[High-purity production of native-conformation sweet protein thaumatin via cell factory fermentation and specific enzymatic cleavage].

Thaumatin is a natural sweet-tasting protein derived from the plant Thaumatococcus daniellii, renowned for its high sweetness intensity, low caloric value, and favorable thermal and pH stability. As it can be degraded into natural amino acids, thaumatin represents a potentially safer alternative sweetener. However, plant-based extraction of this protein is constrained by low endogenous content, as well as geographical and seasonal limitations. Meanwhile, existing microbial cell factories often struggle to simultaneously achieve high expression levels and ensure high product purity with natural

Adaptive Laboratory Evolution of Pichia pastoris by Serial Batch Cultivation.

Adaptive laboratory evolution (ALE) is a research tool widely applied for the improvement and investigation of microorganisms. Multiple yeast species have been effectively targeted with this method, and Komagataella phaffii specifically is now well-suited for ALE experimentation due to advancements in its characterization and genetic manipulation. Here is presented a simple and generalizable protocol for the adaptation of K. phaffii towards growth on minimal media with methanol as the sole carbon source, along with suggestions on how this protocol may be adapted for other purposes. We give det

Systematic perturbation of cultures of K. phaffii by carbon co-feeding show gene signatures associated with production of recombinant proteins.

BACKGROUND: Demand for recombinant proteins is rapidly growing, driven by their use as biotherapeutics, vaccine components, industrial enzymes, and food ingredients. The growing market requires novel strategies for increasing protein production in cellular hosts. Systems-level frameworks have been used to improve production, but have had difficulty relating complex cellular pathways with protein expression. Here, we demonstrate a method for mapping relationships between gene expression signatures and carbon source-related phenotypes related to recombinant protein production. RESULTS: Our appro

De Novo Biosynthesis of Sabinene from Methanol by Multiple Engineered Pichia pastoris.

Sabinene, a high-value monoterpene with broad industrial applications, is facing growing demand. To enable sustainable and cost-efficient production, we engineered the yeast Pichia pastoris for sabinene production from methanol. Through systematic metabolic engineering, we implemented a multipronged strategy to optimize sabinene biosynthesis: (1) enhancing precursor supply and synthase activity via fusion proteins (ERG20WW-tNPPS1 and ERG20WW-t37SabS1 (H561F)), (2) redirecting carbon flux through the phosphoketolase-phosphotransacetylase pathway, and (3) upregulating the mevalonate pathway via

Optimization and scale up of L-malic acid production from methanol by the methylotrophic yeast Ogataea polymorpha.

Industrial production of malic acid remains dependent on fossil resources or, when performed microbiologically, on sugar-based feedstocks. Both routes come with caveats, generating emissions and competing with food supply. The use of CO₂-derived one-carbon substrates offers a promising alternative to circumvent these constraints. In this study, the malic acid production process from methanol in metabolically engineered Ogataea polymorpha NYCY495 LEU-ΔSTE12 Pyc Mdh MAE1 strain was optimized and scaled up. A two-phase cultivation strategy, using glycerol for biomass formation and methanol for pr

Cross Kingdom Metabolic Engineering Paradigm Elevating Sustainable Protein Production.

Confronting the dual crisis of escalating global protein demand and unsustainable agriculture necessitates transformative solutions. Here, we pioneer evolutionary insights from maize nitrogen optimization via asparagine synthetase (ASNS) to rewire metabolism in Pichia pastoris. Empirically, the tri-copy ASNS strain achieved superior protein titers: 62.48% crude protein, 47.86% total amino acids, and 8.05% branched-chain amino acids, nutritionally surpassing conventional protein sources. Genome-scale modeling and transcriptomic studies provided convergent evidence that ASNS overexpression drove

Metabolic Engineering of Komagataella Phaffii for de Novo Synthesis of Retinol From Methanol.

Retinol, a derivative of vitamin A with potent antioxidant and therapeutic properties, is in high market demand. In response to the low productivity of conventional methods, metabolic engineering has been explored for microbial retinol production. However, systematic engineering strategies for high-level retinol synthesis in Komagataella phaffii remain limited. In this study, the methylotrophic yeast K. phaffii was developed as an engineered chassis for efficient de novo retinol biosynthesis. Based on a previously constructed β-carotene-producing strain, β-carotene-15,15'-dioxygenase (Blh) and

Studies in enantioselective microbial deuteration.

This study reports methods for reductive microbial deuteration with a high degree of isotopic labelling, focusing on economical approaches using various yeast strains and inexpensive carbon sources. A strategy of α-hydrogen exchange followed by deuterative microbial reduction gave >95% backbone perdeuteration for crucial chiral building blocks for medicinal and analytical applications, without loss of the enantioselectivity demonstrated in the corresponding protiative processes. Under an air atmosphere, Saccharomyces cerevisiae strain MBG5177 outperformed baker's yeast in the synthesis of (2-2

Rewiring the methanol assimilation pathway in the methylotrophic yeast Pichia pastoris for high-level production of erythritol.

