Interplay of ELF5, GATA-3 and FLI-1 regulate malignant transformation of keratinocytes.

Cutaneous squamous cell carcinoma (cSCC) arises from epidermal keratinocytes and is the second most common epithelial malignancy. E74-like factor 5 (ELF5) is a member of the epithelium specific ETS subfamily of transcription factors and is essential for epithelial development, homeostasis and prevention of epithelial tumourigenesis. Transformation of keratinocytes involves the disruption of the progenitor differentiation program causing keratinocytes to fail to complete the process. This can lead to dysplastic epithelium and precancerous cells in skin. However, the precise mechanisms that caus

IL-22/IL-22R signaling attenuates mitochondrial damage in PCOS by modulating granulosa cell lipid metabolism through ETS1.

OBJECTIVE: This study investigates the therapeutic potential of interleukin-22 (IL-22) in polycystic ovary syndrome (PCOS) and its mechanisms. METHODS: A dehydroepiandrosterone (DHEA)-induced PCOS mouse model and testosterone-treated human KGN granulosa cells were utilized. Mice received IL-22 (50-100 µg/kg) or Diane-35 for 3 weeks. RESULTS: IL-22 administration restored estrous cyclicity, reduced ovarian cysts, and normalized serum testosterone and insulin levels in DHEA or letrozole-induced PCOS mice. DHEA induction led to mitochondrial damage in granulosa cells (GCs), evidenced by impaire

ETS1 Protein Expression May Be Altered by the Complementarity of ETS1 mRNA Sequences with miR-203a-3p and miR-204-3p in Papillary Thyroid Carcinoma.

The expressions of ETS1, miR-203a-3p, and miR-204-3p in papillary thyroid carcinoma (PTC) are poorly described, and their clinical significance is unclear. To determine the prognostic value of ETS1 (E26 transformation-specific), its levels in divergent cell compartments were paired with miR-203a-3p/-204-3p levels and linked to the presence of unfavorable clinical characteristics of PTC patients. Immunohistochemistry and Western blot were performed to evaluate ETS1 protein expression in PTC and matched nonmalignant thyroid tissue (NMT). qPCR was utilized to quantify ETS1 mRNA, miR-203a-3p, and

Dissecting the endothelial cell landscape in meningioma: single-cell insights into PLVAP+ subpopulations and their role in tumor angiogenesis.

BACKGROUND: Meningioma (MEN) is one of the most common intracranial tumors, with a significantly higher incidence rate in females than in males. Although the majority of cases are benign, tumors located in complex anatomical regions or classified as atypical or malignant have a high recurrence rate, underscoring the need to optimize therapeutic strategies to improve patient outcomes. Therefore, this study utilizes single-cell RNA-sequencing technology to investigate the interaction mechanisms between endothelial cells (ECs) and meningiomas, aiming to identify potential therapeutic targets for

Review of Ets1 structure, function, and roles in immunity.

The Ets1 transcription factor is a member of the Ets gene family and is highly conserved throughout evolution. Ets1 is known to regulate a number of important biological processes in normal cells and in tumors. In particular, Ets1 has been associated with regulation of immune cell function and with an aggressive behavior in tumors that express it at high levels. Here we review and summarize the general features of Ets1 and describe its roles in immunity and autoimmunity, with a focus on its roles in B lymphocytes. We also review evidence that suggests that Ets1 may play a role in malignant tra

Fatty acid synthesis promotes mtDNA release via ETS1-mediated oligomerization of VDAC1 facilitating endothelial dysfunction in sepsis-induced lung injury.

Sepsis involves endothelial cell dysfunction leading to the development of lung injury. Fatty acid synthesis contributes to the development of inflammatory injury in sepsis. However, the regulatory mechanisms of fatty acid synthesis-related endothelial activation remain unclear. In this study, we found that fatty acid synthesis in patients with sepsis was greatly disordered. Inhibition of fatty acid synthesis significantly alleviated sepsis-induced endothelial damage and lung injury both in vitro and in vivo. We further found that the release of mtDNA participated in fatty acid synthesis-relat

A Poorly Differentiated Non-keratinizing Sinonasal Squamous Cell Carcinoma with a Novel ETV6-TNFRSF8 Fusion Gene.

Squamous cell carcinoma of the sinonasal tract is relatively rare and morphologically and genetically heterogeneous. We report the case of an adult male with a left sphenoid sinus mass. A biopsy revealed an undifferentiated carcinoma composed of sheets of epithelioid cells lacking keratinization and glandular formation. The tumor was associated with a prominent lymphoplasmacytic inflammatory infiltrate. Immunohistochemical staining demonstrated diffuse expression of pankeratin and p63; it was negative for p16. In addition, EBER was also negative. Morphologically the findings raised the possibi

Opposing functions of the ETS factor family define Shh spatial expression in limb buds and underlie polydactyly.

Sonic hedgehog (Shh) expression during limb development is crucial for specifying the identity and number of digits. The spatial pattern of Shh expression is restricted to a region called the zone of polarizing activity (ZPA), and this expression is controlled from a long distance by the cis-regulator ZRS. Here, members of two groups of ETS transcription factors are shown to act directly at the ZRS mediating a differential effect on Shh, defining its spatial expression pattern. Occupancy at multiple GABPα/ETS1 sites regulates the position of the ZPA boundary, whereas ETV4/ETV5 binding restrict

ARID1A mutations protect follicular lymphoma from FAS-dependent immune surveillance by reducing RUNX3/ETS1-driven FAS-expression.

