Sequential enzymatic hydrolysis of egg yolk proteins: Kinetics, functionality, and bioactivity of hydrolysates.

The lecithin-free egg yolk (LFEY) by-product was hydrolyzed using Alcalase (A) or Flavourzyme (F), or a sequential enzymatic A-F system, under different pH and Flavourzyme concentrations. The hydrolysates' molecular weight (Mw) profile was analyzed, and their functionality and bioactivity were assessed in terms of soluble protein, emulsifying activity index (EAI), emulsion stability index (ESI), and antioxidant and Angiotensin I-converting enzyme (ACEI) inhibitory activities. Hydrolysates were fractionated using 5 and 1 kDa ultrafiltration (UF) membranes. Alcalase efficiently hydrolyzed LFEY p

Repression Effects of Hydrolysates from Hen-Egg Proteins on Amyloid Fibril Formation.

Amyloid fibrils, which are formed from aggregates of aberrant proteins, can cause various forms of amyloidosis (including Alzheimer's disease). Such disorders often occur in elderly populations and are suspected to be lifestyle related. Thus, it has been speculated that some foodstuffs could be beneficial for preventing amyloidosis. In this study, we determine whether fibril formation by the hen egg white lysozyme (HEWL) could be inhibited by conducting a thioflavin T assay followed by fluorescence and electron microscopy observations. The results demonstrated that four peptide specimens prepa

Protein Hydrolysate from Pterygoplichthys disjunctivus, Armoured Catfish, with High Antioxidant Activity.

Pterygoplichthys disjunctivus, locally named the armoured catfish, is a by-catch of tilapia fishing that accounts for up to 80% of total captured fish in the Adolfo Lopez Mateos dam, in Michoacán, México, affecting the economy of its surrounding communities. This invasive fish is discarded by fishermen since native people do not consume it, partly due to its appearance, yet it is rich in protein. The aim of this study was to produce hydrolysates from armoured catfish using food-grade proteases (neutrases HT and PF and alcalase PAL) and investigate the processing conditions (pH and temperature)

Whey protein hydrolysate fibrils-dextran delivery iron oxides: stability and bioavailability.

Iron deficiency anemia (IDA) is one of the most common nutritional disorders worldwide. However, traditional iron supplement often induces adverse gastrointestinal effects (e.g. nausea, vomiting, abdominal pain) due to their gastrointestinal irritation and poor bioavailability. To overcome this defect, we designed an antioxidative whey protein hydrolysate fibril-dextran (WPHF-DEX) complex system, which can delivery low-irritant and highly biocompatible iron oxide by in-situ co-precipitation and enhancing iron bioavailability. After mixing with WPH and grafting with dextran, the antioxidant of

Blood pressure lowering effect of a pea protein hydrolysate in hypertensive rats and humans.

The blood pressure lowering effect of a pea protein hydrolysate (PPH) that contained <3 kDa peptides, isolated by membrane ultrafiltration from the thermolysin digest of pea protein isolate (PPI), was examined using different rat models of hypertension as well as hypertensive human subjects. The PPH showed weak in vitro activities against renin and angiotensin converting enzyme (ACE) with inhibitory activities of 17 and 19%, respectively, at 1 mg/mL test concentration. Oral administration of the PPH to spontaneously hypertensive rats (SHR) at doses of 100 and 200 mg/kg body weight led to a low

Is caffeic acid, as the major metabolite present in Moscatel wine protein haze hydrolysate, involved in protein haze formation?

This work was conducted to identify the major low molecular weight compounds present in the wine precipitate and to assess their potential contribution to wine protein haze formation. The heat-induced protein precipitate from a white Moscatel of Alexandria wine was subjected to alkaline hydrolysis. The major compound present was found to be caffeic acid among other minor, unidentified compounds. Caffeic acid was identified by both UV-vis and 1H NMR spectra. The concentration of caffeic acid in the original wine sample was 1.1mg/L, as quantified by HPLC following SPE. Heat stability tests were

Tripeptide Leu-Pro-Phe from Corn Protein Hydrolysates Attenuates Hyperglycemia-Induced Neural Tube Defect in Chicken Embryos.

Neural tube defect (NTD) is the most common and severe embryopathy causing embryonic malformation and even death associated with gestational diabetes mellitus (GDM). Leu-Pro-Phe (LPF) is an antioxidative tripeptide isolated from hydrolysates of corn protein. However, the biological activity of LPF in vivo and in vitro remains unclear. This study is aimed at investigating the protective effects of tripeptide LPF against NTD in the high glucose exposure condition and delineate the underlying biological mechanism. We found that LPF alleviated NTD in the high glucose-exposed chicken embryo model.
