Plants can be used to produce inexpensive and highly immunogenic vaccines, particularly those aimed against mucosal pathogens. Several plant-derived vaccines have already completed early-phase clinical trials and many more are in the pipeline. The number of products in development has increased as the production technology itself has evolved, reflecting a better understanding of plant molecular biology, more sophisticated genetic engineering techniques, and more recently the development of tools and strategies to increase yields and engineer specific glycan groups on plant-derived glycoprotein

BACKGROUND: We have previously developed a rice-based oral vaccine against cholera diarrhea, MucoRice-CTB. Using Agrobacterium-mediated co-transformation, we produced the selection marker-free MucoRice-CTB line 51A, which has three copies of the cholera toxin B subunit (CTB) gene and two copies of an RNAi cassette inserted into the rice genome. We determined the sequence and location of the transgenes on rice chromosomes 3 and 12. The expression of alpha-amylase/trypsin inhibitor, a major allergen protein in rice, is lower in this line than in wild-type rice. Line 51A was self-pollinated for f

BACKGROUND: β-Conglycinin is the main component of soy protein. However, its potential allergenicity limits the application of soy protein in the food industry. The purpose of this study is to investigate the effects of covalent modification of epigallocatechin gallate (EGCG) on the immune and functional properties of β-conglycinin.
RESULTS: The antigenicity of β-conglycinin covalent complexes was significantly reduced compared to the untreated proteins. The antigenicity reduction of EGCG and β-conglycinin free radical complex was 37.97%, which was better than that of alkali complex (34.21%).

Liposomes were modified with different proportions of β-conglycinin (7S) and glycinin (11S) to form Lip-7S and Lip-11S. The morphology, interaction and in vitro simulated digestion of liposomes were studied. The particle size of Lip-7S was smaller than that of Lip-11S. When the values of Lip-7S and Lip-11S were 1:1 and 1:0.75, respectively, the ζ-potential had the maximum absolute value and the dispersion of the system was good. The results of multispectral analysis showed that hydrogen-bond and hydrophobic interaction dominated protein-modified liposomes, the protein structure adsorbed on the

β-conglycinin (β-CG) induces intestinal damage in piglets; however, its regulatory mechanisms are not fully understood. This study aimed to investigate the molecular mechanisms by which β-CG regulates intestinal injury in piglets through downstream genes and proteins. Our findings revealed that β-CG significantly reduced villus height while increasing the crypt depth. In addition, we analyzed the transcriptome and proteome of jejunum tissues after the β-CG treatment. In total, 382 differentially expressed genes (DEGs) and 292 differentially expressed proteins (DEPs) were identified between the

Gac (Momordica cochinchinensis Spreng.) seed proteins (GSPs) hydrolysate was investigated for angiotensin I-converting enzyme (ACE) inhibitory activities. GSPs were hydrolyzed under simulated gastrointestinal digestion using a combination of enzymes, including pepsin, trypsin, and chymotrypsin. The screening of ACE inhibitory peptides from GSPs hydrolysate was performed using two sequential bioassay-guided fractionations, namely hydrophilic interaction liquid chromatography (HILIC) and reversed-phase high-performance liquid chromatography (RP-HPLC). Then, the peptides in the fraction with the

Bioactive gibberellins (GAs or diterpenes) are essential hormones in land plants that control many aspects of plant growth and development. In flowering plants, 13-OH GAs (having low bioactivity-for example, GA1) and 13-H GAs (having high bioactivity-for example, GA4) frequently coexist in the same plant. However, the identity of the native Arabidopsis thaliana 13-hydroxylase GA and its physiological functions remain unknown. Here, we report that cytochrome P450 genes (CYP72A9 and its homologues) encode active GA 13-hydroxylases in Brassicaceae. Plants overexpressing CYP72A9 exhibited semi-dwa

Proteomic approach is applied for the analysis of seed brans of 14 rice varieties (Oryza sativa L. ssp. indica) which can classify to five aromatic rice and nine nonaromatic rice. The two-dimensional electrophoresis (2-DE) protein patterns for 14 rice varieties were similar within pH ranges of 3-10 and 4-7. To characterize aromatic group-specific proteins, we compared 2-D gels of aromatic rice to nonaromatic rice using PDQUEST image analysis. Four out of six differential spots were identified as hypothetical proteins, but one (SSP 7003) was identified by matrix assisted laser desoption/ionizat

As the demand for plant-based nutrition increases, improving the protein profile of legumes like cowpea has become a breeding priority. Cowpea, a multiuse legume and staple in many low-income regions, provides important dietary protein that can help meet the demand in our growing population. Our research used genome-wide association studies (GWAS) and phenomic tools to investigate the genetic architecture of seed protein content in cowpea and integrated 4 cohorts of undergraduate researchers through a USDA-AFRI REEU program. Using wet chemistry and near-infrared spectroscopy (NIRS), we assesse

