Expectation of reward differentially modulates executive inhibition.

BACKGROUND: Inhibitory control, a key modulatory component of cognition guiding strategy and behaviour, can be affected by diverse contingencies. We explore here the effect of expectation of reward over behavioural adjustment in a Stop Signal Task modulated by reward. We hypothesize that cognitive control is modulated by different expectation of the reward. METHODS: Participants were allocated to two groups differing in their degree of knowledge in what to expect from rewards. Expected Specific Reward participants (N = 21) were informed of the different monetary feedbacks they would receive a

Circuit Mechanisms of Reward, Anhedonia, and Depression.

Pleasure and motivation are important factors for goal-directed behavior and well-being in both animals and humans. Intact hedonic capacity requires an undisturbed interplay between a number of different brain regions and transmitter systems. Concordantly, dysfunction of networks encoding for reward have been shown in depression and other psychiatric disorders. The development of technological possibilities to investigate connectivity on a functional level in humans and to directly influence networks in animals using optogenetics among other techniques has provided new important insights in th

A Neural Circuit for Gut-Induced Reward.

The gut is now recognized as a major regulator of motivational and emotional states. However, the relevant gut-brain neuronal circuitry remains unknown. We show that optical activation of gut-innervating vagal sensory neurons recapitulates the hallmark effects of stimulating brain reward neurons. Specifically, right, but not left, vagal sensory ganglion activation sustained self-stimulation behavior, conditioned both flavor and place preferences, and induced dopamine release from Substantia nigra. Cell-specific transneuronal tracing revealed asymmetric ascending pathways of vagal origin throug

Working Memory Load Strengthens Reward Prediction Errors.

Reinforcement learning (RL) in simple instrumental tasks is usually modeled as a monolithic process in which reward prediction errors (RPEs) are used to update expected values of choice options. This modeling ignores the different contributions of different memory and decision-making systems thought to contribute even to simple learning. In an fMRI experiment, we investigated how working memory (WM) and incremental RL processes interact to guide human learning. WM load was manipulated by varying the number of stimuli to be learned across blocks. Behavioral results and computational modeling co

Lateral hypothalamic circuits for feeding and reward.

In experiments conducted over 60 years ago, the lateral hypothalamic area (LHA) was identified as a critical neuroanatomical substrate for motivated behavior. Electrical stimulation of the LHA induces voracious feeding even in well-fed animals. In the absence of food, animals will work tirelessly, often lever-pressing thousands of times per hour, for electrical stimulation at the same site that provokes feeding, drinking and other species-typical motivated behaviors. Here we review the classic findings from electrical stimulation studies and integrate them with more recent work that has used c

Neurobiology of decision-making: risk and reward.

Neurological patients with bilateral ventromedial (VM) lesions of the prefrontal cortex often deny, or they are not aware that they have a problem. Furthermore, they often pursue actions that bring some reward in the immediate run, despite severe long-term consequences such as the loss of job, home, and family. The somatic marker hypothesis, which provides an account of this defect in decision-making, posits that the impairment is the result of defective activation of somatic markers that normally function as covert or overt signposts for helping with the process of making choices that are adv

Brain neurotransmitters in food and drug reward.

The discovery of brain peptides as neurotransmitters opens a new chapter in the study of feeding from a neural point of view. This article reviews current studies suggesting a limbic system framework of chemical neuroanatomy in which peptides play an important role in the control of eating and behavior reinforcement. In this framework, food intake and a variety of anorectic drugs (amphetamine, phenylpropanolamine, and fenfluramine) act on dopamine, norepinephrine, and serotonin systems that ascend from the hindbrain and midbrain to modulate feeding and satiety systems in the hypothalamus. Reward Behavior Disengagement, a Neuroeconomic Model-Based Objective Measure of Reward Pathology in Depression: Findings from the EMBARC Trial.

The probabilistic reward task (PRT) has identified reward learning impairments in those with major depressive disorder (MDD), as well as anhedonia-specific reward learning impairments. However, attempts to validate the anhedonia-specific impairments have produced inconsistent findings. Thus, we seek to determine whether the Reward Behavior Disengagement (RBD), our proposed economic augmentation of PRT, differs between MDD participants and controls, and whether there is a level at which RBD is high enough for depressed participants to be considered objectively disengaged. Data were gathered as

The Role of the Cerebellum in Drug Reward: A Review.

