Reduction of PCN biosynthesis by NO in Pseudomonas aeruginosa.

Pyocyanin (PCN), a virulence factor synthesized by Pseudomonas aeruginosa, plays an important role during clinical infections. There is no study of the effect of nitric oxide (NO) on PCN biosynthesis. Here, the effect of NO on PCN levels in Pseudomonas aeruginosa strain PAO1, a common reference strain, was tested. The results showed that the NO donor sodium nitroprusside (SNP) can significantly reduce PCN levels (82.5% reduction at 60μM SNP). Furthermore, the effect of endogenous NO on PCN was tested by constructing PAO1 nor (NO reductase gene) knockout mutants. Compared to the wild-type strai

Testing the Luedemann hypothesis: the discovery of novel antimicrobials from slow-growing microbes from nutrient-limited environments.

George Luedemann is known throughout the antimicrobial community as one of the discoverers of the natural product antibiotic gentamicin. He subsequently hypothesized that slow-growing organisms inhabiting inhospitable, nutrient-limited environments may represent an enriched source of previously undescribed microbes that produce novel antimicrobials to create a competitive advantage over faster-growing rival organisms. Accordingly, 750 slow-growing microorganisms were isolated from desert rock surfaces and archived prior to Dr. Luedemann's passing in 2000. Here, we describe the characterization

PA0328 is associated with virulence and immune evasion in carbapenem-resistant Pseudomonas aeruginosa.

BACKGROUND: Pseudomonas aeruginosa is a major nosocomial pathogen. Its persistence in hospital environments and ability to evade host immune responses are closely linked to multiple virulence factors. OBJECTIVE: This study aimed to investigate the regulatory role of PA0328 in controlling key pathogenic traits of carbapenem-resistant Pseudomonas aeruginosa (CRPA), particularly its interactions with host immune cells. METHODS: An extensively drug-resistant CRPA isolate from a COPD patient (BALF sample) was colistin-resistant and biofilm-proficient. It served as the basis for transposon mutagen

Tracing the path of Quorum sensing molecules in cystic fibrosis mucus in a biomimetic in vitro permeability platform.

P. aeruginosa employs specific quorum sensing (QS) mechanisms to orchestrate biofilm formation, enhancing resistance to host defences. In physiological conditions, QS molecules permeate the lung environment and cellular membrane to reach the cytoplasmic Aryl Hydrocarbon Receptor (AhR) that is pivotal for activating the immune response against infection. In pathological conditions like cystic fibrosis (CF) this interkingdom communication is altered, favouring P. aeruginosa persistence and chronic infection. Here, we aim to investigate the molecular journey of QS molecules from CF-like environme

Reprint of: Intracellular Production of Superoxide Radical and of Hydrogen Peroxide by Redox Active Compounds.

Several compounds have been found capable of diverting the electron flow in Escherichia coli and thus causing increased intracellular production of O2- and H2O2. One indication of this electron-shunting action was increased cyanide-resistant respiration and one cellular response was increased biosynthesis of the manganese-containing superoxide dismutase and of catalase. Blocking cytochrome oxidase with cyanide or azide increased the electron flow available for reduction of paraquat and presumably of the other exogenous compounds tested and thus increased their biological effects. Paraquat, pyo

Arsenic-induced modulation of virulence and drug resistance in Pseudomonas aeruginosa.

Arsenic contamination of water sources, whether from natural or industrial origins, represents a significant risk to human health. However, its impact on waterborne pathogens remains understudied. This research explores the effects of arsenic exposure on the opportunistic pathogen Pseudomonas aeruginosa, a bacterium found in diverse environments. The arsenic exposure at concentrations of 0.12-20 mg/L As(III) resulted in rapid growth arrest of P. aeruginosa. Moreover, arsenic exposure significantly reduced the production of key virulence factors such as elastase (by 1.48- to 9.24-fold), pyocyan

Evaluating the Efficacy of Actinidia deliciosa (Kiwi Fruit) Extract in Inhibiting Pseudomonas aeruginosa Biofilm Formation: An In Vitro Study With Therapeutic Implications.

