A Type II CDK6 Degrader Enables Cellular Targeting beyond the Limits of Type II Inhibition.

PROTACs are commonly developed by linking E3 ligase-recruiting ligands to established inhibitors of a protein target, often resulting in degraders that retain enzymatic inhibition. Type II inhibition of cyclin-dependent kinases (CDKs) has been challenging, as reported compounds generally exhibit weak biochemical potency and limited cellular activity. Consistent with these limitations, most reported CDK degraders have been derived from type I ATP-competitive inhibitors. Here, we explored whether targeted protein degradation could enable functional CDK targeting from a type II kinase scaffold. U

Structural diversity and chemical space analysis of a PROTAC database using unsupervised machine learning.

Targeted protein degradation (TPD) mediated by proteolysis-targeting chimeras (PROTACs) has emerged as a powerful therapeutic strategy, enabling the catalytic and selective elimination of disease-relevant proteins via the ubiquitin-proteasome system. Despite their ability to overcome drug resistance and address traditionally undruggable targets, the structural complexity and vast chemical diversity of PROTACs present challenges for systematic analysis and rational design. Here, we present a systematic unsupervised machine-learning framework that represents the first large-scale similarity-driv

Autophagy-tethering compounds (ATTECs) as an emerging and potentially transformative drug discovery approach.

Targeted protein degradation (TPD) strategies leveraging the ubiquitin-proteasome system (UPS), such as proteolysis-targeting chimeras (PROTACs), have gained wide recognition. While the UPS predominantly degrades short-lived soluble proteins, the lysosome-mediated system (LMS) excels at processing larger substrates, including protein aggregates, organelles, and macromolecular complexes. Recently, autophagy-tethering compounds (ATTECs) have emerged as a promising strategy for targeted degradation, harnessing the autophagy-lysosome system (ALS) to enable proximity-induced degradation. These hete

Autonomously Motile Nano-PROTACs Act as Protein-Sweeping Robots to Enhance Targeted Protein Degradation.

Although nanoparticle-based PROTACs (nPROs) provide a modular strategy to improve the biological fate of PROTACs, their degradation performance was fundamentally limited by restricted intracellular diffusivity. Here, we introduced an unconventional solution: Imparting autonomous motility to nPROs to transform them from passive carriers into active protein-seeking degraders. We engineered nano-motoring PROTACs (nMPROs) through asymmetric modification of gold nanoparticles, displaying POI ligands and E3 recruiters on one hemisphere while anchoring catalase on the opposite hemisphere. Targeted protein degradation in pancreatic cancer: advances and challenges of PROTAC-based therapeutics.

Pancreatic cancer remains one of the deadliest malignancies, with limited therapeutic options and a dismal prognosis. Traditional treatment modalities, including chemotherapy and molecular targeted therapies, often face challenges such as drug resistance, limited targetability, and systemic toxicity. The emergence of proteolysis-targeting chimeras (PROTACs) has opened a new frontier in targeted protein degradation, offering a promising approach to overcome these limitations by hijacking the ubiquitin-proteasome system to selectively degrade disease-related proteins. Research progress of PROTACs for neurodegenerative diseases therapy.

Neurodegenerative diseases (NDD) are characterized by the gradual deterioration of neuronal function and integrity, resulting in an overall decline in brain function. The existing therapeutic options for NDD, including Alzheimer's disease, Parkinson's disease, and Huntington's disease, fall short of meeting the clinical demand. A prominent pathological hallmark observed in numerous neurodegenerative disorders is the aggregation and misfolding of proteins both within and outside neurons. These abnormal proteins play a pivotal role in the pathogenesis of neurodegenerative diseases. Targeted degr

Overcoming breast cancer resistance through targeted protein degradation and next generation chimeras.

Proteolysis-Targeting Chimeras (PROTACs) represent a paradigm shift from occupancy-driven inhibition to event-driven protein elimination in cancer therapy. By harnessing the ubiquitin-proteasome system, PROTACs catalytically degrade disease-driving proteins. In breast cancer, the most commonly diagnosed cancer in women globally, PROTAC technology has advanced rapidly from preclinical concept to clinical validation. This review systematically evaluates PROTAC development across multiple therapeutic targets in breast cancer, including estrogen receptor α (ERα), human epidermal growth factor rece

Proteolysis Targeting Chimeric-Based Technology in Myeloma and Lymphoma.

Proteolysis-targeting chimeras (PROTAC) leverage the ubiquitin-proteasome system to selectively degrade oncogenic proteins, including those previously seen as undruggable. Recent preclinical studies indicate that PROTACs may represent a novel therapeutic strategy in lymphoma and myeloma. Indeed, preclinically, PROTACs have shown high efficacy and remarkable selectivity, a favorable safety profile, and lower toxicity compared with conventional therapies. Their catalytic, reusable mechanism enables drug dosing and offers the perspective of long-term low-dose treatment. PROTACs have demonstrated

Targeting the undruggables-the power of protein degraders.

Undruggable targets typically refer to a class of therapeutic targets that are difficult to target through conventional methods or have not yet been targeted, but are of great clinical significance. According to statistics, over 80% of disease-related pathogenic proteins cannot be targeted by current conventional treatment methods. In recent years, with the advancement of basic research and new technologies, the development of various new technologies and mechanisms has brought new perspectives to overcome challenging drug targets. Among them, targeted protein degradation technology is a break

Targeted Degradation of Signal Transduction and Activator of Transcription 3 by Chaperone-Mediated Autophagy Targeting Chimeric Nanoplatform.

