Transcriptional Reinforcement of the HER2-RAS Network in HER2-Positive Gastric Cancer: The RUNX-SOS1 Axis in Context.

HER2-positive gastric cancer represents a distinct molecular subtype characterized by chromosomal instability, variable HER2 amplification, and substantial intratumoral heterogeneity. Although HER2-targeted therapies have improved clinical outcomes, therapeutic resistance commonly develops and limits long-term benefit. Established resistance mechanisms include bypass signaling through alternative receptor tyrosine kinases, reactivation of downstream pathways such as PI3K-AKT and MAPK, and dynamic changes in HER2 expression during treatment. These findings underscore the complexity of signaling

Atrioventricular canal defect in patients with RASopathies.

Congenital heart defects affect 60-85% of patients with RASopathies. We analysed the clinical and molecular characteristics of atrioventricular canal defect in patients with mutations affecting genes coding for proteins with role in the RAS/MAPK pathway. Between 2002 and 2011, 101 patients with cardiac defect and a molecularly confirmed RASopathy were collected. Congenital heart defects within the spectrum of complete or partial (including cleft mitral valve) atrioventricular canal defect were diagnosed in 8/101 (8%) patients, including seven with a PTPN11 gene mutation, and one single subject

Protein persulfidation in plants: a central regulator of multiple signaling pathways.

Protein persulfidation, a redox-based post-translational modification mediated by hydrogen sulfide (H2S) on cysteine thiols, plays a central regulatory role in plant growth, development, and stress adaptation. Elucidating its biological functions and molecular mechanisms depends on accurate detection, identification, and quantification. Over the past decade, various optimized methods for persulfidation detection have been developed, which can be categorized into modified biotin switch assays, mass spectrometry-based high-throughput analysis, antibody-dependent western blotting, electrophilic t

Solid tumours in RASopathies: insights from a large monocentric cohort and systematic review of the literature.

BACKGROUND: Dysregulation of the RAS-mitogen-activated protein kinase signalling pathway underlies RASopathies, a family of neurodevelopmental disorders associated with variable cancer predisposition. However, the prevalence and spectrum of solid tumours and the contribution of specific variants to tumour susceptibility remain poorly defined. METHODS: We assessed solid tumour prevalence and spectrum in the largest single-centre cohort of individuals with RASopathies (n=138), excluding neurofibromatosis type 1 and integrated these findings with a systematic literature review to evaluate tumour

High frequency of hotspot mutation in PTPN11 gene among Moroccan patients with Noonan syndrome.

Noonan syndrome (NS; OMIM 163950) is an autosomal dominant RASopathy with variable clinical expression and genetic heterogeneity. Clinical manifestations include characteristic facial features, short stature, and cardiac anomalies. Variants in protein-tyrosine phosphatase, non-receptor-type 11 (PTPN11), encoding SHP-2, account for about half of NS patients, SOS1 in approximately 13%, RAF1 in 10%, and RIT1 each in 9%. Other genes have been reported to cause NS in less than 5% of cases including SHOC2, RASA2, LZTR1, SPRED2, SOS2, CBL, KRAS, NRAS, MRAS, PRAS, BRAF, PPP1CB, A2ML1, MAP2K1, and CDC4

Targeted therapeutic strategies for KRAS-G12D-mutant non-small cell lung cancer: a narrative review.

BACKGROUND AND OBJECTIVE: Kirsten rat sarcoma viral oncogene homolog (KRAS)-G12D is a common oncogenic mutation in non-small cell lung cancer (NSCLC) and has posed significant challenges in the development of treatment approaches that specifically target this alteration. KRAS-G12D differs from KRAS-G12C in its biochemical and signal transduction features, which limits the use of drugs targeting the KRAS-G12C mutation, creating substantial demands in clinical contexts. This review sought to summarize the major developments in treatment for KRAS-G12D-mutant NSCLC, with a focus on the recent key

Variants of SOS2 are a rare cause of Noonan syndrome with particular predisposition for lymphatic complications.

