Ephrin-A1/EphA2 modulates vascular smooth muscle cell proliferation in vitro, while VSMC-specific ephrin-A1 deletion is not sufficient to alter arteriosclerosis in vivo.

Atherosclerosis, the leading cause of heart attack and stroke, involves plaque formation driven by various cell types, including endothelial cells, immune cells and vascular smooth muscle cells (VSMCs). VSMCs undergo a phenotypic switch from a contractile to a synthetic state, contributing to disease progression. The Eph/ephrin signaling pathway, particularly ephrin-A1 and its receptor EphA2, has been implicated in this phenotypic modulation. Its role in atherosclerosis was explored using in vitro and in vivo models. In plaque-containing arteries, both ephrin-A1 and EphA2 were upregulated comp

EPHA2 Segregates with Microphthalmia and Congenital Cataracts in Two Unrelated Families.

EPHA2 is a transmembrane tyrosine kinase receptor that, when disrupted, causes congenital and age-related cataracts. Cat-Map reports 22 pathogenic EPHA2 variants associated with congenital cataracts, variable microcornea, and lenticonus, but no previous association with microphthalmia (small, underdeveloped eye, ≥2 standard deviations below normal axial length). Microphthalmia arises from ocular maldevelopment with >90 monogenic causes, and can include a complex ocular phenotype. In this paper, we report two pathogenic EPHA2 variants in unrelated families presenting with bilateral microphthalm

MiR-210 promotes bone formation in ovariectomized rats by regulating osteogenic/adipogenic differentiation of bone marrow mesenchymal stem cells through downregulation of EPHA2.

PURPOSE: In osteoporosis, the balance between osteogenic and adipogenic differentiation of mesenchymal stem cells (MSCs) is disrupted. The osteogenic differentiation of bone marrow MSCs (BMSCs) is important for improving osteoporosis. The aim of this study was to explore the role and molecular mechanism of miR-210 in the balance of osteogenic/adipogenic differentiation of BMSCs in postmenopausal osteoporosis. METHODS: Postmenopausal osteoporosis rat models were constructed by ovariectomy (OVX). BMSCs were isolated from the femur in rats of Sham and OVX groups. MiR-210 was overexpressed and su

EPHA2/CD44-directed trafficking enhances endosomal leakiness and antisense therapy delivery.

The potential for using therapeutic antisense oligonucleotides (ASOs) has been hampered by a lack of understanding of how they enter cells and subsequently access their targets. Endocytosis contributes to ASO uptake, but the machinery mediating subsequent ASO trafficking to permit suppression of their target mRNAs has not been described. Here, we show that direct ASO engagement with a scavenger receptor (CD44) activates the ERK-RSK axis to promote serine phosphorylation of a receptor tyrosine kinase (EPHA2). Serine phosphorylation of EPHA2 permits endocytosis, trafficking, and accumulation of

Suppression of CBP/p300 induces growth inhibition and autophagy of hepatocellular carcinoma cells through regulation of EPHA2 by its super-enhancer acetylation.

CREB binding protein (CBP) and its homolog E1A binding protein p300 (p300) are crucial lysine acetyltransferases that have been shown to play key roles in various human malignancies. However, the precise functions and regulatory mechanisms of CBP/p300 in hepatocellular carcinoma (HCC) remain to be explored. Herein, we report that the expression levels of CBP and p300 are upregulated in HCC compared to normal tissues, and high CBP expression is associated with poor overall survival based on TCGA data. Combined knockdown of CBP and p300 or treatment with the CBP/p300 histone acetyltransferases (

A recurrent splice-site mutation in EPHA2 causing congenital posterior nuclear cataract.

UNLABELLED: Intoduction: Inherited cataract, opacification of the lens, is the most common worldwide cause of blindness in children. We aimed to identify the genetic cause of autosomal dominant (AD) posterior nuclear cataract in a four generation British family. METHODS: Whole genome sequence (WGS) was performed on two affected and one unaffected individual of the family and further validated by direct sequencing. Haplotype analysis was performed via genotying. RESULTS: A splice-site mutation c.2826-9G>A in the gene EPHA2, encoding EPH receptor A2 was identified and found to co-segregate wit

Inhibiting Eph/ephrin signaling reduces vascular leak and endothelial cell dysfunction in mice with sepsis.

