Efficacy of Bacteriophages in Propionibacterium acnes-Induced Inflammation in Mice.

BACKGROUND: Bacteriophages have been introduced as living drugs for infectious diseases; thus, they may provide an alternative to conventional acne therapeutics in patients with non-responsive acne. OBJECTIVE: We investigated the effect of bacteriophages using an acne mouse model with Propionibacterium acnes-induced inflammatory nodules by clinical examination, pathology, and immunohistochemical analysis. METHODS: A human-isolated P. acnes suspension (109 colony forming units/µl) was injected into the backs of HR-1 mice. Group A was used as a control, Group B was injected on the back with P.

Hemolysis as a clinical marker for propionibacterium acnes orthopedic infection.

Determining if a Propionibacterium acnes culture is a true infection or a contaminant remains a challenge. We conducted a study to distinguish between a true infection and a contaminated culture based on the P acnes hemolytic phenotype and clinical presentation. All P acnes strains were from orthopedic patients who had undergone arthroplasty or nonarthroplasty shoulder procedures. Hemolysis was determined according to P acnes growth on brucella blood agar plates after 48 to 72 hours. Each patient record that corresponded to the obtained P acnes strains was retrospectively reviewed for clinical

Propionibacterium acnes: an update on its role in the pathogenesis of acne.

In recent years, significant progress has been made in the understanding of the pathophysiological mechanisms of acne and the role of Propionibacterium acnes. With this review, the authors aim to provide an update on the current understanding of the role of P. acnes in the development of acne lesions and analysing the potential implications for future treatments. A total of 188 articles published between January 1980 and March 2013 were searched using key words such as acne, P. acnes, microbiology, Corynebacterium acnes, acne vulgaris, pathogenesis, antibiotic, vaccination and a combination of

The role of Propionibacterium acnes in acne pathogenesis: facts and controversies.

We have come a long way since 1896, when it was first suggested that Propionibacterium acnes, found in acne lesions, was the cause of acne. Although several lines of evidence suggest the direct role of P acnes in acne, the mechanism by which P acnes contributes to the pathogenesis of acne is debated. The importance of P acnes in the induction and maintenance of the inflammatory phase of acne has been established. Emerging data that inflammatory events occur in the very earliest stages of acne development have reopened the debate about the potential involvement of this microorganism in comedoge

Propionibacterium acnes wound contamination at the time of spinal surgery.

UNLABELLED: Bacteria of the normal skin microbiota such as Propionibacterium acnes and coagulase-negative staphylococci often are dismissed as contaminants when detected in clinical samples. Propionibacterium acnes is described as a cause of spinal infection and more recently has been linked to sciatica. To date no researchers formally have examined the incidence of bacterial wound contamination during spinal surgery. Surgical specimens were removed from 79 patients having spinal surgery for analysis using agar culture detection, broth enrichment, and immunofluorescence microscopy. Bacteria we

Propionibacterium acnes as a cause of visually significant corneal ulcers.

PURPOSE: To report Propionibacterium acnes as a cause of vision-threatening infectious keratitis and to discuss culture isolation and antibiotic treatment. METHODS: Retrospective case series presentation collected from three academic medical centers. RESULTS: Six cases of P. acnes infectious keratitis are presented, all of which were associated with a compromised corneal barrier or environment. All cases were culture-positive on thioglycolate broth; none became positive before 7 days of growth. No other organisms were isolated from any culture, and the growth of P. acnes occurred in some cas

Chronic Propionibacterium acnes endophthalmitis as a cause of intermediate uveitis.

Although the pathogenesis in most cases of intermediate uveitis is unknown, a small minority of cases is associated with a variety of specific inflammatory etiologies: sarcoidosis; multiple sclerosis; Lyme disease; syphilis; ocular lymphoma; and as a rare manifestation of Behçet's disease and AIDS. A 61-year-old woman developed pars planitis after cataract surgery. A vitrectomy was performed after ten months when a white capsular plaque and an hypopyon developed. Propionibacterium acnes was isolated. The intermediate uveitis was not controlled until later removal of the intraocular lens and ca

[Porphyrinsynthesis of Propionibacterium acnes in acne and seborrhea (author's transl)].

