The ice-ice disease black box: a disease complex triggered by bacterial associations in an ex situ milieu of biobanked Kappaphycus alvarezii.

Ice-ice disease (IID) is widely associated with environmental stress and microbial presence in seaweed farms, but its expression under ex situ conditions remains poorly understood. This study examined the associated culturable bacterial community in independently maintained Kappaphycus alvarezii stock cultures classified as healthy or diseased based on characteristic symptoms. Bacteria were isolated from healthy tissue, diseased tissue, and their corresponding bulk waters, identified using 16S rRNA gene sequencing, and analyzed through phylogenetic reconstruction, species richness estimation,

Sustainable superhydrophobic nanocellulose coatings derived from Gelidium amansii waste via DES extraction and one-pot functionalization.

Gelidium amansii, a red alga widely utilized for industrial agar production, generates significant residual biomass that is often discarded, leading to resource waste and environmental pollution. This research investigates the valorization of Gelidium amansii waste through the development of superhydrophobic materials. High-purity cellulose was extracted using a choline chloride-lactic acid deep eutectic solvent (DES), achieving a yield of 61.17% under optimized conditions (solid-liquid ratio 1:10, 100 °C, 4 h). The extracted cellulose was converted into TEMPO-oxidized nanocellulose (TONC) via

Reproductive isolation and differential introgression shape the genomic landscape of the red alga Amansia glomerata in the Hawaiian Archipelago.

Reproductive isolation; Genomic divergence; Introgression; Seaweed, Hawai'i. Speciation in marine environments is inherently complex, and the mechanisms underlying the evolution of reproductive isolation remain a fundamental yet understudied challenge in macroalgae. This study investigates two lineages of the marine red alga Amansia glomerata around O'ahu, Hawai'i, to test the hypothesis that lineage boundaries are maintained by strong reproductive barriers. Using genomic sequencing (ddRAD), fine-scale spatial transects, and demographic modeling, we characterized lineage spatial structure and

Functional characterization and periplasmic localization of a phosphorus starvation-induced alkaline phosphatase involved in phosphate acquisition in Pyropia yezoensis.

Expression of the alkaline phosphatase gene in Pyropia yezoensis is transcriptionally induced under phosphorus deprivation. To elucidate its physiological role in phosphate acquisition, we purified the enzyme to homogeneity from phosphorus-starved thalli and characterized its biochemical properties. The alkaline phosphatase exhibited broad substrate specificity toward glycerophosphate, sugar phosphates, and nucleoside phosphates, and its activity was enhanced by Na+, Ca2+, and Mg2+ at concentrations comparable to those in natural seawater. Genomic analyses, together with biochemical characteri

Native proteins from Galdieria sulphuraria to replace fetal bovine serum in mammalian cell culture.

The use of fetal bovine serum (FBS) in cell culture applications causes high costs and unacceptable animal suffering when FBS is extracted from fetal calves. Despite efforts, the exact composition of FBS still remains partially unresolved. Native proteins in FBS, such as growth factors, and their binding to cell receptors seem to be crucial for cell proliferation and differentiation. Recently, algal extracts with high protein content were considered to reduce the FBS demand. Algae extracts yielded promising results as growth serum in mammalian cell culture. Nevertheless, the dependence on resi

Microbiome-informed study of the mechanistic basis of methane inhibition by Asparagopsis taxiformis in dairy cattle.

UNLABELLED: Copious amounts of methane, a major constituent of greenhouse gases currently driving climate change, are emitted by livestock, and efficient methods that curb such emissions are urgently needed to reduce global warming. When fed to cows, the red seaweed Asparagopsis taxiformis (AT) can reduce enteric methane emissions by up to 80%, but the achieved results can vary widely. Livestock produce methane as a byproduct of methanogenesis, which occurs during the breakdown of feed by microbes in the rumen. The ruminant microbiome is a diverse ecosystem comprising bacteria, protozoa, fungi

In vitro fermentation of Bangia fusco-purpurea polysaccharide by human gut microbiota and the protective effects of the resultant products on Caco-2 cells from lipopolysaccharide-induced injury.