Methanol, which is non-food competing, abundant, inexpensive, and potentially renewable, serves as an ideal alternative feedstock for biomanufacturing. Although engineered methylotrophic yeasts have successfully achieved gram-scale production of C2 (acetyl-CoA), C3 (pyruvate), and C6 (fructose-6-phosphate) building blocks from methanol, the production of C4-based (i.e. erythrose-4-phosphate) chemicals remains unexplored. This study demonstrates high-level methanol-to-erythritol production by rewiring the methanol assimilation pathway of Pichia pastoris, achieved through trimming and strengthen

Not all "Candida" are "Candida": Nakaseomyces glabratus (H.W. Anderson) Sugita & M. Takask. 2022 onychomycosis and the need for rapid diagnostic.

Onychomycosis is a common nail disorder typically caused by dermatophytes, with Candida species and non-dermatophytic molds being less frequent agents. Nakaseomyces glabratus (formerly Candida glabrata) is a rare cause of nail infections, notable for its intrinsic resistance to azoles and diagnostic challenges. We report a case of chronic nail discoloration, thickening, and fragility initially misdiagnosed as psoriasis. Microscopy and culture, followed by selective medium analysis, confirmed N. glabratus infection. This case emphasizes the importance of considering uncommon yeast species and h

Biodiesel Production from Waste Oil Through Efficient Enzymatic Synthesis Using Yarrowia lipolytica Lipase 2 in the Presence of Glucose, β-Cyclodextrin, or G50.

In this study, the liquid lipase Yarrowia lipolytica lipase 2 (YLLip2) expressed by Pichia pastoris GS115 was used to produce biodiesel from waste oil. Four signal peptides were compared to express YLLip2 in P. pastoris, among which SP23 exhibited greater secretion performance. In a 1.3-L bioreactor with FM22 medium for 7 days incubation, the maximum YLLip2 activity and total protein content reached 895.44 ± 27.31 U/mL and 3.83 ± 0.31 g/L, respectively. Under the optimal reaction conditions of 30 °C, 20% moisture content, 50 U/g oil of enzyme dosage, and distributed methanol addition, the reac

Unlocking Nature's Toolbox: glutamate-inducible recombinant protein production from the Komagatella phaffii PEPCK promoter.

BACKGROUND: Komagataella phaffii (a.k.a. Pichia pastoris) harbors a glutamate utilization pathway in which synthesis of glutamate dehydrogenase 2 and phosphoenolpyruvate carboxykinase (PEPCK) is induced by glutamate. Glutamate-inducible synthesis of these enzymes is regulated by Rtg1p, a cytosolic, basic helix-loop-helix protein. Here, we report food-grade monosodium glutamate (MSG)-inducible recombinant protein production from K. phaffii PEPCK promoter (PPEPCK) using green fluorescent protein (GFP) and receptor binding domain of SARS-CoV-2 virus (RBD) as model proteins. RESULTS: PPEPCK-RBD/G

The oxygen-tolerant reductive glycine pathway assimilates methanol, formate and CO2 in the yeast Komagataella phaffii.

The current climatic change is predominantly driven by excessive anthropogenic CO2 emissions. As industrial bioprocesses primarily depend on food-competing organic feedstocks or fossil raw materials, CO2 co-assimilation or the use of CO2-derived methanol or formate as carbon sources are considered pathbreaking contributions to solving this global problem. The number of industrially-relevant microorganisms that can use these two carbon sources is limited, and even fewer can concurrently co-assimilate CO2. Here, we search for alternative native methanol and formate assimilation pathways that co-

The In Vitro Activity of Fluconazole, Amphotericin B and Echinocandins Against Cyberlindnera fabianii Planktonic Cells and Biofilms.

Until recently, little was known about the susceptibility pattern of Cyberlindnera fabianii (Cy. fabianii) planktonic cells and biofilms regarding the most frequently administered systemic antifungals, despite the high mortality rate and its potential role in catheter-related infections. In the current study, the activity of fluconazole, amphotericin B and echinocandins (anidulafungin, caspofungin and micafungin) was determined against planktonic and sessile cells of Cy. fabianii clinical isolates (n = 8). Planktonic minimum inhibitory concentrations (MICs) ranged from 1 to 2, from 0.25 to 1,

Characterisation of the secreted apyrase family of Heligmosomoides polygyrus.

Apyrases are a recurrent feature of secretomes from numerous species of parasitic nematodes. Here we characterise the five apyrases secreted by Heligmosomoides polygyrus, a natural parasite of mice and a widely used laboratory model for intestinal nematode infection. All five enzymes are closely related to soluble calcium-activated nucleotidases described in a variety of organisms, and distinct from the CD39 family of ecto-nucleotidases. Expression is maximal in adult worms and restricted to adults and L4s. Recombinant apyrases were produced and purified from Pichia pastoris. The five enzymes

Chromosome Conformation Capture (3C) in Budding Yeast.

Chromosome conformation capture (3C) is a method for studying chromosomal organization that takes advantage of formaldehyde cross-linking to measure the spatial association of two pieces of chromatin. The 3C method begins with whole-cell formaldehyde fixation of chromatin. After cell lysis, solubilized chromatin is digested with a type II restriction endonuclease, and cross-linked DNA fragments are ligated together. Cross-links are reversed by degradation with proteinase K, and chimeric DNA molecules are purified by standard phenol:chloroform extraction. The resulting 3C library represents chr
