This study reports the prevalence, morphological, and molecular identification of Sarcocystis species in the tongues of Florida panthers (Puma concolor coryi). By light microscopy, sarcocysts were found in 46 of 108 (42.6%) panthers and appeared to represent at least 2 morphological types. One morphological type had serrations on the sarcocyst wall and resembled Sarcocystis felis reported from domestic and wild felids; molecular characterization using the ITS1 gene confirmed its identity. The second kind of sarcocyst had a smooth sarcocyst wall without any visible protrusions. The possibility

BACKGROUND: Parasites in the apicomplexan genus Sarcocystis infect cattle worldwide. Assessing the economic importance of each such parasite species requires proper diagnosis. Sarcocystis cruzi, a thin-walled species, infects virtually all cattle. The prevalence of the other thin-walled parasite, Sarcocystis heydorni, remains less well established. The remaining six species all have thick (> 3 µm) cyst walls (Sarcocystis hirsuta, S. hominis, S. bovifelis, S. bovini, S. sigmoideus, and S. rommeli). Thick-walled sarcocysts often induce inflammation in striated muscles (causing bovine eosinophili

Parasites of the genus Sarcocystis can infect several species of animals and cause multiple diseases such as equine protozoal myeloencephalitis. Felines are considered hosts of this protozoa; therefore, the present study aimed to detect anti-Sarcocystis spp.-specific antibodies in domestic cats that were under clinical evaluation, using the indirect immunofluorescence antibody test. Anti-Sarcocystis-specific immunoglobulin Gs were detected in 24 out of 497 (4.82%) cat serum samples. These findings support the fact that natural Sarcocystis infections do occur in cats. Furthermore, it highlights

Toxoplasma gondii has been found to infect almost all warm-blooded animals; however, some hosts lack direct evidence of T. gondii infection. The red panda (Ailurus fulgens) is an endangered species that mainly lives in temperate forests of South Asia. Here, T. gondii infection in red pandas from zoos in China were reported. Antibodies to T. gondii were found in 14.3% (2/14) of red pandas via the modified agglutination test (MAT) with a cut-off titer of 1:25. One viable T. gondii strain was isolated from tissues of red panda and designated as TgRedpandaCHn1. DNA from tachyzoites obtained from c

Reports of Sarcocystis rileyi-like protozoa ('rice breast disease') from anseriform birds had been rare in Europe until the last two decades, when S. rileyi was identified in northern Europe and the UK. However, despite the economic losses resulting from S. rileyi infection, no recent accounts are available on its presence (which can be suspected) in most parts of central, western, southern and eastern Europe. Between 2014 and 2019, twelve mallards (Anas platyrhynchos) were observed to have rice breast disease in Hungary, and the last one of these 12 cases allowed molecular identification of S

A 3-year-old sheep was examined after an acute onset of hind limb paralysis and ataxia. At necropsy, central nervous system, pulmonary and intestinal hyperaemia and ecchymoses in the aortic arch were observed. Main microscopic lesions were confined to the heart, cerebrum and cerebellum. There were a multifocal mild myocarditis and nonsuppurative meningoencephalitis together with protozoal cysts in the heart and the brain. Protozoal cystic structures were observed within many of the myocardial fibers as well as in the cerebrum and cerebellum. Using light microscopy it could not be morphological

Raw horsemeat has the potential to induce food poisoning which often presents with diarrheal symptoms. A sample of horsemeat was found to be infected with Sarcocystis fayeri, and a 15-kDa protein isolated from the cysts of S. fayeri was found to clearly show its diarrhea-inducing activity. A nested polymerase chain reaction was used to clone the cDNA of the 15-kDa protein. The deduced amino acid sequence showed homology to actin-depolymerizing factor (ADF). A recombinant 15-kDa protein depolymerized prepolymerized actins in a test tube. The 15-kDa protein possessed conserved amino acid sequenc

Equine protozoal myeloencephalitis (EPM) is a serious disease of horses, and its management continues to be a challenge for veterinarians. The protozoan Sarcocystis neurona is most commonly associated with EPM. S. neurona has emerged as a common cause of mortality in marine mammals, especially sea otters (Enhydra lutris). EPM-like illness has also been recorded in several other mammals, including domestic dogs and cats. This paper updates S. neurona and EPM information from the last 15 years on the advances regarding life cycle, molecular biology, epidemiology, clinical signs, diagnosis, treat

Sarcocystis infections have been reported from the African buffalo ( Syncerus caffer ), but the species have not been named. Here we propose a new name Sarcocystis cafferi from the African buffalo. Histological examination of heart (92), skeletal muscle (36), and tongue (2) sections from 94 buffalos from the Greater Kruger National Park, South Africa, and a review of the literature revealed only 1 species of Sarcocystis in the African buffalo. Macrocysts were up to 12 mm long and 6 mm wide and were located in the neck muscles and overlying connective tissue. They were pale yellow; shaped like