Polysaccharide extracted from red seaweed Bangia fusco-purpurea (BFP) is a novel sulfated galactan, differed from agarans and carrageenans in fine structure. In this study, in vitro fermentation characteristics of BFP by human gut microbiota and its protective effect on lipopolysaccharide (LPS)-induced injury in Caco-2 cells were investigated. Our results showed that BFP was mainly degraded at transverse colon for 18 h fermentation by gut microbiota with reduced molecular weight. Meanwhile, BFP fermentation was associated with increased short-chain fatty acids (SCFAs) as compared to control gr

Unique ultrastructural characteristics of the tegument of the digenean blood fluke Aporocotyle simplex Odhner, 1900 (Digenea: Aporocotylidae), a parasite of flatfishes.

This paper includes the first transmission electron microscopical (TEM) study of the tegument of a member of the basal digenean family Aporocotylidae. Scanning electron microscopical investigations of the fish blood fluke Aporocotyle simplex show that each boss on the lateral body surface bears 12-15 simple, uniform spines which extend from 0.5-2.7 μm above the surface of the boss. TEM observations revealed that these spines reach deep beneath the distal cytoplasm of the tegument for much of their length (9-12 μm) and are surrounded by a complex of diagonal muscles in each boss. This is the fi

An underwater imagery identification guide for shallow, mesophotic and deep-sea benthos in Maldives.

BACKGROUND: During the 2022 Nekton Maldives Mission, we deployed a variety of platforms (snorkelling, remotely-operated vehicles and manned submersibles) to conduct video surveys of the biodiversity and composition of shallow (< 30 m), mesophotic (30-150 m) and deep-sea (> 150 m) benthos found in the Maldives' central and southern atolls. In total, ~ 80 hrs of stereo-video footage were collected during the benthic transect surveys, which were subsequently processed using annotation software in order to evaluate benthic biodiversity and community composition. Here, we present a photographic gui

Unlocking fine-scale temporal dynamics of coral reef health: a multi-kingdom microbial fingerprinting approach across seawater and sediment.

Coral reefs are rapidly degrading worldwide, creating a need for sensitive biomonitoring tools to detect early ecological shifts before visible collapse. Traditional monitoring often misses these incipient changes. We hypothesized that multi-kingdom microbial fingerprinting could reveal fine-scale temporal dynamics and early bioindicators of reef environmental change. The study was conducted on the fringing reef of Réunion Island (south-west Indian Ocean), a system exposed to local anthropogenic pressures. Seawater and sediment samples were collected bimonthly from April to October 2022 at thr

From waste to resource: Production and characterisation of protein-enriched extracts from Gelidium corneum residue.

This study aimed to develop protocols for extracting proteins from Gelidium corneum agar-extraction residue for their potential use in food-related applications. The residue was mostly composed of carbohydrates (41%), forming highly recalcitrant cell wall structures that hindered protein extraction through alkaline solubilisation. The application of high-pressure (350 MPa, 5 min, 20 °C) and ultrasound (10 min) pre-treatments did not induce significant microstructural changes in the biomass, but promoted the release of highly soluble proteins and peptides, increasing the protein extraction yiel

Identification of a novel natural compound that acts on the membrane progestin receptor α (paqr7) from the marine algae Padina.

Previously, we identified water-soluble compounds with membrane progestin receptor α (mPRα)-binding activity from the marine algae Padina arborescens. The compounds inhibited fish oocyte maturation. These compounds are potentially novel antagonists of mPR and are of interest as novel inhibitors of the nongenomic pathway of steroids. In this study, a compound with mPRα binding activity was purified from the methanol extract of Padina tarries. The structure of one of the major compounds in the fraction was identified as 1-carboxybutyl-2-hydroxypentanoate (1-CB 2-HPNA) using nuclear magnetic reso
