Enantioselective behavioral effects of threo-methylphenidate in rats.

The relative potency of d- and l-threo-methylphenidate (d-MPH and l-MPH) was evaluated using three behavioral paradigms for rats: Responding maintained by a fixed-interval schedule of reinforcement (FI), responding maintained by a concurrent variable-interval schedule of reinforcement (Conc VI VI), and consumption of sweetened condensed milk during a 15-min free-access period. In each case the potency of the d-MPH enantiomer greatly exceeded that of the l-MPH enantiomer. Temporal control of responding was reduced (FI) choice responding was equalized for most rats (Conc VI VI), and milk consump

Effects of chlorpromazine on rats' acquisition of lever-press responding with immediate and delayed reinforcement.

The effects of chlorpromazine (0, 2, 6, and 10 mg/kg) on the acquisition of lever-press responding by rats were examined under conditions where reinforcement (water delivery) was immediate or delayed. Under the immediate reinforcement condition, water-deprived rats were exposed during 8-h sessions to a fixed-ratio 1 (FR 1) schedule of water delivery without prior autoshaping or hand shaping. Under the delayed reinforcement condition, similar rats were exposed to a tandem FR 1 fixed-time 8-s schedule of water delivery. A different squad of eight rats was exposed to each delay condition and drug

Operant behavior and drinking suppression in vasopressin-deficient rats (Brattleboro strain).

A group of Brattleboro rats homozygous for diabetes insipidus (DI) and a group of normal Long-Evans (LE) rats were deprived of water and tested on two tasks thought to measure reinforcement strength: a progressive-ratio schedule in an operant chamber and a quinine-induced drinking-suppression task. Since water-deprived DI rats are in more severe negative water balance than LE rats, it was hypothesized that they would sustain higher ratios than LE rats on the progressive-ratio schedule and would suppress drinking at higher quinine concentrations than LE rats. It was found that DI rats responded

Using high-preference and low-preference music in a synchronous reinforcement treadmill preparation: A further extension.

We conducted a series of studies on the effects of synchronous reinforcement. Study 1 presented 30 participants with their high-preference (HP) music, identified via a conjugate assessment, for walking on a treadmill during three synchronous reinforcement (SYNC) components. The results indicated that HP music produced schedule control of walking speed for 20 participants (66.7%). In addition, 80% of the participants who displayed schedule control also displayed variable walking speeds when music was withheld. Study 2 extended Study 1 by providing 30 new participants with their low-preference (

Cocaine self-administration in adult female and male rhesus monkeys: longitudinal comparison with adolescent behavior and role of early life stress.

A phenomenon involving cocaine use disorders is the "incubation of drug craving" - the drive for the drug increases the longer the abstinence period. The present longitudinal study provided a unique opportunity to test whether an increase in the reinforcing effects of cocaine developed after prolonged abstinence and if early life stress was a risk factor. Fourteen (N = 6 female, 8 male) adult rhesus monkeys, some (N = 7) that were maltreated as infants by their mothers (MALT), had previously self-administered cocaine under a fixed-ratio (FR) schedule of reinforcement as adolescents, but had no

Comparing fixed-amount and progressive-amount DRO Schedules for tic suppression in youth with chronic tic disorders.

Chronic tic disorders (CTDs) involve motor and/or vocal tics that often cause substantial distress and impairment. Differential reinforcement of other behavior (DRO) schedules of reinforcement produce robust, but incomplete, reductions in tic frequency in youth with CTDs; however, a more robust reduction may be needed to affect durable clinical change. Standard, fixed-amount DRO schedules have not commonly yielded such reductions, so we evaluated a novel, progressive-amount DRO schedule, based on its ability to facilitate sustained abstinence from functionally similar behaviors. Five youth wit

The motivational properties of schedule-induced polydipsia.

Schedule-induced polydipsia occurred during initial magazine training to Noyes pellets (45 mg), disappeared when lever-pressing was acquired on a continuous reinforcement schedule (CRF), and reappeared when the food contingency was changed to a 1-min variable interval schedule (VI 1 min). Polydipsia also developed under a VI 1 min food schedule when water was concurrently available on various fixed ratios (FR), rather than being freely available. The level of the polydipsia and its motivating properties allow it to be classified as a form of adjunctive behavior.

