Serotyping Coxiella burnetii isolates from acute and chronic Q fever patients by using monoclonal antibodies.

Four mouse monoclonal antibodies reacting with Coxiella burnetti lipopolysaccharide antigens were produced and used in serotyping 17 C. burnetii isolates from acute Q fever and Q fever endocarditis patients in France. Two monoclonal antibodies (1B2 and 3B6) were considered specific for the Priscilla strain, a representative of Q fever endocarditis isolates, and did not react with the Nine Mile strain, which is representative of acute Q fever isolates. Monoclonal antibodies Nos. 1B2 and 3B6 reacted with 75% (3/4) acute Q fever isolates and 85% (11/13) of endocarditis isolates from France. It is

Streptococcus pneumoniae serotype 33H: a novel serotype with frameshift mutations in the acetyltransferase gene wciG.

BACKGROUND: Streptococcus pneumoniae (the pneumococcus) is a leading cause of community-acquired pneumonia. Pneumococci are categorised into serotypes, based on the type of capsular polysaccharide produced, which has important implications for virulence, vaccine impact and global surveillance. Recently, we identified a novel serotype, which we named 33G, that is comprised of an O-acetylated hexasaccharide repeat unit. In this study, we report and describe variants of 33G, designated 33G-like, which we isolated from the nasopharynx of two adults hospitalised with pneumonia in Mongolia. METHODS

Preexisting inter-serotype immunity drives antigenic evolution of dengue virus serotype 2.

Dengue virus (DENV) infects roughly 400 million people annually, causing febrile and hemorrhagic disease. While preexisting inter-serotype immunity (PISI) provides transient protection, it may drive severe disease over time. PISI's impact on virus evolution, however, is less understood. Retrospective epidemiological analyses suggest that PISI may drive DENV evolution. Using in vitro directed evolution, we explored how DENV2 evolves in the presence of DENV3/4 convalescent serum. Two post-passaging mutations (E-I6M and E-N203D) were then studied for fitness effects in mammalian and insect hosts

The efficacy of a live Listeria monocytogenes combined serotype 1/2a and serotype 4b vaccine.

In each of two experiments, sheep and lambs were vaccinated by a subcutaneous injection of a test vaccine (consisting of a combined serotype 1/2a and serotype 4b live Listeria monocytogenes culture) and challenged 16 days later with a mixture of the homologous wild strains. After challenge, the mortality rate of vaccinated sheep was 28.1% and that of controls 71.9%; that of lambs was 25.0%, although these had been inoculated with the LD70 dose. Furthermore, in each of two field trials in Listeria-infected flocks, primiparous pregnant ewes were vaccinated. In the first field trial, 3 or 110 lam

What is driving the resurgence and persistence of vaccine-targeted pneumococcal serotypes?-Serotype 19F as the paradigm.

Over the past 25 years, pneumococcal conjugate vaccines (PCVs) have markedly reduced both pneumococcal disease and nasopharyngeal carriage caused by vaccine serotypes among the more than 100 known pneumococcal serotypes. In the United States, the transition from the original 7-valent formulation (PCV7) to the 13-valent vaccine (PCV13) occurred approximately a decade after the initial introduction of PCVs, whereas several other countries implemented higher-valency formulations over shorter time intervals. More recently, next-generation PCVs targeting 15 (PCV15) or 20 (PCV20) serotypes have been

Expansion of Dengue Virus Serotype 3 Genetic Diversity and Changing Serotype Composition in Brazil.

Brazil is the most hyperendemic country for dengue, with historically dominant circulation of DENV-1 and DENV-2. Bayesian phylodynamic analyses of 489 Brazilian DENV-3 genomes, including 84 newly generated sequences, reveal a recent and sustained increase in the effective population size of DENV-3 associated with the recent introduction and expansion of the 3III_B.3.2 lineage. This increase coincides with a sharp rise in the proportion of laboratory-confirmed dengue cases attributed to DENV-3 among records with available serotype information from 2024 onwards and a shift in serotype dynamics i

Transcriptomic analysis of Streptococcus pneumoniae serotype 1 reveals serotype-specific gene regulation.