Drug abuse remains a global problem; nonetheless, its mechanism has not yet been fully understood. Recent studies have reported on the non-motor functions of the cerebellum, and evidence from neuroimaging and behavioral studies has suggested the role of cerebellum in drug reward, which has received increasing attention. Furthermore, emerging technological developments have aided in clarifying the various circuits and functions of the cerebellum. Exploring the role of the cerebellum in drug reward can improve our understanding of the mechanism underlying addiction and facilitate the development

The Role of Sensitivity to Reward and Punishment in Aggression.

The study of the risk and protective factors in aggression is of fundamental importance for our society. The aim of this research was to clarify the role of sensitivity to reward/punishment in aggression and provide a better understanding of the mechanisms underlying this relationship, particularly given that previous studies in the literature have yielded mixed results. To this end, two studies were conducted. In Study 1 (484 participants; Mage = 39.09; 48.6s women), we explored the relationship between sensitivity to reward and punishment and four components of aggression: physical, verbal,

Rewarding effects of 2-desoxypipradrol in mice.

Desoxypipradrol (2-DPMP), a new psychoactive substance (NPS), acts as a norepinephrine-dopamine reuptake inhibitor (NDRI). NDRIs can be addictive due to their action mechanisms similar to cocaine and methamphetamine. However, there is a lack of scientific information regarding the exact dependency of 2-DPMP. Thus, the purpose of this study was to evaluate rewarding and reinforcing effects of 2-DPMP in rodents. The effective dose range of 2-DPMP was determined by climbing behavior test. To evaluate rewarding effects of 2-DPMP, conditioned place preference (CPP) test was performed at selected do

A Dedicated Population for Reward Coding in the Hippocampus.

The hippocampus plays a critical role in goal-directed navigation. Across different environments, however, hippocampal maps are randomized, making it unclear how goal locations could be encoded consistently. To address this question, we developed a virtual reality task with shifting reward contingencies to distinguish place versus reward encoding. In mice performing the task, large-scale recordings in CA1 and subiculum revealed a small, specialized cell population that was only active near reward yet whose activity could not be explained by sensory cues or stereotyped reward anticipation behav

Food reward. What it is and how to measure it.

We investigated the contribution of hunger and food liking to food reward, and the relationship between food reward and food intake. We defined liking as the pleasantness of taste of food in the mouth, and food reward as the momentary value of a food to the individual at the time of ingestion. Liking and food reward were measured, respectively, by ratings of the pleasantness of the taste of a mouthful, and ratings of desire to eat a portion, of the food in question. Hunger, which we view as primarily the absence of fullness, was rated without food being present. Study 1 provided evidence that

Monetary rewards modulate inhibitory control.

The ability to override a dominant response, often referred to as behavioral inhibition, is considered a key element of executive cognition. Poor behavioral inhibition is a defining characteristic of several neurological and psychiatric populations. Recently, there has been increasing interest in the motivational dimension of behavioral inhibition, with some experiments incorporating emotional contingencies in classical inhibitory paradigms such as the Go/NoGo and Stop Signal Tasks (SSTs). Several studies have reported a positive modulatory effect of reward on performance in pathological condi

The endocannabinoid system and nondrug rewarding behaviours.

Rewarding behaviours such as sexual activity, eating, nursing, parenting, social interactions, and play activity are conserved strongly in evolution, and they are essential for development and survival. All of these behaviours are enjoyable and represent pleasant experiences with a high reward value. Remarkably, rewarding behaviours activate the same brain circuits that mediate the positive reinforcing effects of drugs of abuse and of other forms of addiction, such as gambling and food addiction. Given the involvement of the endocannabinoid system in a variety of physiological functions of the

Probing biomarkers and clinical utility of reward learning across species using the Probabilistic Reward Task: 20 years of findings.

Disrupted reward processing, including reward learning, is a prominent feature of various neuropsychiatric disorders. Such abnormalities are particularly consequential in major depressive disorder, in which anhedonia (reduced reactivity to pleasurable stimuli) predicts a host of negative outcomes, including poor response to treatments, worse disease course, impaired psychosocial functioning, and suicide. Despite substantial research in this area, there are no approved treatments for anhedonia; reasons for such modest progress include the use of vastly disparate approaches to assess anhedonic b
