Enhanced Biosynthesis of Fatty Acids Is Associated with the Acquisition of Ciprofloxacin Resistance in Edwardsiella tarda.

Misuse and overuse of antibiotics drive the selection and spread of antibiotic-resistant bacteria. Although genetic mutations have been well defined for different types of antibiotic resistance, ways to revert antibiotic resistance are largely unexplored. Here, we adopted a proteomics approach to investigate the mechanism underlying ciprofloxacin resistance in Edwardsiella tarda, a representative pathogen that infects both economic animal species and human beings. By comparing the protein expression profiles of ciprofloxacin-sensitive and -resistant E. tarda, a total of 233 proteins of differe

Functional Differences between Synaptic Mitochondria from the Striatum and the Cerebral Cortex.

Mitochondrial dysfunction has been shown to play a major role in neurodegenerative disorders such as Huntington's disease, Alzheimer's disease and Parkinson's disease. In these and other neurodegenerative disorders, disruption of synaptic connectivity and impaired neuronal signaling are among the early signs. When looking for potential causes of neurodegeneration, specific attention is drawn to the function of synaptic mitochondria, as the energy supply from mitochondria is crucial for normal synaptic function. Mitochondrial heterogeneity between synaptic and non-synaptic mitochondria has been

PBS3 and EPS1 Complete Salicylic Acid Biosynthesis from Isochorismate in Arabidopsis.

Salicylic acid (SA) is an important phytohormone mediating both local and systemic defense responses in plants. Despite over half a century of research, how plants biosynthesize SA remains unresolved. In Arabidopsis, a major part of SA is derived from isochorismate, a key intermediate produced by the isochorismate synthase, which is reminiscent of SA biosynthesis in bacteria. Whereas bacteria employ an isochorismate pyruvate lyase (IPL) that catalyzes the turnover of isochorismate to pyruvate and SA, plants do not contain an IPL ortholog and generate SA from isochorismate through an unknown me

Metabolic Regulation of Carotenoid-Enriched Golden Rice Line.

Vitamin A deficiency (VAD) is the leading cause of blindness among children and is associated with high risk of maternal mortality. In order to enhance the bioavailability of vitamin A, high carotenoid transgenic golden rice has been developed by manipulating enzymes, such as phytoene synthase (psy) and phytoene desaturase (crtI). In this study, proteome and metabolite analyses were carried out to comprehend metabolic regulation and adaptation of transgenic golden rice after the manipulation of endosperm specific carotenoid pathways. The main alteration was observed in carbohydrate metabolism

Metabolic adaptability in liver and gastrocnemius muscle of mice following subacute lead toxicity.

Lead (Pb) is one of several environmental pollutants that adversely affect human health by producing toxicity at the tissue level. The aim of the study was to understand the effects of Pb on the metabolic profiles of liver and gastrocnemius muscle of mice in relation to carbohydrate and fat metabolisms. Swiss albino mice were chosen and divided into two groups, control and Pb-treated. The Pb-treated animals were exposed to Pb at a dose of 5 mg/kg body weight for 30 days orally, which resulted in hypoglycemia, glycosuria, and increased glycogenolysis in the liver and gastrocnemius muscle of tre

Melatonin-regulated heat shock proteins and mitochondrial ATP synthase induce drought tolerance through sustaining ROS homeostasis in H2S-dependent manner.

Drought is thought to be one of the major global hazards to crop production. Understanding the role of melatonin (Mel) during plant adaptive responses to drought stress (DS) was the aim of the current investigation. Involvement of hydrogen sulfide (H2S) was also explored in Mel-regulated mechanisms of plants' tolerance to DS. A perusal of the data shows that exposure of tomato plants to DS elevated the activity of mitochondrial enzymes viz. pyruvate dehydrogenase, malate dehydrogenase, and citrate synthase. Whereas the activity of ATP synthase and ATPase was downregulated under stress conditio

Formaldehyde induces ferroptosis in hippocampal neuronal cells by upregulation of the Warburg effect.

The mechanisms underlying formaldehyde (FA)-induced neurotoxicity have not yet been fully clarified. Ferroptosis is a novel regulatory cell death and the Warburg effect is involved in regulating neural function. In this study, we investigated whether FA-induced neurotoxicity is implicated in neuronal ferroptosis and determined whether the Warburg effect mediates FA-induced neuronal ferroptosis. We found that FA (0.1, 0.5 and 1.0 mM, 6 h) induced cell death in HT22 cells (a cell line of mouse hippocampal neuron), as evidenced by a decrease in cell viability and an increase in cell mortality; en

Entamoeba histolytica: oxygen resistance and virulence.