Streptococcus pneumoniae (Sp) is an opportunistic pathogen that colonizes the mucosal surfaces of the human upper respiratory tract. While transcriptomic studies of Sp have become more common, most have focused on laboratory-adapted strains such as D39 or TIGR4. These strains, though widely used in research, may not fully capture the biology of clinical isolates, particularly the hypervirulent serotype 1 (S1). S1 is clinically significant due to its association with invasive disease, epidemic outbreaks and a distinct global distribution, particularly in regions with a high pneumococcal disease

[Emergence of invasive nonvaccine pneumococcal serotypes: 2 cases involving 19A and 10A serotypes].

The authors report 2 cases of invasive pneumococcal disease. One case consisted in occult bacteremia due to a 19A serotype in a child who had received 3 doses and a booster dose of the heptavalent pneumococcal conjugate vaccine (Prevenar). The 2nd case was pneumococcal meningitis due to a 10A serotype. The incidence of nonvaccine serotypes is increasing, notably the 19A serotype. They must be closely monitored because of the high antimicrobial resistance of certain strains.

Association of meningococcal serotypes with the course of disease: serotypes 2a and 2b in the Netherlands, 1959-1981.

Case histories of 692 patients with meningococcal disease due to serogroup B, C, or W (W-135) were reviewed to study the association of the serotypes 2a and 2b with the course of disease. The case-fatality rate in group B disease was significantly associated with serotype 2b (B:2b) strains (P = 0.03). Age and year of admission did not account for this association. Septicemia was also found more frequently with B:2b than with other B serotypes, but neurological complications and sequelae were not. Neither C:2a nor W:2a was associated with a higher case-fatality rate, with more cases of septicem

Population structure analysis of Salmonella serovar Muenchen to redefine geno-serotyping using genome indexing approaches.

Accurate identification of Salmonella serovars for source attribution in foodborne illness outbreaks. Traditional serotyping, which relies on antigenic properties, continues to serve as gold standard; however, advances in whole-genome sequencing (WGS) have enabled to the development of in-silico serotyping tools such as SeqSero2 and Salmonella In Silico Typing Resource (SISTR). Genome-indexing methods, such as bettercallsal, integrate DNA sketching and genome proximity analysis, have emerged as a promising tool for improving serovar resolution. This study examines the performance of DNA sketch

Fourier-transform infrared spectroscopy for rapid Streptococcus pneumoniae serotyping in a tertiary care general hospital.

Streptococcus pneumoniae is the leading cause of community-acquired pneumonia and remains a significant contributor to bacteremia and meningitis, collectively known as invasive pneumococcal disease (IPD). Certain serotypes are more strongly associated with severe illness and antimicrobial resistance. Accurate serotyping is essential for effective IPD surveillance and vaccine development. Fourier-transform infrared (FTIR) spectroscopy has emerged as a valuable tool for differentiating among serotypes across various isolates. We analyzed 150 pneumococcal strains isolated from a tertiary hospital

Towards decentralization of Salmonella serotyping and risk assessment in poultry production environments with nanopore sequencing.

Salmonella enterica is a highly diverse group of organisms with over 2,500 serovars described to date. However, not all serovars are pathogenic to humans and animals. Many serovars typically cause self-limiting gastrointestinal infections, but certain host-adapted serovars, as well as those expressing specialized virulence factors (e.g., Vi capsule) have an increased potential to cause bacteremia and trigger systemic responses, necessitating immediate medical attention. Timely identification of Salmonella at the serovar level is, therefore, critical for informing risk assessments, guiding clin

Evaluation of Serotyping of Environmental and Clinical Isolates of Legionella pneumophila using MALDI-TOF MS.

Legionella pneumophila (L. pneumophila) is responsible for most Legionnaire's disease cases diagnosed worldwide. The species includes 16 serogroups, but most Legionnaire's disease cases (85.7% in Europe, 87.0% in Japan) are caused by L. pneumophila serogroup 1. Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) can be used to identify the L. pneumophila serogroup. In this study, we compared three sample preparation methods that are compatible with MALDI-TOF MS: the direct colony transfer method (DCTM), on-target extraction method (OTEM), and in-tube ext

Comparison of sequential multiplex PCR, sequetyping and whole genome sequencing for serotyping of Streptococcus pneumoniae.

