Safety and efficacy of CyTisine for smoking cessation in a hOSPital context (CITOSP): study protocol for a prospective observational study.

Tobacco addiction is the primary preventable factor contributing to global mortality, and nicotine is one of the substances with the greatest potential for addiction. With a strong affinity for the α4β2 subtype receptor, cytisine (CYT) functions as a partial agonist of the acetylcholine nicotinic cholinergic receptor. It counteracts the effects of nicotine without causing any withdrawal symptoms. These features, combined with its limited mild adverse effects and minimal drug-drug interactions, make cytisine a cost-effective treatment for smoking cessation. The current protocol describes a pros

Non-competitive Inhibition of Nicotinic Acetylcholine Receptors by Ladybird Beetle Alkaloids.

Ladybird beetles (Family Coccinellidae) secrete an alkaloid rich venom from their leg joints that protects them from predators. Coccinellines, the major venom constituents, are alkaloids composed of three fused piperidine rings that share a common nitrogen atom. Although many coccinellines have been isolated and chemically characterized, their pharmacological properties are essentially unknown. Using radioligand binding and functional assays we investigated the actions of several coccinellines on skeletal muscle and α7 nicotinic acetylcholine receptors (nAChRs). The alkaloids were shown to dis

Aloperine treatment attenuates acute spinal cord injury by reducing oxidative, inflammatory, and apoptotic responses via PI3K/AKT/NF-κB signaling in a rat contusion model.

Spinal cord injury (SCI) is a severe condition that can result in nerve damage, impaired motor or sensory function, and ultimately a high mortality rate for injured individuals. High oxidative and inflammatory responses are closely linked to poor prognosis and can influence the recovery of neurological functions. Therefore, overcoming these processes early is a valuable therapy approach for SCI. Aloperine (ALO) is a quinolizidine-type alkaloid with numerous pharmacological activities, including antioxidant, anti-inflammatory, and neuroprotective effects. However, the role of ALO in SCI recover

Matrine alleviates early brain injury after experimental subarachnoid hemorrhage in rats: possible involvement of PI3K/Akt-mediated NF-κB inhibition and Keap1/Nrf2-dependent HO-1 inductionn.

Matrine is a quinolizidine alkaloid derived from the herb Radix Sophorae Flavescentis, and possesses anti-oxidant, anti-inflammatory and anti-tumoral properties. However, its effectiveness against subarachnoid hemorrhage (SAH) is not well known. In this study, we investigated the effects of matrine on early brain injury (EBI) and the related potential mechanisms following SAH in rats. Our results showed that matrine pretreatment partially alleviated SAH-induced EBI, including neurological deficit, severity of SAH grade, brain edema, and blood-brain barrier (BBB) disruption in rats. In addition

[Treatment for smoking cessation : situation in 2025].

In Switzerland, smoking has slightly declined in recent years, while the use of new nicotine products such as electronic cigarettes and snus is increasing, especially among young people. All these products cause addiction, with some being potentially less harmful than conventional cigarettes, which remain the most widely used product. Nicotine replacement therapy and bupropion are currently the only approved medications for smoking cessation in Switzerland. Since varenicline was withdrawn, cytisine-an older drug with a similar mechanism of action-has seen renewed interest. Recent studies confi

Measuring functional brain recovery in regenerating planarians by assessing the behavioral response to the cholinergic compound cytisine.

Planarians are traditional model invertebrates in regeneration and developmental biology research that also display a variety of quantifiable behaviors useful to screen for pharmacologically active compounds. One such behavior is the expression of seizure-like movements (pSLMs) induced by a variety of substances. Previous work from our laboratory showed that cocaine, but not nicotine, induced pSLMs in intact but not decapitated planarians. Interestingly, as decapitated planarians regenerated their heads, they gradually recovered their sensitivity to cocaine. These results suggested a method to

Role of training dose in discrimination of nicotine and related compounds by rats.

Rats were trained to discriminate nicotine from saline in a two-bar operant conditioning procedure with food reinforcement. There was partial generalization to the nicotine analogues anabasine and cytisine in rats trained to discriminate either 0.2 or 0.4 mg/kg nicotine from saline. However, generalization was complete in rats trained to discriminate 0.1 mg/kg nicotine and, in a novel procedure, any one of three doses of nicotine (0.1, 0.2, or 0.4 mg/kg). There was no generalization to the muscarinic-cholinergic agonist oxotremorine (0.0025-0.04 mg/kg). Additional experiments were carried to f

Aloperine Attenuates Hepatic Ischemia/Reperfusion-Induced Liver Injury via STAT-3 Signaling in a Murine Model.

