Effects of Pinacidil and Calcium on Succinate-Energized Rat Heart Mitochondria in the Presence of Rotenone.

The effect of pinacidil was studied on calcium-loaded rat heart mitochondria (RHM) in the presence of succinate and rotenone. In experiments with pinacidil, the swelling of these mitochondria increased in media with NH4NO3 or K-acetate, but the inner membrane potential (ΔΨmito) and the respiration in 3 or 2,4-dinitrophenol-stimulated states of these organelles decreased due to the opening of the mitochondrial permeability transition pore (MPTP) in their inner membrane. These effects were inhibited by cyclosporin A and ADP. It was concluded that the protective effect of pinacidil in the cardiac

Haplogroup J mitogenomes are the most sensitive to the pesticide rotenone: Relevance for human diseases.

There is growing evidence that the sequence variation of mitochondrial DNA (mtDNA), which clusters in population- and/or geographic-specific haplogroups, may result in functional effects that, in turn, become relevant in disease predisposition or protection, interaction with environmental factors and ultimately in modulating longevity. To unravel functional differences between mtDNA haplogroups we here employed transmitochondrial cytoplasmic hybrid cells (cybrids) grown in galactose medium, a culture condition that forces oxidative phosphorylation, and in the presence of rotenone, the classic

Investigation of tyrosine hydroxylase and BDNF in a low-dose rotenone model of Parkinson's disease.

Tyrosine hydroxylase (TH, the rate limiting-enzyme in catecholamine synthesis) is regulated acutely via phosphorylation of 3 serine residues--Ser19, 31 and 40, and chronically via changes in TH protein levels. In this study, we aimed to investigate how TH is regulated in the brain, gut and adrenal gland as well as changes in mature brain-derived neurotrophic factor (mBDNF) and proBDNF levels in a low-dose (2 mg/kg, 5 days/week for 4 weeks) rotenone model of Parkinson's disease (PD). Rearing behaviour decreased by week 3 in the rotenone group (p<0.01), with further decreases in rearing by week

Susceptibility to rotenone is increased in neurons from parkin null mice and is reduced by minocycline.

Parkinson's disease is a neurodegenerative disorder which is in most cases of unknown etiology. Mutations of the Park-2 gene are the most frequent cause of familial parkinsonism and parkin knockout (PK-KO) mice have abnormalities that resemble the clinical syndrome. We investigated the interaction of genetic and environmental factors, treating midbrain neuronal cultures from PK-KO and wild-type (WT) mice with rotenone (ROT). ROT (0.025-0.1 microm) produced a dose-dependent selective reduction of tyrosine hydroxylase-immunoreactive cells and of other neurons, as shown by the immunoreactivity to

Differential effects of ginsenosides on Ca2+ regulation in rotenone-treated neuronal and microglial cells.

Parkinson's disease is one of the most common neurodegenerative disorders, and the pesticide rotenone is widely used to model Parkinson's disease in experimental studies. Rotenone-induced mitochondrial dysfunction is associated with oxidative stress and intracellular Ca2+ ([Ca2+]ᵢ) dysregulation in neuronal systems. L-type Ca2+ channels (LTCCs) contribute to Ca² ⁺ influx under oxidative stress conditions, but pharmacological inhibition of LTCCs is limited by cardiovascular side effects. Phospholipase D (PLD) has also been implicated in cellular stress-associated signaling pathways. Ginsenoside

Dysregulation of Metabolic Peptides Precedes Hyperinsulinemia and Inflammation Following Exposure to Rotenone in Rats.

Rotenone, a naturally occurring compound derived from the roots of tropical plants, is used as a broad-spectrum insecticide, piscicide, and pesticide. It is a classical, high-affinity mitochondrial complex I inhibitor that causes not only oxidative stress, α-synuclein phosphorylation, DJ-1 (Parkinson's disease protein 7) modifications, and inhibition of the ubiquitin-proteasome system but it is also widely considered an environmental contributor to Parkinson's disease (PD). While prodromal symptoms, such as loss of smell, constipation, sleep disorder, anxiety/depression, and the loss of dopami

Effects of Enteric Environmental Modification by Coffee Components on Neurodegeneration in Rotenone-Treated Mice.

