Protein microgels tend to adsorb at the surface of dispersed droplets in water-in-water (W/W) emulsions. It was hypothesized that suspensions of stable protein microcapsules (MC) can be produced by spontaneous crosslinking of the microgels at the interface in a process akin so-called cold gelation. Rapeseed or whey protein microgels added to W/W emulsions formed by mixing polyethylene oxide (PEO) and Dextran (DEX) adsorbed at the W/W interface. The rapeseed protein microgels were found to spontaneously crosslink and formed MC suspensions that were stable to dilution and washing. The size of th

The nucleation of cholesterol crystals is an obligatory precursor to cholesterol gallstone formation. Nucleation, in turn, is believed to be preceded by aggregation and fusion of cholesterol-rich vesicles. We have investigated the effects of two putative pro-nucleating proteins, a concanavalin A-binding protein fraction and a calcium-binding protein, on the stability of sonicated small unilamellar cholesterol-lecithin vesicles. Vesicle aggregation is followed by monitoring absorbance, and upon addition of the concanavalin A-binding protein fraction the absorbance of a vesicle dispersion increa

Synaptic function and plasticity depend on the precise control of protein abundance and turnover, governed by the balance of synthesis and degradation. This review examines the regulatory mechanisms that maintain synaptic protein stability, focusing on the ubiquitin-proteasome system, autophagy-lysosomal pathways, and related proteolytic systems. We detail how key enzymes, including E3 ligases such as Nedd4-1, Mdm2, and Parkin, and deubiquitinating enzymes like USP46 and USP8, dynamically regulate the degradation of critical synaptic components from AMPA and NMDA receptors to scaffolds like PS

Intervening proteins (inteins) interrupt host protein sequences and are removed by a self-mediated protein splicing reaction. Inteins are abundant in the microbial world and are often found within essential genes involved in DNA replication, recombination, and repair. Compelling examples of conditional protein splicing, where intein removal and subsequent host protein activation are highly dependent on environmental cues, suggest that some inteins serve as post-translational regulatory elements. Within the cellular context, however, the factors influencing protein splicing are still largely un

DJ-1 is a conserved, disease-associated protein that protects against oxidative stress and mitochondrial damage in multiple organisms. Human DJ-1 contains a functionally essential cysteine residue (Cys106) whose oxidation is important for regulating protein function by an unknown mechanism. This residue is well-conserved in other DJ-1 homologues, including two (DJ-1α and DJ-1β) in Drosophila melanogaster. Because D. melanogaster is a powerful model system for studying DJ-1 function, we have determined the crystal structure and impact of cysteine oxidation on Drosophila DJ-1β. The structure of

Enhancing thermal stability in high-content soy protein solutions (SPs) is crucial. Cyclodextrins (CDs), with their distinctive hydrophobic cavities, are commonly used as stabilizers in food systems. This study investigates the effects of cyclodextrins with different cavity sizes (α-CD, β-CD, and γ-CD) and preheating on the thermal stability of soy proteins (SPs). The results show that all CDs improved reheating stability, but their effects were cavity-size dependent. Larger cavities enhanced binding to exposed hydrophobic groups and increased electrostatic repulsion; γ-CD reduced surface hydr

BACKGROUND: Metabolic dysfunction-associated steatotic liver disease (MASLD) is a progressive hepatic disorder that is closely associated with metabolic syndrome. Zinc finger matrin-type protein 3 (ZMAT3), an RNA-binding protein implicated in various malignancies, has not been fully clarified in the context of MASLD.
METHODS: To investigate the role of ZMAT3 in MASLD, a murine model was established using a choline-deficient amino acid (CDAA) diet, combined with hepatocyte-specific ZMAT3 overexpression. An in vitro model was generated by treating AML12 hepatocytes with free fatty acids to eval

Prion protein (PrP) adopts either a helical conformation (PrPC) or an alternative, beta sheet-rich, misfolded conformation (PrPSc). The PrPSc form has the ability to "infect" PrPC and force it into the misfolded state. Accumulation of PrPSc is associated with a number of lethal neurodegenerative disorders, including Creutzfeldt-Jacob disease (CJD). Knockout of PrPC protects cells and animals from PrPSc infection; thus, there is interest in identifying factors that regulate PrPC stability, with the therapeutic goal of reducing PrPC levels and limiting infection by PrPSc. Here, we assembled a sh

Prions and other self-propagating protein aggregates can exist as distinct strains, which are thought to represent different conformations of aggregates. There is growing evidence that protein aggregate strains may be important for understanding the biology of common neurodegenerative disorders, such as Alzheimer's disease and Parkinson's disease. While methodology for discriminating prion strains is in widespread use, there is a paucity of tools for comparing the conformational properties of aggregates composed of β-amyloid (Aβ) peptide or α-synuclein protein, particularly when present in com

Glutamate carboxypeptidase II (GCPII) is known to be implicated in brain diseases such as schizophrenia and bipolar disorder, and dramatically increases in prostate cancer. Here, we investigated the regulation of GCPII expression in astrocytes and examined whether GCPII is epigenetically regulated through histone modification. In this study, valproic acid (VPA), a drug used for bipolar disorder and epilepsy and a known histone deacetylase (HDAC) inhibitor was used. We found that acute exposure of VPA for 4-6h increased the GCPII protein level in human astrocyte U87MG cells but did not have a s

