The role of medial prefrontal cortex in context-specific inhibition during reversal learning of a visual discrimination.

Rats with medial prefrontal cortex or sham lesions were trained on a visual discrimination task designed for the eight-arm radial maze. After reaching asymptotic performance on this task, both groups were divided into sub-groups that would experience reversal learning in the same or different context from original training. The results showed that both groups reversed in the different context had accelerated learning compared to the groups reversed in the same context. Reversal learning in rats with medial prefrontal cortex damage was faster than sham animals in the same context. These and oth

Reversal of age-related learning deficits and brain oxidative stress in mice with superoxide dismutase/catalase mimetics.

Oxidative stress has been implicated in cognitive impairment in both old experimental animals and aged humans. This implication has led to the notion that antioxidant defense mechanisms in the brain are not sufficient to prevent age-related increase in oxidative damage and that dietary intake of a variety of antioxidants might be beneficial for preserving brain function. Here we report a dramatic loss of learning and memory function from 8 to 11 months of age in mice, associated with marked increases in several markers of brain oxidative stress. Chronic systemic administration of two synthetic

Impairment of olfactory, auditory, and spatial serial reversal learning in rats recovered from pyrithiamine-induced thiamine deficiency.

Rats that had recovered from pyrithiamine-induced thiamine deficiency (PTD) were compared with controls for spatial, auditory, and olfactory serial reversal learning (SRL); spatial matching to sample (MTS); auditory go-no-go discrimination; and open-field exploration. PTD rats made more errors reaching criterion for SRL in all modalities but showed normal transfer effects between problems. PTD rats were also impaired in learning the go-no-go and MTS tasks and showed consistent alterations in exploratory activity. It is argued that the PTD rat, like human Korsakoff patients, have impairments of

Learning curve of augmented reality surgical application for reverse shoulder arthroplasty.

BACKGROUND: Accurate positioning of the glenoid component in reverse total shoulder arthroplasty is important since it reduces prosthesis-related complications. Conventional instrumentation is still the gold standard. However, patient-specific instrumentation using 3-D printed guides, as well as computer-assisted navigation, is gaining more and more importance. Augmented reality has been established to enable the surgeon to have more precision in the positioning and inclination of the glenoid component. A new technique called "NextAR" (©Medacta) supports the surgeon during operation by guidanc

Reversible second-order conditional sequences in incidental sequence learning tasks.

In sequence learning tasks, participants' sensitivity to the sequential structure of a series of events often overshoots their ability to express relevant knowledge intentionally, as in generation tasks that require participants to produce either the next element of a sequence (inclusion) or a different element (exclusion). Comparing generation performance under inclusion and exclusion conditions makes it possible to assess the respective influences of conscious and unconscious learning. Recently, two main concerns have been expressed concerning such tasks. First, it is often difficult to desi

Decoding face identity: A reverse-correlation approach using deep learning.

Face recognition is crucial for social interactions. Traditional approaches primarily rely on subjective judgment, utilizing a pre-selected set of facial features based on literature or intuition to identify critical facial features for face recognition. In this study, we adopted a reverse-correlation approach, aligning responses of a deep convolutional neural network (DCNN) with its internal representations to objectively identify facial features pivotal for face recognition. Specifically, we trained a DCNN, namely VGG-FD, to possess human-like capability in discriminating facial identities.

Exercise reverses learning deficits induced by hippocampal injury by promoting neurogenesis.

Hippocampal atrophy and cognitive decline are common sequelae of many neurodegenerative disorders, including stroke. To determine whether cognitive decline can be ameliorated by exercise-induced neurogenesis, C57BL/6 mice in which a unilateral hippocampal injury had been induced by injecting the vasoconstrictor endothelin-1 into their right hippocampus, were run voluntarily for 21 days on a running-wheel. We found the severe deficits in spatial learning, as detected by active place-avoidance task, following injury were almost completely restored in animals that ran whereas those that did not r

A complication-based learning curve from 200 reverse shoulder arthroplasties.

