SNHG12/NFYC-AS1 Acted as the Sponge for hsa-miR-199a-5p to Promote the Expression of S100A8/S100A7/XDH and was Involved in the Progression of Diabetic Foot Ulcers.

Traditional Chinese medicine has been used to treat diabetic foot ulcer (DFU) for a long time. However, the underlying mechanism of Radix arnebiae seu lithospermi ointment (RAS-ointment) has not been revealed. Effects of RAS-ointment treatment were observed in DFU patients. The endogenous competitive RNA mechanism was constructed based on micro-array sequencing and bioinformatics analysis. RT-PCR was used to detected the expression of genes in DFU ulcerated skins and non-ulcerated skins. Dual luciferase and RT-PCR experiments were used to investigate the endogenous competitive RNA mechanism. B

Mechanistic Landscape and Therapeutic Opportunities of Non-Coding RNAs in Modulating Fibroblast-Like Synoviocytes in Rheumatoid Arthritis.

Current disease-modifying antirheumatic drugs (DMARDs) for rheumatoid arthritis (RA) predominantly target systemic immune-inflammatory pathways, yet fail to effectively reverse progressive articular structural damage driven by fibroblast-like synoviocytes (FLS), representing a core unmet clinical need in the field. Non-coding RNAs (ncRNAs) are key epigenetic regulators of pathological FLS activation; however, the field is constrained by three core limitations: fragmented evidence, mechanistic homogenization, and a persistent basic-to-clinical translational gap. Adopting a structured evidence-g

Circular RNAs in tumor immunity and immunotherapy of breast cancer.

As the stable and conserved circular non-coding RNAs, circular RNAs (circRNAs) have assumed growing significance in regulating breast cancer immunity and immunotherapy. This review focuses on elucidating the molecular mechanisms through which circRNAs influence the efficacy of breast cancer immunotherapy. Specifically, circRNAs modulate the levels and stability of critical immune checkpoints, including PD-L1 and various cytokines. They achieve this by acting as competing endogenous RNAs (ceRNAs) to sequester miRNAs, functioning as protein scaffolds, and thereby modulating gene expression and t

Small nucleolar RNA host genes in hepatocellular carcinoma: an evidence-weighted and etiology-aware framework for mechanistic and translational interpretation.

Small nucleolar RNA host genes (SNHGs) are a distinctive subgroup of long non-coding RNAs whose loci can generate both host lncRNA transcripts and intronic small nucleolar RNAs. In hepatocellular carcinoma (HCC), dysregulated SNHGs have been linked to tumor growth, epithelial-mesenchymal transition, metastasis, stemness, immune remodeling, extracellular vesicle communication, and therapeutic resistance. However, the clinical meaning of these associations remains uneven because many reported mechanisms are based on limited cell-line experiments, retrospective cohorts, or non-stratified HCC mode

Regulatory roles of microRNAs in epigenetic and immune gene networks in psoriasis.

Psoriasis is a chronic, immune-mediated skin disorder characterized by keratinocyte hyperproliferation, impaired differentiation, and sustained inflammation driven by complex interactions between the immune system and the epidermis. Emerging evidence indicates that microRNAs (miRNAs) are central post-transcriptional regulators orchestrating these processes, linking environmental cues, genetic predisposition, and immune signaling. Dysregulated miRNAs in keratinocytes, immune cells, and the circulation contribute to disease onset and progression. Specific miRNAs, such as miR-21, miR-31, miR-155,

Competing Endogenous RNAs Crosstalk in Hippocampus: A Potential Mechanism for Neuronal Developing Defects in Down Syndrome.

Down syndrome (DS) is the most example of aneuploidy, resulting from an additional copy of all or part of chromosome 21. Competing endogenous RNAs (ceRNAs) play important roles in neuronal development and neurological defects. This study aimed to identify hub genes and synergistic crosstalk among ceRNAs in the DS fetal hippocampus as potential targets for the treatment of DS-related neurodegenerative diseases. We profiled differentially expressed long non-coding RNAs (DElncRNAs), differentially expressed circular RNAs (DEcircRNAs), differentially expressed microRNAs (DEmiRNAs), and differentia

Small extracellular vesicle-encapsulated circSATB1 in seminal plasma promotes ovine embryo implantation through the miR-200c/GATA2 pathway.

Impaired embryo implantation is a major cause of early gestational failure and represents a significant reproductive challenge. Seminal plasma-derived small extracellular vesicles (SP-sEVs) have been demonstrated as critical regulator in embryo implantation. Circular RNAs (circRNAs) constitute key SP-sEVs cargo that modulate intercellular communication via competitive endogenous RNA (ceRNA) network, providing novel and comprehensive insights into implantation defects. However, the functions and molecular mechanisms of SP-sEVs encapsulated circRNAs in this process remain to be elucidated. This

Circular RNA-centered regulatory networks in breast cancer: From ceRNA crosstalk to immune microenvironment reprogramming.