The oesophagus, diaphragm and heart of 103 camels slaughtered at Al-Ahsa abattoir were investigated for infection with sarcocysts. Sarcocystis cysts were found in 91 camels (88.35%): in the diaphragm of 79.6%, oesophagus of 72.8% and heart of 71.8%. Two morphologically distinct sarcocysts were found. Thin-walled cysts (141-400 x 70.5-188 microns) were in all the three indicator organs while the thick-walled cysts (170-194 x 117.5-188 microns) were only found in the oesophagus. Experimental feeding of four cats and two dogs with sarcocysts-infected meat showed that cats were refractory to the i

The closely related apicomplexa protozoa, Toxoplasma gondii, Neospora spp., and Sarcocystis neurona, have all been associated with neurological and reproductive diseases in horses. However, there is limited data regarding the presence of these three parasites in equine placental tissues and amniotic fluid. The aim of the present report was to investigate the presence of the DNA of T. gondii, Neospora spp. and S. neurona in placentas and amniotic fluid in mares. Anti-T. gondii, anti- S. neurona and anti- Neospora spp. antibody titers were evaluated in 31 mares in the final third of pregnancy by

A free-ranging juvenile fisher (Martes pennanti) with ataxia, lethargy, stupor, and intermittent, whole-body tremors was examined postmortem. Microscopically, the fisher had protozoal meningoencephalitis caused by Sarcocystis neurona, which was confirmed by immunohistochemistry, polymerase chain reaction (PCR) and restriction fragment length polymorphism testing, and genetic sequencing. Sarcocysts found in the skeletal muscle of the fisher were negative for S. neurona by PCR, but were morphologically similar to previous light and electron microscopy descriptions of S. neurona. This is the firs

Old World psittacines experienced an acute fatal illness in outdoor breeding collections in South Florida. Toxoplasma-like organisms were found histologically in pulmonary capillaries and elsewhere. Because the organisms underwent schizogony and could not be transmitted to mice, we looked for a cause other than Toxoplasma gondii. An opossum was trapped on the premises of 1 facility and was found to be shedding sporocysts similar to Sarcocystis falcatula in its feces. Cockroaches were prevalent and suspected as transport hosts. Cockroaches that had ingested opossum feces and subsequently were f

Twelve anticoccidial or antimalarial drugs were tested for their efficacy against various development stages of Sarcocystis muris in NMRI-mice. Schizogonic stages present in the liver from day 11 to 17 p.i. showed to be most sensitive to drug action. Sulfaquinoxaline plus pyrimethamine, zoalene and Bay g 7183 completely eliminated these stages. A strong though not 100 per cent efficacy was observed in experiments with primaquine. The other drugs tested were less (halofuginone, sulfadoxine plus trimethoprim) or not effective (sulfadimethoxine, amprolium, monensin, aprinocid, sulfaquinoxaline pl

This study reports the first identification of Sarcocysits medusiformis in sheep (Ovis aries) in China, based on integrated morphological and molecular analyses. Macroscopic S. medusiformis sarcocysts were detected in 4 of 92 sheep (4.3%), measuring 2490–4796 × 248–405 μm, and featuring thin, smooth walls (1–2 μm thick). Ultrastructurally, the cyst wall was covered by trapezoidal villar protrusions, each lined by an electron-dense layer and containing scattered microtubules extending from the apex to the base. Genetic characterization of three isolates targeted four markers: the nuclear 18 S r

Sarcocystis calchasi is the causative agent of Pigeon Protozoal Encephalitis, a neurological disease in pigeons. The biphasic disease is characterized by neurological signs in the chronic phase. Parasite stages are generally not associated with inflammatory brain lesions and the parasite has been suggested to modulate the host’s immune system. To test this hypothesis, pigeons experimentally infected with S. calchasi were T-cell immunosuppressed beginning from 14 days post infection (dpi) until the end of the experiment (59/60 dpi) and compared with immunocompetent animals. When scored histolog

PURPOSE: Despite of classically acting as definitive hosts of different Sarcocystis species, domestic cats have been pointed out as possible intermediate hosts of S. neurona and S. felis. Nonetheless, details concerning natural sarcocyst development in cats without Sarcocystis-associated disease are scarce. This study aimed to characterize the natural occurrence of muscular sarcocysts in a random group of cats submitted for necropsy.
METHODS: One hundred cats necropsied at a Veterinary Pathology Service were included. Nine different muscular tissues from each cat were sampled for histological

The inner membrane complex (IMC) of apicomplexan parasites contains a network of intermediate filament-like proteins. The 14 alveolin domain-containing IMC proteins in Toxoplasma gondii fall into different groups defined by their distinct spatiotemporal dynamics during the internal budding process of tachyzoites. Here, we analyzed representatives of different IMC protein groups across all stages of the Toxoplasma life cycle and during Sarcocystis neurona asexual development. We found that across asexually dividing Toxoplasma stages, IMC7 is present exclusively in the mother's cytoskeleton, whe
