Transcriptional up-regulation of the human androgen receptor by androgen in bone cells.

Androgen regulation of androgen receptor (AR) expression has been observed in a variety of tissues, generally as inhibition, and is thought to attenuate cellular responses to androgen. AR is expressed in osteoblasts, the bone-forming cell, suggesting direct actions of androgens on bone. Here we characterized the effect of androgen exposure on AR gene expression in human osteoblastic SaOS-2 and U-2 OS cells. Treatment of osteoblastic cells with the nonaromatizable androgen 5alpha-dihydrotestosterone increased AR steady state messenger RNA levels in a time- and dose-dependent fashion. Reporter a

Wide variation in androgen receptor dysfunction in complete androgen insensitivity syndrome.

Androgen insensitivity syndrome (AIS) is a disorder of male sexual differentiation caused by mutations in the androgen receptor (AR) gene. The partial form (PAIS), associated with varying degrees of receptor dysfunction, presents with a range of undervirilization phenotypes. The complete form (CAIS) is characterized by normal female external appearance at birth. In these cases the receptor is often absent or inactive. However, cases have been described where the mutant receptor concerned has considerable residual activity in in vitro assays. Here we describe the effects of five mutations, Gly7

Partial androgen resistance due to a distinctive qualitative defect of the androgen receptor.

Using whole genital skin fibroblasts, we have characterized a novel androgen receptor mutation in a family with partial androgen resistance. The proposita was born with bilateral labioscrotal folds and a single perineal urogenital orifice. Her similarly affected maternal aunt was raised as a female with the support of gonadectomy and vaginoplasty. The mutant androgen receptor has a normal maximum binding capacity (Bmax), but an increased apparent equilibrium dissociation constant (Kd) with 5 alpha-dihydrotestosterone (DHT) and 2 synthetic androgens, methyltrienolone (MT) and mibolerone (MB). Androgen receptors and sociosexual behaviors in mammals: The limits of generalization.

Circulating testosterone is easily aromatized to estradiol and reduced to dihydrotestosterone in target tissues and elsewhere in the body. Thus, the actions of testosterone can be mediated either by the estrogen receptors, the androgen receptor or by simultaneous action at both receptors. To determine the role of androgens acting at the androgen receptor, we need to eliminate actions at the estrogen receptors. Alternatively, actions at the androgen receptor itself can be eliminated. In the present review, I will analyze the specific role of androgen receptors in male and female sexual behavior

Androgen receptors are frequently expressed in mammary and extramammary Paget's disease.

Mammary Paget's disease and extramammary Paget's disease are rare intraepithelial neoplasms. Mammary Paget's disease is almost exclusively associated with underlying invasive breast carcinoma or high-grade ductal carcinoma in situ (DCIS G3). Extramammary Paget's disease arises in areas rich in apocrine glands and is suspected to have apocrine gland origin. The aim of the study was to investigate the presence of estrogen receptor (ER), progesterone receptor (PR), androgen receptor (AR) and Her2/neu in a large number of cases. We investigated 58 cases of mammary and 23 cases of extramammary Page

Possible role of androgen receptors in amyotrophic lateral sclerosis. A hypothesis.

Androgen receptors have been demonstrated in both cranial nerve and spinal motor neurons. This article proposes that amyotrophic lateral sclerosis (ALS) may be a disease in which androgen receptors in motor neurons are lost or not functioning. This is suggested by the male-to-female ratio of the disease, the age of onset, and the sparing of neurons of cranial nerves III, IV, and VI that coincidentally lack androgen receptors. The hypothesis is that ALS may be due to a loss of androgen receptors that results in an inability to respond to a variety of insults including axonal damage.

Expression of Androgen and Estrogen Receptors in the Human Lacrimal Gland.

Lacrimal gland dysfunction causes dry eye disease (DED) due to decreased tear production. Aqueous-deficient DED is more prevalent in women, suggesting that sexual dimorphism of the human lacrimal gland could be a potential cause. Sex steroid hormones are a key factor in the development of sexual dimorphism. This study aimed to quantify estrogen receptor (ER) and androgen receptor (AR) expression in the human lacrimal gland and compare it between sexes. RNA was isolated from 35 human lacrimal gland tissue samples collected from 19 cornea donors. AR, ERα, and ERβ mRNA was identified in all sampl

Immunoexpression of androgen receptors in the reproductive tract of the stallion.

The objective of this study was to visualize androgen receptors (ARs) in the testis, epididymis, and prostate of the stallion by means of immunohistochemistry. Nuclear immunostaining was found in all somatic cells in the testis--Leydig, Sertoli, and peritubular myoid cells; in both types of epithelial cells of the epididymis; and in the secretory cells of the prostate. These results indicate that ARs are distributed throughout the reproductive tract cells of the stallion.

Conversion of transformed androgen and glucocorticoid receptors to higher molecular forms.

The 4 S transformed androgen receptor from rat prostate and thymus converted to a higher molecular form (5-7 S) on low-salt conditions. The converted receptor retained the DNA-binding capacity as well as the 4 S transformed receptor. This conversion was also demonstrated on glucocorticoid receptor from rat liver and thymus. The sedimentation coefficients of both the converted receptors were affected by sodium molybdate, i.e., the receptors converted to a relatively smaller molecule in the presence of molybdate than in the absence of the reagent. These observations suggest that molybdate direct

Ontogeny of mesenchymal androgen receptors in the embryonic mouse mammary gland.