The prolamin peptides in wheat gluten and in the homologous storage proteins of barley and rye cause painful chronic erasure of microvilli of the small intestine epithelium in celiac patients. If untreated, it can lead to chronic diarrhea, abdominal distension, osteoporosis, weight-loss due to malabsorption of nutrients, and anemia. In addition to congenital cases, life-long exposure to gluten proteins in bread and pasta can also induce development of celiac sprue in adults. To date, the only effective treatment is life-long strict abstinence from the staple food grains. Complete exclusion of

Perilla (Perilla frutescens (L.) Britt.) seed protein, an underutilized resource derived from medicinal and edible plants, was subjected to ultrasound (400 W, 20 min), pH-shifting (pH 12.0, 1 h), and high-pressure homogenization (90 MPa, 3 cycles), applied individually or in combination, and subsequently fermented with probiotics to induce gel formation. The triple combination (pH-shifting + high-pressure homogenization + ultrasound) produced the highest solubility increase (129%), smallest particle size, greatest random coil content, and maximal surface hydrophobicity and free sulfhydryl leve

BACKGROUND: Nut allergy varies from pollen cross-allergy, to primary severe allergy with life-threatening symptoms. The screening of IgE antibodies to a wide spectrum of allergens, including species-specific and cross-reactive allergens, is made possible via microarray analysis.
OBJECTIVE: We sought to study the association of variable IgE sensitization profiles to clinical response in peanut-challenged children and adolescents in a birch-endemic region. In addition, we studied the avoidance of tree nuts and species-specific sensitizations.
METHODS: We studied 102 peanut-sensitized patients

High-quality seeds necessitate both appropriate dormancy to prevent preharvest sprouting and robust storability to preserve germination vigor, which serve as the fundamental guarantee for the successful production and safe storage of crops. As complex and quantitative traits, the improvement of seed quality has been impeded by limited understanding of their regulatory genes. Here, in rice, we identify SEED-AGING GENE 9 (SAG9), encoding a nuclear protein, as a negative regulator underlying the major seed-storability quantitative trait locus qSS-9, which also exerts a moderate effect on dormancy

Soy protein is widely used in plant-based foods; however, heating often induces large and irregular aggregates that compromise sensory quality. A distinct thermal aggregation behavior was observed in soy β-conglycinin (7S), contributing to soluble aggregates with limited size and minor sensory impacts. In this study, the mechanism of thermal limited aggregation in 7S was revealed by focusing on the effects of its main fractions, N-terminal extension region (ER) and core region (CR). Their evolutions in turbidity, solubility and dynamic light scattering, and conformational transition during hea

Peanut is one of the most popular foods in the world due to its high nutrition; however, it contains multiple seed storage proteins which are identified as allergens and hence are the most common cause of life-threatening, IgE-mediated anaphylaxis among the hypersensitive individuals. Three peanut proteins, Arachis hypogaea allergy 1, 2, 3 (Ara h 1, Ara h 2 and Ara h 3), which have the common biochemical characteristics like resistance to proteases and heat, are considered as the major allergens because they are recognized by serum IgE from a peanut-allergic patient population. The linear IgE-

Background. Tree nut and peanut allergy are common, and often persistent conditions, that significantly affect quality of life. Multiple nut avoidance is common due to cross-reactivity concerns but may cause unnecessary restrictions. Data regarding multiple nut oral food challenges (MN-OFC) remains limited. Our aim was to evaluate the safety and clinical impact of MN-OFC in the management of tree nut and/or peanut allergy. Methods. Retrospective analysis of 25 patients who underwent MN-OFCs between 2022 and 2024 at a tertiary-level Allergy Department. Patients with profilin and/or PR-10 sensit

β-Conglycinin (β-CG), an anti-nutritional factor, is a major allergen in soybeans to induce intestinal dysfunction and diarrhea in neonatal animals, including piglets and human infants. This study with a piglet model determined the effects of N-acetylcysteine (NAC) on intestinal function and autophagy in response to β-CG challenge. Twenty-four 12-day-old piglets (3.44 ± 0.28 kg), which had been weaned at 7 days of age and adapted for 5 days after weaning, were randomly allocated to the control, β-CG, and β-CG + NAC groups. Piglets in the control group were fed a liquid diet containing 10% case

AIMS: Formalin-fixed paraffin-embedded (FFPE) samples, routinely used in neuropathology, represent an invaluable resource for studying rare diseases like transmissible spongiform encephalopathies (TSE). Despite fixation-induced protein cross-linking, prion seeding activity can be effectively detected using the seeding amplification assays. In this study, we employed the second-generation real-time quaking-induced conversion (RT-QuIC) assay to analyse and quantify human prion seeding activity in FFPE brain tissues.
METHODS: FFPE frontal brain tissues were deparaffinised in xylene, followed by

BACKGROUND: Cucumber plants suffer from a serious threatening disease, downy mildew, throughout the growing seasons irrespective of the weather temperature. The causal agent, Pseudoperonospora cubensis, tends to evolve rapidly upon sequential applications of chemical fungicides and generate new progeny possessing tolerance to such fungicides. Glycoproteins represent an environmentally safe alternative for chemically synthetized fungicides and do not trigger fungicide resistance. We studied the antifungal activity of four glycoproteins namely soybean β-conglycinin, chickpea vicilin, duck egg ov
