Synergistic Remodeling of Tumor Immune Microenvironment via a DNA Nanodevice Integrating STING Activation and Lysosome-Targeted PD-L1 Degradation.

The cGAS-STING pathway is a cornerstone of innate antitumor immunity; however, its therapeutic potential is often limited by the inefficient cytosolic delivery of agonists and the immunosuppressive tumor microenvironment (TME). Here, we report a DNA nanostructure-based platform that simultaneously induces lysosomal degradation of immune checkpoint protein PD-L1 and activates the STING pathway. We demonstrate that STING activation further enhances PD-L1 degradation, resulting in strong synergistic immune activation. In murine models of triple-negative breast cancer, the DNA nanodevice effective

eIF4A3 inhibits pseudorabies virus replication by facilitating antiviral immune response.

Eukaryotic translation initiation factor 4A3 (eIF4A3)-mediated RNA metabolism is essential for cellular homeostasis and viral replication. However, its role in regulating antiviral innate immunity during pseudorabies virus (PRV) infection remains unknown. Here, we demonstrate that eIF4A3 protein expression was significantly downregulated both in vitro and in vivo during PRV infection. Functional assays showed that eIF4A3 overexpression suppressed PRV replication, whereas its knockdown enhanced viral replication. Mechanistically, eIF4A3 inhibited m6A modification of STING mRNA, thereby increasi

Targeting STING attenuates ROS induced intervertebral disc degeneration.

OBJECTIVE: DNA damage induced by ROS is considered one of the main causes of nucleus pulposus (NP) cells degeneration during the progression of intervertebral disc degeneration (IVDD). cGAS-STING pathway acts as DNA-sensing mechanism for monitoring DNA damage. Recent studies have proved that cGAS-STING contributes to the development of various diseases by inducing inflammation, senescence, and apoptosis. This work explored the role of STING, the main effector of cGAS-STING signaling pathway, in NP degeneration. METHOD: Immunohistochemistry was conducted to measure STING protein levels in the

STING Signaling Promotes Inflammation in Experimental Acute Pancreatitis.

BACKGROUND & AIMS: Acute pancreatitis (AP) is characterized by severe inflammation and acinar cell death. Transmembrane protein 173 (TMEM173 or STING) is a DNA sensor adaptor protein on immune cells that recognizes cytosolic nucleic acids and transmits signals that activate production of interferons and the innate immune response. We investigated whether leukocyte STING signaling mediates inflammation in mice with AP. METHODS: We induced AP in C57BL/6J mice (control) and C57BL/6J-Tmem173gt/J mice (STING-knockout mice) by injection of cerulein or placement on choline-deficient DL-ethionine sup

Double-Stranded DNA Sensing cGAS-STING Immune Signaling in a Rat Co-Culture Model of the Blood-Brain Barrier.

Double-stranded DNA coming from, for example, viruses, bacteria, or apoptotic cells is recognized by the cGAS-STING signaling pathway comprising the cyclic GMP-AMP synthase (cGAS) and the stimulator of interferon genes (STING) receptors. The pathway induces type I interferon response and activates transcription of interferon-stimulated genes and proinflammatory cytokines. Though the brain is an immune-privileged site, the blood-brain barrier (BBB) elicits inflammatory immune response in neurodegenerative diseases. Parkinson's disease is characterized by α-synuclein oligomer (αSO) aggregates, n

Gelsevirine is a novel STING-specific inhibitor and mitigates STING-related inflammation in sepsis.

BACKGROUND: Stimulation of IFN genes (STING) is central to the production of interferon and proinflammatory cytokines in response to microbial DNA or self-DNA in the cytosol. The detrimental role of the activation of STING during sepsis has been well documented. METHODS: Here, we found that gelsevirine (GS) potently inhibit interferon and inflammatory cytokine induction in macrophages exposed to STING agonists (2'3'-cGAMP, IFN stimulatory DNA (ISD), and poly(dA:dT)). I n silico docking analysis and surface plasmon resonance binding study showed that GS bonds with high affinity to the cyclic d

Brucella suppress STING expression via miR-24 to enhance infection.

