating poxviruses infections. Natural bioactive components, particularly polyphenolics, are promising for creating pow

Monkeypox mutants arising spontaneously or after serial, high multiplicity passage were characterized phenotypically and by restriction endonuclease mapping. Some resemble "whitepox" and variola viruses in several of the markers tested but all are distinguishable phenotypically from these. None resembles "whitepox" viruses in genome structure although near-terminal deletions or symmetrical, terminal rearrangements, relative to parental monkeypox, occurred. "Whitepox" viruses isolated from animals closely resemble variola in both phenotype and genome structure.

Monkeypox virus (MPXV) is a DNA virus belonging to the Orthopoxvirus genus within the Poxviridae family which can cause a zoonotic infection. The unexpected non-endemic outbreak of mpox in 2022 is considered as a new global threat. It is imperative to take proactive measures, including enhancing our understanding of MPXV's biology and pathogenesis, and developing novel antiviral strategies. The host immune responses play critical roles in defensing against MPXV infection while the virus has also evolved multiple strategies for immune escape. This review summarizes the biological features, anti

The unprecedented global spread of monkeypox (mpox) has reinvigorated the pursuit of next-generation poxvirus vaccines. Given the success of the mRNA-lipid nanoparticle (LNP) vaccine platform to vaccinate against SARS-CoV-2, candidate poxvirus mRNA vaccines, as recently reported by Zuiani et al. and Mucker et al., demonstrate success in nonhuman primate (NHP) models and provide insights into the basis of the protective immune response.

Mass mortality events of common carp (Cyprinus carpio, carp) associated with carp edema virus (CEV) alone or in coinfections with koi herpesvirus (KHV), is an emerging issue. Despite recent outbreaks of CEV in wild carp populations, the host range of North American species has not been well studied. To that end, we intensively sampled carp (n = 106) and co-habiting native fish species (n = 5 species; n = 156 total fish) from a CEV-suspect mass-mortality event of carp in a small Minnesota lake (Lake Swartout). Additionally, fecal and regurgitant samples (n = 73 each) from double-crested cormora

A test for monkeypox-specific antibodies is described. Monkeypox immune sera can be made type-specific by immunoabsorption with heterotypic poxvirus extracts. Monkeypox-specific antibodies were demonstrated in sera from 9 cynomolgus monkeys (Macaca fascicularis) that had previously been experimentally infected with monkeypox. Monkeypox-specific antibodies were found in 3 wild-caught African monkeys (Cercopithecus spp.) and in 3 human sera collected from Africans in the Ivory Coast and Nigeria 3(1/2)-4 years after they had suffered a pox-like infection. Monkeypox had been recognized in one of t

An immunocompromised status has been associated with more odds of being infected with Mpox virus (MPXV) and progressing to severe disease. This aligns with the importance of immune competence for MPXV control and clearance. We and others have previously reviewed parallels between MPXV and other viruses belonging to the Poxviridae in affecting the immune system. This article reviews studies providing direct evidence of the MPXV-immune interactions. The wide-ranging effects of MPXV on the immune system, from stimulation to modulation to memory, are broadly categorised, followed by a detailing of

Vaccinia virus (VACV) is the prototypic member of the Orthopoxvirus genus of the Poxviridae. It is also the live vaccine that was used to eradicate smallpox. Like other poxviruses, VACV replicates in the cytoplasm and has a large double-stranded (ds)DNA genome and a complex virion. Approximately, half of the VACV genes are nonessential for virus replication in cell culture but encode a remarkable array of immunomodulators that antagonise the innate immune response to virus infection. This short review concerns one such protein, C6, that is a multifunctional inhibitor of interferon. C6 can both

Natural infections of Vaccinia virus (VACV)-the prototype species of the Orthopoxvirus genus, from the family Poxviridae and subfamily Chordopoxvirinae-cause an occupational emergent zoonotic disease that is primarily associated with the handling of infected dairy cattle. In humans, VACV infection is characterized by skin lesions, primarily on the hands, and accompanied by systemic symptoms such as fever, myalgia, headache, and lymphadenopathy. The diagnosis of VACV is usually performed according to the methods described for other orthopoxviruses. This unit describes the methods utilized to ob

Poxviruses are large double-stranded DNA viruses that replicate exclusively in the cytoplasm. Their life cycle is closely associated with various membrane-related cellular processes. This review summarizes current findings on the complex interplay between poxviruses and autophagy, as well as the endo-lysosomal network. However, due to the large diversity of poxvirus species and the limited number of relevant studies, it remains challenging to draw definitive conclusions regarding the bidirectional regulatory relationship between poxviruses and the autophagy-lysosome system. In addition, poxvir

In the last 4 years, the world has experienced two pandemics of bat-borne viruses. Firstly, in 2019 the SARS-CoV-2 pandemic started and has been causing millions of deaths around the world. In 2022, a Monkeypox pandemic rose in various countries of the world. Those pandemics have witnessed movements and initiatives from healthcare and research institutions to establish a worldwide understanding to battle any future pandemics and biological threats. One Health concept is a modern, comprehensive, unifying ways to improve humans, animals, and ecosystems' health. This concept shows how much they a

Molluscum contagiosum (MC) is a benign, self-limiting infectious lesion that primarily affects children and young adults, especially those with immunodeficiency. MC is caused by a poxvirus from the Molluscipoxvirus genus of the Poxviridae family. Diagnosis is mainly clinical, with lesions presenting as dome-shaped, spherical, solid, flesh-colored, pearly papules with central umbilication. MC commonly appears on the limbs, face, trunk, and axillary regions. The differential diagnosis includes inflammatory, infectious, and neoplastic causes, varying by age and immunological condition. This case

This article reviews the different types of poxvirus infections. Smallpox, although eradicated, must continue to be monitored because of the potential risk of accidental or voluntary (by bioterrorism) reintroduction. Monkeypox and cowpox viruses are considered to be emergent today ; their high risk of dissemination is due to the increase in international transport as well as trends for new animals as pets and the loss of vaccinal protection against smallpox. Molluscum contagiosum (molluscipoxvirus) causes mild infections, is particularly frequent in children ; in adults it is a marker of the r