Hepatic ischemia/reperfusion (I/R) damage is one of the most common side effects of liver surgery. This pathophysiological process may lead to excessive hepatic damage. Aloperine is an active ingredient isolated from Sophora alopecuroides Linn and has a variety of therapeutic effects, including organ protection. However, the hepatoprotective effect of aloperine against hepatic I/R damage has not yet been determined. C57BL/6 mice were allocated to the sham-operated (sham), hepatic ischemia/reperfusion (I/R), and aloperine groups. The mice were exposed to 30 min of hepatic hilum occlusion. Then

An evaluation of cytisinicline for smoking cessation in adult smokers.

INTRODUCTION: Cigarette smoking is the world's leading preventable cause of death. With no new smoking cessation pharmacotherapy approved since 2006, the 29 million US adults who smoke need new treatment options. Cytisinicline, a partial agonist at α4β2 neuronal nicotinic acetylcholine receptors, is the name of a new dosing regimen of cytisine, a smoking cessation pharmacotherapy that has been sold for decades in Eastern Europe but is currently unavailable in the US. The Food and Drug Administration is evaluating a New Drug Application for cytisinicline. AREAS COVERED: We summarize the histor

Study on the Analgesic Mechanism of Aloperine in Mice with Neuropathic Pain Based on the p38MAPK/FKN/CX3CR1 Pathway.

A lesion or disease of the somatosensory nervous system causes neuropathic pain (NP). Previous experiments have demonstrated that aloperine (ALO) exhibits antiallodynic effects on neuropathic pain in mice; however, the mechanism underlying its analgesic action remains unclear. This study investigated the analgesic mechanism of ALO on neuropathic pain in mice using a chronic constriction injury (CCI) model. Experiments were conducted using the ALO and p38MAPK inhibitor SB 203580. In vivo experiments demonstrated that ALO played an analgesic role by inhibiting p38MAPK phosphorylation, and it imp

Beneficial effects of aloperine on inflammation and oxidative stress by suppressing necroptosis in lipopolysaccharide-induced acute lung injury mouse model.

RATIONALE: Alveolar epithelial cell death, inflammation, and oxidative stress are typical features of acute lung injury (ALI). Aloperine (Alo), an alkaloid isolated from Sophora alopecuroides, has been reported to display various biological effects, such as anti-inflammatory, immunoregulatory, and anti-oxidant properties. In this study, we investigated the effects and mechanisms of Alo in treating a lipopolysaccharide (LPS)-induced ALI in a murine model. METHODS: The effects of Alo in LPS-induced ALI were investigated in C57BL/6 mice. The RIPK1 inhibitor (Nec-1) and the RIPK3 inhibitor (GSK'8

Topical application of aloperine improves 2,4-dinitrofluorobenzene-induced atopic dermatitis-like skin lesions in NC/Nga mice.

Aloperine has been shown to inhibit 2,4-dinitrofluorobenzene (DNFB) induced allergic contact dermatitis in BALB/c mice. In the present study, we further investigated the effect of aloperine on DNFB-induced atopic dermatitis-like skin lesions in NC/Nga mice. NC/Nga mice elicited atopic dermatitis-like skin lesions after the topical application of DNFB. Aloperine treatment significantly inhibited dermatitis index and ear thickness in DNFB-treated NC/Nga mice in a dose-dependent manner. Eosinophils, mast cells infiltration into the ears and plasma level of immunoglobulin (Ig) E were also suppress

The anticonvulsant effect of sparteine on pentylenetetrazole-induced seizures in rats: a behavioral, electroencephalographic, morphological and molecular study.

Abnormal synchronous activity in neurons generates epileptic seizures. Antiepileptic drugs (AEDs) are effective in 70% of patients, but this percentage is drastically lower in developing countries. Sparteine is a quinolizidine alkaloid synthesized from most Lupine species and has a probable anticonvulsive effect. For this reason, the objective of the present work was to study the anticonvulsant effect of sparteine using a dose-effect curve and to determine its effectiveness against seizures using behavioral, electroencephalographic, morphological and molecular data. Wistar rats were grouped in

Genetically Encoded Constitutive Upregulation of β2 Subunit-Containing Neuronal Nicotinic Acetylcholine Receptors Is Neuroprotective in Female Parkinsonian Mice.

Parkinson's disease is projected to rise to pandemic proportions by 2050, which has resulted in an urgent need for disease-modifying treatments. We previously showed that in a mouse model of parkinsonism with unilateral 6-hydroxydopamine (6-OHDA) injection into the dorsolateral striatum, low doses of the neuronal nicotinic acetylcholine receptor (nAChR) partial agonist and smoking cessation drug, cytisine, exert neuroprotection in substantia nigra pars compacta (SNc) dopaminergic (DA) neurons of only female mice by reducing endoplasmic reticulum (ER) stress and that cytisine-mediated neuroprot
