The severe phenotype of human females whose karyotype includes tiny ring X chromosomes has been attributed to the inability of the small ring X chromosome to inactivate. The XIST locus is expressed only from the inactive X chromosome, resides at the putative X inactivation center, and is considered a prime player in the initiation of mammalian X dosage compensation. Using PCR, Southern blot analysis, and in situ hybridization, we have looked for the presence of the XIST locus in tiny ring X chromosomes from eight females who have multiple congenital malformations and severe mental retardation.

Among 61 human pleomorphic adenomas subjected to chromosomal study by banding methods, two rare cases were detected, one having a dicentric marker chromosome and the other a ring chromosome included in the stemline karyotype. Excepting meningiomas, such marker types have previously only been observed in different types of malignant neoplasms. However, as in meningiomas, the occurrence of dicentrics or rings has no implications as regards the question of malignancy. Both our mixed tumours showed structural changes affecting one chromosome No. 8. These findings, along with similar observations i

We report here a case of myelodysplastic syndrome (MDS) that developed in a 60-year-old female with dehydrated hereditary stomatocytosis 1 (DHS1) with PIEZO1 gene mutation (p. E2496ELE). The patient had a non-transfused status until the age of 60, when her anemia progressed. Bone marrow examination revealed multilineage MDS (no increase in blasts) with a non-complex karyotype showing two types of ring chromosomes (RCs) confirmed by the G-banding method. A targeted next-generation sequencing (NGS) assay revealed Tet methylcytosine dioxygenase 2 (TET2) gene mutations (p. Y592fs and p. R1261C), a

Two ring chromosomes 13 were studied by means of various cytogenetic techniques in two psychomotorically retarded male infants. Common features of our patients include microcephalia, hypertelorism, wide and prominent nose bridge, and cryptorchism. Various configurations of the aberrant chromosomes could be identified in cultured skin fibroblasts and peripheral lymphocytes from both patients. Chromosome heteromorphisms and analysis of silver stained nucleolus organizer regions (Ag-NOR) substantiated the parental origin of the ring chromosomes. The more severely affected patient showed a break p

UNLABELLED: <p>Introduction: Ring chromosomes (RCs) are acquired circular structural abnormalities associated with gain or loss of genetic material, which are thought to be associated with inferior outcomes in patients with myeloid neoplasms (MNs). Responses of patients with MN and RC to the standard therapeutic options have not been previously reported.
METHODS: We analyzed the demographics and outcomes of 31 consecutive patients with an MN and RC, comparing overall survival (OS) and progression-free survival (PFS) of patients who received supportive care (n = 9), cytotoxic chemotherapeutic

Background/Objectives: Ring chromosomes (RCs) can be rare or common depending on the chromosome involved. With interest in the historical RCs identified by our laboratory, we curated instances to provide further information to this research field. Methodology: We carried out a retrospective, single-center study of constitutional RCs identified starting in the late 1980s. Details for 40 RCs with a modal number of 46 chromosomes are featured here. Results: Mosaic and non-mosaic RCs are identified, with a preponderance of pediatric-aged females at first ascertainment. We corroborated an enrichmen

Human ring chromosomes are often unstable during mitosis, and daughter cells can be partially or completely aneuploid. We studied the mitotic stability of four ring chromosomes, 8, 13, 18, and 22, in long-term cultures of skin fibroblasts and induced pluripotent stem cells (iPSCs) by GTG karyotyping and aCGH. Ring chromosome loss and secondary aberrations were observed in all fibroblast cultures except for r(18). We found monosomy, fragmentation, and translocation of indexed chromosomes. In iPSCs, aCGH revealed striking differences in mitotic stability both between iPSC lines with different ri

