Fungal species identification in FFPE tissues: A comparative evaluation of droplet digital PCR, ITS sequencing, and metagenomic next-generation sequencing.

Accurate histological diagnosis of fungal infections is challenging due to morphological similarities among fungi, which can affect treatment outcomes. This study evaluated the performance of droplet digital PCR (ddPCR), internal transcribed spacer (ITS) sequencing, and metagenomic next-generation sequencing (mNGS) using 111 formalin-fixed, paraffin-embedded tissue samples with histologically confirmed fungal infections. All three methods showed comparable detection rates for filamentous fungi (84.2%-94.7%; P = .2). For yeast-like fungi, however, mNGS demonstrated significantly higher detectio

[Synthetic Study of Polycyclic Natural Products Based on Development of New Strategy].

On the occasion of receiving the Pharmaceutical Society of Japan Award 2020, I explained our research activities on the total syntheses of polycyclic alkaloids as an invited review. The structure of lundurine B, which has an unstable cyclopropane fused indoline skeleton, was proved firstly by the total synthesis. I also describe the total syntheses of optically active lundurine B and rapidilectine B utilizing asymmetric desymmetrization of the spiro intermediate. We developed an intramolecular bond formation reaction between the 2-position of the furan ring to the iminium cation (furane-iminiu

Biological Activity of Endophytic Fungi from the Roots of the Medicinal Plant Vernonia anthelmintica.

Endophytic fungi were first isolated from the fresh root of the Chinese medicinal plant Vernonia anthelmintica collected from the Hotan Prefecture within the Xinjiang Autonomous region of the People's Republic of China. This plant has been used in Uyghur traditional medicine to treat vitiligo, a skin condition characterized by patches of the skin losing their pigment. In total, fifteen fungal strains were isolated. Among these, four endophytic fungi were identified by their DNA sequences and registered to GenBank with accession numbers. The isolates were identified as Schizophyllum commune XJA

Rapidly progressive mucus plugs in allergic bronchopulmonary mycosis.

INTRODUCTION: Allergic bronchopulmonary mycosis (ABPM) is a chronic airway disease characterized by the presence of fungi that trigger allergic reactions and airway obstruction. Here, we present a unique case of ABPM in which a patient experienced sudden respiratory failure due to mucus plug-induced airway obstruction. The patient's life was saved by venovenous extracorporeal membrane oxygenation (VV-ECMO) and bronchoscopic removal of the plug. This case emphasizes the clinical significance of mucus plug-induced airway obstruction in the differential diagnosis of respiratory failure in patient

Chitosan-glucan complex hollow fibers reinforced collagen wound dressing embedded with aloe vera. II. Multifunctional properties to promote cutaneous wound healing.

This study presents an innovative multifunctional system in fabricating new functional wound dressing (FWD) products that could be used for skin regeneration, especially in cases of infected chronic wounds and ulcers. The innovation is based on the extraction, characterization, and application of collagen (CO)/chitosan-glucan complex hollow fibers (CSGC)/aloe vera (AV) as a novel FWS. For the first time, specific hollow fibers were extracted with controlled inner (500-900 nm)/outer (2-3 µm) diameters from mycelium of Schizophyllum commune. Further on, research and evaluation of morphology, hyd

Successful treatment with inhaled corticosteroid/long-acting β2-agonist in a case of allergic bronchopulmonary mycosis caused by Schizophyllumcommune.

Allergic bronchopulmonary mycosis (ABPM) is a chronic immune-mediated pulmonary disease, which is caused by fungal infection of the airways. Aspergillus species are the main causative fungi and standard treatment typically comprises systemic corticosteroid therapy with or without adjunct antifungal agents. We describe our experience with a case of ABPM caused by Schizophyllum commune (S. commune), with satisfactory response to treatment with a combination of an inhaled corticosteroid and a long-acting β 2-agonist. The patient was a 61-year-old man who was referred to our hospital with dry coug

Dietary schizophyllan reduces mitochondrial damage by activating SIRT3 in mice.

Schizophyllan (SPG), produced by Schizophyllum commune, is an exopolysaccharide with multiple academic and commercial uses, including in the food industry and for various medical functions. We previously demonstrated that SPG conjugated with c-Src peptide exerted a significant therapeutic effect on mouse models of the acute inflammatory diseases polymicrobial sepsis and ulcerative colitis. Here we extended these results by investigating whether SPG exerted a protective effect against mitochondrial damage in the liver via sirtuin 3 (SIRT3) induction, focusing on the deacetylation of succinate d

The role of fungal sensitisation in clinical presentation in patients with chronic obstructive pulmonary disease.

Atopic patients with chronic obstructive pulmonary disease (COPD) demonstrate more severe symptoms than their non-atopic counterparts. Also, Aspergillus hypersensitivity is known in COPD. However, allergic sensitisation to non-Aspergillus fungi has never been studied in COPD patients. To evaluate the prevalence of fungal sensitisation and its impact on the clinical presentation and outcome of COPD patients. Sensitisation to 17 fungi was studied in 55 COPD patients through skin prick tests, fungus-specific IgE, precipitating antibodies, total IgE and eosinophil counts. The clinical symptoms of

Integrated Metabolomic and Bioreporter Screening of Five Thai Mushroom Extracts: Exploring Antibiofilm and Biocompatibility Profiles.

