A capsaicin-receptor homologue with a high threshold for noxious heat.

Pain-producing heat is detected by several classes of nociceptive sensory neuron that differ in their thermal response thresholds. The cloned capsaicin receptor, also known as the vanilloid receptor subtype 1 (VR1), is a heat-gated ion channel that has been proposed to mediate responses of small-diameter sensory neurons to moderate (43 degrees C) thermal stimuli. VR1 is also activated by protons, indicating that it may participate in the detection of noxious thermal and chemical stimuli in vivo. Here we identify a structurally related receptor, VRL-1, that does not respond to capsaicin, acid o

The expression of P2X3 purinoreceptors in sensory neurons: effects of axotomy and glial-derived neurotrophic factor.

We have studied the distribution and regulation of the P2X3 receptor (a ligand-gated ion channel activated by ATP) in adult dorsal root ganglion (DRG) neurons using a polyclonal antibody. P2X3 receptor immunoreactivity was normally present in about 35% of L4/5 DRG neurons, virtually all small in diameter. In the dorsal horn, P2X3 receptor expression was restricted to the terminals of sensory neurons terminating in lamina IIinner. P2X3 receptors were expressed in approximately equal numbers of sensory neurons projecting to skin and viscera but in very few of those innervating skeletal muscle. P

Neurotransmission and neuromodulation in the chemosensory carotid body.

The mammalian carotid body is a small chemosensory organ that helps maintain the chemical composition of arterial blood via reflex control of ventilation. Thus, in response to decreased PO2 (hypoxia), increased PCO2 (hypercapnia), or decreased pH (acidity), chemoreceptor glomus or type I cells become stimulated and release neuroactive agents that excite apposed sensory terminals of the carotid sinus nerve. The resulting increase in afferent discharge ultimately leads to corrective changes in ventilation so as to maintain blood gas and pH homeostasis. Recent evidence that the organ can also sen

Sensory behaviors in marine organisms.

Marine environments are vast, dynamic, and complex, posing significant challenges for the release and perception of sensory information that differ significantly from their terrestrial counterparts. Despite increasing interest, our knowledge about sensory behaviors in marine organisms remains relatively narrow. Recent advances in marine sensory biology have started to investigate how animals adapt to the specific constraints of aquatic environments to gather and respond appropriately to sensory information, ensuring their survival. New information about sensory organs, their underlying neurona

Replacement of receptor neurones after section of the vomeronasal nerves in the adult mouse.

Eight days after vomeronasal nerve section or removal of the accessory olfactory bulb, the majority of receptor cells of the vomeronasal neuroepithelium degenerate and disappear, leaving a regular framework consisting of supporting cells and their radial processes. The cell clusters at the boundaries of the epithelial sheet (which have been shown to be actively dividing in the normal, unoperated adult mouse) are also spared. The epithelium is subsequently repopulated by receptor cells appearing first in the basal part of the receptor cell layer and later occupying the full width of the recepto

TEX15 safeguards neuronal diversity in the mouse olfactory system.

Animals detect diverse chemicals using hundreds of chemoreceptor proteins. In many organisms, sensory chemoreceptors obey a one-receptor-type-per-cell rule, which organizes information gathered by sensory neurons1. A key question remains how receptor expression gets diversified so that hundreds of chemoreceptors get expressed across the population of sensory neurons2-4. Here we show that diverse odorant receptor (OR) expression in mice depends on testis expressed gene 15 (Tex15). We find that Tex15 regulates OR expression in neuronal precursor cells. In Tex15 deficient (Tex15-/-) mice, OR tran

Enteric P2X receptors as potential targets for drug treatment of the irritable bowel syndrome.

The irritable bowel syndrome (IBS) is a gastrointestinal motility disorder affecting millions of patients. IBS symptoms include diarrhea, constipation and pain. The etiology of IBS is due partly to changes in the function of nerves supplying the gastrointestinal tract, immune system activation and to psychological factors. P2X receptors are multimeric ATP-gated cation channels expressed by neuronal and non-neuronal cells. Sensory nerve endings in the gastrointestinal tract express P2X receptors. ATP released from gastrointestinal cells activates P2X receptors on sensory nerve endings to stimul

Kainate Receptors in Sensory Neurons: Molecular Identity and Functional Relevance to Pain.

Kainate receptors (KARs) are members of the ionotropic glutamate receptor family and possess diverse structural and functional properties that play critical roles in synaptic signaling, plasticity, and neural development. Several KAR subunits are strongly expressed in dorsal root ganglion neurons, and while their involvement in pain has been suggested, their precise role remains unclear. This review re-evaluates the roles of KARs in sensory physiology, with a particular focus on pain mechanisms. By combining recent single-cell transcriptomic data from dorsal root ganglia neurons with experimen

[Androgen-mediated pain-regulatory mechanisms targeting primary sensory neurons].

Given the sex differences in pain sensitivity and the prevalence of pain disorders, there is a strong need to elucidate their molecular basis. Because sex differences arise from the interactions between genetic factors and postnatal influences mediated by sex hormones, understanding how sex steroid hormones regulate pain is of particular importance. The mechanical pain threshold is lower in females than in males, whereas no sex differences are observed in thermal, chemical or inflammatory pain, consistent with findings in humans. In gonadectomized male mice, the mechanical threshold is decreas

Pioneer statoacoustic neurons guide neuroblast behaviour during otic ganglion assembly.