We report the case of an 84 years old prostate cancer patient with severe side effects after radiotherapy in 2006. He was cytogenetically analysed in 2009 and in 2012 in a comparative study for individual radiosensitivity of prostate cancer patients. No other patient had clonal aberrations, but this patient showed ring chromosomes in the range of 21-25% of lymphocytes. He received 5 cycles of 5-fluorouracil/folic acid for chemotherapy of sigmoid colon carcinoma in 2003, three years before radiotherapy of prostate cancer. Blood samples were irradiated ex vivo with Cs-137 γ-rays (0.7Gy/min) in t

We describe a 77-year-old woman who had with what appears to be the first case of plasma cell leukemia (PCL) with a ring chromosome of variable size. Of the three clones evident, two of them were abnormal: a 41,X,-X hypodiploid clone with rings of two different sizes, and a 81-82,XX hypotetraploid clone with a double-sized dicentric ring or variable double rings. Additional structural and numerical abnormalities included a 14q+ marker, deletions of two chromosomes 1 and monosomies 8 and 13, all previously reported in PCLs. Ring chromosomes have been associated with a poor prognosis, especially

Ring chromosome 6 is a rare structural chromosomal anomaly with a variable phenotype that includes growth restriction, congenital malformations, and neurological impairment. We present a term newborn female with a prenatal history of fetal growth restriction (FGR) and central nervous system malformations. At birth, she exhibited dysmorphic facies, bilateral corneal opacity, hypotonia, and a congenital heart defect. Cytogenetic analysis revealed a karyotype of 46,XX,r(6)(p25q27)[27]. The infant died on day 18 days of life due to a respiratory complication. Conclusions: Ring chromosome 6 is asso

ABSTRACT: In brief: Extensive remodelling of the fat-tailed dunnart endometrium accompanies gestation, highlighting the importance of endometrial glandular contributions to the marsupial mode of reproduction. Abstract: Eutherian mammal embryos typically implant at the blastocyst stage of development. In contrast, marsupial pregnancy is characterised by a prolonged pre-implantation period and an advanced stage of embryonic development before implantation and placentation. During this early extended phase of pre-implantation growth and development, nutrition is provided predominantly by secretio

Ring chromosomes, which are formed through the fusion of the telomeric ends of a chromosome, present with a spectrum of symptoms whose severity depends on the amount of genetic material lost. Ring chromosome 13 cases present with symptoms similar to that of deletion 13q syndrome, and can be classified depending on whether several critical regions are involved in the deletion. An important region to consider is locus 13q32, whose deletion is known to cause severe phenotypes and major malformations. In contrast, deletions of the more distal locus 13q34 have been shown to be involved in symptoms

Chromosomal anomalies occur in 0.4% of live births. Ring chromosomes have been found for all human chromosomes and when it is replacing a normal chromosome, it results as a partial monosomy. The phenotype often overlaps that seen in comparable deletion syndromes of the same chromosomes. In the present report the authors describe the clinical manifestations of a girl with ring chromosome 18 (46XX,r18) including dysmorphic features, failure to thrive, global delay of development, hypothyroidism, atopic dermatitis, bilateral chronic otitis media, aortic regurgitation with patent foramen ovale and

We report on a woman with Muellerian aplasia, renal and skeletal anomalies, and minor dysmorphic signs. Conventional cytogenetic analysis revealed mosaicism for a small supernumerary, undefinable ring chromosome. Chromosome microdissection and reverse painting demonstrated that this marker contained pericentric material from chromosome 8 (8p12q12). Thus, we identified a Muellerian aplasia phenotype with partial trisomy 8 mosaicism.

INTRODUCTION: Ring chromosome 20 syndrome (C20A) is characterised by mental retardation, behavioural disorders, dysmorphias and refractory epilepsy with polymorphic seizures. It should therefore be treated with broad-spectrum antiepileptic drugs (AEDs), such as valproate (VPA) and topiramate (TPM). The relatively frequent hypersensitivity reactions to aromatic AEDs, not to VPA, the hyperammonemic encephalopathy (HAE) caused by the combination of VPA and TPM and the chromosome disorder described, affecting the same patient, do not appear in the literature we reviewed.