This study evaluated the biomedical potential of ethanolic extracts from five native Thai mushrooms, revealing distinct biological activities associated with their metabolomic profiles. Termitomyces clypeatus R. Heim demonstrated exceptional biocompatibility, effectively promoting human keratinocyte (HaCaT) proliferation without causing cytotoxicity to the cells. Its primary metabolome, rich in essential amino acids and nucleosides, supports its potential as a candidate for topical wound healing, pending further functional validation. Conversely, Lentinus squarrosulus exhibited strong antibiof

Dissociation between sensitizing and colonizing fungi in patients with allergic bronchopulmonary aspergillosis.

BACKGROUND: Because allergic bronchopulmonary aspergillosis (ABPA) does not require the presence of Aspergillus fumigatus for diagnosis, serological and radiological findings without cultures usually confirm this condition. OBJECTIVE: To determine which fungi colonize the airways of patients with definitive ABPA. METHODS: We enrolled 11 patients (ages 57.5 ± 17.1 years; male: female, 4:7) with ABPA diagnosed by serological and radiological criteria. Fungi colonizing the airway were identified from mucous plugs that were naturally expectorated or obtained by fiberoptic bronchoscopy. RESULTS:

Growth and Cultural Characteristics of Cordyceps cardinalis Collected from Korea.

Cordyceps cardinalis was reported in Japan and the USA in 2004, and its fruiting bodies have recently been cultured in Korea. Herbarium specimens preserved at the Cordyceps Research Institute, Mushtech, Korea were revised and identified as C. cardinalis, based on morphological characters and conidial structures. Most of the C. cardinalis specimens were collected from Mt. Halla in Jeju-do. The effects of various nutritional sources and environmental conditions such as temperature and pH on mycelial growth of C. cardinalis were studied. Oatmeal agar, Martin's peptone dextrose agar, and Schizophy

Antifungal and proteolytic activities of endophytic fungi isolated from Piper hispidum Sw.

Endophytes are being considered for use in biological control, and the enzymes they secrete might facilitate their initial colonization of internal plant tissues and direct interactions with microbial pathogens. Microbial proteases are also biotechnologically important products employed in bioremediation processes, cosmetics, and the pharmaceutical, photographic and food industries. In the present study, we evaluated antagonism and competitive interactions between 98 fungal endophytes and Alternaria alternata, Colletotrichum sp., Phyllosticta citricarpa and Moniliophthora perniciosa. Bilateral allergic fungal rhinosinusitis caused by Schizophillum commune and Aspergillus niger. A case report.

Schizophillum commune (S. commune) is a rare type of basidiomycetous fungus that has being reported as a cause of allergic fungal rhinosinusitis (AFRS), invasive type of fungal sinusitis and allergic bronchopulmonary mycosis (ABPM). However, it is believed that S. commune was often misdiagnosed to Aspergillus sp. We report a case of bilateral nasal polyps and maxillary, ethmoidal and sphenoidal involvement within the context of S. commune and Aspergillus niger associated AFRS. Our patient was suffering from a chronic disease with periods of remission and exacerbation and was treated successful

Integrated genetic and process engineering of Pichia pastoris for high-yield ergothioneine production via methanol-free cultivation.

Ergothioneine (EGT) is a histidine-derived thiol antioxidant with broad applications in food, pharmaceuticals and cosmetics, but its industrial production is limited by scarce natural sources and low heterologous host titers. Here, a high-yield, methanol-free EGT production platform was established in Pichia pastoris X33 via integrated genetic and process engineering: de novo pathway assembly first generated strain P4 with a shake-flask EGT titer of 99.6 ± 3.2 mg/L via screening of the strong constitutive promoter PGCW14, site-directed engineering of Schizophyllum commune methyltransferase ScE

Nasopharyngeal metagenomics of symptomatic healthcare workers provides insights into the respiratory microbiome and antimicrobial resistance.

Respiratory infections represent a significant risk for healthcare workers (HCWs), particularly during viral outbreaks. This study applied metagenomic sequencing to characterize microbial communities and antimicrobial resistance (AMR) genes in nasopharyngeal swabs from HCWs presenting respiratory symptoms. Samples from 161 HCWs collected at a tertiary hospital in 2020-2021 were screened using FilmArray; negative samples were analyzed by metagenomic sequencing. After removal of human reads, sequences were taxonomically classified into viral, bacterial, and eukaryotic groups, and AMR genes were

Dysbiosis in the Nasal Mycobiome of Infants Born in the Aftermath of Hurricane Maria.

Hurricanes and flooding events substantially elevate indoor fungal spore levels, which have been associated with increased risks of developing childhood asthma and other adverse respiratory outcomes. Although environmental fungal compositions following major hurricanes have been well characterized, the fungal communities within the nasal cavity (i.e., the nasal mycobiome) of exposed individuals remain unexplored. We collected nasal swab samples from infants following Hurricane Maria in San Juan, Puerto Rico, during two periods (March to August 2018 and February to September 2019). We processed
