Singapore grouper iridovirus (SGIV) always causes high transmission efficiency and mortality in the larval and juvenile stages of grouper in aquaculture industry. Although inactivated virus and recombinant DNA vaccines administered via intraperitoneal injection have shown efficacy in protection against SGIV, their potential applications in field testing were limited due to the vaccine delivery methods. Here, we developed an immersion vaccine containing inactivated virus and Montanide IMS 1312 adjuvant (IMS 1312) and evaluated its protective efficacy against SGIV infection. Compared to the PBS

Tiger frog virus (TFV), in the genus Ranavirus of the family Iridoviridae, causes high mortality of cultured tiger frog tadpoles in China. To explore the cellular entry mechanism of TFV, HepG2 cells were treated with drugs that inhibit the main endocytic pathways. We observed that TFV entry was inhibited by NH(4)Cl, chloroquine, and bafilomycin, which can all elevate the pH of acidic organelles. In contrast, TFV entry was not influenced by chlorpromazine or overexpression of a dominant-negative form of Esp15, which inhibit the assembly of clathrin-coated pits. These results suggested that TFV

In recent years, the viral disease caused by largemouth bass virus (LMBV) has inflicted severe economic losses on China's largemouth bass (Micropterus salmoides) aquaculture, significantly restricting its development. As targeted antiviral drugs and commercial vaccines remain in the investigational stage, the discovery of novel antiviral agents to control LMBV infections is urgently needed. Consequently, this study systematically evaluated the antiviral efficacy and underlying immunopharmacological mechanisms of cepharanthine (CE), a natural bisbenzylisoquinoline alkaloid, against LMBV infecti

Amphibian populations are undergoing dramatic global declines, with infectious diseases recognized as major contributors. While chytridiomycosis, caused by Batrachochytrium dendrobatidis (Bd), and disease caused by ranaviruses are well known, herpesviruses in amphibians remain comparatively neglected. We conducted a passive survey using carcasses collected between 2022 and 2025 in Schleswig-Holstein, northern Germany. Dead amphibians (n = 187) were dissected. Skin, kidney, liver, and brain samples were screened for Bd, ranaviruses, bufonid herpesvirus 1 (BfHV1), and ranid herpesvirus 3 (RaHV3)

Increasing disease challenges represent a critical bottleneck for the sustainable development of largemouth bass farming. Vaccination can effectively protect fish from pathogens, with dominant immunogenic epitope identification being essential for vaccine development. This research documents a case of largemouth bass virus (LMBV) infection and identifies the dominant immunogenic epitope of the major capsid protein of LMBV (LMBV-MCP). Samples showing clinical signs, such as skin lesions and hepatic bleeding, were collected from naturally infected largemouth bass. Histopathological analysis show

Largemouth bass virus (LMBV) is an infectious pathogen that causes high mortality rates in largemouth bass, and outbreaks of this virus can significantly harm the aquaculture industry. Currently, no vaccine has been developed that can effectively prevent the transmission of LMBV. In this study, we constructed a recombinant Lactobacillus plantarum (L. plantarum) strain capable of expressing the MCP gene of LMBV and displaying this protein on its surface; then, we evaluated the immunoprotective effect of this recombinant bacterium on largemouth bass. Western blotting, immunofluorescence, and flo

Singapore grouper iridovirus (SGIV) is a highly pathogenic double-stranded DNA virus, and the fatality rate of SGIV-infected grouper is more than 90%. Up to now, there is no effective methods to control the disease. Long non-coding RNAs (lncRNAs) might play an important role in individual growth and development, immune regulation and other life processes. In this study, lncRNAs were identified in Epinephelus coioides, an important economic aquaculture marine fish in China and Southeast Asia, and the regulatory relationships of lncRNAs and mRNA response to SGIV infection were analyzed. BACKGROUND: Behavioural plasticity may contribute to the ability of wild animals to survive disease outbreaks. In absence of endogen heat control, ectothermic animals adjust their body temperature behaviourally. While many studies reported behavioural fever, its opposite, behavioural cooling-when infected animals lower their body temperature by using cool microenvironments-remains poorly documented.
RESULTS: Here, we report the first experimental evidence of behavioural cooling in an ectothermic vertebrate as a response to pathogenic infection. We investigated thermoregulatory responses in ta

While amphibians around the globe are facing catastrophic declines, in part because of infections with pathogens such as the Frog Virus 3 (FV3) ranavirus; the mechanisms governing amphibian susceptibility and resistance to such pathogens remain poorly understood. The type I and type III interferon (IFN) cytokines represent a cornerstone of vertebrate antiviral immunity, while our recent work indicates that tadpoles and adult frogs of the amphibian Xenopus laevis may differ in their IFN responses to FV3. In this respect, it is notable that anuran (frogs and toads) tadpoles are significantly mor

