Cytochrome P450 (P450) from Rhodococcus rhodochrous have been characterized through circular dichroism and nuclear magnetic resonance (NMR) spectroscopy, both in the substrate-free and substrate-bound forms. The data are compared with those of P450cam and indicate a close similarity of the structure of the active site in the two proteins. The substrate-free species contains low-spin iron(III), while the 2-ethoxyphenol bound species contains high-spin iron(III). The substrate is in slow exchange on the NMR time scale. The binding of CN- has been investigated and the final adduct characterized t

Rhodococcus sp. strain B4, isolated from a soil sample contaminated with polycyclic aromatic hydrocarbons, grows with naphthalene as the sole source of carbon and energy. Salicylate and gentisate were identified as intermediates in the catabolism of naphthalene. In contrast to the well-studied catabolic pathway encoded by the NAH7 plasmid of Pseudomonas putida, salicylate does not induce the genes of the naphthalene-degradative pathway in Rhodococcus sp. strain B4. The key enzymes of naphthalene degradation in Rhodococcus sp. strain B4 have unusual cofactor requirements. The 1,2-dihydroxynapht

The microbial accumulation of heavy metals and phosphate is of interest for the bioremediation of polluted waters. In this work, we showed that at cultivation of the bacterium Rhodococcus ruber SiAl in the medium with 2.0 mM Fe³⁺ for stationary growth stage, up to 99% of the iron was associated with the biomass. Magnesium ion accumulation from the medium with 2 mM Mg²⁺ did not exceed 5% of the initial content. The cells did not remove manganese ions from the medium; moreover, the presence of MnSO4 inhibited growth. The cells of Rhodococcus ruber SiAl removed phosphate from the medium: 75, 20,

The wide ecological distribution of actinobacteria suggests that they have developed efficient mechanisms to adapt to extremely nutritionally deficient (oligotrophic) conditions. The impact of nutrient limitation typically observed in oligotrophic areas on bacteria remains to be assessed for many species. The non-model Rhodococcus aetherivorans L13can grow under oligotrophic conditions, even without an added carbon source. Oligotrophic cells of L13 undergo physiological and morphological changes compared to glucose-grown cells, including forming short-fragmenting cells, producing an extracellu

Rhodococcus (R.) equi is a pathogen primarily known for infections in equine foals, but is also present in numerous livestock species including New World camelids. Moreover, R. equi is considered an emerging zoonotic pathogen. In this report, we describe in detail a fatal rhodococcal infection in an alpaca (Vicugna pacos), to our best knowledge, for the first time. The alpaca died due to a septicemic course of an R. equi infection resulting in emaciation and severe lesions including pyogranulomas in the lungs and pericardial effusion. The onset of the infection was presumably caused by aspirat

Isoprene (2-methyl-1,3-butadiene) is a climate-active gas released to the atmosphere in large quantities, comparable to methane in magnitude. Several bacteria have been isolated which can grow on isoprene as a sole carbon and energy source, but very little information is available about the degradation of isoprene by these bacteria at the biochemical level. Isoprene utilization is dependent on a multistep pathway, with the first step being the oxidation of isoprene to epoxy-isoprene. This is catalyzed by a four-component soluble diiron monooxygenase, isoprene monooxygenase (IsoMO). IsoMO is a

Rhodococcus equi is one of the most important causes of disease in foals. Infection is typically characterized by pyogranulomatous pneumonia although extrapulmonary infections occur occasionally. Uveitis and polysynovitis have been reported in foals naturally infected with R. equi and are thought to be the result of an immune-mediated process. However, the pathogenesis of these conditions is poorly understood. The objectives of this study were to document the occurrence of uveitis and polysynovitis after experimental infection with R. equi and to determine if these disorders are the direct res

Some of the noxious atmospheric pollutants such as nitrogen and sulfur dioxides come from the fossil fuel combustion. Biodesulfurization and biodenitrogenation are processes which remove those pollutants through the action of microorganisms. The ability of sulfur and nitrogen removal by the strain Rhodococcus erythropolis ATCC 4277 was tested in a biphasic system containing different heavy gas oil concentrations in a batch reactor. Heavy gas oil is an important fraction of petroleum, because after passing through, the vacuum distillation is incorporated into diesel oil. This strain was able to

Rhodococcus equi is an important pathogen of foals that causes severe pneumonia. To date, there is no licensed vaccine effective against R. equi pneumonia of foals. The objectives of our study were to develop an electron beam (eBeam) inactivated vaccine against R. equi and evaluate its immunogenicity. A dose of eBeam irradiation that inactivated replication of R. equi while maintaining outer cell wall integrity was identified. Enteral administration of eBeam inactivated R. equi increased interferon-γ production by peripheral blood mononuclear cells in response to stimulation with virulent R. e

