[The effect of L-DOPA and 6-hydroxydopamine on the intracellular cAMP level in the kidneys of rats with impaired neurotrophic support of the organs].

The administration to rats surviving reflectory renal dystrophy of L-DOPA causing the disturbance in the function of mineralocorticoid receptors was demonstrated to be accompanied by an increase of cAMP concentration in the kidneys whereas 6-oxidopamine alleviating the damage of these receptors led to a reduction of cAMP content in the organ. It is assumed that catecholamine-activated adenylate-cyclase system and the processes initiated thereby which determine protein metabolism in renal tubule cells, among them protein subunits of mineralocorticoid receptors may take part in the mechanisms of

Differential regulation of neuropeptide Y and catecholamine production in superior cervical ganglion cultures.

Primary cultures of newborn rat superior cervical ganglion (SCG) neurons grown in complete serum-free medium have been used to study the regulation of catecholamine and neuropeptide Y (NPY) production. The levels of mRNA for preproNPY and for tyrosine hydroxylase were determined, and the rates of synthesis and accumulation of NPY and catecholamines were measured. Drugs which effectively blocked accumulation of newly synthesized catecholamines (tyramine, reserpine) had no effect on NPY synthesis and accumulation. The catecholamine uptake blocker desipramine and the alpha(2)-adrenergic receptor

Biochemical studies on the release of catecholamines from the rat carotid body in vitro.

The effects of hypoxia and carbachol on the release of newly synthesized catecholamines from superfused rat carotid bodies have been examined. Hypoxic superfusion medium was found to evoke catecholamine release which was dependent on the extracellular calcium concentration and was reduced by nitrendipine and atropine. Superfusion with the muscarinic agonist, carbachol, stimulated catecholamine release independently of the oxygen tension of the medium. The effect of carbachol on catecholamine release was abolished by atropine, suggesting that it was mediated by activation of cholinergic recepto

Inhibition of lymphocyte activation by catecholamines: evidence for a non-classical mechanism of catecholamine action.

The effects of noradrenaline and other adrenergic agonists on lymphocyte activation were studied. Spleen and thymus cells from BALB/c mice were stimulated by mitogens and lymphocyte activation was monitored by measuring the incorporation of [methyl-3H]thymidine into DNA. Noradrenaline, adrenaline, isoproterenol and dopamine all inhibited the activation of spleen and thymus cells by concanavalin A, a T-cell specific mitogen, and the activation of spleen cells by lipopolysaccharide, a T-independent B-cell mitogen. The various catecholamines were approximately equipotent, having IC50 of approxima

Characterization of muscarinic receptors of bovine adrenal chromaffin cells: binding, secretion and anti-microtubule drug effects.

1. Binding of [3H]QNB to adrenal membranes is saturable, specific and to a single class of receptors. 2. Tubulozole, and not other microtubule drugs, inhibits [3H]QNB binding. 3. Pretreating cultured chromaffin cells with oxotremorine, a muscarinic receptor agonist, has no effect on either basal, nicotine (10 microM) or K(+)-stimulated catecholamine release and failed to enhance secretion of submaximal concentrations of nicotine (3-5 microM). 4. These results confirm that binding of [3H]QNB is associated with muscarinic receptors on bovine adrenal medullary tissue. 5. These studies also demons

Rapid antidepressant response to alprazolam in depressed patients with high catecholamine output and heterologous desensitization of platelet adenylate cyclase.

The present study examined the relationship between 24-hr urinary catecholamine (norepinephrine and epinephrine) output and measures of platelet adenylate cyclase (AC) activity in depressed patients (n = 17) and control subjects (n = 10). In both groups, significant inverse correlations were observed when 24-hr urinary catecholamine levels were examined in relation to measures of both receptor-mediated (prostaglandin D2 and alpha 2-adrenergic) and postreceptor-mediated (NaF) platelet AC enzyme activities, suggesting that circulating catecholamines may regulate platelet AC by heterologous (agon

Effects of dopamine agonists and antagonists in Tourette's disease.