Circular RNAs (circRNAs) have emerged as a pervasive and functionally diverse class of regulatory molecules in breast cancer. While early studies predominantly described circRNAs as competing endogenous RNAs that modulate gene expression through microRNA sequestration, accumulating evidence indicates that this paradigm captures only a limited aspect of circRNA biology. Recent advances reveal that circRNAs function as central organizers of multilayered regulatory networks, integrating post-transcriptional control, RNA-protein interactions, noncanonical translation, metabolic adaptation, and imm

Identification of a circRNA-mediated immune-related ceRNA network and circRNAs as diagnostic biomarkers in acute ischemic stroke.

BACKGROUND: Research has demonstrated that circular RNAs (circRNAs) play important roles in acute ischemic stroke (AIS). However, the functions of circRNA-mediated competitive endogenous RNA (ceRNA) in AIS-related immunological inflammation are not well understood. In our study, we aimed to construct a circRNA-mediated immune-related ceRNA network and identify diagnostic circRNAs for AIS. METHODS: R software was used to analyze the microarray data obtained from the GEO database. The bioinformatics database was then used to develop the circRNA-mediated ceRNA network. Functional dynamics of lncRNAs and miRNAs in cancer metabolism and neuroinflammation.

Non-coding RNAs (ncRNAs) influence gene expression and diverse physiological and pathological processes. Long non-coding RNAs (lncRNAs) and microRNAs (miRNAs) play roles in cancer and neurodegenerative diseases. LncRNAs, such as HOTAIR, MALAT1, and NEAT1, regulate chromatin remodeling through histone modification. Dysregulated expression leads to oncogenesis and neuronal dysfunction. Conversely, miRNAs such as miR-21 and miR-155, as post-transcriptional regulators, act in oncogenic signaling and neuroinflammation. MiR-21 targets PTEN and PDCD4, leading to aberrant activation of the PI3K/AKT an

Cross-organ communication based on non-coding RNA networks leads to cognitive dysfunction in mice.

BACKGROUND: Beyond regulation of mRNAs by individual ncRNA types, like miRNAs, circRNAs or lncRNAs, their interplay as competitive endogenous RNAs (ceRNAs) appears pivotal in diseases. ceRNAs are promising biomarkers in diagnosing diseases as well as in understanding off-target drug mechanisms or developing specific therapeutics. Many diseases are influenced at different molecular levels and across multiple organs, making cross-organ multi-omics interaction studies necessary to holistically understand diseases and their regulation. In our study we applied this approach to the cognitive dysfunc

Maternally Expressed Gene 3 (MEG3) LncRNA: The silent guardian against solid and hematological malignancies.

Maternally Expressed Gene 3 (MEG3), a long non-coding RNA (lncRNA) that plays a crucial role in regulating cellular processes, including apoptosis, proliferation, and tumor suppression. MEG3 is located at the DLK1-MEG3 locus on human chromosome 14q32.3 and is implicated in multiple cancers and pathological conditions. MEG3 expression is often downregulated in various solid and liquid malignancies due to promoter hypermethylation, leading to tumor progression and poor prognosis. Yet, it showed paradoxical expression in other malignancies. MEG3 interacts with multiple molecular pathways, includi

Implications of LINC01094 for human malignancies.

Long non-coding RNAs (lncRNAs) constitute a large class of ribonucleic acids, participating in multiple biological events within tumor cells, especially the regulation of transcription. A growing body of literature has revealed that abnormalities of lncRNA expression could result in carcinogenesis and oncogenesis by exerting inhibition or oncogenic effects. LINC01094 is a recently identified lncRNA found to be dys-regulated in an assortment of cancer tissues and control multiple biological processes via competing endogenous RNA (ceRNA) mechanisms. Specifically, LINC01094 functions as a ceRNA t

LncRNA Gm16023 limits M1 macrophage-mediated hyperglycemia-exacerbated septic intestinal injury via miR-377-3p/Sirt1 axis.

Diabetes is closely associated with inflammation and sepsis, but its clinical significance and underlying mechanisms remain obscure. Leveraging epidemiological data from 404,184 individuals in the UK Biobank cohort, we found that hyperglycemia was significantly associated with increased sepsis risk. Phenotypically, hyperglycemia enhanced macrophage infiltration and exacerbated intestinal inflammation in both septic patients and murine models. Using both a genetic CD11b-DTR model and clodronate liposome-mediated chemical ablation, we demonstrated that macrophage depletion markedly attenuated hy
