Alternative morphs and plasticity of vulval development in a rhabditid nematode species.

The nematode species Rhabditis sp. SB347 (Family Rhabditidae) in standard culture conditions displays two developmental morphs with distinct modes of sexual reproduction: (1). females and males that develop through four feeding juvenile ("larval") stages; (2). self-fertile protandric hermaphrodites that develop through an obligatory non-feeding third juvenile stage, the "dauer" larva. In females and males, somatic gonad development begins in the first larval stage, whereas in hermaphrodites it is delayed to the second larval stage. Vulval development also differs between females and hermaphrod

An evolutionary framework for the study of developmental evolution in a set of nematodes related to Caenorhabditis elegans.

Nematodes are known to be a useful system for studies of comparative development. Here we perform a molecular phylogenetic analysis to allow for the independent interpretation of the developmental and morphological changes observed among a selected set of nematode species. Our molecular phylogenetic analysis is based on coding regions of the genes for RNA polymerase II, the small subunit rRNA and an expansion segment of the large subunit rRNA. Sequences were compared from five species in the family (Rhabditidae) that includes the developmental model organism Caenorhabditis elegans and from an

Scent of death: Emission and behavioral role of 1-nonene in entomopathogenic nematode Steinernema kraussei.

Entomopathogenic nematodes (EPNs) provide a natural alternative to synthetic pesticides for pest control. Understanding how EPNs respond to volatile cues including those released by infected cadavers is crucial for determining the behavior control necessary for improving their effectiveness. In this study, we tested the response of Steinernema kraussei, a cruiser foraging species, to 1-nonene. Additionally, we analyzed the emission dynamics of this compound from Galleria mellonella larvae infected by three different EPN species: S. kraussei, S. carpocapsae, and S. feltiae. Concentrations of 1-

Host defense or parasite cue: Skin secretions mediate interactions between amphibians and their parasites.

Amphibian skin secretions (substances produced by the amphibian plus microbiota) plausibly act as a first line of defense against parasite/pathogen attack, but may also provide chemical cues for pathogens. To clarify the role of skin secretions in host-parasite interactions, we conducted experiments using cane toads (Rhinella marina) and their lungworms (Rhabdias pseudosphaerocephala) from the range-core and invasion-front of the introduced anurans' range in Australia. Depending on the geographical area, toad skin secretions can reduce the longevity and infection success of parasite larvae, or

Molecular characterization of Ditylenchus destructor voltage-gated calcium channel α1 subunits and analysis of the effect of their knockdown on nematode activity.

Voltage-gated calcium channels (VGCCs) mediate the entry of Ca2+ ions into cells in response to membrane depolarization and play fundamental roles in the nervous system, and the α1 subunits are the main subunits of Ca2+ channels. Caenorhabditis elegans possesses genes encoding α1 subunits; however, very few of these genes have been cloned in plant-parasitic nematodes (PPNs). Ditylenchus destructor is a PPN that has been proposed as a new model for studying the biology and control of PPNs. To understand the structure and function of the VGCCs of this PPN, we first cloned and identified three fu

Radiculomeningomyelitis due to Halicephalobus gingivalis in a horse.

An adult horse was euthanatized following a clinical diagnosis of cauda equina neuritis. Significant gross postmortem and histopathologic findings were limited to the sacral spinal cord and cauda equina. The sacral spinal cord, meninges, and spinal nerve roots were expanded and partially effaced by sclerosing granulomatous inflammation with necrosis. The lesion contained numerous nematode larvae and fewer adults with a rhabditiform esophagus having a corpus, isthmus, and valved bulb. Female nematodes were amphidelphic and didelphic with reflexed ovaries. These morphologic features confirm Hali

Entomopathogenic nematodes associated with organic honeybush (Cyclopia spp.) cultivation in South Africa.

Entomopathogenic nematodes (EPNs) of the genera Steinernema and Heterorhabditis have demonstrated great potential as useful bio-control agents in the management of certain important soil-inhabiting insect pests of agricultural crops. In a survey of nematodes associated with organic honeybush cultivation, soil samples were obtained from nine organic honeybush plots, which are located in the Bredasdorp area of the Western Cape province of South Africa. The EPNs were isolated from soil by baiting with larvae of Galleria mellonella (greater wax moth) and identified by the amplification of the inte

Effects of tetracycline on entomopathogenic nematodes and their bacterial symbionts.

Among the new contaminants relevant for environment, one of the most significant roles is played by pharmaceuticals like antibiotic products for either human or veterinary use. Their presence could cause serious damage to bacteria and microfauna, like nematodes. Within the widely investigated nematodes, very little is known about the interaction between antibiotics and entomopathogenic nematodes (EPN). EPNs have been used for biological control of crops, due to their ability to penetrate arthropod pests and kill their hosts thanks to a complex symbiotic mechanism with specific gram-negative ba

Routes of penetration of the entomopathogenic nematode steinernema feltiae attacking larval and adult houseflies (Musca domestica).

The way in which Steinernema feltiae (Nematoda: Rhabditida: Steinernematidae) infects the housefly, Musca domestica (Diptera: Muscidae), was investigated. Adult flies were confined in petri dishes containing 1 million S. feltiae on capillary matting and individuals were dissected at hourly intervals. First, second, and third instar larvae were placed on filter papers with 100,000 infective juveniles, and then 2 larvae were examined at 30-min intervals. Infective juveniles were aggregated on the proboscis and anal aperture of male and female houseflies after 1 h. The nematodes penetrated female

Case report: Halicephalobus gingivalis in a Tennessee pony.

