Anti-acne activities of pulsaquinone, hydropulsaquinone, and structurally related 1, 4-quinone derivatives.

Quinone type compound, pulsaquinone 1, isolated from the aqueous ethanol extract of the roots of Pulsatilla koreana exhibited antimicrobial activities against an anaerobic non-spore-forming gram-positive bacillus, Propionibacterium acnes, which is related with the pathogenesis of the inflamed lesions in a common skin disease, acne vulgaris. Compound 1 was unstable on standing and thus converted to more stable compound 2, namely hydropulsaquinone by hydrogenation, whose activity was comparable to mother compound 1 (MIC for 1 and 2 against P. acnes: 2.0 and 4.0 microg/mL, respectively). Organic cation transporter 1 and cytochrome P450s play crucial roles in coptisine- and worenine-induced hepatotoxicity.

Coptisine and worenine, isoquinoline alkaloids prevalent in traditional herbal medicines from Papaveraceae (eg, Corydalis Rhizoma) and Ranunculaceae (eg, Coptidis Rhizoma), exhibit significant pharmacologic effects, including cardioprotective and anticancer properties. However, little information is available concerning coptisine- or worenine-induced toxicity, particularly hepatotoxicity, and the underlying mechanisms. Coptisine and worenine contain methylenedioxyphenyl groups, which may be metabolized by cytochrome P450s (P450s) to form reactive metabolites and subsequently induce toxicity. I

In vitro antimalarial activity of extracts of some plants from a biological reserve in Costa Rica.

Treatment with the usual antimalarial drugs, have induced parasite resistance, reinforcing the need to finding natural antimalarial components that would be found on plants from the forest. Therefore, we decided to look for these components in Costa Rican plants from a protected forest area. Fresh and dry extracts of roots, bark, leaves, flowers and fruits of 25 plants from a biological reserve in Costa Rica, Reserva Biol6gica Alberto Manuel Brenes (REBAMB), were studied in vitro for the presence of substances with antimalarial activity. By studying the inhibition of P berghei schizogony, we a

Photochemistry and photocytotoxicity of alkaloids from Goldenseal (Hydrastis canadensis L.) 1. Berberine.

Goldenseal is an herb which is widely used for many medical applications such as in eyewashes and skin lotions and which is currently undergoing testing by the National Toxicology Program. The main alkaloid constituent of Goldenseal is berberine. The topical application of Goldenseal or berberine to the skin or eyes raises the possibility that an adverse phototoxic reaction may result from an interaction between the alkaloid and light. We have therefore studied the photochemistry of berberine in different solvents and its phototoxicty to HaCaT keratinocytes. Irradiation of berberine in aqueous

Adverse and beneficial effects of plant extracts on skin and skin disorders.

Plants are of relevance to dermatology for both their adverse and beneficial effects on skin and skin disorders respectively. Virtually all cultures worldwide have relied historically, or continue to rely on medicinal plants for primary health care. Approximately one-third of all traditional medicines are for treatment of wounds or skin disorders, compared to only 1-3% of modern drugs. The use of such medicinal plant extracts for the treatment of skin disorders arguably has been based largely on historical/anecdotal evidence, since there has been relatively little data available in the scienti

Integrative management of gastric cardia cancer and precancerous lesions: prospects and mechanistic rationale for Banxia Xiexin Tang.

Gastric cardia cancer is a biologically heterogeneous malignancy arising at the gastroesophageal junction that is, the upper opening of the stomach rather than the heart. It develops along an inflammation-metaplasia-dysplasia-carcinoma sequence and remains clinically challenging because early lesions are often occult and advanced disease is shaped by a treatment-resistant tumor microenvironment. A clinically useful review therefore needs to distinguish gastric cardia cancer from non-cardia gastric cancer and from esophageal adenocarcinoma, while also clarifying where current evidence is direct

Nigella sativa Oil Reduces LPS-Induced Microglial Inflammation: An Evaluation on M 1/M 2 Balance.

OBJECTIVES: The immune system plays a critical defence role against infections, injuries, and carcinogenic stimuli. As the macrophages of the brain resides in the innate immune system, microglia and their polarisation (M 1/M 2) play regulatory roles in inflammation in CNS, such as Parkinson's, Alzheimer's, dementia complex, and multiple sclerosis. Nigella sativa belongs to the Ranunculaceae family and has different anti-inflammatory and antioxidant effects. We conducted this study to evaluate the anti-inflammatory and protective properties of N. sativa oil (NSO) on the microglial cells and the