Chaperone-mediated autophagy (CMA) is a lysosomal-dependent proteolysis pathway for the degradation of cytosolic proteins. However, exploiting CMA-mediated proteolysis to degrade proteins of interest in cancer therapy has not been widely applied. In this study, we develop a CMA-targeting chimera (CMATAC) to efficiently and specifically degrade signal transduction and activator of transcription 3 (STAT3) in tumor cells. CMATAC consists of STAT3 and heat shock cognate 70 kDa protein (HSC70) targeting peptides connected by a linker. To efficiently deliver CMATACs into tumor cells, lipid nanoparti

PROTACs for central nervous system disorders: challenges and opportunities.

Central nervous system (CNS) disorders, particularly neurodegenerative diseases and brain tumors, pose substantial and long-standing challenges to the global public health system. Current therapeutic approaches for CNS disorders are primarily confined to symptomatic relief and generally fail to halt or reverse disease progression. However, proteolysis-targeting chimeras (PROTACs), as an emerging therapeutic strategy, have introduced new prospects for effective intervention in this domain. Accelerate Your Science: Direct-to-Biology Strategies in Medicinal Chemistry.

Direct-to-biology (D2B) is a powerful strategy that accelerates early drug discovery. It enables compounds to be synthesized in miniaturized formats and evaluated directly as crude reaction mixtures. This bypasses the need for purification during the initial design-make-test cycle. Advances in robust synthetic methodologies, automation, reaction miniaturization, and biological screening have transformed D2B from a proof-of-concept approach into a versatile medicinal chemistry platform. This platform is applicable to fragment optimization, covalent ligands, macrocycles, proteolysis-targeting ch

BacPROTAC-induced protein degradation as a new antibiotic concept.

Antibiotic resistance is spreading worldwide, associated with more than 5 million deaths annually, yet few new antibiotics have emerged in recent years. This gap highlights the need for alternative strategies that enable the rational design of antibacterial agents. One promising approach is the development of proteolysis-targeting chimeras in bacteria (BacPROTACs), which can hijack endogenous degradation pathways. BacPROTACs tether selected proteins to caseinolytic proteases (Clp) to promote their degradation. Thus, unlike conventional inhibitors, they eliminate their targets, act in a catalyt

Progress in targeting the untouchables: emerging approaches for hard-to-drug cancer targets.

Innovation in drug development is an evolving concept that can take many forms and is often confused with iteration, i.e. new versions of existing drug classes targeting familiar oncogenes. Yet meaningful innovation increasingly lies in translating approaches to historically 'untouchable' targets: transcription factors, tumor suppressors, and lineage-defining proteins, which have long resisted conventional pharmacologic approaches. At the European Society for Medical Oncology (ESMO) Targeted Anticancer Therapies (TAT) 2025 Congress, a growing body of early-phase trials has continued to test an

Viral infections pose a persistent threat to human health. The relentless epidemic of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has become a global health problem, with millions of infections and fatalities so far. Traditional approaches such as random screening and optimization of lead compounds by organic synthesis have become extremely resource- and time-consuming. Various modern innovative methods or integrated paradigms are now being applied to drug discovery for significant resistance in order to simplify the drug process. This review provides an overview of newly emer

Targeted Protein Degradation Strategies for Lung Cancers: Focusing on Epidermal Growth Factor Receptor.

Lung cancer remains the leading cause of cancer-related mortality worldwide, with dysregulation of epidermal growth factor receptor (EGFR) playing a pivotal role in its pathogenesis and progression. Although EGFR-targeted tyrosine kinase inhibitors (TKIs) initially provide clinical benefits, resistance inevitably develops. Targeted protein degradation (TPD) offers a paradigm shift in cancer therapy by eliminating pathogenic proteins entirely, rather than merely inhibiting their function. This review comprehensively examines TPD strategies for lung cancer treatment, focusing on EGFR as an exemp

In situ self-assembly of PROTACs for precision cancer therapy.

Proteolysis-targeting chimeras (PROTACs) are heterobifunctional molecules that degrade pathogenic proteins via the ubiquitin-proteasome system. Their event-driven mechanism enables targeting of traditionally undruggable proteins and overcomes acquired resistance through complete protein degradation. Despite these advantages in cancer therapy, clinical translation of PROTACs is hampered by high molecular weight, poor solubility, off-target effects, and the hook effect. To address these challenges, integrating bioorthogonal in situ self-assembly of PROTACs with advanced nanodelivery platforms ha

A nuclear-targeted PROTAC enables subcellular-selective degradation of AURKA.

Aurora kinase A (AURKA) exerts its oncogenic effects partly through aberrant nuclear accumulation, thereby activating mechanisms that differ from its canonical cytoplasmic roles in cell cycle regulation. However, existing small molecule compounds, such as kinase inhibitors or degraders, are unable to selectively target nuclear AURKA. In this study, we developed a novel class of AURKA proteolysis targeting chimeras (PROTACs) that incorporate the UNC1215 warhead, which has been shown to recruit the nuclear L3MBTL3-DCAF5 E3 ligase complex in prior work. Through linker optimization, we synthesized

Integrative Proteomics and Ubiquitomics Reveal on-Targets and off-Targets of PROTAC dBET1.

Proteolysis-targeting chimeras (PROTACs) are heterobifunctional molecules that induce selective degradation of target proteins by hijacking the ubiquitin-proteasome system (UPS). Despite their transformative potential in eliminating disease-associated proteins, comprehensively identifying off-target degradation events remains technically challenging. Here, we employed an integrated proteomic and ubiquitinomic strategy to systematically profile the degradation landscape of the PROTAC molecule dBET1 in Jurkat T cells. By capturing the upstream ubiquitination events─which serve as earlier and mor
