Resistance against Ralstonia solanacearum in tomato depends on the methionine cycle and the γ-aminobutyric acid metabolic pathway.

Bacterial wilt caused by Ralstonia solanacearum is a complex and destructive disease that affects over 200 plant species. To investigate the interaction of R. solanacearum and its tomato (Solanum lycopersicum) plant host, a comparative proteomic analysis was conducted in tomato stems inoculated with highly and mildly aggressive R. solanacearum isolates (RsH and RsM, respectively). The results indicated a significant alteration of the methionine cycle (MTC) and downregulation of γ-aminobutyric acid (GABA) biosynthesis. Furthermore, transcriptome profiling of two key tissues (stem and root) at t

Isolation and characterization of Pseudomonas brassicacearum J12 as an antagonist against Ralstonia solanacearum and identification of its antimicrobial components.

A bacterial strain, J12, isolated from the rhizosphere soil of tomato plants strongly inhibited the growth of phytopathogenic bacteria Ralstonia solanacearum. Strain J12 was identified as Pseudomonas brassicacearum based on its 16S rRNA gene sequence. J12 could produce 2,4-diacetylphloroglucinol (2,4-DAPG), hydrogen cyanide (HCN), siderophore(s) and protease. The maximum growth and antagonistic activity were recorded at 30°C and pH 8. Glucose and tryptone were used as the most suitable carbon and nitrogen sources, respectively. Strain J12 significantly suppressed tomato bacteria wilt by 45.5%

Differential occurrence of oxidative burst and antioxidative mechanism in compatible and incompatible interactions of Solanum lycopersicum and Ralstonia solanacearum.

Striking increase in reactive oxygen species (ROS) such as hydrogen peroxide (H(2)O(2)) has been demonstrated to occur in plants in response to pathogen attack. The aim of this study was to investigate the biochemical aspects of ROS generation, antioxidative mechanism and cell wall reinforcement as responses of tomato cultivars Arka Meghali (AM; susceptible) and BT-10 (BT; resistant) against Ralstonia solanacearum (Ralsol). While the oxidative burst was characterized by a single phase ROS increase in AM, there was a clear bi-phasic ROS generation in BT. The first significant increase of H(2)O(

Antagonistic activity and biocontrol potential of Pseudomonas sp. T19G isolated from the tomato rhizosphere against Ralstonia solanacearum.

Tomato bacterial wilt, caused by the soilborne bacterium Ralstonia solanacearum, remains difficult to manage, creating a need for biologically derived control agents. Here, we isolated Pseudomonas sp. T19G from the rhizosphere soil of healthy tomato plants and evaluated its antagonistic activity, protective effects, genomic basis, and influence on rhizosphere function. T19G fermentation broth inhibited R. solanacearum, and sterile cell-free supernatant caused a dose-dependent decline in viable pathogen populations. T19G significantly reduced viable root-associated pathogen populations and path

Assembly processes and network structure of rhizosphere fungal communities in healthy and Ralstonia solanacearum-infected tomato plants.

INTRODUCTION: Bacterial wilt caused by Ralstonia solanacearum is a major constraint to tomato production worldwide. Rhizosphere microbial community is closely linked to plant health. However, how R. solanacearum invasion alters the assembly processes and co-occurrence network in tomato rhizosphere fungal communities remains largely unclear. METHODS: Here, we collected rhizosphere soil samples from three major tomato-producing regions in China to investigate how R. solanacearum invasion affects fungal community composition, network topology, and assembly processes in tomato rhizosphere. Auxin-responsive SlXTH3 promotes Ralstonia solanacearum infection by modulating lateral root development and root immunity in tomato.

SlXTH3 promotes tomato lateral root development and cell wall remodeling, which facilitate early root colonization by Ralstonia solanacearum and increase susceptibility to bacterial wilt. Ralstonia solanacearum is a highly destructive soil-borne bacterial pathogen whose infection typically initiates in the root system and is accompanied by remodeling of host root development and defense responses. The results of this study showed that, during the early infection of tomato roots, R. solanacearum preferentially colonized lateral root emergence sites and promoted lateral root development, thereby

Silica nanoparticles activate defense responses by reducing reactive oxygen species under Ralstonia solanacearum infection in tomato plants.