BACKGROUND: Reported early complication rates in reverse total shoulder arthroplasty have widely varied from 0% to 75% in part due to a lack of standard inclusion criteria. In addition, it is unclear whether revision arthroplasty is associated with a higher rate of complications than primary arthroplasty. QUESTIONS/PURPOSE: We therefore (1) determined the types and rates of early complications in reverse total shoulder arthroplasty using defined criteria, (2) characterized an early complication-based learning curve for reverse total shoulder arthroplasty, and (3) determined whether revision a

Fear learning in unmedicated patients with anxiety disorders: a comparison of delay conditioning, fear reversal, and trace conditioning.

Anxiety disorders are common and impairing, yet their underlying mechanisms remain incompletely understood. Fear learning provides a critical translational framework for investigating pathological anxiety, bridging laboratory models and clinical phenomena. Prior studies have been limited by important methodological issues, including the inclusion of non-anxiety diagnoses, high comorbidity, and medication use. Here we examined three forms of fear learning- delay conditioning, fear reversal, and trace conditioning-in unmedicated adults with minimally comorbid primary anxiety disorders (Generaliz

Novel application potential of lisdexamfetamine for visual discrimination and reversal learning improvement in rats: A translational cognitive research.

Impairments in visual discrimination and cognitive flexibility are prevalent in several neuropsychiatric disorders. Here, we explored the effects of lisdexamfetamine, the prodrug of d-amphetamine, on visual discrimination and cognitive flexibility using touchscreen-based visual discrimination and reversal learning task, a translational cross-species cognitive paradigm in rats. A single administration of 1.0 mg/kg lisdexamfetamine significantly improved the accuracy of visual discrimination. Furthermore, repeated treatment with 1.0 mg/kg lisdexamfetamine during the visual discrimination trainin

The role of 5-HT2C receptors in touchscreen visual reversal learning in the rat: a cross-site study.

RATIONALE: Reversal learning requires associative learning and executive functioning to suppress non-adaptive responding. Reversal-learning deficits are observed in e.g. schizophrenia and obsessive-compulsive disorder and implicate neural circuitry including the orbitofrontal cortex (OFC). Serotonergic function has been strongly linked to visual reversal learning in humans and experimental animals but less is known about which receptor subtypes are involved. OBJECTIVES: The objectives of the study were to test the effects of systemic and intra-OFC 5-HT2C-receptor antagonism on visual reversal

Prefrontal cortex and hippocampus in behavioural flexibility and posttraumatic functional recovery: reversal learning and set-shifting in rats.

Within one experiment and one T-maze, we examined the consequences of (i) bilateral lesions of the anteromedial prefrontal cortex (PFC), (ii) bilateral transections of the fimbria-fornix (FF), or (iii) combined lesions of both PFC and FF (COMB) on rats' ability to perform reversal or set-shifting. Postoperatively, the animals were trained to perform a spatial discrimination go-right task. This was followed by (1) a spatial reversal go-left task (reversal learning), or (2) a visual pattern discrimination task (set-shift). Neither single (PFC or FF) lesion nor combined (COMB) lesions affected th

Dysfunction of Orbitofrontal GABAergic Interneurons Leads to Impaired Reversal Learning in a Mouse Model of Obsessive-Compulsive Disorder.

Cognitive inflexibility is a cardinal symptom of obsessive-compulsive disorder (OCD) and often manifests as impaired reversal learning. Abnormal recruitment of the orbitofrontal cortex (OFC)-striatal circuit is implicated in reversal learning deficits in patients with OCD. However, the precise circuitry mechanism underlying normal and impaired reversal learning remains elusive. Using fiber photometry and optogenetics, we demonstrated cell-type-specific activity dynamics in the OFC-striatal circuit underlying normal reversal learning and cell-type-specific dysfunctions that causally lead to imp

Increased perseverative errors following high-definition transcranial direct current stimulation over the ventrolateral cortex during probabilistic reversal learning.