Mangiferin, a natural bioactive compound with multiple physiological functions, faces limitations in its oral bioavailability due to poor stability and solubility in the gastrointestinal environment. Liposome-based delivery systems have shown promise in enhancing the bioavailability of such compounds, but their stability under physiological conditions remains a challenge. In this study, mangiferin-loaded liposomes were prepared via the ethanol injection method. Using entrapment efficiency as the evaluation index, the optimal preparation conditions were determined through a combination of singl

FBXW7, the substrate recognition component of the SCFFBXW7 E3 ubiquitin ligase complex, functions as a tumor suppressor by regulating cell cycle progression, stem cell maintenance, and cellular differentiation. Loss of FBXW7 contributes to hematopoietic malignancies and promotes monocyte-to-macrophage differentiation. Here, we identify the lineage-determining myeloid transcription factor MAFB as a previously unrecognized substrate of FBXW7. MAFB is a master regulator of monocyte-macrophage lineage commitment, terminal differentiation, and anti-inflammatory macrophage identity. We demonstrate t

Programmed cell death is essential for maintaining cellular homeostasis, and its evasion is a defining hallmark of cancer. The threshold for apoptosis is largely determined by the stability of pro- and anti-apoptotic proteins, which is tightly regulated by the ubiquitin-proteasome system (UPS). In this system, E1, E2, and E3 enzymes sequentially attach ubiquitin chains that mark substrates for degradation by the 26S proteasome, whereas deubiquitinating enzymes (DUBs) reverse these modifications to stabilize specific substrates. The opposing actions of E3 ubiquitin ligases and DUBs therefore fo

Lipid nanoparticles (LNPs) have become pivotal in advancing modern medicine, from mRNA-based vaccines to gene editing with CRISPR-Cas9 systems. Though LNPs based therapeutics offer promising drug delivery with satisfactory clinical safety profiles, concerns are raised regarding their potential nanotoxicity. Here, we explore the impacts of LNPs on protein stability in buffer and cellular protein homeostasis (proteostasis) in HepG2 cells. First, we show that LNPs of different polyethylene glycol (PEG) molar ratios to total lipid ratio boost protein aggregation propensity by reducing protein stab

Endothelial dysfunction (ED) serves as the pathological basis for various cardiovascular diseases. Guanosine triphosphate cyclopyrrolone 1 (GCH1) emerges as a pivotal protein in sustaining nitric oxide (NO) production within endothelial cells, yet it undergoes degradation under oxidative stress, contributing to endothelial cell dysfunction. Citronellal (CT), a monoterpenoid, has been shown to ameliorate endothelial dysfunction induced by in atherosclerosis rats. However, whether CT can inhibit the degradation of GCH1 protein is not clear. It has been reported that ubiquitination may play a cru

Amyloid precursor protein (APP) is a transmembrane protein that plays a crucial role in the production of amyloid-β peptides. Any disruption in APP protein production, its mRNA decay rate or processing may result in abnormal production of amyloid-β peptides and subsequent development of protein aggregation diseases. Therefore, the equilibrium is crucial for neuronal function. An association study of heterogeneous nuclear ribonucleoprotein (hnRNP)-F and hnRNP H1 with APP was carried out in Neuro-2a (N2a) cells. In the present study, we found that hnRNP F and hnRNP H1 were significantly upregula

Polysorbates (PS) are commonly used surfactants in biopharmaceutical protein formulations. However, they are susceptible to a variety of degradation pathways, including chemical hydrolysis, oxidation, and enzymatic hydrolysis. Polysorbates are also heterogeneous mixtures, and it has been observed that the patterns of degradation can be strikingly different between the different pathways. Polysorbates (PS20 and PS80) were fractionated, and the fractions were characterized for their physicochemical properties, such as surface tension, micelle size, critical micelle concentration (CMC), and agita

Commercially available, uncoated elastomeric closures were examined in regard to a potential contribution of extracted compounds from the rubber stoppers to protein aggregation under worst-case conditions. All rubber stoppers were confirmed to comply with Ph. Eur. quality requirements. Extraction with 2-propanol under reflux-conditions for 3 h led to closure-specific extraction profiles of the tested samples. One type of rubber stopper exhibited a considerably greater number and higher content of extractables. Four extracted compounds were identified as trialkyl benzene-1, 2, 4-tricarboxylates

A previous computational analysis of missense mutations linked to monogenic disease found a high proportion of missense mutations affect protein stability, rather than other aspects of protein structure and function. The purpose of this study is to relate the presence of such stability damaging missense mutations to the levels of a particular protein present under "in vivo" like conditions, and to test the reliability of the computational methods. Experimental data on a set of missense mutations of the enzyme phenylalanine hydroxylase (PAH) associated with the monogenic disease phenylketonuria

Effects of protein kinase C on protein stability and activity of rat AANAT were investigated in vitro and in vivo. When COS-7 cells transfected with AANAT cDNA were treated with phorbol 12-myristate 13-acetate (PMA), both the activity and protein level of AANAT were increased. These effects of PMA were blocked by GF109203X, a specific inhibitor of PKC. Moreover, PMA increased the phosphorylation of AANAT and induced the formation of AANAT/14-3-3zeta complex. PMA did not affect the basal level of cAMP and did not involve the potentiation of the cAMP production by forskolin, indicating that PKC-