Control of schedule-induced polydipsia: type, size, and spacing of meals.

Rats were given daily 1-min variable-interval sessions for several types of food delivered in various amounts per reinforcement and the concurrent, schedule-induced polydipsia was measured. Dry, solid food was neither a necessary nor sufficient condition for the development of polydipsia. Small portions of liquid Standard Monkey Diet produced polydipsia, but 45-mg dextrose or sucrose pellets did not. Within the range studied, smaller portions of both solid and liquid foods produced more drinking than larger portions per reinforcement. Two-min variable-interval sessions produced a greater polyd

Caregiver-Implemented Interventions to Improve Daily Living Skills for Individuals With Developmental Disabilities: A Systematic Review.

Many individuals with intellectual and developmental disabilities (DD) encounter difficulties in performing daily living skills, which limits their self-sufficiency and autonomy. Caregivers, such as parents, are often the individuals who interact with individuals with DD the most during daily living skills routines. Therefore, it is critical to teach caregivers to implement daily living skills interventions with their children. A systematic review of the literature related to caregiver-implemented daily living skills interventions yielded 38 articles. Of those, 20 articles, with 21 experiments

Prize contingency management for smoking cessation: a randomized trial.

BACKGROUND: Adjunctive behavioral smoking cessation treatments have the potential to improve outcomes beyond standard care. The present study had two aims: (1) compare standard care (SC) for smoking (four weeks of brief counseling and monitoring) to SC plus prize-based contingency management (CM), involving the chance to earn prizes on days with demonstrated smoking abstinence (carbon monoxide (CO) ≤6 ppm); and (2) compare the relative efficacy of two prize reinforcement schedules-one a traditional CM schedule, and the second an early enhanced CM schedule providing greater reinforcement magnit

Chronic ruminative vomiting: a comparison of four treatment procedures.

An experimental analysis of a case of chronic ruminative vomiting in a 15-year-old, profoundly retarded, Down's syndrome boy addressed the consequences of vomiting in the postmealtime environment. The experiment compared four treatments in each hour-long session using a multiple reinforcement schedule with order of the following procedures counterbalanced over days in a Latin square design: differential reinforcement of any behaviors other than vomiting (DRO) in which periods of no vomiting were followed by reinforcement and vomiting postponed reinforcement; differential reinforcement of speci

β2* nAChR sensitivity modulates acquisition of cocaine self-administration in male rats.

Signaling through nicotinic acetylcholine receptors (nAChRs) plays a role in cocaine reward and reinforcement, suggesting that the cholinergic system could be manipulated with therapeutics to modulate aspects of cocaine use disorder (CUD). We examined the interaction between nAChRs and cocaine reinforcement by expressing a hypersensitive β2 nAChR subunit (β2Leu9'Ser) in the ventral tegmental area of male Sprague Dawley rats. Compared to control rats, β2Leu9'Ser rats acquired (fixed ratio) intravenous cocaine self-administration faster and with greater likelihood. By contrast, β2Leu9'Ser rats w

Behavioural analysis of the anorectic effects of fluoxetine and fenfluramine.

Two sets of experiments were carried out to compare the effects of fenfluramine and fluoxetine on consummatory and operant behaviour. In food-deprived rats allowed access to a 35% sucrose solution, an initial period of sucrose consumption was followed by a short period of grooming and exploratory behaviour, later superceded by resting. This "behavioural satiety sequence" was advanced by fluoxetine, but disrupted by dl-fenfluramine, which suppressed post-prandial resting, even at sub-anorectic doses. Fluoxetine also elicited resting behaviour following water drinking. However, this did not appe

Reinforcement magnitude modulation of rate dependent effects in pigeons and rats.