The actions of haloperidol, dextroamphetamine sulfate, levamfetamine succinate, apomorphine, and piribedil were studied in two patients with Giles de la Tourette's disease in an attempt to clarify the catecholamine mechanisms involved in this condition. Both dextroamphetamine and levamfetamine increased the severity of the symptoms; dextroamphetamine was more potent. Haloperidol controlled the symptoms and also antagonized the effect of dextroamphetamine. Apomorphine injections reduced the severity of symptoms, even in the presence of dextroamphetamine. We conclude that dopamine rather than no

Suppression of pre-B cell colony formation by catecholamine oxidation.

The sympathetic nervous system regulates many aspects of lymphocyte function by stimulating adrenergic receptors with catecholamines. To determine the effects of β-adrenergic receptor (β-AR) signaling on B cell development, we utilized the pre-B cell colony forming unit (CFU-Pre-B) assay and found that isoproterenol (ISO), a β-AR selective catecholamine, potently suppresses pre-B cell colony formation. Further investigation using selective agonists and knockout mice showed that the colony suppression was independent of adrenergic receptor signaling. This colony suppression was replicated by tr

[Effects of dopamine on the coronary vessels of swine].

The pharmacological responses of dopamine, a catecholamine which is used as a circulatory active drug in shock treatment, were investigated in isolated coronary arteries of pigs from the slaughter house. The effects are compared with those obtained after application of adrenaline and noradrenaline in the same in vitro system. The aim of the experiments was to discover whether the three catecholamines have an effect on the same adrenergic receptors and to characterize the specific effect of dopamine. Phentolamine and propranolol were used as adrenergic alpha- und beta-blockers, azaperone and ha

Trophoblastic cells of the hydatidiform mole contain a beta 1-subtype adrenergic receptor.

Both beta 1- and beta 2-adrenergic receptors have been previously described in normal human placental homogenates; the cells upon whose surface membranes these receptors reside have not been identified. In order to show that a beta 1-adrenergic receptor is present on trophoblastic cells, the cells which mediate maternal-fetal transport and produce placental hormones, beta-adrenergic receptors were demonstrated in membrane fractions of human hydatidiform mole. Microscopic sections of the mole samples used demonstrated edematous villi lined by trophoblastic cells with minimal nontrophoblastic (s

Sympathetic neural activation evoked by mu-receptor blockade in patients addicted to opioids is abolished by intravenous clonidine.

BACKGROUND: Mu-opioid receptor blockade by naloxone administered for acute detoxification in patients addicted to opioids markedly increases catecholamine plasma concentrations, muscle sympathetic activity (MSA), and is associated with cardiovascular stimulation despite general anesthesia. The current authors tested the hypothesis that the alpha2-adrenoceptor agonist clonidine (1) attenuates increased MSA during mu-opioid receptor blockade for detoxification, and (2) prevents cardiovascular activation when given before detoxification. METHODS: Fourteen mono-opioid addicted patients received n

Age-associated alterations in catecholaminergic concentrations, neuronal activity, and alpha 1 receptor densities in female rats.

The density of alpha 1-adrenergic receptors and catecholamine concentrations and neuronal activity were assessed in selected estrogen-responsive regions of the hypothalamus and pineal glands. The results of this study demonstrate that the densities of alpha 1-adrenergic receptors and norepinephrine and epinephrine concentrations and activity rate constants are altered in aged rats. The direction and extent of the change depend on the specific brain region and reproductive state of the animals. No widespread decline with increased age was observed in any of the parameters measured.

Effects of testosterone on fat cell lipolysis. Species differences and possible role in polycystic ovarian syndrome.

Testosterone is a potent regulator of lipolysis by influencing catecholamine signal transduction in fat cells. Major species differences exist as regards the testosterone effect. In rodents testosterone increases beta-adrenergic receptor mediated signals to lipolysis at multiple steps in the lipolytic cascade. The sex hormone also increases alpha2-adrenoceptor antilipolytic signalling in hamster which unlike rat express this receptor in their fat cells. In humans the region of adipose tissue is critical. Visceral fat cell lipolysis is not responsive to testosterone but this sex hormone decreas

Induction of M2 regulatory macrophages through the β2-adrenergic receptor with protection during endotoxemia and acute lung injury.