Epidemiological studies have shown that coffee consumption decreases the risk of Parkinson's disease (PD). Caffeic acid (CA) and chlorogenic acid (CGA) are coffee components that have antioxidative properties. Rotenone, a mitochondrial complex I inhibitor, has been used to develop parkinsonian models, because the toxin induces PD-like pathology. Here, we examined the neuroprotective effects of CA and CGA against the rotenone-induced degeneration of central dopaminergic and peripheral enteric neurons. Male C57BL/6J mice were chronically administered rotenone (2.5 mg/kg/day), subcutaneously for

Rotenone induces gastrointestinal pathology and microbiota alterations in a rat model of Parkinson's disease.

While people are often aware of the motor symptoms in Parkinson's disease (PD), few know of the many non-motor symptoms, which patients report have a greater impact on their quality of life. Gastrointestinal (GI) dysfunction is one of the most common non-motor symptoms, which can occur at any stage of PD, even years prior to diagnosis, and can affect all sections along the GI tract causing a range of symptoms including drooling, gastroparesis and constipation. We have investigated whether a neurotoxin model of PD induced by rotenone, a mitochondrial complex I inhibitor, is capable of reproduci

Preparation of rotenone derivatives and in vitro analysis of their antimalarial, antileishmanial and selective cytotoxic activities.

Six derivatives of the known biopesticide rotenone were prepared by several chemical transformations. Rotenone and its derivatives showed differential in vitro antiparasitic activity and selective cytotoxicity. In general, compounds were more active against Plasmodium falciparum than Leishmania panamensis. Rotenone had an EC50 of 19.0 µM against P. falciparum, and 127.2 µM against L. panamensis. Although chemical transformation does not improve its biological profile against P. falciparum, three of its derivatives showed a significant level of action within an adequate range of activity with E

Empagliflozin Halts NLRP3 Inflammasome-Mediated Neurodegeneration in Parkinson's Disease in a Rotenone Rat Model.

The NLRP3 inflammasome is a crucial booster of pyroptosis and neuroinflammation in Parkinson's disease (PD). Although empagliflozin, a sodium-glucose cotransporter-2 (SGLT2) inhibitor, has shown neuroprotective ability, its impact on NLRP3 inflammasome stimulation in PD remains undefined. This study examined the neuroprotective properties of empagliflozin in the PD rat model induced by rotenone, with an emphasis on its modulation of NLRP3 inflammasome-mediated pyroptosis. PD was induced via daily subcutaneous administration of rotenone for 14 consecutive days. During this period, animals recei

Pharmacological modulation of p75 neurotrophin receptor in microglial cells improves resilience to rotenone cytotoxicity.

INTRODUCTION: Parkinson's disease (PD) is a progressive neurodegenerative disorder, characterized by dopaminergic neuronal loss, mitochondrial dysfunction, oxidative stress, and chronic neuroinflammation. Microglial activation plays a primary role in disease progression, by amplifying inflammatory response and redox imbalance. The low-affinity neurotrophin receptor p75NTR has recently emerged as a potential therapeutic target in neurodegenerative disorders, due to its involvement in cell survival, apoptosis, and inflammatory signaling. In the present study, we investigated whether pharmacologi

Loganin Attenuates Rotenone-Induced Parkinsonism-like Features in Rats Through Multi-Target Neuroprotective Mechanisms.

Background/Objectives: Rotenone (RT)-induced neurotoxicity is widely used to model Parkinsonism-like nigrostriatal injury and recapitulates several PD-relevant pathological features, including oxidative stress, mitochondrial dysfunction, neuroinflammation, and dopaminergic neurochemical disturbance. Loganin (LG), an iridoid glycoside isolated from Cornus officinalis, has been reported to possess antioxidant, anti-inflammatory, anti-apoptotic, and neuroprotective properties. However, its protective effects in a unilateral stereotaxic RT lesion model have not been fully elucidated. This study ai

Nicotinamide counteracts Rotenone-induced mitochondrial and neuronal dysfunction in a translational early-life model.

Mitochondrial function is essential for key neurodevelopmental processes, including cellular proliferation, differentiation, migration, synaptogenesis, and synaptic pruning. Previous research from our group demonstrated that neonatal administration of Rotenone (Rot), a mitochondrial complex I inhibitor, led to mitochondrial dysfunction and schizophrenia-like behavioral phenotypes. In this study, we aimed to identify possible cellular pathways disrupted by Rot exposure that may underlie these behavioral alterations. Thus, primary cortical neurons were treated with 1.325 nM Rot for 24 h, a conce

GM1-Oligosaccharide Rescues Rotenone-Impaired Neuronal Polarization Through RhoA/ROCK Modulation and Mitochondrial Protection.

