Mixed Skin and Soft Tissue Infection Caused by Rhodotorula glutinis and Enterococcus faecalis in a Diabetic Patient: A Case Report.

Rhodotorula is a widely distributed fungus that has evolved as an important pathogen, especially in immunocompromised individuals, causing fungemias, central nervous system infections, peritoneal dialysis-associated peritonitis and keratitis. Even though there are a few reports of skin and soft tissue infections caused by Rhodotorula, all these cases involve immunocompromised individuals. Enterococcus faecalis is a Gram-positive bacterium known to cause several infections such as bloodstream, urinary tract and skin and soft tissue infections. We report a mixed Rhodotorula glutinis and Enteroco

Identification, antifungal susceptibility and scanning electron microscopy of a keratinolytic strain of Rhodotorula mucilaginosa: a primary causative agent of onychomycosis.

Onychomycosis is a dermatological problem of high prevalence that mainly affects the hallux toenail. Onychomycosis caused by the yeast Rhodotorula mucilaginosa was identified using colony morphology, light microscopy, urease and carbohydrate metabolism in a 57-year-old immunocompetent patient from Rio de Janeiro, Brazil. High-resolution scanning electron microscopy of nail fragments, processed by a noncoating method, led to the observation with fine detail of the structures of both nail and fungus involved in the infection. Yeasts were mainly found inside grooves in the nail. Budding yeasts pr

Optimization of the culture medium for an iron-sensitive oleaginous yeast, Rhodotorula toruloides NBRC 0559, through functional iron deficiency.

A complete iron deficiency in iron-sensitive oleaginous yeast showed insufficient biomass, resulting in a lower lipid amount, although lipid accumulation was greater compared to deficiency in other ions. In this study, the effect of functional iron deficiency on lipid production on Rhodotorula toruloides NBRC 0559 was examined. Two supplements, an iron-added (growth) supplement and an iron-free (lipid-producing) supplement were tested for detecting functional iron deficiency. The addition of iron-added supplement increased the biomass by 1.5-fold. Furthermore, the addition of iron-free supplem

Supercritical carbon dioxide extraction of lipids and carotenoids from Rhodotorula toruloides CBS 14 in comparison with conventional extraction methods.

BACKGROUND: Oil from oleaginous yeasts has the potential to replace non-sustainable vegetable oil as raw material to produce food, feed, biofuels, or biochemicals. Co-produced compounds like carotenoids may be helpful to obtain economically viable bioprocesses. Identifying appropriate extraction methods is a bottleneck both for establishing oleaginous yeasts as cell factories for both oil and carotenoids production and for analysis of intracellular compounds like lipids and carotenoids. We conducted extractions using supercritical carbon dioxide (SC-CO2) and conventional solvent methods to ext

Systematic evaluation of the DNA repair machinery facilitated the development of efficient genome editing tools for the red yeast Rhodotorula toruloides.

The red yeast Rhodotorula toruloides is a robust lipid producer, and its biotechnology applications have been expanded through genetic engineering based on the non-homologous end joining (NHEJ) pathway. As the NHEJ pathway leads to random integration, it remains laborious in phenotypic assessment and challenging for rational strain design. To develop more efficient genetic tools for R. toruloides based on the homologous recombination (HR) pathway, we conducted a systematic evaluation of its DNA repair machinery. Overexpression of HR-related proteins, such as ScRad52, RtRad54 and ScSae2, increa

Isolation, purification, and characterization of nitrate reductase from a salt-tolerant Rhodotorula glutinis yeast strain grown in the presence of tungsten.

The salt-tolerant Rhodotorula glutinis yeast strain grew in medium containing nitrate, 1 mM tungsten, and trace amounts of molybdenum (as impurities from the reagents used). Isolation of electrophoretically homogenous preparation of nitrate reductase from the Rh. glutinis cells grown under these growth conditions is described. The isolated nitrate reductase is a molybdenum-containing homodimer with molecular mass of 130 kD, containing 0.177 mol of Mo per mol of the enzyme. The activity of the enzyme is maximal at pH 7.0 and 35-45 degrees C and is inhibited by low concentrations of azide and cy

Characterization of the overall quality, flavor compounds, sensory analysis, and microbial community of huangjiu co-fermentation with Rhodotorula mucilaginosa and Saccharomyces cerevisiae.

