Development of a self-contained accelerometry based system for control of functional electrical stimulation in hemiplegia.

For stroke patients, functional electrical stimulation (FES) has been shown in the past to greatly reduce gait impairments. A critical element of the success of this intervention is accurate and reliable triggering of the stimulation for step initiation. Foot switches are the most commonly used devices for triggering hemiplegic FES gait, but they have been known to produce unreliable results and degrade over time. This paper outlines the development of a self-contained accelerometry-based gait stimulation system that can be worn around the waist and unlike other systems, adds no additional har

Disturbances of self-other distinction after stimulation of the extrastriate body area in the human brain.

In a recent experiment with functional magnetic-resonance imaging, we found that brain activity in the extrastriate body area (EBA) distinguished between observed self- and other-generated movements, being significantly higher during observation of someone else's movement. Here, we investigated further the role of EBA in self-other distinctions using low-frequency repetitive transcranial magnetic stimulation (rTMS). As compared with rTMS applied over a control site, rTMS applied over the EBA increased reaction times, without affecting accuracy, for the detection of other-generated movements. P

A pen injection device for self-administration of recombinant follicle-stimulating hormone for fertility treatments.

A pen injection device modeled on the insulin pen used by diabetic patients has been introduced to deliver recombinant follicle-stimulating hormone to women undergoing controlled ovarian stimulation for in vitro fertilization or other forms of assisted reproductive technology. The pen device makes it easier and less painful for women to self-administer daily injections. The high precision and accuracy of the device enables physicians to fine-tune dosage. The woman simply inserts the multidose cartridge into the Puregon (Follistim) Pen, attaches a BD Micro-Fine Pen Needle, selects the prescribe

The role of parietal cortex in awareness of self-generated movements: a transcranial magnetic stimulation study.

Awareness of self-generated movements arises from comparing motor plans, and the accompanying (hypothetical) efference copy, with the visual and proprioceptive consequences of movement. Here we used repetitive transcranial magnetic stimulation (rTMS) to investigate the role of a posterior region in the superior parietal lobule (SPL) in this process. Nine healthy volunteers performed a finger extension actively and passively while wearing a CyberGlove; the glove recorded these (actual) finger movements and used this information in real time to move a virtual hand displayed on a computer screen.

The appetite suppressant, d-fenfluramine, decreases self-stimulation at a feeding site in the lateral hypothalamus.

In prior studies rats showed a relative shift from self-stimulation to escape (i.e., from reward to aversion) following a large meal, obesity or anorectic doses of insulin. Racemic fenfluramine, on the other hand, decreased both self-stimulation and escape suggesting it had a general behavior suppressant property. To avoid the depressive, antidopaminergic effects of the l-isomer, this study tested the d-isomer which is primarily serotonergic. Rats were screened for stimulation-induced feeding and then trained to self-stimulate with one lever in 5-min periods that alternated with 5-min periods

[Interrelationship between the lateral hypothalamic self-stimulation reaction and the periodic motor activity of the stomach].

In 26 dogs, self-stimulation of the dorsal portion of the lateral hypothalamus at the level of anterior parts of ventromedial nuclei inhibits completely the stomach motor activity. Self-stimulation of the lateral portion of the middle hypothalamus at the level of posterior parts of ventro--medial nuclei does not affect at all the periodic contractions of the stomach. The intensity of self-stimulation depends on phase of the stomach periodic activity: during the contractions the number of lever-pressings is by 20% higher on the average than at resting. Bilateral supra-diaphragmal vagotomy incre

Self-stimulation of the brain; its use to study local effects of hunger, sex, and drugs.

My conclusions are these: (i) The cells which mediate primary rewarding effects are located in a midline system running from the midbrain through the hypothalamus and midline thalamus and into the subcortical and cortical groups of the rhinencephalon. (ii) The cell groups which mediate primary rewarding effects are different from those which mediate primary punishing effects. (iii) Despite this relative independence, there are, undoubtedly, relationships of mutual inhibition existing between these two systems. Rewards do, among other things, tend to reduce sensitivity to pain, and punishments

Examining the influence of brain stimulation to the medial prefrontal cortex on the self-reference effect in memory.

Past work shows that processing information in relation to the self improves memory which is known as the self-reference effect in memory. Other work suggests that transcranial direct current stimulation (tDCS) can also improve memory. Given recent research on self-reference context memory effects (improved memory for contextual episodic details associated with self-referential processing), we were interested in examining the extent stimulation might increase the magnitude of the self-reference context memory effect. In this investigation, participants studied objects superimposed on different

Differential effects of nomifensine and imipramine on motivated behavior in the runway model of intracranial self-stimulation.

A motivational deficit (the loss of pleasure or interest in previously rewarding stimuli) is one of the core symptoms of major depression, and valid models evaluating the motivational effects of drugs are needed. It was recently demonstrated that the priming stimulation effect in the runway model of intracranial self-stimulation (ICSS) can be used as a model system to study the motivational effects of drugs. However, the characteristics of this novel experimental model have not been fully clarified. In this study, we investigated the effects of nomifensine and imipramine in the runway ICCS mod

[The extended amygdala system and self-stimulation of the lateral hypothalamus in rats: modulation with opiates and opioids].