Brucellosis, caused by a number of Brucella species, remains the most prevalent zoonotic disease worldwide. Brucella establish chronic infections within host macrophages despite triggering cytosolic innate immune sensors, including Stimulator of Interferon Genes (STING), which potentially limit infection. In this study, STING was required for control of chronic Brucella infection in vivo. However, early during infection, Brucella down-regulated STING mRNA and protein. Down-regulation occurred post-transcriptionally, required live bacteria, the Brucella type IV secretion system, and was indepen

Discovery of isoindoline-2(1H)-carboxamide STING inhibitors as anti-inflammatory agents.

STING (stimulator of interferon genes) is an endoplasmic reticulum-resident membrane-spanning protein that is widely expressed in mammalian cells and functions as a central regulator for the innate immunity. Aberrant activation of the STING axis due to loss-of-function or gain-of-function mutation leads to various autoimmune and autoinflammatory disorders such as Aicardi-Goutières syndrome, systemic lupus erythematosus, and STING-associated vasculopathy with onset in infancy. Here we report the design, synthesis, and structure-activity relationship (SAR) of the isoindoline-2(1H)-carboxamide ST

STING agonists in combination with epigenetic drugs potentiate ZNFX1-driven inflammatory necroptosis in TP53 -mutated AML.

UNLABELLED: TP53 -mutated acute myeloid leukemia (AML) has dismal outcomes with current treatments and represents a critical unmet need. TP53 -mutated AML is proposed to be susceptible to immunotherapeutic approaches but, to date, there is no established immunotherapy for this sub-group. Expression of stimulator of interferon genes (STING), a key innate immune driver that activates interferon (IFN) signaling, is decreased by epigenetic silencing or mutation in many cancers, including those with TP53 mutations. Here, we report that response to the next-generation synthetic STING agonist C92 is

STING-NF-κB signaling builds an influenza spillover barrier.

Influenza pandemics are often traced back to the spillover of avian influenza A viruses (IAVs) to humans. However, barriers against IAV transmission remain elusive. We demonstrated human stimulator of interferon genes (STING) as a transmission barrier against IAVs. STING activated nuclear factor κB (NF-κB) and downstream NF-κB-stimulated genes (NSGs) through a specific domain. Among these NSGs, growth arrest and DNA damage-inducible protein 34 (GADD34) was crucial for IAV restriction. Some IAVs have evolved to evade activating human STING by mutating residue 115 in their matrix protein 1 (M1),

A defect in COPI-mediated transport of STING causes immune dysregulation in COPA syndrome.

Pathogenic COPA variants cause a Mendelian syndrome of immune dysregulation with elevated type I interferon signaling. COPA is a subunit of coat protein complex I (COPI) that mediates Golgi to ER transport. Missense mutations of the COPA WD40 domain impair binding and sorting of proteins targeted for ER retrieval, but how this causes disease remains unknown. Given the importance of COPA in Golgi-ER transport, we speculated that type I interferon signaling in COPA syndrome involves missorting of STING. We show that a defect in COPI transport causes ligand-independent activation of STING. Furthe

LPS-Induced Activation of the cGAS-STING Pathway is Regulated by Mitochondrial Dysfunction and Mitochondrial DNA Leakage in Endometritis.

INTRODUCTION: Chronic endometritis is a common disease in women of childbearing age and can cause pelvic inflammatory disease. The cGAS-STING pathway plays an important role in many inflammatory diseases. PURPOSE: The aim of this study was to investigate the relationship between the cGAS-STING pathway and endometritis. METHODS: We collected endometrium samples from patients with endometritis to detect changes in the cGAS-STING pathway. In vitro, human endometrial stromal cells (HESC) were stimulated with lipopolysaccharide (LPS), and a mouse STING gene-knockout model was established by CRISP

PRRSV Nsp2 promotes inflammatory responses by restricting ER-to-Golgi translocation of STING through HMGB1.