A 17-year-old female grade pony presented to University of Tennessee Veterinary Medical Center in May of 2021 for evaluation of multifocal, firm, sessile, circular lesions of various diameters on the ventrum and flank. The lesions had been present for two weeks at presentation. An excisional biopsy found numerous adult and larval rhabditid nematodes most consistent with Halicephalobus gingivalis. PCR targeting a portion of the large ribosomal subunit confirmed this diagnosis. The patient was treated with a high dose course of ivermectin followed by fenbendazole. The patient began showing neuro

Dimethyl sulfoxide and ascarosides improve the growth and yields of entomopathogenic nematodes in liquid cultures.

Increasing the infective juvenile (IJ) yields of entomopathogenic nematodes in monoxenic culture systems would reduce their production cost for the market. Ascarosides act as universal nematode pheromones with developmental and behavioral effects of nematodes. Dimethyl sulfoxide (DMSO) is unexpectedly found to enhance the IJ yields of entomopathogenic nematodes on fortified nutrient broth plates. In this study, the influence of selected ascarosides (ascr#7, ascr#9 and ascr#11) and DMSO in three concentrations on the IJ yields of S. carpocapsae All and H. bacteriophora H06 in liquid culture fla

Efficacy and residual activity of commercially available entomopathogenic nematode strains for Mediterranean fruit fly control and their ability to infect infested fruits.

BACKGROUND: Entomopathogenic nematodes (EPN) show potential in controlling larvae of the Mediterranean fruit fly (medfly) Ceratitis capitate, but previous studies mainly concern species and strains that are not commercially available. The use of EPN for control of Mediterranean fruit fly is further hampered by the cost of using nematodes. In this study, the efficacy and residual activity of commercial strains of three EPN species, Steinernema carpοcapsae, S. feltiae and Heterοrhabditis bacteriοphοra medfly) C. capitata, in the soil substrate and inside fruits were evaluated. Laboratory and field evaluation of Korean entomopathogenic nematode isolates against the oriental beetle Exomala orientalis (Coleoptera: Scarabaeidae).

The oriental beetle Exomala (Anomala) orientalis (Waterhouse) is an important pest of turfgrass in Korean golf courses, and although a few chemical insecticides are registered for insect pest control, they are not very effective against scarab larvae. There is also a growing concern in Korea about the run-off of insecticides into sensitive habitats and the potential for groundwater contamination. A safe and environmentally sound alternative is needed to conventional insecticides. We therefore evaluated six Korean entomopathogenic nematode isolates: S. carpocapsae Pocheon, S. glaseri Dongrae, S

Apoptosis and change of competence limit the size of the vulva equivalence group in Pristionchus pacificus: a genetic analysis.

BACKGROUND: To understand how alterations in the molecular mechanisms underlying developmental processes generate a diversity of biological forms, comparative developmental biology can be combined with genetic analysis. The formation of the nematode vulva is one tractable system for such evolutionary developmental analysis, as much is understood about its development in Caenorhabditis elegans. In Caenorhabditis, six of twelve ventral epidermal cells form the 'vulva equivalence group'; although all six cells are competent to adopt vulval cell fates in response to an inductive signal, only three

Biological control of sciarid and phorid pests of mushroom with predatory mites from the genus Hypoaspis(Acari: Hypoaspidae) and the entomopathogenic nematode Steinernema feltiae.

In small-scale experiments, the predatory mites, Hypoaspis aculeifer (Canestrini) and H. miles Berlese, applied at 700 mites m(-2), and the entomopathogenic nematode, Steinernema feltiae (Filipjev) applied at 3 x 10(-6) nematodes m(-2) controlled sciarids and phorids in mushroom compost and casing substrates. For both mite species, earliest application to the growing substrate following sciarid infestation reduced sciarid emergence. In contrast, later application of each biological control agent provided more effective control of phorid emergence. The behaviour of adult mites suggested that H.

An eight-year survey of the intestinal parasites of carnivores, hoofed mammals, primates, ratites and reptiles in the Ljubljana zoo in Slovenia.

Problems with parasitic infections and their interspecies transmissions are common in zoological gardens and could pose serious health damage to captive animals. This study presents results of eight-year monitoring of intestinal parasites in animals from Zoo Ljubljana, Slovenia. A total of 741 faecal samples from 40 animal species were collected two to four times per year and examined microscopically. Intestinal parasites were detected in 45% of samples, with detection of helminths (Cestoda, Nematoda - Ascaridida, Enoplida, Strongylida, Oxyurida, Rhabditida and Trichurida) and protists (Apicom

Histologic characterization of eosinophilic encephalitis in horses in Florida.

Eosinophils within the central nervous system are abnormal and are usually associated with fungal or parasitic infections in horses. Causative agents include Halicephalobus gingivalis, Sarcocystis neurona, and Neospora hughesi. Confirmation of these organisms via specific testing is typically not performed, and final diagnoses are often presumptive. With molecular technology, many of these organisms can now be confirmed. This is important for emerging and zoonotic pathogens, including Angiostrongylus cantonensis, an emerging parasite of interest in the southeastern United States. We retrospect