Sepsis is a life-threatening disease caused by a dysregulated host response to infection, resulting in 11 million deaths globally each year. Vascular endothelial cell dysfunction results in the loss of endothelial barrier integrity, which contributes to sepsis-induced multiple organ failure and mortality. Erythropoietin-producing hepatocellular carcinoma (Eph) receptors and their ephrin ligands play a key role in vascular endothelial barrier disruption but are currently not a therapeutic target in sepsis. Using a cecal ligation and puncture (CLP) mouse model of sepsis, we showed that prophylac

Progranulin Regulates Protein Synthesis in Myocytes Through an Ephrin Type A Receptor 2-Dependent Pathway.

AIM: Sarcopenia is associated with metabolic dysregulation, yet the molecular mediators remain poorly defined. This study aimed to identify relevant regulators of muscle mass and to elucidate the role and underlying mechanism of progranulin in skeletal muscle protein synthesis. METHODS: We combined transcriptomic profiling of muscles from high-fat diet-fed mice with human genetic data from the HugeAMP Type 2 Diabetes Knowledge Portal to identify potential regulators. Clinically, 172 participants were stratified into low muscle mass (LMM) and normal muscle mass (NMM) groups according to the As

Targeting EphA2 with a novel EFNA3-mimetic peptide enhances vaginal epithelial barrier function: Running title: EFNA3-MAP enhancing the vaginal epithelial barrier.

Barrier protection provided by vaginal stratified squamous epithelium (SSE) is supported by estrogen (E)-mediated stimulation of ephrin-Eph signaling that promote desmosome expression and function. Conversely, loss of E induced by use of the contraceptive depot-medroxyprogesterone acetate (DMPA) or menopause reduces desmosome expression and weakens this epithelial barrier. We previously reported treating mice with DMPA lowers expression of the desmosomal cadherin desmoglein-1 (DSG1) in vaginal SSE by dampening EFNA3-mediated signaling whereas these effects are partially reversed by intravagina

Inhibitor of DNA-Binding 3 Is a Novel Regulator of Limbal Epithelial Cell Migration Via the EphA2/Akt Signaling Pathway.

PURPOSE: Upon corneal injury, early and late transit amplifying (TA) cells, the progeny of epithelial stem cells, migrate to the site of the wound, which facilitates its closure. Understanding the targetable signals that guide such cell migration is essential for the development of novel wound-healing strategies. METHODS: Single-cell RNA sequencing (scRNA-seq) was conducted in wild-type cornea. To investigate the role of inhibitor of DNA binding 3 (ID3) in the limbal epithelium, bulk RNA-seq was conducted in limbal epithelial cells with ID3 knockdown. hTCEpi cells were transfected with small

The status of intercellular junctions in established lens epithelial cell lines.

PURPOSE: Cataract is the major cause of vision-related disability worldwide. Mutations in the crystallin genes are the most common known cause of inherited congenital cataract. Mutations in the genes associated with intercellular contacts, such as Nance-Horan Syndrome (NHS) and Ephrin type A receptor-2 (EPHA2), are other recognized causes of congenital cataract. The EPHA2 gene has been also associated with age-related cataract, suggesting that intercellular junctions are important in not only lens development, but also in maintaining lens transparency. The purpose of this study was to analyze

To escape or not to escape, that is the question.

Marco et al. (https://doi.org/10.1083/jcb.202507217) identify a tumor-selective endocytic route in which CD44 and EPHA2 co-traffic antisense oligonucleotides (ASOs) to nuclear-proximal, recycling-endosome-like compartments that become leaky through lipid peroxidation and show that stress granule-mediated repair of these compartments limits productive ASO escape.

Proteomic Analysis of PTEN-Deficient Cells Reveals Src-Mediated Upregulation of EphA2 and Therapeutic Potential of Dual Inhibition.

Loss of the tumor suppressor phosphatase and tensin homolog (PTEN) is frequently observed in various cancers and promotes tumorigenesis by activating the PI3K-AKT pathway. However, the effectiveness of therapies targeting this pathway is limited by complex signaling crosstalk and compensatory mechanisms. Here, we employed quantitative proteomic and phosphoproteomic analyses using MCF10A PTEN KO models to comprehensively map the signaling alterations induced by PTEN loss. Our analyses revealed that PTEN deficiency not only activates canonical PI3K-AKT signaling but also induces widespread chang

A new ephrin-A1 isoform (ephrin-A1b) with altered receptor binding properties abrogates the cleavage of ephrin-A1a.