BACKGROUND: The prefrontal cortex regulates behavioural adaptation in response to feedback. However, the causal role of different prefrontal regions remains unclear, based on indirect evidence derived from functional neuroimaging. Neuroimaging studies show dorsomedial prefrontal activation during feedback monitoring, whereas the ventrolateral prefrontal cortex engages during behavioural adaptation (shifting). OBJECTIVE: We used high-definition transcranial direct current stimulation (HD-tDCS) to elucidate the roles of the ventrolateral prefrontal cortex (vlPFC) and the dorsomedial prefrontal

Altered reversal and extinction learning in the DMSXL mouse model of type I myotonic dystrophy (DM1): An exploratory study.

BackgroundCognitive changes in type 1 myotonic dystrophy (DM1) have a pronounced negative effect on quality of life measures. Despite this, the neural basis of these changes is poorly understood. DM1 patients demonstrate deficits in motivation and cognitive flexibility, reflective of apathy and obsessive-compulsive / autistic-like traits.ObjectiveThese traits can be readily assessed using reversal learning and appetitive extinction tasks. Reversal learning assesses the ability to learn following a change in a rule and can evaluate cognitive flexibility and habitual responding, while appetitive

EphB2 reverse signaling regulates learned opiate tolerance via hippocampal function.

Despite significant progress, many uncertainties remain regarding molecular and cellular mechanisms governing opiate tolerance. We report that loss of EphB2 receptor reverse signaling results in a marked acceleration of morphine tolerance in vivo. EphB2 null mice exhibited no significant difference in brain or blood morphine metabolism, mu opiate receptor affinity or binding capacity. Motor and sensory performance for EphB2 null mice was also comparable to controls for both morphine naïve or tolerized states. Regional distributions of mu opioid receptor, CGRP and substance P were also unaltere

Predicting Micropollutant Removal by Reverse Osmosis and Nanofiltration Membranes: Is Machine Learning Viable?

Predictive models for micropollutant removal by membrane separation are highly desirable for the design and selection of appropriate membranes. While machine learning (ML) models have been applied for such purposes, their reliability might be compromised by data leakage due to inappropriate data splitting. More importantly, whether ML models can truly understand the mechanisms of membrane separation has not been revealed. In this study, we evaluate the capability of the XGBoost model to predict micropollutant removal efficiencies of reverse osmosis and nanofiltration membranes. Our results dem

COVID-19 Coronavirus Vaccine Design Using Reverse Vaccinology and Machine Learning.

To ultimately combat the emerging COVID-19 pandemic, it is desired to develop an effective and safe vaccine against this highly contagious disease caused by the SARS-CoV-2 coronavirus. Our literature and clinical trial survey showed that the whole virus, as well as the spike (S) protein, nucleocapsid (N) protein, and membrane (M) protein, have been tested for vaccine development against SARS and MERS. However, these vaccine candidates might lack the induction of complete protection and have safety concerns. We then applied the Vaxign and the newly developed machine learning-based Vaxign-ML rev

Concord grape juice reverses the age-related impairment in latent learning in rats.

OBJECTIVE: Two experiments were conducted to determine if dietary supplementation with Concord grape juice could reverse the latent learning impairment normally observed in middle-aged male rats. METHODS: Both experiments utilized the latent cue preference (LCP) task, in which water-replete rats sample water in one compartment of a three-compartment box, and are subsequently given a compartment preference test when water-deprived to determine if they remember the compartment cue previously associated with water. In the first experiment, 40 male Sprague-Dawley rats (9, 10, 11, or 12 months old

Effects of meta-chlorophenylpiperazine (mCPP) on a T-maze acquisition and reversal learning task in male and female rats.

Behavioral flexibility is critical for adaptive functioning and is often impaired in neuropsychiatric disorders such as obsessive-compulsive disorder (OCD). The serotonergic agent meta-chlorophenylpiperazine (mCPP), a non-selective 5-HT receptor agonist, is commonly used to model OCD-like behaviors. However, its effects on behavioral flexibility, particularly in females, remain poorly understood. This study investigated sex differences and the effects of mCPP on acquisition and reversal learning in adult Wistar rats using the T-maze task. Experiment 1 compared male and female rats during the a