Silica nanoparticles (SNPs) play an important positive role in enhancing stress resistance of plants. However, their absorption and the mechanisms of resistance in plants are not yet fully understood. In this study, we investigated the uptake of SNPs in tomato plants and explored the physiological and molecular mechanisms of SNPs-mediated bacterial wilt resistance. Folia application of SNPs significantly increased silicon content in tomato leaves and roots by 5.4-fold and 1.8-fold compared with healthy control, respectively. Moreover, foliar-applied SNPs mainly accumulated in the shoots of pla

Overexpression of a novel peanut NBS-LRR gene AhRRS5 enhances disease resistance to Ralstonia solanacearum in tobacco.

Bacterial wilt caused by Ralstonia solanacearum is a ruinous soilborne disease affecting more than 450 plant species. Efficient control methods for this disease remain unavailable to date. This study characterized a novel nucleotide-binding site-leucine-rich repeat resistance gene AhRRS5 from peanut, which was up-regulated in both resistant and susceptible peanut cultivars in response to R. solanacearum. The product of AhRRS5 was localized in the nucleus. Furthermore, treatment with phytohormones such as salicylic acid (SA), abscisic acid (ABA), methyl jasmonate (MeJA) and ethephon (ET) increa

In vivo application of potent probiotics for enhancing potato growth and controlling Ralstonia solanacearum and Fusarium oxysporum infections.

Plant probiotics are live microbial cells or cultures that support plant growth and control plant pathogens through different mechanisms. They have various effects on plants, including plant growth promotion through the production of indole acetic acid (IAA), biological control activity (BCA), and production of cellulase enzymes, thus inducing systemic resistance and increasing the availability of mineral elements. The present work aimed to study the potential of Achromobacter marplatensis and Bacillus velezensis as plant probiotics for the field cultivation of potatoes. In vitro studies have

Disease-stage-associated transcriptional responses underlying bacterial wilt resistance in tobacco cultivar Yunyan 87 caused by Ralstonia solanacearum.

Bacterial wilt caused by Ralstonia solanacearum severely limits tobacco production, yet the molecular basis of resistance to vascular wilt diseases remains poorly understood. We integrated field phenotyping, pathogen isolation, and transcriptomic analyses across different disease stages to investigate root responses of the resistant tobacco cultivar Yunyan 87 at healthy, moderately infected, and severely infected stages. Disease progression was accompanied by marked root biomass loss, with fresh and dry weights reduced by approximately 60% and 70%, respectively. Transcriptomic analyses identif

Spatiotemporal reprogramming of auxin signaling and resistance outputs across model plants and solanaceae crops under Ralstonia solanacearum infection.

Bacterial wilt is a devastating vascular disease caused by the soil-borne bacterium Ralstonia solanacearum, which invades and blocks the xylem of susceptible plants, especially economically important solanaceous crops. In addition to classical plant defense hormones, recent research has focused on understanding the role of auxin as a key and continuous regulator of infection and development. Auxin connects changes during infection to root development and the progression of bacterial wilt disease. The auxin related shift in plant hormone patterns associated with root development has been observ

Trichloroethylene degradation by Ralstonia sp. KN1-10A constitutively expressing phenol hydroxylase: transformation products, NADH limitation, and product toxicity.

Ralstonia sp. KN1-10A, which was constructed by inserting the tac promoter upstream of the phenol hydroxylase (PH) gene in the chromosomal DNA of the wild-type strain, Ralstonia sp. KN1, is a useful recombinant strain for eliminating trichloroethylene (TCE) from contaminated sites because it exhibits constitutive TCE oxidation activity. During TCE degradation by Ralstonia sp. KN1-10A, noxious chlorinated compounds, such as dichloroacetic acid, trichloroacetic acid, 2,2,2-trichloroethanol, and chloral, were not detected, and more than 95% of chlorine in TCE was released as chloride ions. Metabolic rewiring and transcriptional interference underlie the biocontrol efficacy of Burkholderia arboris BB417 against Ralstonia solanacearum species complex.

Ralstonia solanacearum species complex (RSSC) is a globally destructive plant pathogen with high ecological persistence, making bacterial wilt management a long-standing challenge. In this study, we identified Burkholderia arboris BB417 as a potent biocontrol agent and systematically elucidated its multilevel antagonistic mechanisms against RSSC. BB417 exhibited broad-spectrum inhibition against diverse RSSC strains and achieved greenhouse control efficacy comparable to chemical pesticides within the 10-day observation period, while also promoting plant growth through nitrogen fixation, phosph

A YopJ-family effector from Ralstonia solanacearum is associated with auxin-responsive transcriptional reprogramming and bacterial wilt suppression.

