[Possibilities of multiphoton microscopy for the diagnosis of the vulvar lichen sclerosus].

BACKGROUND: Vulvar lichen sclerosus (VLS) is a chronic and recurrent dermatosis of an inflammatory nature with severe focal atrophy of the skin. Connective tissue changes are polymorphic and are still not taken into account in histological diagnostics due to the difficulty of interpreting routine histological methods. In this work, we use multiphoton microscopy (MPM) as a new imaging technique that provides detailed information about the organization of collagen fibers in the dermis based on a non-linear second harmonic generation (SHG) process. OBJECTIVE: To determine the degree of connectiv

Fibrillar collagen scoring by second harmonic microscopy: a new tool in the assessment of liver fibrosis.

BACKGROUND & AIMS: Imaging of supramolecular structures by multiphoton microscopy offers significant advantages for studying specific fibrillar compounds in biological tissues. In this study, we aimed to demonstrate the relevance of Second Harmonic Generation (SHG) for assessing and quantifying, without staining, fibrillar collagen in liver fibrosis. METHODS: We first showed the relationship between SHG signal and collagen forms over-produced and accumulated during fibrosis progression. Taking this property into consideration, we developed an innovative method to precisely quantify the fibros

Analysis of forward and backward Second Harmonic Generation images to probe the nanoscale structure of collagen within bone and cartilage.

Collagen ultrastructure plays a central role in the function of a wide range of connective tissues. Studying collagen structure at the microscopic scale is therefore of considerable interest to understand the mechanisms of tissue pathologies. Here, we use second harmonic generation microscopy to characterize collagen structure within bone and articular cartilage in human knees. We analyze the intensity dependence on polarization and discuss the differences between Forward and Backward images in both tissues. Focusing on articular cartilage, we observe an increase in Forward/Backward ratio from

Harmonic Imaging of Stem Cells in Whole Blood at GHz Pixel Rate.

The pre-clinical validation of cell therapies requires monitoring the biodistribution of transplanted cells in tissues of host organisms. Real-time detection of these cells in the circulatory system and identification of their aggregation state is a crucial piece of information, but necessitates deep penetration and fast imaging with high selectivity, subcellular resolution, and high throughput. In this study, multiphoton-based in-flow detection of human stem cells in whole, unfiltered blood is demonstrated in a microfluidic channel. The approach relies on a multiphoton microscope with diffrac

Nonlinear Microscopy of ECM Remodeling in Renal and Vascular Tissues: A Systematic Review Integrating Human AVF Imaging.

Background and Objectives: Extracellular matrix (ECM) and collagen remodeling contribute to chronic kidney disease (CKD) progression and vascular access dysfunction. Conventional histological techniques rely on staining and provide limited sensitivity for detecting early or subtle ECM alterations. Nonlinear optical imaging modalities, including second-harmonic generation (SHG), third-harmonic generation (THG), and multiphoton fluorescence (MPF) microscopy, enable label-free, high-resolution visualization of fibrillar collagen and may offer additional structural information. This study aimed to

Intrinsic ferroelectric CuVP2S6 for potential applications in neuromorphic recognition and translation.

Two-dimensional ferroelectric materials hold promises for non-volatile memory and integrated electronic circuits. However, direct synthesizing and understanding the ferroelectric mechanism of two-dimensional room-temperature ferroelectrics remains challenging. Here, we report the synthesis of intrinsic room-temperature ferroelectric CuVP2S6 via spatially confined chemical vapor deposition method, as demonstrated by second harmonic generation and piezoresponse force microscopy measurements. Importantly, we experimentally uncover the ferroelectric mechanism of CuVP2S6, which originates from the

Homochiral Multiferroic Cyanido-Bridged Dimetallic Complexes Assembled by C-F⋅⋅⋅K Interactions.

Cyanido-bridged dimetallic complexes are attracting attention due to their varied structures and properties. However, homochiral cyanido-bridged dimetallic complexes are rare, and making them ferroelectric is a great challenge. Introducing C-F⋅⋅⋅K interactions between the guest chiral cations and the host [KFe(CN)6 ]2- framework, gives three-dimensional cyanido-bridged dimetallic multiferroics, [R- and S-3-fluoropyrrolidinium]2 [KFe(CN)6 ] (R- and S-3-FPC). The mirror-symmetric vibrational circular dichroism (VCD) signal shows their enantiomeric nature. R- and S-3-FPC crystallize in the same c

Persistent bone resorption lacunae on necrotic bone distinguish bisphosphonate-related osteonecrosis of jaw from denosumab-related osteonecrosis.

