Tissue parameters of the sea urchin (Paracentrotus lividus): the effects of magnesium and transport stress.

The present study investigates some biochemical and biological variables in sea urchins (Paracentrotus lividus) that were stressed in the course of collection, transportation and laboratory conditioning and the effect thereon of pre-treatment with a test compound; magnesium chloride (MgC12). Handling and transportation for 5h resulted in significantly decreased gonadal indices due to unplanned spawning. The pre-treatment of sea urchins with 5 mmol 1(-1) MgCl2 reduced unplanned spawning during all of the experiments. Protein content and humidity were significantly higher (p < 0.05) in sea urchi

Cyclic GMP-specific phosphodiesterase-5 regulates motility of sea urchin spermatozoa.

Motility, chemotaxis, and the acrosome reaction of animal sperm are all regulated by cyclic nucleotides and protein phosphorylation. One of the cyclic AMP-dependent protein kinase (PKA) substrates in sea urchin sperm is a member of the phosphodiesterase (PDE) family. The molecular identity and in vivo function of this PDE remained unknown. Here we cloned and characterized this sea urchin sperm PDE (suPDE5), which is an ortholog of human PDE5. The recombinant catalytic domain of suPDE5 hydrolyzes only cyclic GMP (cGMP) and the activity is pH-dependent. Phospho-suPDE5 localizes mainly to sperm f

Mastoparan induces Ca2+-independent cortical granule exocytosis in sea urchin eggs.

In most species, cortical granule exocytosis is characteristic of egg activation by sperm. It is a Ca(2+)-mediated event which results in elevation of the vitelline coat to block permanently the polyspermy at fertilization. We examined the effect of mastoparan, an activator of G-proteins, on the sea urchin egg activation. Mastoparan was able to induce, in a concentration-dependent manner, the egg cortical granule exocytosis; mastoparan-17, an inactive analogue of mastoparan, had no effect. Mastoparan, but not sperm, induced cortical granule exocytosis in eggs preloaded with BAPTA, a Ca(2+) che

Measurement of the intracellular pH threshold for sperm aster formation in sea urchin eggs.

In the fertilization of sea urchin eggs, intracellular [Ca2+] (Cai) increases transiently and intracellular pH (pHi) elevates accordingly. Unlinking these two activating factors experimentally, the requirement of the increase in pHi for sperm aster formation in the sea urchin, Clypeaster japonicus, was investigated. When the eggs were injected with an EGTA or BAPTA solution, they incorporated sperm but did not organize the sperm aster. Using these sperm-incorporated eggs under the condition that an increase in Cai was blocked, pHi was regulated by two methods: (i) perfusing ammonium acetate-co

Calcium influx mediates the voltage-dependence of sperm entry into sea urchin eggs.

Sperm entry was monitored in voltage-clamped sea urchin eggs following insemination in a variety of artificial seawaters. In regular seawater, maintaining the membrane potential at increasingly negative values progressively inhibits sperm entry. Reducing [Ca(2+)](o) relieves the inhibition, shifting the sperm entry vs voltage relationship toward more negative potentials. Raising [Ca(2+)](o) shifts the relationship in the other direction. Large changes in [Na(+)](o) or [Mg(2+)](o) do not affect sperm entry although changing [Na(+)](o) dramatically changes the currents following sperm attachment

Recombinant SpTrf proteins illustrate putative functions of native SpTrf proteins: from immune response to resolution in sea urchins.

The complex and sophisticated sea urchin innate immune system relies on conserved immune receptor and effector molecules plus the euechinoid-specific Transformer family. Native SpTransformer (natSpTrf) proteins in Strongylocentrotus purpuratus respond to immune challenge by augmenting phagocytosis of microbes. These proteins show significant sequence diversity both within and among sea urchins with the predicted benefit of host protection through the recognition of a broad range of pathogens. Prior reports show that 6 recombinant (r)SpTrf proteins from insect cells induce phagocytosis when cro

Shewanella harboring antimicrobial and copper resistance genes in sea urchins (Paracentrotus lividus) from the Crozon peninsula (Brittany, France).

Shewanella is a genus of aquatic non-fermenting Gram-negative bacteria with increasing numbers of reports of infections in humans and appearance of antimicrobial resistant strains. Cases of infection show a relatively strong association with seafood consumption or exposure to seawater. This study aimed to analyze Shewanella spp. isolated from the sea urchin Paracentrotus lividus collected from the Crozon peninsula (France) with the intention of obtaining insights into the role of this genus as a reservoir of antimicrobial and heavy metal resistance genes. Five among seven Shewanella isolates w

Estimating carbon storage and flux in sea urchin barrens following kelp forest collapse.

Mitigating climate change impacts due to excess carbon dioxide will require knowledge of blue carbon sources and sinks. Marine ecosystems including seagrass meadows, mangroves, salt marshes and kelp forests store organic carbon while inorganic carbon is bound as calcium carbonate (CaCO3) in shells. Kelp forest ecosystems however can shift from carbon bound in kelps to sea urchin barrens composed of CaCO3. We examine this ecosystem shift with respect to carbon by estimating purple, Strongylocentrotus purpuratus, and red sea urchin, Mesocentrotus franciscanus, population abundances (in 2022-2023

Unravelling Hidden Trophic Interactions Among Sea Urchin Juveniles and Macroinvertebrates by DNA Amplification.

Rocky reefs may shift between two distinct stable states: productive algal forests, characterised by high abundance and biodiversity of macrofauna, and impoverished barrens, dominated by overgrazing sea urchins. Barren states may persist despite the recovery of adult sea urchin predators, suggesting additional stabilising mechanisms. Sea urchin settlers equally colonise forests and barrens in large numbers, but in forests only a few of them reach adult size, suggesting that post-settlement predation might play a crucial role in determining sea urchin population density. Visual assessment of pr

Impact of hyposalinity on the barrens-forming sea urchin Centrostephanus rodgersii in context with flooding events.