Effector-triggered immunity (ETI) can reshape hormone-associated transcriptional programs during plant-pathogen interactions. However, how bacterial effectors are associated with hormone-responsive defense outputs in potato remains poorly understood. This study identifies RipBR, a previously uncharacterized YopJ-family effector from Ralstonia solanacearum, as an avirulence-associated determinant in the wild potato accession Solanum albicans ALB28-1. Loss- and gain-of-function analyses demonstrated that RipBR contributes to reduced bacterial wilt symptoms and restricts bacterial colonization in

A novel plant immune inducer esculetin activates systemic acquired resistance against Ralstonia solanacearum via the SA signaling pathway.

BACKGROUND: Ralstonia solanacearum poses a severe threat to global agriculture due to its broad host range, high dispersal capacity, and limited chemical control options. Plant immune inducers provide an effective strategy for controlling bacterial wilt disease. RESULTS: This study demonstrated a natural-derived compound esculetin (ES) serves as a novel plant immune inducer against tobacco bacterial wilt. Foliar application of ES exhibits considerable control effect on tobacco bacterial wilt, with control efficacy is high as 45.41%-68.69%, significantly higher than positive control treatment-

CaWRKY58, encoding a group I WRKY transcription factor of Capsicum annuum, negatively regulates resistance to Ralstonia solanacearum infection.

WRKY transcription factors are encoded by large gene families across the plant kingdom. So far, their biological and molecular functions in nonmodel plants, including pepper (Capsicum annuum) and other Solanaceae, remain poorly understood. Here, we report on the functional characterization of a new group I WRKY protein from pepper, termed CaWRKY58. Our data indicate that CaWRKY58 can be localized to the nucleus and can activate the transcription of the reporter β-glucuronidase (GUS) gene driven by the 35S core promoter with two copies of the W-box in its proximal upstream region. In pepper pla

Optimization and characterization of a low-toxicity trans-anethole nanoemulsion for controlling the soil-borne bacterial pathogen Ralstonia solanacearum.

Plant-derived trans-anethole (TAN) exhibits favorable antibacterial activity and biocompatibility, but poor solubility, photolysis, and volatility limit its use. Here, we prepared a TAN nanoemulsion (TAN-NE) via low-energy emulsification using non-ionic surfactants (polyethylene glycol tert-octylphenyl ether and polyoxyethylenated castor oil) and a co-surfactant (anhydrous ethanol). TAN-NE had spherical morphology, particle size of 20.68 nm, formulation yield (92.16%), encapsulation efficiency (88.49%), and loading efficiency (3.60%). It showed improved water solubility, photostability, and st

Integrated transcriptomic and metabolomic analysis revealed the response of resistant variety Zhonghua 6 to the infection of Ralstonia solanacearum in peanut.

Peanut (Arachis hypogaea L.) is an important oil and economic crop, and its production has long been severely threatened by soil-borne bacterial wilt (BW) disease. However, the molecular mechanism of host resistance to it has not yet been systematically elucidated. In this study, the highly resistant peanut variety Zhonghua 6 was used as the research object. Through transcriptomic analysis, a total of 1,122 differentially expressed genes (DEGs) were identified between carefully designed treatment and control groups. WGCNA analysis led to the discovery of 14 hub genes, including two cytochrome

Carbamoylphosphate serves as the source of CN(-), but not of the intrinsic CO in the active site of the regulatory [NiFe]-hydrogenase from Ralstonia eutropha.

Within the catalytic centre of [NiFe]-hydrogenases one carbonyl and two cyanide ligands are covalently attached to the iron. To identify the metabolic origins of these ligands, the regulatory [NiFe] hydrogenase in conjunction with the indigenous Hyp maturation proteins of Ralstonia eutropha H16 were heterologously overproduced in E. coli grown in the presence of L-[ureido-(13)C] citrulline and NaH(13)CO(3). Infrared spectroscopy of purified hydrogenase provided direct evidence that only the cyanide ligands, but not the CO ligand, originate from CO(2) and carbamoylphosphate. Incorporation of la