We demonstrate the presence of a high affinity androgen-binding site in the embryonic mouse mammary gland, and describe its appearance and development from the initial formation of the gland bud (day 12) through the androgen-responsive stage (day 14) until term (day 19). Binding assays were done on entire organ rudiments exposed to the ligand [( 3H] testosterone, [3H]5 alpha-dihydrotestosterone, or [3H]methyltrienolone) in vitro, and receptor levels are expressed per gland. In competition experiments, the binding site shows a ligand specificity of a typical rodent androgen receptor. Scatchard

Androgen receptor immunohistochemistry in genitourinary neoplasms.

PURPOSE: Androgen receptor (AR) is a recognized immunohistochemical marker of prostate cancer. However, the sensitivity and specificity of AR for prostate cancer in the setting of other genitourinary neoplasms has not been rigorously studied. METHODS: We employed tissue microarrays containing prostate carcinomas, urothelial carcinomas, renal cell carcinomas, and testicular neoplasms. Slides were stained immunohistochemically for AR. RESULTS: Androgen receptor was positive in 95% of prostate carcinomas (n=230), but 19% of invasive urothelial carcinomas of the bladder (n=190) and 33% of non-in

Modulators of androgen and estrogen receptor activity.

This review focuses on significant recent findings regarding modulators of androgen and estrogen receptor activity. Selective androgen receptor modulators (SARMs) interact with androgen receptors (ARs), and selective estrogen receptor modulators (SERMs) interact with estrogen receptors (ERs), with variable tissue selectivity. SERMs, which interact with both ERб and ERв in a tissue-specific manner to produce diverse outcomes in multiple tissues, continue to generate significant interest for clinical application. Development of SARMs for clinical application has been slower to date because of po

Intranuclear organization and function of the androgen receptor.

We have entered an exciting era for androgen-receptor (AR) research that should provide a detailed description of how the AR functions as a ligand-regulated transcription factor. That AR activity is regulated by subcellular compartmentalization was first established a decade ago with the finding that binding of androgen to the AR induces its translocation from the cytoplasm to the nucleus. The contribution of compartmentalization to AR activity is, however, likely to extend beyond simple delivery to the nucleus. Defects in AR and coregulator compartmentalization in the nucleus have been demons

[Clinical consequences of androgen receptor malfunction].

The androgen receptor (AR), the mediator of the effects of the male sex hormones testosterone and dihydrotestosterone, plays a crucial role in development of male sex and in the function of male sexual organs. Pathological alterations of AR structure and function are a major cause of androgen insensitivity in male pseudohermaphroditism and the accompanying deformations of genital organs, or result in spinal and bulbar muscular atrophy (SBMA). In addition, AR alterations that generate a hyperreactive receptor contribute to the development of resistance to hormone ablation therapy in prostate ca

Recent advances in androgen receptor action.

Androgens, principally testosterone and 5 alpha-dihydrotestosterone, play critical roles in the development and growth of the male reproductive and nonreproductive systems. Their biological actions are mediated by the androgen receptor (AR), a ligand-dependent transcription factor, belonging to the nuclear receptor superfamily. These androgen-AR complexes interact with various factors (e.g. coactivators or corepressors) to modulate transcription of androgen target genes via specific DNA sequences. Many lines of evidence have also correlated AR with several mammalian disorders. Finally, recent

Repressors of androgen and progesterone receptor action.

Androgen and progesterone receptors (AR and PR) are two determining factors in gonadal differentiation that are highly expressed in developing and mature gonads. Loss of AR results in XY sex reversal and mutations causing reduced AR activity lead to varying degrees of defects in masculinization. Female PR knockout mice are infertile due to ovarian defects. While much has been discovered about positive regulation of these receptors by coactivators little is known about repression of the transcriptional activity of AR and PR in the presence of agonists. In this study we assessed the effect of SM

Molecular basis of androgen receptor diseases.

androgens act through a single intracellular androgen receptor (AR) which is encoded by a single-copy gene in the X chromosome. Disruption of the AR by genetic mutation results in complete androgen insensitivity syndrome (CAIS) and the female phenotype in otherwise healthy 46XY individuals. Although CAIS is the best known phenotype, recent studies from our laboratory and elsewhere show that malfunction of the AR is associated with many androgen-regulated diseases or conditions that cross traditional clinical disciplines ranging from paediatrics (ambiguous genitalia), gynaecology (primary ameno

Androgen receptor content in human endometrium.

Human endometrium changes morphologically and biochemically during the various phases of the menstrual cycle, influenced by not only estrogens and progesterone, but probably also by androgens. The purpose of this study was to investigate the androgen receptor (AR) content in human endometrium and myometrium and its significance. AR immunocytochemistry was performed on 30 paraffin-embedded uterine sections of pre-menopausal women who were scheduled for hysterectomy because of benign gynaecologic abnormalities. AR receptor content was seen in all cell types of the human endometrium and myometriu