BACKGROUND: Bisphosphonate and denosumab are widely used for the treatment of osteoporosis and bone metastasis of cancer to prevent excessive bone resorption. Osteonecrosis of the jaw is a serious adverse effect of bisphosphonate or denosumab referred to as bisphosphonate-related osteonecrosis of the jaw (BRONJ) or denosumab-related osteonecrosis of the jaw (DRONJ), respectively. Since bisphosphonate and denosumab inhibit bone resorption by different mechanism, we evaluated whether these drug types result in different histopathological characteristics related to bone resorption. Fast Fourier transform-based analysis of second-harmonic generation image in keratoconic cornea.

PURPOSE: To qualify and quantify structural alterations in keratoconic corneas ex vivo by the use of second-harmonic generation (SHG) microscopy and two-dimensional fast Fourier transform (2D-FFT) analysis. METHODS: Two keratoconic and three normal human corneal specimens were imaged with a multiphoton microscope. Forward and backward second-harmonic generation images (FSHG and BSHG) of corneal stroma were obtained at high resolution and at different depths. The SHG images were further analyzed with a 2D-FFT algorithm to quantify the texture and orientation changes of collagen fibers. RESULT

Magnetic-Order-Mediated Second Harmonic Generation in a Room-Temperature van der Waals Ferromagnet Fe3GaTe2.

Magnetic order coupled second harmonic generation (SHG) provides a unique nonlinear probe of symmetry breaking in low dimensional magnets. However, its capability to resolve nonuniform magnetic domain structures and identify domain boundaries remains largely unexplored. Here, we demonstrate optical SHG imaging of labyrinthine magnetic domains in room temperature Fe3GaTe2, showing high sensitivity to magnetic configurations consistent with magnetic force microscopy. Magnetic field dependent SHG measurements reveal coherent interference between structural and magnetization coupled contributions,

The role of FXR and TGR5 in reversing and preventing progression of Western diet-induced hepatic steatosis, inflammation, and fibrosis in mice.

Nonalcoholic steatohepatitis (NASH) is the most common chronic liver disease in the US, partly due to the increasing incidence of metabolic syndrome, obesity, and type 2 diabetes. The roles of bile acids and their receptors, such as the nuclear receptor farnesoid X receptor (FXR) and the G protein-coupled receptor TGR5, on the development of NASH are not fully clear. C57BL/6J male mice fed a Western diet (WD) develop characteristics of NASH, allowing determination of the effects of FXR and TGR5 agonists on this disease. Here we show that the FXR-TGR5 dual agonist INT-767 prevents progression o

Quantitative characterization of duodenal gastrinoma autofluorescence using multiphoton microscopy.

BACKGROUND: Duodenal gastrinomas (DGASTs) are neuroendocrine tumors that develop in the submucosa of the duodenum and produce the hormone gastrin. Surgical resection of DGASTs is complicated by the small size of these tumors and the tendency for them to develop diffusely in the duodenum. Endoscopic mucosal resection of DGASTs is an increasingly popular method for treating this disease due to its low complication rate but suffers from poor rates of pathologically negative margins. Multiphoton microscopy can capture high-resolution images of biological tissue with contrast generated from endogen

Blind deconvolution of second harmonic microscopy images of the living human eye.

Second harmonic generation (SHG) imaging microscopy of thick biological tissues is affected by the presence of aberrations and scattering within the sample. Moreover, additional problems, such as uncontrolled movements, appear when imaging in-vivo. Deconvolution methods can be used to overcome these limitations under some conditions. In particular, we present here a technique based on a marginal blind deconvolution approach for improving SHG images obtained in vivo in the human eye (cornea and sclera). Different image quality metrics are used to quantify the attained improvement. Collagen fibe

Quantification of collagen structural changes during chick corneal development.

As the most abundant structural mammalian protein, collagen has been implicated in the pathogenesis of numerous diseases such as osteogenesis imperfecta, and cancer. In the case of cornea, abnormal cornea development can lead to conditions such as agenesis, megalocornea, microcornea, and cornea plana. Therefore, understanding the mechanisms of collagen assembly during development may contribute to the prevention or treatment of corneal diseases. In this study, we applied fast Fourier transform second harmonic generation microscopy to quantify parameters of corneal structures during chick devel

Deformation-Potential-Driven Photostriction in Layered Ferroelectrics.

The coupling between electronic excitations and lattice deformation in van der Waals ferroelectrics is governed by a competition between the electron deformation potential and the inverse piezoelectric effect. While theory predicts that piezoelectric screening should drive a polar-axis contraction in monolayer group-IV monochalcogenides, we demonstrate that, in multilayer tin monosulfide (SnS), the deformation potential provides the dominant contribution, driving a polar-axis expansion even within ferroelectric domains. By correlating polarization-resolved second-harmonic generation microscopy
