OBJECTIVES: Cell-specific antibodies were used to identify immunocompetent cells in a comparison of valves from patients who had symptomatic tricuspid aortic stenosis with subjects who had no evidence of valvular heart disease.
BACKGROUND: Nonrheumatic valvular aortic stenosis is the most common valvular heart disease among adults. The biologic processes involved in the development of this disease are poorly understood.
METHODS: Tricuspid stenotic aortic valves were obtained from 19 patients undergoing surgery for nonrheumatic valvular aortic stenosis, and 10 control valves were collected at

OBJECTIVE: Soluble interleukin-2 receptor (sIL-2R) is a biomarker for T cell activity. T cells are involved in neuromyelitis optica spectrum disorders (NMOSD) and myelin oligodendrocyte glycoprotein antibody-associated disease (MOGAD) pathogenesis. However, sIL-2R has so far not been evaluated in these conditions. Here, we compared sIL-2R levels in serum and cerebrospinal fluid (CSF) of patients with aquaporin-4-IgG-seropositive and seronegative (AQP4-IgG+/-) NMOSD, MOGAD, and noninflammatory neurologic disorders (NINDs), and assessed the prognostic value of sIL-2R for future attacks.

Endocytosis controls many functions including nutrient uptake, cell division, migration and signal transduction. A clathrin- and caveolin-independent endocytosis pathway is used by important physiological cargos, including interleukin-2 receptors (IL-2R). However, this process lacks morphological and dynamic data. Our electron microscopy (EM) and tomography studies reveal that IL-2R-pits and vesicles are initiated at the base of protrusions. We identify the WAVE complex as a specific endocytic actor. The WAVE complex interacts with IL-2R, via a WAVE-interacting receptor sequence (WIRS) present

Human T-lymphotropic virus type 1 (HTLV-1) is the etiological agent of adult T-cell leukemia/lymphoma, and it encodes a number of nonstructural proteins that are involved in virus replication and immune evasion. The viral protein p12 previously has been characterized to interfere with major histocompatibility complex class, ICAM-1, and ICAM-2 expression, and it activates STAT5. Using a previously established T-cell line immortalized with an HTLV-1 molecular clone deleted for p12, we assessed the role of p12 in regulating cellular growth and virus transmission. These cells were complemented for

Alopecia areata affects 1%-2% of the population and is hypothesized to be an autoimmune, organ specific T-cell mediated reaction directed against the human hair follicle. It is characterized by loss of hair in patches (alopecia areata) with progression in some individuals to total loss of scalp hair (alopecia totalis) or to loss of all scalp and body hair (alopecia universalis). The interleukin-1 receptor antagonist (IL-1RN) gene was found to be associated with more severe clinical outcome in several chronic inflammatory diseases, including alopecia areata. The IL-1RN*2 allele was found to be

OBJECTIVE: To observe serumal soluble interleukin-2 receptor levels in patients with tonsillitis.
METHOD: SIL-2R levels in serum were detected in 68 patients with acute or chronic tonsillitis and 68 normal controls.
RESULT: The SIL-2R concentrations in patients with acute tonsillitis and in patients with chronic tonsillitis within attacking period were (642.2 +/- 87.2) x 10(3) U/L and (762.3 +/- 90.5) x 10(3) U/L, respectively. Both were significantly higher than those of the controls (285.5 +/- 49.6) x 10(3) U/L (P < 0.001). Moreover the latter was higher than the former (P < 0.001). There

To explore the regulatory mechanism of the soluble interleukin-2 receptor (SIL-2R) in patient allergic rhinitis, SIL-2R levels in serum and nasal secretion were detected in 39 patients with allergic rhinitis, 19 patients with chronic rhinitis and 15 normal controls, The determination of SIL-2R was done by a sandwich ELISA method. The results showed that the SIL-2R concentrations in patients with allergic rhinitis were significantly higher than those of the controls (P < 0.001). These results suggest that SIL-2R may play an important role in allergic rhinitis.

The levels of soluble interleukin 2 receptor alpha (sIL-2R alpha) were measured in pleural fluid from 38 patients including 16 cases with tuberculous pleurisy, 14 with malignant pleural effusions and 8 with transudative effusions by using ELISA technique. The level of sIL-2R alpha in tuberculous pleurisy is markedly higher than in malignant pleural effusions (P < 0.01). The levels of sIL-2R alpha in tuberculous pleurisy and in malignant pleural effusions are significantly higher than in transudative effusions (P < 0.01). The clinical significance is discussed.