Ephrins are ligands for the Eph receptor tyrosine kinases, which play important roles in patterning nervous and vascular systems. Ephrin-A1 is a glycosylphosphatidylinositol-anchored ligand that binds to the EphA receptor tyrosine kinases. In the present study, we have identified a new ephrin-A1 isoform, denoted ephrin-A1b (ephrin-A1 isoform b). Compared with the originally described ephrin-A1 sequence, ephrin-A1a [Holzman, Marks and Dixit (1990) Mol. Cell. Biol. 10, 5830-5838], ephrin-A1b lacks a segment of 22 amino acids (residues 131-152). At the transcript level, exon 3 is spliced out in t

Eph receptors in pancreatic cancer: Biological roles and clinical implications.

Pancreatic ductal adenocarcinoma (PDAC) is one of the most aggressive malignancies with a 5-year survival rate of approximately 12%. Treatment options, including surgical resection and chemotherapy, are often ineffective in advanced stages due to late detection. This review examines the role of Erythropoietin-producing human hepatocellular (Eph) receptors in PDAC, emphasizing their potential both as therapeutic targets and biomarkers. Eph receptors, a family of receptor tyrosine kinases, are frequently overexpressed in various cancers and contribute to tumor growth, angiogenesis, and metastasi

A Human Three-Dimensional In Vitro Model of Lens Epithelial Cells as a Model to Study Mechanisms of Drug-Induced Posterior Subcapsular Cataracts.

Purpose: Cataract is a pathological opacification of the lens, which is still one of the leading causes of blindness in the world. Several etiologies are described, among them drug-induced cataract, for example, posterior subcapsular cataract (PSC) after steroid treatment. To investigate different mechanisms of drug-induced cataract a human three-dimensional (3D) lens in vitro model was developed, consisting of immortalized human lens epithelial cells. Methods: These cells were cultivated on 96-well, ultralow attachment plates, where they rapidly form spheroids. By gene expression analysis dif

In Silico Redesign of Epstein-Barr Virus Entry Glycoproteins to Redirect Cellular Tropism as a Foundation for a Novel B Cell Oncolytic Therapy.

B cell acute lymphoblastic leukemia (B-ALL) affects thousands of patients each year, with relapse occurring in some cases due to residual malignant B cells following chemotherapy or immunotherapy. Relapsed disease has a median survival of only a few months under conventional salvage therapy and a markedly elevated mortality rate.Oncolytic viruses offer an alternative treatment that works by specifically replicating within cancer cells, inducing cytolysis, and replicating and amplifying the viral particles at the site of infection until the tumor is eliminated, a mechanism valuable for eliminat

Universal Solid-Phase Reversible Sample-Prep for Concurrent Proteome and N-Glycome Characterization.

We describe a novel solid-phase reversible sample-prep (SRS) platform that enables rapid sample preparation for concurrent proteome and N-glycome characterization for nearly all protein samples. SRS utilizes a uniquely functionalized, silica-based bead that has strong affinity toward proteins with minimal to no affinity for peptides and other small molecules. By leveraging this inherent size difference between proteins and peptides, SRS permits high-capacity binding of proteins, rapid removal of small molecules (detergents, metabolites, salts, peptides, etc.), extensive manipulation including

Multi-receptor targeted therapy of breast cancer and brain metastases with a novel QUAD-drug conjugate.

BACKGROUND: Identifying treatments for triple-negative breast cancer (TNBC) remains a critical medical need. We have found that Interleukin 13 receptor alpha 2 (IL-13RA2), EphA2, EphA3 and EphB2 receptors are over-expressed collectively in majority of patients with breast cancer and its brain metastases. We are pursuing the novel idea of targeting these four tumor-associated receptors identified by us with one pharmaceutical compound. A compound, called QUAD, was designed and constructed, which binds all four targeted receptors. METHODS: We have examined the presence of IL-13RA2, EphA2, EphA3

Penicophenone F from an Arctic Fungus Against UVB-Induced Corneal Damage via Inhibiting the ROS-EphA2 Pathway.

Ultraviolet B (UVB) radiation-induced corneal injury poses a significant public health challenge. However, its underlying molecular mechanisms remain incompletely understood, hindering the development of effective interventions. This study identified a key molecular pathway in UVB-induced corneal damage, revealing that UVB exposure triggers a rapid intracellular burst of reactive oxygen species (ROS), which in turn upregulates and aberrantly activates the receptor tyrosine kinase Ephrin type-A receptor 2 (EphA2), thereby collectively accelerating DNA damage and photoaging in corneal epithelial