Wistar male rats were implanted with bipolar electrodes in the lateral hypothalamus to study self-stimulation reaction in the Skinner box. Simultaneously, the microcanules were implanted into the central nucleus of the amygdala to inject the drugs studied (1 microl in volume for each injection). The blockade of CRF receptors (astressin 1 microg) or sodium influx ionic currents (xycaine, or lidocain 1 microg) by means of intrastructural administration of drugs into the amygdala descreased self-stimulation reaction of the lateral hypothalamus in rats by 29-55%. A randomized comparison of thalamic stimulation and lesion on self-paced finger movement in essential tremor.

Deep brain stimulation (DBS) and ablation (thalamotomy) of the motor thalamus reduce tremor and improve function of the contralateral hand in patients with essential tremor (ET). Neuroimaging and electrophysiological evidence suggest that unlike a focal lesion, high frequency stimulation affects widespread neural networks that include those involved in motor timing. The purpose of this pilot study was to compare the effects of thalamic stimulation and lesion on the timing of simple, self-paced finger movements in patients with ET. Twenty-one subjects with advanced ET were randomized to unilate

Isoosmotic shrinkage by self-stimulated outward Na-K-Cl cotransport in quail erythrocytes.

In mammalian erythrocytes, outward fluxes by the Na-K-Cl cotransporter NKCC have been clearly characterized, but NKCC fluxes are small and their physiological role, if any, is poorly understood. Avian erythrocytes are nucleated cells, in which a physiologically relevant NKCC acts as a cell volume regulator. Therefore, we further investigated outward cotransport and its relation to cell volume by using quail erythrocytes. Unlike human or rat erythrocytes, quail erythrocytes exhibit outward cotransport fluxes: (1) of high magnitude [maximal rate of bumetanide-sensitive Li+ efflux=12.3+/-1.1 mmol

AAV-only targeting of ventral tegmental area dopamine neurons for optical self-stimulation studies in mice.

Studies employing optogenetic approaches in rodent models have highlighted the important contribution of ventral tegmental area (VTA) dopamine (DA) neurons to reward, learning, and motivation. Selective manipulation of VTA DA neurons is generally achieved in these studies using transgenic mouse or rat lines that express Cre recombinase under the control of a promoter active in DA neurons, combined with intra-VTA infusion of adeno-associated virus (AAV) vectors harboring Cre recombinase-dependent expression cassettes. Reliance on transgenic Cre driver lines is expensive and decreases study effi

Bioactive Self-Adaptive Hydrogel Platform for Integrated Electrical Stimulation Therapy and Clinical Monitoring of Postoperative Wound.

Postoperative wound management remains clinically challenging due to delayed healing, subcutaneous infections, and the absence of systems capable of real-time therapeutic and diagnostic integration. Addressing this need, we developed a bioactive, self-adaptive hydrogel platform that combines electrical stimulation (ES) therapy with resistance-based bioelectronic sensing for personalized postoperative wound care. The hydrogel is constructed from dynamic borate ester-cross-linked poly(vinyl alcohol) (PVA), with wool keratin for hemostasis and tissue remodeling and tannic acid for adhesion, antio

Inflammation-, immunothrombosis,- and autoimmune-feedback loops may lead to persistent neutrophil self-stimulation in long COVID.

Understanding the pathophysiology of long COVID is one of the most intriguing challenges confronting contemporary medicine. Despite observations recently made in the relevant molecular, cellular, and physiological domains, it is still difficult to say whether the post-acute sequelae of COVID-19 directly correspond to the consequences of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection. This work hypothesizes that neutrophils and neutrophil extracellular traps (NETs) production are at the interconnection of three positive feedback loops which are initiated in the acute pha

Can self-powered piezoelectric materials be used to treat disc degeneration by means of electrical stimulation?

Intervertebral disc degeneration (IDD) due to multiple causes is one of the major causes of low back pain (LBP). A variety of traditional treatments and biologic therapies are currently used to delay or even reverse IDD; however, these treatments still have some limitations. Finding safer and more effective treatments is urgent for LBP patients. With increasing reports it has been found that the intervertebral disc (IVD) can convert pressure loads from the spine into electrical stimulation in a variety of ways, and that this electrical stimulation is of great importance in modulating cell beha

Allogeneic offspring produced by induction of PD-L1 in spermatogonial stem cells via self-renewal stimulation.

The testis is an immune-privileged organ. It is considered that the testis somatic microenvironment is responsible for immune suppression. However, immunological properties of spermatogonial stem cells (SSCs) have remained unknown. Here, we report the birth of allogeneic offspring by enhanced expression of immunosuppressive PD-L1 in SSCs. In vitro supplementation of GDNF and FGF2 increased expression of PD-L1 in SSCs. Cultured SSCs maintained allogeneic spermatogenesis that persisted for >1 year. However, depletion or gene editing of Pd-l1 family genes in SSCs prevented allogeneic spermatogene

High-Frequency Stimulation at the Subthalamic Nucleus Suppresses Excessive Self-Grooming in Autism-Like Mouse Models.

Approximately one quarter of individuals with an autism spectrum disorder (ASD) display self-injurious behavior (SIB) ranging from head banging to self-directed biting and punching. Sometimes, these behaviors are extreme and unresponsive to pharmacological and behavioral therapies. We have found electroconvulsive therapy (ECT) can produce life-changing results, with more than 90% suppression of SIB frequency. However, these patients typically require frequent maintenance ECT (mECT), as often as every 5 days, to sustain the improvement gained during the acute course. Long-term consequences of s