Response rate can influence the behavioral effects of many drugs. Reinforcement magnitude may also influence drug effects. Further, reinforcement magnitude can influence rate-dependent effects. For example, in an earlier report, we showed that rate-dependent effects of two antidepressants depended on reinforcement magnitude. The ability of reinforcement magnitude to interact with rate-dependency has not been well characterized. It is not known whether our previous results are specific to antidepressants or generalize to other drug classes. Here, we further examine rate-magnitude interactions b

A laboratory comparison of two variations of differential-reinforcement-of-low-rate procedures.

We compared 2 variations of differential-reinforcement-of-low-rate (DRL) procedures: spaced-responding DRL, in which a reinforcer was delivered contingent on each response if a specified interval had passed since the last response, and full-session DRL, in which a reinforcer was presented at the end of an interval if the response rate was below criterion within the specified interval. We used a human-operant procedure and analyzed within-session responding to assess any similarities or differences between procedures. Data revealed a positive contingency between responding and reinforcement und

Schedule control of quantal and graded dose-effect curves in a drug-drug-saline discrimination.

Pigeons were trained to discriminate among 5 mg/kg pentobarbital, 5 mg/kg morphine, and saline when responding was maintained under fixed-interval (FI) or fixed-ratio (FR) reinforcement schedules. After the discrimination was established, other drugs were substituted for the training drugs. After low doses of pentobarbital and chlordiazepoxide, responding shifted from the saline key to the pentobarbital key under both FR and FI schedules. After low doses of morphine and methadone, responding shifted from the saline key to the morphine key under both reinforcement schedules. After all doses of

Metabotropic glutamate receptor (mGluR5) antagonist MPEP attenuated cue- and schedule-induced reinstatement of nicotine self-administration behavior in rats.

Previous studies suggested that metabotropic glutamate 5 (mGlu5) receptors play an important role in the reinforcing effects of abused drugs. The present experiments evaluated the effects of the mGlu5 receptor antagonist, MPEP (2-methyl-6-(phenylethynyl)-pyridine hydrochloride; 1-10 mg/kg, salt, i.p.), in rat models of nicotine-seeking behavior that may have relevance to relapse to drug-taking. Male Wistar rats (with restricted access to food) were trained to nose-poke to receive intravenous infusions of nicotine (0.03 mg/kg per infusion, base) under a fixed ratio 5 time out 60 s schedule of r

Resurgence of behavior during extinction depends on previous rate of response.

In two experiments, food-deprived rat subjects leverpressed for food in three successive training phases. In the first phase of both experiments, rats were exposed to a multiple schedule, one component of which produced a high rate of response, and the other of which produced a lower rate of response (multiple random ratio [RR], random interval [RI] in Experiment 1, and multiple differential reinforcement of high rate, differential reinforcement of low rate in Experiment 2). Rats were then transferred to a multiple fixed interval (FI; 60-sec, 60-sec) schedule, until the effects of the first ph

Schedule-induced polydipsia as a function of percent of body weight in the Mongolian gerbil.

Three male Mongolian gerbils displayed schedule-induced polydipsia on a fixed-time 3-min food reinforcement schedule at 85% body weight (BW). When the gerbils' body weights were gradually increased to ad lib levels, the polydipsia disappeared. When the gerbils were subsequently reduced to 85% BW again, the previous polydipsia levels of water intake were recaptured. These data replicate previous findings with rats, and suggest that schedule-induced polydipsia may be as robust in gerbils and other species, as it is with rats, once the optimal generating conditions are established.

The Long-Acting D3 Partial Agonist MC-25-41 Attenuates Motivation for Cocaine in Sprague-Dawley Rats.

The dopamine D3 receptor is a prime target for developing treatments for cocaine use disorders (CUDs). In this study, we conducted a pre-clinical investigation of the therapeutic potential of a long-acting, D3 receptor partial agonist, MC-25-41. Male rats were pre-treated with MC-25-41 (vehicle, 1.0, 3.0, 5.6, or 10 mg/kg, intraperitoneal (IP)) five minutes prior to tests of cocaine or sucrose intake on either a progressive ratio schedule of reinforcement or a variable interval 60 s multiple schedule consisting of 4, 15-min components with sucrose or cocaine available in alternating components