A Rhodococcus sp. UKMP-5M isolate was shown to detoxify cyanide successfully, suggesting the presence of an intrinsic property in the bacterium which required no prior cyanide exposure for induction of this property. However, in order to promote growth, Rhodococcus sp. UKMP-5M was fully acclimatized to cyanide after 7 successive subcultures in 0.1 mM KCN for 30 days. To further shorten the lag phase and simultaneously increase the tolerance towards higher cyanide concentrations, the bacterium was induced with various nitrile compounds sharing a similar degradatory pathway to cyanide. Two Rhodococcus strains which were isolated from a trichloroethylene (TCE)-degrading bacterial mixture and Rhodococcus rhodochrous ATCC 21197 mineralized vinyl chloride (VC) and TCE. Greater than 99.9% of a 1-mg/liter concentration of VC was degraded by cell suspensions. [1,2-C]VC was degraded by cell suspensions, with the production of greater than 66% CO(2) and 20% C-aqueous phase products and incorporation of 10% of the C into the biomass. Cultures that utilized propane as a substrate were able to mineralize greater than 28% of [1,2-C]TCE to CO(2), with approximately 40% appearing in C-aque

Rhodococcus equi is a gram-positive coccobacillus which, like other members of the order Actinomycetales, is increasingly reported as an opportunistic pathogen in patients with AIDS. The use of combinations of antibiotics that include imipenem (IMP) has been suggested for the treatment of patients infected with R. equi. An antagonism between IMP, meropenem, cefoxitin, ceftriaxone, moxalactam, and oxacillin and other beta-lactams, such as penicillin, amoxicillin, cephalothin, and ticarcillin, was detected in vitro both on Mueller-Hinton agar and in broth for all 10 IMP-susceptible R. equi strai

The catabolism of propane by "Nocardia paraffinicum" (Rhodococcus rhodochrous) has been shown to involve CO(2) fixation after its oxidation to propionic acid. "N. paraffinicum" failed to grow on either propane or 1-propanol in the absence of CO(2). The rate of propane utilization was directly related to the initial CO(2) concentration, and Warburg respirometry suggested that CO(2) was required for the catabolism of 1-propanol, propionaldehyde, and propionate but not for 2-propanol. These data also suggested that the predominant pathway for the utilization of propane by "N. paraffinicum" was th

Extremophilic microorganisms offer an untapped potential for producing unique bioactive metabolites with therapeutic applications. In the current study, bacterial isolates were obtained from samples collected from Chamalang cave located in Kohlu District, Balochistan, Pakistan. The cave-derived isolate C1 (Rhodococcus jialingiae) exhibits prominent antibacterial activity against multidrug-resistant pathogens (MDR), including Escherichia coli, Staphylococcus aureus, and Micrococcus luteus. It also demonstrates substantial antioxidant activity, with 71% and 58.39% DPPH radical scavenging. Optimi

Rhodococcus equi is a well-known intracellular facultative bacterium that is opportunistic in nature, and a contagious disease-causing agent of pyogranulomatous infections in humans and multihost animals. Feline rhodococcosis is an uncommon or unnoticed clinical condition, in which the organism is usually refractory to conventional antimicrobial therapy. The pathogenicity of the agent is intimately associated with plasmid-governed infectivity, which is attributed to the presence of plasmid-encoded virulence-associated proteins (Vap). Three host-adapted virulence plasmid types (VAPs) have been

Cometabolic degradation of TCE by toluene-degrading bacteria has the potential for being a cost-effective bioremediation technology. However, the application of toluene may pose environmental problems. In this study, several plant essential oils and their components were examined as alternative inducer for TCE cometabolic degradation in a toluene-degrading bacterium, Rhodococcus sp. L4. Using the initial TCE concentration of 80 microM, lemon and lemongrass oil-grown cells were capable of 20 +/- 6% and 27 +/- 8% TCE degradation, which were lower than that of toluene-grown cells (57 +/- 5%). Rhodococcus qingshengii IGTS8 is a model strain well-known for its ability to remove sulfur from dibenzothiophene (DBT) and its derivatives via the 4S pathway, a process that catalyzes the conversion of DBT to 2-hydroxybiphenyl (2-HBP) and sulfite without losses in the energy content of the fuel. The desulfurization ability of the wild-type (wt) strain is limited due to repression of the dsz operon in the presence of bioavailable sulfur sources, including methionine and in some cases, sulfates, whereas genetic deletion of the reverse transsulfuration pathway enzymes, Cbs and MetB, alleviates t

BACKGROUND: Polyethylene (PE) is the most widely produced polyolefin and represents a major contributor to plastic waste, largely due to its chemical stability and resistance to biological degradation. PE biodegradation is typically characterized as a series of oxidative metabolic processes rather than mineralization. An urgent priority is the development of integrated experimental setups that bring together physiological, biochemical, and molecular analyses under controlled cultivation conditions to unlock the metabolic traits of PE biodegradation. This study aims to elucidate the molecular m

Quorum sensing (QS) enables bacteria to coordinate collective behaviors, including the production of secondary metabolites with potential biotechnological applications, through the synthesis and detection of small signaling molecules. In Streptomyces, QS is well-described and mainly mediated by 2,3-disubstituted γ-butyrolactones (GBLs), which play key roles in the regulation of secondary metabolism and spore production. Despite their importance, GBL-based QS systems remain poorly characterized in other actinomycetal genera. Here, we investigated the distribution, structure, and function of GBL