Cranial ganglia are aggregates of sensory neurons that mediate distinct types of sensation. The statoacoustic ganglion (SAG) develops into several lobes that are spatially arranged to connect appropriately with hair cells of the inner ear. To investigate the cellular behaviours involved in the 3D organization of the SAG, we use high-resolution confocal imaging of single-cell, labelled zebrafish neuroblasts (NBs), photoconversion, photoablation, and genetic perturbations. We show that otic NBs delaminate out of the otic epithelium in an epithelial-mesenchymal transition-like manner, rearranging

Sensory neurons drive pancreatic cancer progression through glutamatergic neuron-cancer pseudo-synapses.

Cancers thrive on neuronal input. Here, we demonstrate the presence of pseudo-synaptic connections between sensory nerve endings and cancer cells in an extracerebral cancer, i.e., pancreatic ductal adenocarcinoma (PDAC). These synaptic sites exhibit a selective enrichment of the glutamatergic N-methyl-D-aspartate receptor (NMDA) receptor subunit NMDAR2D (GRIN2D) on the cancer cells, which turns PDAC cells responsive to neuron-derived glutamate and promotes tumor growth and spread. Intriguingly, neurons transform a subset of co-cultured PDAC cells into calcium-responsive cells via GRIN2D-type g

Receptor cell regeneration and connectivity in olfaction and taste.

The capacity of adult mammalian gustatory and olfactory receptor cells to regenerate and make synaptic reconnections provides examples that may be useful in initiating replacement of other kinds of sensory receptor cells. The sensory code for taste quality may not be degraded by taste receptor cell turnover because axons probably recouple to the appropriate type of new receptor cell by axon-receptor cell affinity. Experiments on the development and regeneration of taste receptor cells suggest that they regenerate and turn over by recapitulating the late but not the early steps in taste bud dev

TRPV1 and TRPA1 in cutaneous neurogenic and chronic inflammation: pro-inflammatory response induced by their activation and their sensitization.

Cutaneous neurogenic inflammation (CNI) is inflammation that is induced (or enhanced) in the skin by the release of neuropeptides from sensory nerve endings. Clinical manifestations are mainly sensory and vascular disorders such as pruritus and erythema. Transient receptor potential vanilloid 1 and ankyrin 1 (TRPV1 and TRPA1, respectively) are non-selective cation channels known to specifically participate in pain and CNI. Both TRPV1 and TRPA1 are co-expressed in a large subset of sensory nerves, where they integrate numerous noxious stimuli. It is now clear that the expression of both channel

Comparison of neurogenesis and cell replacement in the hamster olfactory system with and without a target (olfactory bulb).

The olfactory sensory neurons are unique in the vertebrate nervous system in that they are replaced following experimentally induced degeneration. Unilateral removal of the olfactory bulb in hamster results in degeneration of all mature receptor neurons followed by a neurogenesis and partial replacement of the receptor cell population. To determine if full recovery requires the presence of normal target tissue, a study of sensory neuron replacement was made following a nerve transection procedure, which leaves the olfactory bulb (target) intact. A comparison of quantitative measurements of cel

Extrasynaptic receptors on cell bodies of neurons in central nervous system of the leech.

1. A systematic study has been made of the sensitivity of identified sensory and motoneurons in the leech central nervous system to chemical transmitter substances. 2. The following substances elicited responses from the cell bodies of individual neurons: acetylcholine, 5-hydroxytryptamine, gamma-aminobutyric acid, glutamic acid, glycine, dopamine, and norepinephrine. Since the cell bodies of leech neurons are free of synapses, the receptors that give rise to these responses are extrasynaptic. 3. Sensory and motoneurons of different function had characteristic complements of extrasynaptic rece

Pathophysiological Roles of Ion Channels in Epidermal Cells, Immune Cells, and Sensory Neurons in Psoriasis.

Psoriasis is a chronic inflammatory skin disease characterized by the rapid abnormal growth of skin cells in the epidermis, driven by an overactive immune system. Consequently, a complex interplay among epidermal cells, immune cells, and sensory neurons contributes to the development and progression of psoriasis. In these cellular contexts, various ion channels, such as acetylcholine receptors, TRP channels, Ca2+ release-activated channels, chloride channels, and potassium channels, each serve specific functions to maintain the homeostasis of the skin. The dysregulation of ion channels plays a

Satellite cells of sensory neurons after various types of sciatic nerve trauma in the rat.

Sciatic nerve crushing, transection, and ligation models were used in rats to study the reactions of and changes in the numbers of satellite cells (SC) in spinal dorsal root ganglia in the lumbar segment. Nerve transection was followed by the appearance of neurons surrounded by two layers of SC. The thickness of SC processes and the areas of contacts with neurons increased as a result of invaginations into neuron perikarya. After nerve ligation, SC and their processes were located around parts of large and intermediate neurons in several tightly appressed layers; the area of contact between SC

[Development of human olfactory receptors].

The development of the olfactory epithelium in 7-, 8-, 9-, 10- and 11-week old human fetuses was followed using electron microscopy. The differentiation of receptor and supporting cells and the establishment of Bowman's glands and the formation of olfactory nerve have been observed. In 7 week old fetuses the development of peripheral processes with olfactory knobs, lacking hairs, and of central processes with growth cones penetrating the connective tissue has been followed. A typical olfactory knobs possessing hairs were first found in 9 week old fetuses, the number of those progressively incr