The porphyrin productions of P a.-strains, isolated on the one hand from inlfammatory acne lesions, on the other hand from sebaceous follicle filaments from 10 persons each were compared quantitatively and qualitatively, after the appropriate material had been pooled, With respect to the acne strains one could observe a somewhat increased porphyrin production as well as the discharging of a precentually greater portion of porphyrins to the surrounding material. When separated by TLC the acne material showed a more intense coproporphyrin band.

Overview: the role of Propionibacterium acnes in nonpyogenic intervertebral discs.

Propionibacterium acnes (P. acnes), an important opportunistic anaerobic Gram-positive bacterium, causes bone and joint infections, discitis and spondylodiscitis. Accumulated evidence suggested that this microbe can colonise inside intervertebral discs without causing symptoms of discitis. Epidemiological investigation shows that the prevalence ranges from 13 % to 44 %. Furthermore, colonisation by P. acnes inside nonpyogenic intervertebral discs is thought to be one pathogen causing sciatica, Modic changes and nonspecific low back pain. Specially, patients can attain significant relief of low

Typing of Propionibacterium acnes: a review of methods and comparative analysis.

Propionibacterium acnes is a major commensal of the human skin. However, it is also the pathogen responsible for acne vulgaris and other diseases, such as medical-device infections. Strains of Propionibacterium acnes have long been classified into several different types. Recently, typing systems for this bacterium have taken on an increased importance as different types of P. acnes have been found to be associated with different disease states, including acne. Genetic approaches based on individual or multiple genes have classified P. acnes into types, which have been supported by the sequenc

Analysis of the Surface, Secreted, and Intracellular Proteome of Propionibacterium acnes.

Propionibacterium acnes , plays an important role in acne vulgaris and other diseases. However, understanding of the exact mechanisms of P. acnes pathogenesis is limited. Few studies have investigated its proteome, which is essential for vaccine development. Here, we comprehensively investigate the proteome of P. acnes strain ATCC 6919, including secreted, cell wall, membrane, and cytosolic fractions in three types of growth media. A total of 531 proteins were quantified using an Orbitrap mass spectrometer and bioinformatically categorized for localization and function. Several, including PPA1

Effect of Garcinia mangostana on inflammation caused by Propionibacterium acnes.

The present study was aimed to investigate the activity of Thai medicinal plants on inflammation caused by Propionibacterium acnes in terms of free radical scavenging and cytokine reducing properties. P. acnes have been recognized as pus-forming bacteria triggering an inflammation in acne. Antioxidant activity was determined by DPPH scavenging and NBT reduction assay. The result showed that Garcinia mangostana possessed the most significant antioxidant activity and reduced reactive oxygen species production. Houttuynia cordata, Eupatorium odoratum, and Senna alata had a moderate antioxidant ef

Molecular Mechanisms of Intervertebral Disc Degeneration Induced by Propionibacterium acnes.

Intervertebral disc degeneration (IVDD), a prevalent degenerative disorder with substantial socioeconomic impacts, is closely linked to endplate inflammation and chronic low back pain. Its pathogenesis involves multifactorial mechanisms, including long-term chronic mechanical loading, external trauma, and hereditary factors. Emerging evidence highlights Propionibacterium acnes (P. acnes), a gram-positive bacterium with potent proinflammatory properties, as a key contributor to IVDD progression. This review systematically analyses the latest literature on related studies, focusing on the molecu

Recent advances in understanding Propionibacterium acnes ( Cutibacterium acnes) in acne.

The skin commensal Propionibacterium acnes, recently renamed Cutibacterium acnes, along with the other major pathophysiological factors of increased seborrhea, hyperkeratinization of the pilosebaceous unit, and inflammation, has long been implicated in the pathogenesis of acne. Recent advances have contributed to our understanding of the role of P. acnes in acne. Although there are no quantitative differences in P. acnes of the skin of patients with acne compared with controls, the P. acnes phylogenic groups display distinct genetic and phenotypic characteristics, P. acnes biofilms are more fr