RASopathies are caused by variants in genes encoding components or modulators of the RAS/MAPK signaling pathway. Noonan syndrome is the most common entity among this group of disorders and is characterized by heart defects, short stature, variable developmental delay, and typical facial features. Heterozygous variants in SOS2, encoding a guanine nucleotide exchange factor for RAS, have recently been identified in patients with Noonan syndrome. The number of published cases with SOS2-related Noonan syndrome is still limited and little is known about genotype-phenotype correlations. We collected

Variable phenotypic expression in a large Noonan syndrome family segregating a novel SOS1 mutation.

Noonan syndrome (NS) is an autosomal dominant multisystem condition with a variable phenotype. The most characteristic features are short stature, congenital heart defects, and recognizable facial features. Mutations in SOS1 are found in 10-20% of patients with NS. Different genotype-phenotype studies mention correlations between SOS1 mutations and some features, such as ectodermal abnormalities and specific facial features. We present a large NS family with a novel pathogenic mutation; SOS1 c.3134C>G, p.Pro1045Arg. Ten family members with NS are included with genetically confirmed mutation an

Noonan syndrome spectrum disorders in real life: patient characteristics and response to growth hormone therapy in a genetically defined single-country multicenter cohort.

The variety of genes associated with Noonan syndrome spectrum disorders (NSSD) is expanding, and real-life experience with its management is increasing; however, phenotypic differences among genotypes remain poorly defined. We aimed to assess clinical characteristics and response to growth hormone (GH) therapy in a genetically confirmed, single-country multicentre NSSD cohort. We included 101 patients with NSSD (56 males) from six centres: 76 with (likely) pathogenic PTPN11 variants, 7 with SOS1 variants, and 18 with other gene variations. All completed at least one year of GH therapy; 23 reac

Evidence for aberrant regulation of the p21Ras pathway in PBMCs of patients with chronic idiopathic urticaria.

BACKGROUND: Recent data suggest that a subpopulation of patients with chronic urticaria has an autoimmune disorder. Aberrant expression and regulation of the p21Ras pathway has been reported in lymphoid cells in a variety of systemic autoimmune diseases but not in chronic idiopathic urticaria (CIU). OBJECTIVES: The aim of this study was to examine the expression, regulation, and function of the p21Ras pathway in patients with CIU. METHODS: Twenty-four patients with CIU and 14 control subjects were enrolled. All patients and 9 control subjects were intradermally injected with autologous serum

Transcriptional hallmarks of Noonan syndrome and Noonan-like syndrome with loose anagen hair.

Noonan syndrome (NS) is among the most common nonchromosomal disorders affecting development and growth. NS is genetically heterogeneous, being caused by germline mutations affecting various genes implicated in the RAS signaling network. This network transduces extracellular signals into intracellular biochemical and transcriptional responses controlling cell proliferation, differentiation, metabolism, and senescence. To explore the transcriptional consequences of NS-causing mutations, we performed global mRNA expression profiling on peripheral blood mononuclear cells obtained from 23 NS patie

An Allometric Approach to Fine-Mapping Subcortical Volume Alterations in RASopathies.

Structural neuroimaging in youth with RASopathies has shown alterations in total brain volume (TBV) and subcortical structures. However, whether these subcortical differences scale proportionately with TBV or follow nonlinear allometric relationships remains unclear, limiting accurate interpretation of neuroanatomical findings and their relevance to cognition. Using an allometric framework, this study aimed to determine whether (1) subcortical volumes exhibit deviations from normative scaling in RASopathies and (2) methods used to account for TBV influence the detection of subcortical volume a

Noonan Syndrome Growth Charts and Genotypes: 15-Year Longitudinal Single-Centre Study.

INTRODUCTION: Growth patterns in Noonan syndrome (NS) remain relatively unknown. The objective of this study was to provide growth reference curves for patients with NS and identify correlations between their growth, genotype, and clinical features. METHODS: This was a 15-year-long, monocentric, observational, retrospective, non-interventional study. Children with NS followed up between 2005 and 2022 at "Bambino Gesù" Children's Hospital, Italy, were included and excluded if they had received growth hormone treatment. Comparison of growth curves of participants with NS versus the general Ital

Role of primary protectors of plant cells in salinity tolerance: molecular mechanisms and adaptive strategies.