Non-Saccharomyces played a crucial role in huangjiu brewing. To explore the influence of aroma-producing Rhodotorula mucilaginosa H04, huangjiu samples fermented with S. cerevisiae (CK) and produced through co-fermentation with R. mucilaginosa and S. cerevisiae (COF) were evaluated. The total organic acid and amino acid content in COF were significantly reduced (P < 0.05). E-tongue revealed that the umami response value of the COF samples was highest and the sourness was lowest (P < 0.05). The esters and alcohols were significantly enhanced in the COF, endowing floral, fruity, and alcoholic ar

Identification of Irpex and Rhodotorula on surveillance bronchoscopy in a pediatric lung transplant recipient: A case report and review of literature of these atypical fungal organisms.

BACKGROUND: Invasive fungal disease (IFD) is a frequent complication in pediatric lung transplant recipients, occurring in up to 12% of patients in the first year. Risk factors for infection include impaired lung defenses and intense immunosuppressive regimens. While most IFD occurs from Aspergillus, other fungal conidia are continuously inhaled, and infections with fungi on a spectrum of human pathogenicity can occur. CASE REPORT: We report a case of a 17-year-old lung transplant recipient in whom Irpex lacteus and Rhodotorula species were identified during surveillance bronchoscopy. She was

Application of adaptive laboratory evolution to improve the tolerance of Rhodotorula strain to methanol in crude glycerol and development of an effective method for cell lysis.

Rhodotorula toruloides can utilize crude glycerol as the low-cost carbon source for lipid production, but its growth is subjected to inhibition by methanol in crude glycerol. Here, transcriptome profiling demonstrated that 1004 genes were significantly regulated in the strain R. toruloides TO2 under methanol stress. Methanol impaired the function of membrane transport and subsequently weakened the utilization of glycerol, activities of the primary metabolism and functions of nucleus and ribosome. Afterwards the tolerance of TO2 to methanol was improved by using two-round adaptive laboratory ev

An Evolved Strain of the Oleaginous Yeast Rhodotorula toruloides, Multi-Tolerant to the Major Inhibitors Present in Lignocellulosic Hydrolysates, Exhibits an Altered Cell Envelope.

The presence of toxic compounds in lignocellulosic hydrolysates (LCH) is among the main barriers affecting the efficiency of lignocellulose-based fermentation processes, in particular, to produce biofuels, hindering the production of intracellular lipids by oleaginous yeasts. These microbial oils are promising sustainable alternatives to vegetable oils for biodiesel production. In this study, we explored adaptive laboratory evolution (ALE), under methanol- and high glycerol concentration-induced selective pressures, to improve the robustness of a Rhodotorula toruloides strain, previously selec

Long-chain fatty acid esters produced by Sporidiobolus pararoseus or Rhodotorula mucilaginosa enhance the fat flavor of soy sauce fermented by defatted soybeans.

Two unique oleaginous strains, Sporidiobolus pararoseus and Rhodotorula mucilaginosa, were identified during soy sauce fermentation. Both strains demonstrated robust growth under extreme conditions (15 % NaCl, pH 5, 3 % ethanol). Lipidomics revealed 689 and 678 lipid species in S. pararoseus and R. mucilaginosa, with 671 shared components. Among the common components, Triglyceride (TG, 180 species), Diglyceride (DG, 72 species), and Ceramides (Cer, 52 species) were significantly enriched, directly participating in the biosynthesis of characteristic aroma substances by releasing fatty acids wit

Intraluminal gas in deep vein thrombosis associated with Rhodotorula mucilaginosa fungemia and Prevotella intermedia bacteremia: a unique clinical case report and literature overview.

Intravenous drug use (IVDU) is associated with multiple complications, including thrombus and infection with rare pathogens. Here, we present a unique case of septic thrombophlebitis with gas present within the lumen of the thrombus that was identified from an otherwise unassuming wound via point-of-care ultrasound (POCUS). In conjunction with past literature, this finding raises the possibility of POCUS detection of intraluminal gas as a sensitive indicator of septic thrombophlebitis. Furthermore, the patient's polymicrobial infection with Rhodotorula mucilaginosa and Prevotella intermedia ex

Hydrolysis of Gluten-Derived Celiac Disease-Triggering Peptides across a Broad pH Range by RmuAP1: A Novel Aspartic Peptidase Isolated from Rhodotorula mucilaginosa.

An aspartate peptidase with proteolytic activity toward gluten was identified from an isolated red yeast Rhodotorula mucilaginosa strain. This peptidase consists of 425 amino acids, comprising an N-terminal signal peptide, a propeptide, and a C-terminal catalytic domain. The catalytic domain, termed RmuAP1CD, could be secreted by the recombinant oleaginous yeast Yarrowia lipolytica, whose genome contains the expression cassette for RmuAP1CD. RmuAP1CD exhibited optimum activity at pH 2.5 when acting on bovine serum albumin. Moreover, it facilitated the hydrolysis of gluten-derived immunogenic p