Neuronal polarization is a fundamental process in the formation of functional neural circuits, relying on the precise coordination between cytoskeletal regulatory signals and mechanisms that sustain cellular integrity. Disruption of these processes compromises neuronal differentiation and survival, and various neurotoxic compounds, including certain pesticides, have been associated with such dysfunctions. In this context, identifying molecules that counteract these detrimental effects is of significant therapeutic interest. Neuronal polarization is essential for the establishment of functional

The phenyl carbamates JBPOS0101 and JBPOS0607 attenuate rotenone-induced dopaminergic neuronal death in mice.

Parkinson's disease (PD) is a progressive neurodegenerative disease that causes motor abnormalities such as tremors, rigidity, and posture imbalance. The loss of dopaminergic neurons and Lewy bodies containing the aggregation of α-synuclein are the main neuropathological features of PD. Rotenone, a widely used pesticide and mitochondrial complex I inhibitor, induces dopaminergic neuronal death and is used to model PD in rodents. We previously reported that JBPOS0101, a phenyl carbamate compound, attenuates amyloid β accumulation and memory impairment in the 5xFAD mouse model of Alzheimer's dis

Mechanism analysis of rotenone toxic exposure inducing neurotoxicity through the MAPK/MMPs signaling pathway.

Rotenone can enter the animal body directly through the skin and stomach, leading to neurotoxic effects. In this study, a mouse model of rotenone exposure was established to investigate the pathological changes and underlying mechanisms of rotenone-induced neurotoxicity. Behavioral tests, molecular biology techniques, gut microbiota analysis, and short-chain fatty acid (SCFA) content measurements were employed. The results demonstrated that rotenone exposure significantly reduced body weight, impaired motor coordination, and resulted in the loss of dopaminergic neurons (TH+) in the substantia

Urolithin improves α-synuclein aggregation and DNMT1 expression in rotenone model of Parkinson's disease.

α-synuclein aggregation is a key hallmark of Parkinson's disease (PD), dementia with Lewy bodies (DLB), and multiple system atrophy (MSA). We examined the multi-targeting effects of urolithin (UA, UB, UC, UD, UE, UM5, and UM6) against α-synuclein aggregation using an in-silico and in-vitro approach. For in-silico analysis, several potential targets were selected like 1XQ8 (α-synuclein monomer), 1H1D (catechol-o-methyltransferase), 2BK3 (monoamine oxidase-B), 3IAM (NADH dehydrogenase), 4I5I (Sirtuin-1), and 5WVO [DNA methyltransferase-1], which play key role in α-synuclein aggregation, levodopa

ECHS1-NOX4 interaction suppresses rotenone-induced dopaminergic neurotoxicity through inhibition of mitochondrial ROS production.

BACKGROUND: Parkinson's disease (PD) is the most common neurodegenerative movement disorder with uncleared mechanisms. Short-chain enoyl-CoA hydratase 1 (ECHS1) is a mitochondrial enzyme critical for the β-oxidation of fatty acids and ATP production. This study aims to explore the roles of ECHS1 in PD by using rotenone-induced experimental PD models. METHODS: To evaluate the role of ECHS1 in rotenone-induced dopaminergic neurodegeneration, adeno-associated virus (AAV)-ECHS1 was stereotactically injected into the substantia nigra region of mice to overexpress ECHS1. Rotenone Induces a Neuropathological Phenotype in Cholinergic-like Neurons Resembling Parkinson's Disease Dementia (PDD).

Parkinson's disease with dementia (PDD) is a neurological disorder that clinically and neuropathologically overlaps with Parkinson's disease (PD) and Alzheimer's disease (AD). Although it is assumed that alpha-synuclein ( α -Syn), amyloid beta (A β ), and the protein Tau might synergistically induce cholinergic neuronal degeneration, presently the pathological mechanism of PDD remains unclear. Therefore, it is essential to delve into the cellular and molecular aspects of this neurological entity to identify potential targets for prevention and treatment strategies.