Salinity stress is among the most pervasive abiotic factors limiting plant growth and agricultural productivity worldwide, currently affecting more than 20% of irrigated croplands. Elevated salt concentrations induce osmotic imbalance, ionic toxicity, and oxidative stress, leading to disruptions in photosynthesis, metabolism, and plant development. To counteract these deleterious effects, plants have evolved intricate, highly coordinated defense systems comprising structural and functional cellular protectors. At the structural level, the cell wall, plasma membrane, vacuole, and peroxisome pla

Acute lymphoblastic leukemia in the context of RASopathies.

Noonan syndrome is associated with a range of malignancies including acute lymphoblastic leukemia (ALL). However, little information is available regarding the frequency, natural history, characteristics and prognosis of ALL in Noonan syndrome or RASopathies in general. Cross-referencing data from a large prospective cohort of 1176 patients having a molecularly confirmed RASopathy with data from the French childhood cancer registry allowed us to identify ALL in 6 (0.5%) patients including 4/778 (0.5%) with a germline PTPN11 mutation and 2/94 (2.1%) with a germline SOS1 mutation. None of the pa

Hereditary gingival hyperplasia associated with amelogenesis imperfecta: a case report.

Hereditary gingival fibromatosis (HGF) and amelogenesis imperfecta (AI) are two rare oral conditions with genetic etiologies. The case of a 17-year-old boy affected by HGF, AI, anterior open bite, and pyramidal impaction of the maxillary molars is reported. Internal bevel gingivectomies were carried out to reduce gingival overgrowth. Clinical examination of the family revealed the presence of HGF and AI in his 12-year-old sister (both in milder forms) and of HGF in his older half brother. Genetic sequencing analyses were performed to detect any of the known mutations leading to HGF and AI. His

Hereditary gingival fibromatosis: a three-generation case and pathogenic mechanism research on progress of the disease.

BACKGROUND: Hereditary gingival fibromatosis (HGF) is a rare benign disorder characterized by progressive overgrowth of gingiva. Although the clinical and histopathologic characteristics of HGF are explicit, the pathogenic mechanism remains unclear. The goal of this article is to describe a three-generation HGF case and discuss the diagnosis, treatment, and inheritance of the disease. The known cellular and molecular features of HGF are also emphasized. METHODS: Family and medical histories of the patients were recorded, and a series of preliminary examinations, including clinical, histologic

RASGRP4 is a key factor in the KRAS activation mediated by SOS in tumor Y1 adrenocortical cell lines.

The Y1 mouse adrenocortical carcinoma cell line presents amplification of the KRas oncogene and high-basal levels of KRAS-GTP mediated by the GEF SOS. In this research, we developed a dynamic model based on ordinary differential equations of the KRAS-GTP activation mediated by SOS in Y1 cells, which showed that SOS alone is not sufficient to reach the high-basal levels of KRAS-GTP experimentally observed for this cell line. Interestingly, a modification in this system, which added another GEF in the model, made the model reach the expected levels of KRAS activation, leading to the hypothesis t

SOS1: a new player in the Noonan-like/multiple giant cell lesion syndrome.

Noonan-like/multiple giant cell lesion syndrome is a rare condition with phenotypic overlap with Noonan syndrome (NS) and cherubism. PTPN11 gene mutations were described in several individuals with this phenotype, and it is recently considered as a variant phenotype of NS. Gain-of-function mutations in the SOS1 gene were recently described as the second major cause of NS. Here, we report for the first time the involvement of SOS1 gene in a family with the Noonan-like/multiple giant cell lesion phenotype.

Impact of RASopathy subtype on the early disease course of RASopathy-associated hypertrophic cardiomyopathy: clinical outcomes and genetic insights.

BACKGROUND: Genetic mutations causing dysregulation of the RAS/mitogen-activated protein kinase pathway contribute to hypertrophic cardiomyopathy (HCM). Genotype-phenotype associations in this population remain poorly understood. This study aimed to evaluate the impact of RASopathy subtype on cardiovascular outcomes. METHODS: We included 24 patients diagnosed with RAS signaling pathway mutations and HCM (RAS-HCM) before age 18. Data on sociodemographic, prenatal, clinical, genetic, and echocardiographic parameters were retrospectively collected from medical records spanning 2004-2024. Death,