Long non-coding RNAs (lncRNAs) play important roles in the infection of pathogen. In this study, the role of the lncRNA lnc32039 in Epinephelus coioides, the precious marine fish in China, was studied during Singapore grouper iridovirus (SGIV) infection. The results demonstrated that SGIV infection upregulated the expression of lnc32039. Overexpressing lnc32039 could significantly exacerbate viral induced CPE, upregulate the expressions of viral key genes (MCP, ICP18, VP19, LITAF), and lnc32039 also significantly increased the emergence of progeny viruses by TCID50, while the opposite results

Grouper iridovirus (GIV) is regarded as a prominent viral pathogen of grouper, particularly during the larval and juvenile stages. The aim of this study was to comprehensively characterize the GIV-120L gene during viral infection. The results of sequence analysis suggest that GIV-120L is a 1,470 bp gene encoding a Ranavirus-specific viral protein. Recombinant GIV-120L protein was purified using a nickel-affinity column, and its molecular weight was found to be 59.1 kDa. To obtain antibodies against GIV, mice were immunized with recombinant GIV-120L protein, the spleen was harvested 8 weeks lat

Grouper Rana-iridovirus (GIV-R) and nervous necrosis virus (NNV) are two distinct yet very important causative agents in several fish species. However, as of now, no effective treatment has been developed to simultaneously prevent these two pathogens. In this study, we developed GIV-R-NNV chimeric vaccine candidates by using GIV-R to chimerically express the full length and P domain of the coat protein of NNV, and designated as GIV-R-Δ51-NNV-CP-FL, GIV-R-Δ51-NNV-CP-P, respectively. Firstly, the orf51 deletion recombinant GIV-R (GIV-R-Δ51) was constructed and characterized. The results showed t

Largemouth bass ranavirus (LMBV) inflicts substantial economic losses on largemouth bass aquaculture; however, the contributions of long non-coding RNAs (lncRNAs) and competing endogenous RNA (ceRNA) networks to the host antiviral immune response remain poorly understood. In this study, we performed a comprehensive analysis of lncRNA-mediated ceRNA regulatory networks during LMBV infection, aiming to elucidate their functional roles in modulating immune defenses in largemouth bass.We performed whole transcriptome sequencing (RNA-seq and miRNA-seq) on head kidney tissues from LMBV-infected fish

As the main highly pathogenic pathogen of grouper, Singapore grouper iridovirus (SGIV) can give rise to significant economic losses in grouper aquaculture. Bacillus amyloliquefaciens, is a probiotic with proven positive effects on grouper and Trachinotus ovatus, protecting those species against several bacterial pathogens. However, its antiviral activity remains to be investigated. We have previously isolated B. amyloliquefaciens E35, a probiotic with a desirable effect on pathogenic bacteria. We aimed to evaluate the anti-SGIV activity of E35 and extracts in vivo and in vitro, and the current

The spleen is an important immune organ in adult Xenopus laevis, supporting the differentiation of B cells and acting as the main peripheral lymphoid organ. Key to these processes are the supporting non-hematopoietic cells, or stromal cells, within the spleen tissue. Despite the importance of the spleen to frog immunity, few frog cell lines originating from spleen tissue have been reported. In this study, we report on the establishment and characterization of two cell lines originating from X. laevis spleen tissue, Xela S5F and Xela S5E. Morphological observations and gene expression profiling

Pathogens such as herpesviruses, Mycoplasma spp., and frog virus 3-like ranavirus have contributed to morbidity and mortality in many species of free-living and zoo-maintained chelonians. However, their prevalence is understudied in Blanding's turtles (Emydoidea blandingii) across North America. To assess the presence of these pathogens, Blanding's turtles were sampled in Lake County, Illinois, in 2017 (N = 213) and 2018 (N = 160). DNA from cloacal-oral swabs was assayed for four ranaviruses, three Mycoplasma spp., two Salmonella spp., Emydoidea herpesvirus 1 (EBHV1), and tortoise intranuclear

Singapore grouper iridovirus (SGIV) causes severe systemic infections and high mortality in farmed groupers, leading to substantial economic losses. Natural products with immunomodulatory properties have emerged as promising eco-friendly alternatives for antiviral prevention and control in aquaculture. In this study, the antiviral activity and underlying mechanisms of 3-indoleacetonitrile against SGIV were investigated in grouper spleen (GS) cells and in hybrid grouper. The results showed that 3-indoleacetonitrile exhibited no cytotoxicity at concentrations up to 62.5 μg/mL in GS cells, and si

Amphibians have been disappearing at an unprecedented rate worldwide. Among the proposed contributing factors are infectious diseases. Investigations have focused mainly on ranavirus and chytrids; however, additional agents may be relevant stressors. Two novel batrachoviruses have been discovered (ranid herpesvirus 3 [RaHV-3] and bufonid herpesvirus 1 [BfHV-1]). Their clinical role is still to be clarified; however, both have been associated with obvious skin lesions in their respective hosts. Herein we present 2 consensus PCR protocols that can be used to detect all of the known and, possibly