The main drivers of acute inflammation are macrophages, which are known to have receptors for catecholamines. Based on their function, macrophages are broadly categorized as having either M1 (proinflammatory) or M2 phenotypes (anti-inflammatory). In this study, we investigated catecholamine-induced alterations in the phenotype of activated macrophages. In the presence of lipopolysaccharide (LPS), mouse peritoneal macrophages acquired an M1 phenotype. However, the copresence of LPS and either epinephrine or norepinephrine resulted in a strong M2 phenotype including high levels of arginase-1 and

Effect of various pharmacological agonists on the rat caecum-appendix preparation: a preliminary study.

A preliminary study with various pharmacological agents revealed the presence of contractile muscarinic and H1-histamine receptors as well as inhibitory alpha- and beta-adrenoceptors and purinoceptors in rat caecum-appendix. Histamine also produced indirect actions mediated through the release of catecholamines. 5-Hydroxytryptamine produced variable response, the contractile response was mediated through 5-HT receptors and the relaxant response through the release of catecholamines.

Beta adrenergic receptors in keratinocytes.

Beta2 adrenergic receptors were identified in keratinocytes more than 30 years ago, but their function in the epidermis continues to be elucidated. Abnormalities in their expression, signaling pathway, or in the generation of endogenous catecholamine agonists by keratinocytes have been implicated in the pathogenesis of cutaneous diseases such as atopic dermatitis, vitiligo, and psoriasis. New studies also indicate that the beta2AR also modulates keratinocyte migration, and thus can function to regulate wound reepithelialization. This review focuses on the function of these receptors in keratin

Isoproterenol-initiated beta-adrenergic receptor diacytosis in cultured cells.

The kinetics of the return of internalized beta-adrenergic receptors to the plasma membrane were measured in human astrocytoma cells. The movement of [125I]iodopindolol-labeled receptors back to the plasma membrane was measured directly and was shown to occur with a t1+2 of 3-4 min. Unlabeled receptors appeared to exhibit the same kinetics of externalization. The process was not inhibited by low concentrations (1-10 microM) of propranolol or even high concentrations of isoproterenol (0.1-1.0 mM). Higher concentrations of propranolol (0.1-1.0 mM) and other lipophilic amines inhibited externaliz

Pathological cardiac hypertrophy: the synergy of adenylyl cyclases inhibition in cardiac and immune cells during chronic catecholamine stress.

Response to stressors in our environment and daily lives is an adaptation conserved through evolution as it is beneficial in enhancing the survival and continuity of humans. Although stressors have evolved, the drastic physiological response they elicit still remains unchanged. The chronic secretion and circulation of catecholamines to produce physical responses when they are not required may result in pathological consequences which affect cardiac function drastically. This review seeks to point out the probable implication of chronic stress in inducing an inflammation disorder in the heart.

Induction of glycogenolysis in cultured Ewing's sarcoma cells by dopamine and beta-adrenergic agonists.

This study describes hormonal regulation of glycogen metabolism in Ewing's sarcoma cells. 3H-Glycogen synthesized in cultured Ewing's sarcoma WE-68 cells from 3H-glucose was hydrolyzed in a concentration-dependent manner by various catecholamines. The order of potency for the glycogenolytic effects of catecholamines was isoproterenol greater than or equal to dopamine greater than norepinephrine greater than epinephrine. The concentrations giving half-maximal effectiveness (EC50) were about 2 x 10(-8) M, 3 x 10(-8) M, 8 x 10(-8) M, and 5 x 10(-7) M for isoproterenol, dopamine, norepinephrine, a

Lithium inhibits function of voltage-dependent sodium channels and catecholamine secretion independent of glycogen synthase kinase-3 in adrenal chromaffin cells.

Lithium has been proven to be effective in the therapy of bipolar disorder, but its mechanism of pharmacological action is not clearly defined.
