Effects of semaglutide on subclinical cardiovascular health in people with HIV.

DESIGN AND OBJECTIVE: Lipohypertrophy, characterized by central adipose tissue accumulation, is common in people with HIV (PWH) and contributes to cardiometabolic risk. In a recent randomized, double-blind, placebo-controlled phase 2b trial, semaglutide significantly improved weight, total fat, visceral adiposity, blood pressure, glucose metabolism, and lipids over 32 weeks in PWH with lipohypertrophy. This prespecified secondary analysis evaluated semaglutide's effects on subclinical cardiovascular health. METHODS: Virologically suppressed adults with HIV without known diabetes, receiving st

Semaglutide improves markers of cardiovascular risk in people with HIV.

OBJECTIVE: Semaglutide improves cardiovascular disease (CVD) risk in people who are diabetic, overweight, or obese through incompletely understood mechanisms. To address this, we explored novel lipidomic and lipoprotein/glycoprotein profiling with semaglutide therapy. DESIGN: Secondary analysis of SLIM LIVER (ACTG A5371), an open-label, phase 2b, single-arm trial of 1 mg semaglutide weekly in adult people with HIV (PWH) and metabolic dysfunction-associated steatotic liver disease. METHODS: Participants ( n  = 36) experiencing clinical response (>5 lb weight loss) to semaglutide were included

Cost-Effectiveness of Semaglutide in Patients With Obesity and Cardiovascular Disease.

BACKGROUND: Randomized clinical trials have shown that semaglutide is associated with a clinically relevant reduction in body weight and a lower risk of adverse cardiovascular events in those who are overweight or obese with a history of cardiovascular disease but no diabetes. The objective of this study was to assess the cost-effectiveness of semaglutide for this indication. METHODS: A decision analytic Markov model was used to compare the lifetime benefits and costs of semaglutide 2.4-mg subcutaneous weekly vs standard care in a hypothetical cohort of patients who were overweight or obese w

Sociodemographic disparities in semaglutide and tirzepatide prescription for obesity treatment.

INTRODUCTION: Semaglutide and tirzepatide, the 2 most recently approved glucagon-like peptide-1 receptor agonists, have demonstrated substantial effectiveness in weight management. However, evidence on treatment inequities remains limited. This study assesses temporal trends and sociodemographic disparities in semaglutide and tirzepatide prescription. METHODS: Using claims data from Komodo Health, we identified commercially insured individuals aged 18-64 years with overweight or obesity but without type 2 diabetes. We estimated 6-month cumulative incidence of semaglutide and tirzepatide presc

Angiogenesis and inflammation in the retinopathy risk of insulin and semaglutide - a review.

PURPOSE: To critically evaluate evidence supporting the angiogenesis-inflammation hypothesis in diabetic retinopathy (DR); to examine the roles of angiogenic growth factors and inflammatory cytokines in the biphasic effects of insulin on DR - early worsening followed by long-term risk reduction; and to explore broader implications of this hypothesis, including potential biphasic effects of semaglutide due to its insulinotropic action and strategies to mitigate the transient DR exacerbation associated with insulin and insulin secretagogues. METHODS: Literature review. RESULTS: Hyperglycemia and

Transient Hyperprolactinemia Associated With Semaglutide in a Patient With Hashimoto's Thyroiditis.

Semaglutide, a glucagon-like peptide-1 (GLP-1) receptor agonist, is widely used for weight management and glycemic control. While its metabolic and gastrointestinal effects are well characterized, its potential influence on pituitary function remains underexplored. To our knowledge, this appears to be among the first published reports of transient hyperprolactinemia associated with semaglutide use in a middle-aged woman with well-controlled Hashimoto's thyroiditis. The patient developed prolactin levels approaching 100 ng/mL without clinical symptoms or MRI evidence of pituitary adenoma. Prola

Targeting neuroinflammation in neurodegenerative disorders: the emerging potential of semaglutide.

BACKGROUND: Chronic neuroinflammation is increasingly recognized not as a secondary effect but as a primary driver of neurodegenerative disease progression. In conditions such as Alzheimer's disease (AD), Parkinson's disease (PD), multiple sclerosis (MS), amyotrophic lateral sclerosis (ALS), frontotemporal dementia (FTD), Huntington's disease (HD), and Lewy body dementia (LBD), dysregulated glial activity, marked by sustained microglial and astrocytic activation, initiates a cascade of cytokine release, oxidative stress, and impaired neuronal support. This review synthesizes recent advances in

Mechanism of Semaglutide in MASLD Treatment: Where Is the Master Key?

BACKGROUND: Semaglutide, a glucagon-like peptide-1 receptor agonist (GLP-1 RA), is used for weight reduction and metabolic improvement. Semaglutide effectively improves MASH but not confers histological fibrosis resolution. The key question remains whether semaglutide exerts direct, independent effects in ameliorating MASH or if its benefits are merely secondary to improvements in weight, insulin resistance, and glycemic control. The exact mechanisms of action through which semaglutide or other GLP-1 RAs offer liver protection are not entirely clear. METHODS: A comprehensive search in PubMed

The Efficacy of Semaglutide on Hypothalamic Obesity Caused by Craniopharyngioma Surgery.

BACKGROUND: Hypothalamic obesity, weight gain dominantly due to hypothalamic dysfunction, is a challenge for patients with craniopharyngioma surgery. Semaglutide, a long-acting GLP-1 receptor agonist, has shown promise in obesity management. However, the efficacy in this specific patient population remains underexplored. OBJECTIVE: This study aimed to evaluate the efficacy of semaglutide on weight reduction in patients with craniopharyngioma surgery. METHODS: A prospective clinical study was conducted involving 23 patients with obesity after craniopharyngioma surgery who attended the outpati

Blinded by the drug? A vigilyze signal linking semaglutide to NAION.

INTRODUCTION: Semaglutide, available widely as Ozempic, Wegovy, and Rybelsus, has significantly improved the management of type 2 diabetes and obesity due to its beneficial adverse drug reactions and safer use in diverse population. However, as its use grows widely, rare but serious side effects are coming to light. One such emerging concern is Non-Arteritic Anterior Ischemic Optic Neuropathy (NAION), a condition causing sudden, often irreversible vision loss. METHODOLOGY: A retrospective, post-market observational study was conducted using data collected from VigiBase® until June 2025, using

Population pharmacokinetic of paracetamol and atorvastatin with co-administration of semaglutide.

Semaglutide is a glucagon-like-peptide-1 (GLP-1) analogue marketed for once-weekly subcutaneous administration for type 2 diabetes mellitus. Like other long-acting GLP-1 analogues, semaglutide reduces gastric emptying and, potentially, alters the rate of absorption of orally co-administered drugs. The objective of the current analysis was to evaluate the effects on the gastric emptying rate caused by semaglutide on pharmacokinetic model parameters of paracetamol and atorvastatin in healthy subjects. Non-linear mixed effect modeling was used to estimate population pharmacokinetic model paramete