The sea urchin Centrostephanus rodgersii is an ecologically important species in southeastern Australia, where its grazing contributes to the formation of macroalgal-free barrens habitat. While climate warming has facilitated this species southward range extension, climate change is also causing an increase in the frequency of storm-driven hyposalinity events. These events can expose marine organisms to acute or gradual decrease salinity stress, depending on the rate and duration of freshwater influx. The tolerance of C. rodgersii to below ambient salinity was investigated using a range of tre

Understanding osmoregulation, cryoprotectant toxicity, and cold tolerance in sea urchin eggs: Implications for cryopreservation.

This study investigates the challenges and potential solutions for cryopreserving sea urchin (Paracentrotus lividus) eggs. It focuses on understanding the thresholds and osmoregulatory mechanisms, cryoprotectant toxicity, and cold tolerance in these eggs, which are especially difficult to preserve due to high sensitivity to cryoprotectants and low-temperature injury. Key findings suggest that stepwise addition of cryoprotectants, particularly dimethyl sulfoxide (Me2SO) combined with other agents like DMF or methanol, reduces toxicity and enhances larval survival. The study also highlights that

Under the Skin: A Review of Sea Urchin Granulomas and Delayed Dermal Reactions to Echinoid Spines.

Sea urchin injuries are common in coastal and tropical environments, often resulting in immediate pain and mechanical trauma. In a subset of patients, retained spine fragments trigger delayed granulomatous reactions that present diagnostic and therapeutic challenges. These "sea urchin granulomas" encompass a histopathologic spectrum including foreign body, sarcoidal, suppurative, and necrobiotic subtypes. This review synthesizes clinical, immunologic, diagnostic, and therapeutic insights from 36 peer-reviewed studies describing over 120 cases. Granulomas typically emerge weeks to months after

Screening and purification of antimicrobial materials from coelomic fluid of sea urchin, Heliocidaris crassispina.

Marine organisms, such as sea urchins like Heliocidaris crassispina, produce bioactive substances with antimicrobial activity to protect themselves from the high density of microorganisms in their habitats. One such substance, Echinochrome A (Ech A), has been isolated from various sea urchins' shells and spines using strong acidic solutions and organic solvents. Ech A, however, has not been reported from the coelomic fluid of H. crassispina. In this study, we report the antimicrobial activity of H. crassispina coelomic fluid extract against various microbes, evaluating its potential for purify

The Impact of Sea Urchin as an Ingredient on the Physicochemical, Microbiological, and Sensory Properties of Fish Sauce Fermentation.

The consequences of using 25% whole or shelled sea urchin as an ingredient in anchovy sauce on its fermentation and development of its physicochemical properties after 20 days fermentation was studied. Two varieties of fish and sea urchin sauce were made with or without shell at 1:2:1 ratio (salt:fish:sea urchin) plus a control fish sauce at 1:3 ratio (salt:fish). All sauces were fermented at 40-50 °C for 20 days, where for the first 7 days the preparation remained in a static phase. During their fermentation, pH, salt concentration, aw, TVB-N, TMA, total nitrogen, formaldehyde nitrogen, amino

Toxicity of tire particle leachates on early life stages of keystone sea urchin species.

Particles from tires are a major fraction of microplastic pollution. They contain a wide range of chemical additives that can leach into the water and be harmful to aquatic organisms. In this study, we investigated the acute toxicity of tire particle leachates in early life stages of three keystone echinoderm species (Paracentrotus lividus, Arbacia lixula, Diadema africanum). Embryos were exposed for 72 h to a range of leachate dilutions, prepared using a concentration of 1 g L-1. Larval growth, abnormal development, and mortality were the measured endpoints. Furthermore, we estimated the acti

Comparison of feeding preferences of herbivorous fishes and the sea urchin Diadema antillarum in Little Cayman.

On Caribbean coral reefs, losses of two key groups of grazers, herbivorous fishes and Diadema antillarum, coincided with dramatic increases in macroalgae, which have contributed to decreases in the resilience of these coral reefs and continued low coral cover. In some locations, herbivorous reef fishes and D. antillarum populations have begun to recover, and reductions in macroalgal cover and abundance have followed. Harder to determine, and perhaps more important, are the combined grazing effects of herbivorous fishes and D. antillarum on the structure of macroalgal communities. Surprisingly

Effects of neurotransmitter receptor antagonists on sea urchin righting behavior and tube foot motility.

PURPOSE: Although abnormal peripheral and central pain processing has been observed in fibromyalgia (FM) patients, the biomechanics and pathophysiology, surrounding the peripheral mechanism are not well understood. An intermediate conductance channel, KCa3.1, is expressed in peripheral sensory nerve fibers where it maintains the resting membrane potential and controls nerve firing, making it a plausible target for peripheral therapeutic interventions. ASP0819, a KCa3.1 channel opener, is an orally available molecular entity and is used in this investigation to elucidate the role of KCa3.1 in s

The multiple functions of the wild type Huntington's disease protein of the sea urchin Hemicentrotus pulcherrimus (Hp-Htt) have been examined using the anti-Hp-Htt antibody (Ab) raised against synthetic oligopeptides. According to immunoblotting, Hp-Htt was detected as a single band at around the 350 kDa region at the swimming blastula stage to the prism larva stage. From the 2-arm pluteus stage (2aPL), however, an additional smaller band at the 165 kDa region appeared. Immunohistochemically, Hp-Htt was detected in the nuclei and the nearby cytoplasm of the ectodermal cells from the swimming b