Entamoeba histolytica virulence has been attributed to several amoebic molecules such as adhesins, amoebapores and cysteine proteinases, but supporting evidence is either partial or indirect. In this work we compared several in vitro and in vivo features of both virulent E. histolytica (vEh) and non-virulent E. histolytica (nvEh) axenic HM-1 IMSS strains, such as complement resistance, proteinase activity, haemolytic, phagocytic and cytotoxic capacities, survival in mice caecum, and susceptibility to O(2). The only difference observed was a higher in vitro susceptibility of nvEh to O(2). The m

Effect of prolonged undernutrition on rat diaphragm mitochondrial respiration.

Previous studies have shown that undernutrition induces an impairment of the respiratory muscle function in patients with chronic lung disease. To explain this, we hypothesized that undernutrition could decrease oxidative metabolism in the diaphragm. We therefore examined the effect of prolonged undernutrition on diaphragm mitochondrial oxygen uptake with pyruvate and palmitate as substrates in adult rats. Ten rats served as controls (CTL). Ten nutritionally deprived rats (ND) received 40% of their estimated daily nutrition. Five weeks of undernutrition induced a 33% decrease in state 3 respir

Qiliqiangxin Improves Cardiac Glucose Metabolism After Myocardial Infarction Through a HIF-1α/MIF/AMPK Axis.

Cardiac glucose metabolism is critically involved in the pathophysiology of myocardial infarction. This study investigated whether Qiliqiangxin (QL), a traditional Chinese medicine, improves myocardial glucose uptake and aerobic oxidation after myocardial infarction (MI) and explored the underlying mechanisms. QL treatment significantly increased LVEF, enhanced myocardial glucose uptake, reduced fibrosis, apoptosis, ROS production and elevated ATP generation along with activity of pyruvate dehydrogenase (PDH) and citrate synthase (CS). These effects were abolished by HIF-1α-shRNA transduction.

Molecular basis of photoinduced seasonal energy rheostasis in Japanese quail (Coturnix japonica).

Seasonal rhythms in photoperiod are a predictive cue used by many temperate-zone animals to time cycles of lipid accumulation. The neuroendocrine regulation of seasonal energy homeostasis and rheostasis are widely studied. However, the molecular pathways underlying tissue-specific adaptations remain poorly described. We conducted two experiments to examine long-term rheostatic changes in energy stability using the well-characterized photoperiodic response of the Japanese quail. In experiment 1, we exposed quails to photoperiodic transitions simulating the annual photic cycle and examined the m

Quantitative proteomics reveals energy metabolism remodeling in Bacillus cereus spores under pulsed light treatment.

Pulsed light (PL) is an emerging non-thermal sterilization technology. However, its molecular inactivation mechanisms against the highly resistant dormant spores of the foodborne pathogen Bacillus cereus remain poorly understood, representing a critical research gap. To address this, our specific contribution involved employing a data-independent acquisition (DIA)-based quantitative proteomic approach to elucidate the dynamic protein-level responses of B. cereus spores to PL treatment. A total of 489 proteins exhibiting differential abundance (270 demonstrating increased abundance, 219 demonst

Multi-omics dissection of CO2-induced metabolic reprogramming in broccoli: key regulators of growth and nutritional quality.

The rising global CO2 level influences plant growth, productivity, and nutritional quality, and elevated CO2 (ECO2) is commonly utilized in greenhouses to enhance vegetable crop yields. However, the molecular mechanism underlying plant responses to ECO2 remains unclear. This study examined the impacts of ECO2 on the nutritional quality and growth characteristics of two widely cultivated broccoli varieties, 'Yanxiu' and 'Qiuli'. A pot experiment was carried out in environmentally auto-controlled growth chambers over a period of 100 days, under both ambient CO2 (ACO2, 420/470 ppm, daytime/nightt

Ataxin-1 regulates the cerebellar bioenergetics proteome through the GSK3β-mTOR pathway which is altered in Spinocerebellar ataxia type 1 (SCA1).

A polyglutamine expansion within the ataxin-1 protein (ATXN1) underlies spinocerebellar ataxia type-1 (SCA1), a neurological disorder mainly characterized by ataxia and cerebellar deficits. In SCA1, both loss and gain of ATXN1 biological functions contribute to cerebellar pathogenesis. However, the critical ATXN1 functions and pathways involved remain unclear. To further investigate the early signalling pathways regulated by ATXN1, we performed an unbiased proteomic study of the Atxn1-KO 5-week-old mice cerebellum. Here, we show that lack of ATXN1 expression induces early alterations in protei