The cell death receptor FAS and its ligand (FASLG) play crucial roles in the selection of B cells during the germinal center (GC) reaction. Failure to eliminate potentially harmful B cells via FAS can lead to lymphoproliferation and the development of B cell malignancies. The classic form of follicular lymphoma (FL) is a prototypic GC-derived B cell malignancy, characterized by the t(14;18)(q32;q21)IGH::BCL2 translocation and overexpression of antiapoptotic BCL2. Additional alterations were shown to be clinically relevant, including mutations in ARID1A. ARID1A is part of the SWI/SNF nucleosome

All-Trans Retinoic Acid Modulates MicroRNA-129-5p/Hypoxia-Inducible Factor-1α/Vascular Endothelial Growth Factor Signaling and Suppresses Ets-1 in Retinal Pigment Epithelial ARPE-19 Cells.

Purpose: Proliferative vitreoretinopathy (PVR) is a vision-threatening complication of retinal detachment or ocular trauma characterized by the formation of contractile fibrotic membranes. Retinal pigment epithelium (RPE) cells are central to PVR pathogenesis, driving maladaptive wound-healing responses. This study investigated the effects of all-trans retinoic acid (ATRA) on RPE cell proliferation, vascular endothelial growth factor (VEGF) secretion, and miR-129-5p biogenesis, alongside the downstream regulation of Ets-1 and the hypoxia-inducible factor-1α (HIF-1α)/VEGF axis. Methods: Human AR

A targeted CRISPR screen identifies ETS1 as a regulator of HIV-1 latency.

Human Immunodeficiency virus (HIV) infection is regulated by a wide array of host cell factors that combine to influence viral transcription and latency. To understand the complex relationship between the host cell and HIV-1 latency, we performed a lentiviral CRISPR screen that targeted a set of host cell genes whose expression or activity correlates with HIV-1 expression. We further investigated one of the identified factors - the transcription factor ETS1, and found that it is required for maintenance of HIV-1 latency in both latently infected cell lines and in a primary CD4 T cell latency m

MYB and ELF3 differentially modulate labor-inducing gene expression in myometrial cells.

Spontaneous uterine contractions are initiated when smooth muscle cells (SMCs) within the uterine muscle, or myometrium, transition from a functionally dormant to an actively contractile phenotype at the end of the pregnancy period. We know that this process is accompanied by gestational time point-specific differences in the SMC transcriptome, which can be modulated by the activator protein 1 (AP-1), nuclear factor kappa beta (NF-κβ), estrogen receptor (ER), and progesterone receptor (PR) transcription factors. Less is known, however, about the additional proteins that might assist these fact

Intravitreal Ets1 siRNA alleviates choroidal neovascularization in a mouse model of age-related macular degeneration.

Choroidal neovascularization (CNV) is the basic feature of neovascular age-related macular degeneration (AMD), the leading cause of blindness in elders. Macrophages and microglia promote CNV via producing pro-angiogenic factors and inflammatory cytokines. Transcription factor E26 transformation specific-1 (Ets1) plays a pro-angiogenic role via its pro-inflammatory function. In this study, Ets1 increased and localized in the macrophages and microglia of a mouse laser-induced CNV region. Ets1 siRNA intravitreal injection ameliorated the leakage and area of CNV, as well as inhibiting the dysfunct

ELF3 suppresses gallbladder cancer development through downregulation of the EREG/EGFR/mTOR complex 1 signalling pathway.

The prognosis of gallbladder cancer (GBC) remains poor, and a better understanding of GBC molecular mechanisms is important. Genome sequencing of human GBC has demonstrated that loss-of-function mutations of E74-like ETS transcription factor 3 (ELF3) are frequently observed, with ELF3 considered to be a tumour suppressor in GBC. To clarify the underlying molecular mechanisms by which ELF3 suppresses GBC development, we performed in vivo analysis using a combination of autochthonous and allograft mouse models. We first evaluated the clinical significance of ELF3 expression in human GBC tissues

E74-like factor 3 (ELF3) impacts on matrix metalloproteinase 13 (MMP13) transcriptional control in articular chondrocytes under proinflammatory stress.

Matrix metalloproteinase (MMP)-13 has a pivotal, rate-limiting function in cartilage remodeling and degradation due to its specificity for cleaving type II collagen. The proximal MMP13 promoter contains evolutionarily conserved E26 transformation-specific sequence binding sites that are closely flanked by AP-1 and Runx2 binding motifs, and interplay among these and other factors has been implicated in regulation by stress and inflammatory signals. Here we report that ELF3 directly controls MMP13 promoter activity by targeting an E26 transformation-specific sequence binding site at position -78

SLC45A3-ELK4 is a novel and frequent erythroblast transformation-specific fusion transcript in prostate cancer.

Chromosomal rearrangements account for all erythroblast transformation-specific (ETS) family member gene fusions that have been reported in prostate cancer and have clinical, diagnostic, and prognostic implications. Androgen-regulated genes account for the majority of the 5' genomic regulatory promoter elements fused with ETS genes. TMPRSS2-ERG, TMPRSS2-ETV1, and SLC45A3-ERG rearrangements account for roughly 90% of ETS fusion prostate cancer. ELK4, another ETS family member, is androgen regulated, involved in promoting cell growth, and highly expressed in a subset of prostate cancer, yet the

ETS1-EGR2 facilitates renal cell carcinoma progression by activating NOTCH signalling.

BACKGROUND: Renal cell carcinoma (RCC) is known as one of the three major malignant tumors of the urinary system, and clear-cell RCC (ccRCC) is the most frequent and aggressive pathological subtype and is correlated with high mortality. Despite diverse treatments, the clinical prognosis of patients with RCC remains poor. Therefore, further characterization of the mechanisms underlying ccRCC progression is urgently required. METHODS: Different RCC celllines were used to study cell malignancies in vitro and to elucidate mechanisms. Nude mice were used for validating results in vivo. Sequencing,