STING serves as a central adaptor in innate immunity, with its trafficking from the endoplasmic reticulum (ER) to the Golgi apparatus determining its signaling outcomes. Retention of STING within the ER is associated with enhanced inflammatory responses, whereas its localization to the Golgi apparatus facilitates the production of type I interferon. In this study, we demonstrate that porcine reproductive and respiratory syndrome virus (PRRSV), an economically significant swine pathogen characterized by causing interstitial pneumonia, promotes the retention of STING within the ER, thereby enhan

The herpesviral antagonist m152 reveals differential activation of STING-dependent IRF and NF-κB signaling and STING's dual role during MCMV infection.

Cytomegaloviruses (CMVs) are master manipulators of the host immune response. Here, we reveal that the murine CMV (MCMV) protein m152 specifically targets the type I interferon (IFN) response by binding to stimulator of interferon genes (STING), thereby delaying its trafficking to the Golgi compartment from where STING initiates type I IFN signaling. Infection with an MCMV lacking m152 induced elevated type I IFN responses and this leads to reduced viral transcript levels both in vitro and in vivo This effect is ameliorated in the absence of STING Interestingly, while m152 inhibits STING-media

Discovery of 3,4-dihydroisoquinoline-2(1H)-carboxamide STING inhibitors as anti-inflammatory agents.

Stimulator of interferon genes (STING) is an endoplasmic reticulum-localized adaptor protein that plays a vital role in mediating the cytosolic DNA-sensing pathway to prime the innate immune responses. Overactivated STING axis causes excessive accumulation of interferons and proinflammatory cytokines, leading to autoimmune and autoinflammatory diseases such as STING-associated vasculopathy of infancy (SAVI) and Aicardi-Goutières syndrome (AGS). Inhibiting the aberrant STING signaling with its inhibitors has proven to alleviate the inflammatory symptoms of the autoimmune and autoinflammatory di

1,25(OH)2 D3 blocks IFNβ production through regulating STING in epithelial layer of oral lichen planus.

Stimulator of interferon genes (STING) is reported to exert vital functions in inflammatory responses and autoimmune diseases. Nevertheless, the status and roles of STING in oral lichen planus (OLP) remain elusive. Here, we state that STING and its downstream cytokine interferon-β (IFNβ) expression is boosted in the oral keratinocytes from patients suffering OLP in comparison with those from healthy participants. Mechanistically, transcription factor GATA-binding protein 1 (GATA1) which is highly increased in diseased samples specifically interacts with its element in the promoter of STING to

Exercise training ameliorates high-fat diet-induced skeletal muscle atrophy and ferroptosis via downregulation of STING.

BACKGROUND: High-fat diet (HFD)-induced sarcopenic obesity can lead to reductions in muscle fiber diameter, enhanced protein degradation, and various forms of cell death. Exercise training has been shown to alleviate HFD-induced muscle atrophy. However, the underlying mechanism remains unclear. Stimulator of interferon genes (STING) is involved in ferroptosis and various forms of muscle atrophy. This study aimed to investigate the role of STING in exercise training against HFD-induced skeletal muscle atrophy. Quercetin induces molecular hallmarks of immunogenic cell death and cGAS-STING pathway activation via mitochondrial DNA release in hepatocellular carcinoma cells.

Immunotherapy for hepatocellular carcinoma (HCC) is constrained by the immunosuppressive tumor microenvironment. Inducing immunogenic cell death (ICD) and activating the cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) pathway are promising strategies to reinvigorate anticancer immunity. Quercetin (QUR) exerts extensive anti-HCC effects, yet whether it can provoke ICD and STING pathway activation remains unknown. Human HCC cell lines SMMC‑7721 and Huh‑7 were treated with various concentrations of QUR. Cell proliferation, colony formation, migration, invasion, and apoptosis