Background Gram-negative Pseudomonas aeruginosa is a common bacteria that is well-known for its capacity to build biofilms, which are organized cell communities encased in a self-produced polymeric matrix. Treating infections becomes more challenging due to biofilms' capacity to provide immunity and resistance to antibiotics. The search for novel anti-biofilm agents has gained significant momentum, and the diverse range of bioactive compounds found in natural products offers a promising avenue. Rich in vitamins, antioxidants, and various phytochemicals, Actinidia deliciosa (kiwi fruit) has dem

Myricetin as a potential therapeutic agent against Pseudomonas aeruginosa: inhibition of biofilm formation, quorum sensing, and virulence factor production.

Multidrug-resistant pathogenic bacteria, particularly Pseudomonas aeruginosa (P. aeruginosa), pose a significant threat to human health. Despite the huge persistence of antibiotics, there remains a lack of effective natural compounds capable of simultaneously disrupting quorum sensing (QS), biofilm formation, and virulence in this pathogen. This study aimed to investigate the inhibitory potential of myricetin against P. aeruginosa focusing on its ability to interfere with QS-regulated virulence traits. Antibacterial, antibiofilm, anti-QS, and virulence factor activities were evaluated using cr

Effects of quercetin, baicalein, azithromycin, and their combination on biofilm formation, virulence factors and gene expression associated with Pseudomonas aeruginosa quorum sensing.

BACKGROUND: Pseudomonas aeruginosa is a Gram-negative opportunistic pathogen commonly associated with acute and chronic hospital-acquired infections. Its ability to form biofilms, regulated in part by quorum sensing, contributes to its persistence and resistance. Classified as a critical priority pathogen by the World Health Organization, there is an urgent need for new therapeutic strategies. In this study, we evaluated the effects of quercetin, baicalein and azithromycin, alone and in combination, on biofilm formation, virulence factor production, and quorum sensing gene expression in P. aer

Synthesis and Biological Evaluation of New Quinoline and Anthranilic Acid Derivatives as Potential Quorum Sensing Inhibitors.

Inhibiting quorum sensing (QS), a central communication system, is a promising strategy to combat bacterial pathogens without antibiotics. Here, we designed novel hybrid compounds targeting the PQS (Pseudomonas quinolone signal)-dependent quorum sensing (QS) of Pseudomonas aeruginosa that is one of the multidrug-resistant and highly virulent pathogens with urgent need of new antibacterial strategies. We synthesized 12 compounds using standard procedures to combine halogen-substituted anthranilic acids with 4-(2-aminoethyl/4-aminobuthyl)amino-7-chloroquinoline, linked via 1,3,4-oxadiazole. Thei

Design, synthesis and evaluation of oxazolopyridinone derivatives as quorum sensing inhibitors.

The antibiotic crisis is associated with the appearance of multidrug resistant (MDR) pathogens, which has caused severe bacterial infections and imposed a huge burden on modern society. Therefore, there is an urgent need to develop new antibacterial drugs with novel mechanism of action. Here we designed and synthesized three series of benzoxazolone, oxazolopyridinone and 3-(2-hydroxyphenyl)hydantoin derivatives and evaluated their activity as novel quorum sensing (QS) inhibitors. We found that benzoxazolone and oxazolopyridinone derivatives had promising QS inhibitory activity in the minimum i

A novel stress response pathway mediates biofilm architecture in Pseudomonas aeruginosa.

Pseudomonas aeruginosa is a multidrug-resistant opportunistic pathogen, with chronic infections often associated with biofilm formation. Here, we investigate the previously uncharacterized gene PA3049, which is upregulated under biofilm conditions, to determine its role in infection, biofilm formation, and antimicrobial sensitivity. We show that the small uncharacterised protein PA3049, renamed as Biofilm architecture Regulator (BatR), promotes biofilm establishment and enhances biofilm survival in sub-inhibitory concentrations of antibiotics. Proteomic analysis revealed that BatR influences t

Increased oxidative stress in AOA3 cells disturbs ATM-dependent DNA damage responses.

Ataxia telangiectasia (AT) is caused by a mutation in the ataxia-telangiectasia-mutated (ATM) gene; the condition is associated with hyper-radiosensitivity, abnormal cell-cycle checkpoints, and genomic instability. AT patients also show cerebellar ataxia, possibly due to reactive oxygen species (ROS) sensitivity in neural cells. The ATM protein is a key regulator of the DNA damage response. Recently, several AT-like disorders have been reported. The genes responsible for them are predicted to encode proteins that interact with ATM in the DNA-damage response. Ataxia with oculomotor apraxia type