It is known that the affinity cross-linking study of the human high-affinity Interleukin 2 (IL-2) receptor reveals triplet bands consisting of 70 kDa alpha chain(Tac)-IL-2 and the 90/80 kDa doublet. We found the cell lines lacking the lower band of the doublet in spite of the expression of both alpha and beta chains. No IL-2 binding was detectable in the presence of anti-Tac antibody in these cells. Immunoprecipitation from the cell extract of [125 I] IL-2-cross-linked T cells with anti-beta chain polyclonal IgG detected the upper band, but not lower band of the doublet. These data suggest tha

The serum of twenty-four patients with histopathological confirmation of cutaneous T-cell lymphoma, including three with Sezary's syndrome, and twenty age matched controls was examined for soluble interleukin-2 receptor using a commercially available ELISA kit. Whilst there was no significant difference in the mean concentration of the interleukin-2 receptor between patients with mycosis fungoides and controls, six of twenty-one patients had interleukin-2 receptor levels outside the mean plus two standard deviations of the age matched cohort. Patients with Sezary's syndrome had markedly elevat

The nature of the interleukin 2 (IL-2) receptor on purified human B lymphocytes was examined. Both normal and malignant cells showed evidence of a 70-75,000 mol. wt (p75) IL-2 binding molecule as assessed by 125I-labeled IL-2 binding and receptor cross-linking. On normal, Tac-negative B lymphocytes the estimated number of p75 binding sites was 1100 per cell and the dissociation constant (Kd) was 1.7 nM. Consistent with this, cross-linking experiments demonstrated the presence of an IL-2 binding molecule of 70-75,000 mol. wt. Purified B cells from patients with hairy cell leukemia and chronic l

The expression of interleukin-2 (IL-2) receptor (CD25) has been investigated in 165 cases of hematologic neoplasia by alkaline phosphatase-antialkaline phosphatase (APAAP) labeling of cell smears using two monoclonal anti-IL-2 receptor antibodies (anti-Tac and DAKO-IL2-R). IL-2 receptor was found in 2 of 16 (13%) T-cell malignancies (1 T-ALL, 1 Sézary syndrome). In contrast, among B-cell disorders, IL-2 receptor was expressed in the majority of hairy cell leukemia cases (14 of 16, i.e., 88%) and a smaller proportion of cases of common acute lymphoblastic leukemia (7 of 44, i.e., 16%), B-cell c

The objective was to assess the impact of genetic variation on cervical cytokine concentrations of interleukin (IL)-1α, IL-1β, IL-6, IL-8 and tumor necrosis factor alpha (TNF-α), and first, to determine if these variants interact with polymorphisms in Toll-like receptor 4 (TLR4) that were previously shown to associate with pro-inflammatory cervical cytokine concentrations, and second, to determine if findings are affected by bacterial vaginosis (BV). We examined 183 single nucleotide polymorphisms (SNPs) in 13 cytokine genes and receptors for associations with cervical cytokine levels in 188 A

High-affinity interleukin 2 (IL2) receptors on human T lymphocytes are multimeric complexes containing two IL2-binding polypeptides, alpha and beta chains of 50-55 and 70-75 kDa, respectively, associated by noncovalent bonds. IL2 binds to high-affinity IL2 receptors on the surface of T lymphocytes, mediates cell growth, and is internalized. In this paper, we used a biochemical method to directly identify the receptors components internalized together with the ligand. 125I-IL2-receptor complexes were solubilized with the detergent 3-[(3-cholamidopropyl)dimethylammonio]-1-propane-sulfonate, and

Kawasaki disease is a febrile disease of childhood that is associated with increased inflammatory cytokines and immunoregulatory abnormalities. While the serum concentrations of soluble IL-2 receptor can change under such pathologies, the relevance of the soluble IL-2 receptor concentration in patients with Kawasaki disease has not been specified. We aimed to summarize the existing studies that reported the soluble IL-2 receptor concentrations in patients with Kawasaki disease. Original articles that were published up to July 2016 were collected using a PubMed/Medline-based search engine. A to

Serum soluble interleukin-2 receptor (sIL-2r) is an important disease marker in hemophagocytic lymphohistiocytosis (HLH), but there are no published data on its diagnostic value in adults. We conducted a single-center retrospective study of 78 consecutive adults who had sIL-2r measured for suspected HLH. Serum sIL-2r levels were measured by enzyme-linked immunosorbent assay (adult reference range, 241-846 U/mL). There were 38 patients with HLH and 40 with a non-HLH diagnosis (such as sepsis, liver disease, histiocyte disorders, autoimmune disease, leukemia, or lymphoma). The receiver operating