Streptococcus pneumoniae is one of the major causes of pneumonia, meningitis and other pneumococcal infections in young children and elders. Determination of circulating S. pneumoniae serotypes is an essential service by public health laboratories for the monitoring of putative serotype replacement following the introduction of pneumococcal conjugate vaccines (PCVs) and of the efficacy of the immunization program. The Quellung method remains the gold standard for typing S. pneumoniae. Although this method is very effective, it is also costly, time consuming and not totally reliable due to its

Molecular serotyping and antimicrobial resistance profiles of Actinobacillus pleuropneumoniae isolated from pigs in South Korea.

BACKGROUND: Actinobacillus pleuropneumoniae (APP) causes porcine pleuropneumonia (PP). OBJECTIVE: Serotypes and antimicrobial resistance patterns in APP isolates from pigs in Korea were examined. METHODS: Sixty-five APP isolates were genetically serotyped using standard and multiplex PCR (polymerase chain reaction). Antimicrobial susceptibilities were tested using the standardized disk-agar method. PCR was used to detect β-lactam, gentamicin and tetracycline-resistance genes. The random amplified polymorphic DNA (RAPD) patterns were determined by PCR. RESULTS: Korean pigs predominantly carr

Multi-serotype pneumococcal nasopharyngeal carriage prevalence in vaccine naïve Nepalese children, assessed using molecular serotyping.

Invasive pneumococcal disease is one of the major causes of death in young children in resource poor countries. Nasopharyngeal carriage studies provide insight into the local prevalence of circulating pneumococcal serotypes. There are very few data on the concurrent carriage of multiple pneumococcal serotypes. This study aimed to identify the prevalence and serotype distribution of pneumococci carried in the nasopharynx of young healthy Nepalese children prior to the introduction of a pneumococcal conjugate vaccine using a microarray-based molecular serotyping method capable of detecting multi

Molecular characterization, serotyping, and antibiotic susceptibility profile of Leptospira interrogans serovar Copenhageni isolates from Brazil.

Leptospira interrogans serogroup Icterohaemorrhagiae is the major serogroup infecting humans worldwide, and rodents and dogs are the most significant transmission sources in urban environments. Knowledge of the prevalent serovars and their maintenance hosts is essential to understand the epidemiology of leptospirosis. In this study, 20 Leptospira isolates were evaluated by pulsed-field gel electrophoresis (PFGE), variable number tandem-repeat analysis (VNTR), serotyping, and determination of antimicrobial resistance profile. Isolates, originated from bovine, canine, human, and rodent sources,

Serotyping and antibiotic susceptibility of Streptococcus pneumoniae strains isolated in Algeria from 2001 to 2010.

INTRODUCTION: Pneumococcal infections are a major public health problem because of the virulence of this bacterium and its ability to develop resistance. MATERIAL AND METHOD: Two hundred and ninety-four strains of Streptococcus pneumoniae were isolated from sterile (56.8%) and non-sterile samples (43.2%), from January 2001 to July 2010. RESULTS: The interpretation of antibiotic susceptibility testing, according to CLSI criteria (M100-S21 2011), yielded a 25.2% overall resistance to penicillin, with 23.5% of strains isolated from CSF (meningitis), and only 1.7% in other samples. Multiplex PCR assay simplifies serotyping and sequence typing of Listeria monocytogenes associated with human outbreaks.

Listeria monocytogenes serotypes 1/2a and 4b are responsible for the majority of cases of human listeriosis worldwide. In this study, a multiplex PCR assay was developed to allow rapid identification and easily interpretable differentiation of serotypes 1/2a and 4b from other serotypes of L. monocytogenes by simultaneously targeting two virulence genes (inlB and inlC) and two serotype-specific genes (ORF2372 and Imo0171). A subsequent gel extraction and sequence typing analysis of the highly polymorphic intragenic regions in inlB and inlC simplified a previously developed multi-virulence-locus

An epidemiological survey of human listeriosis in France during 1987, using serotyping and phage typing.

During 1987 the French National Reference Center for Listeria received, from this country, 384 Listeria monocytogenes strains isolated from human listeriosis cases. A constant exchange of information and strains between the Reference Center and the Bacterial Ecology Unit of the Institute Pasteur of Paris allowed complete characterization of these isolates, using serotyping and phage typing. Among the strains studied 22%, 13% and 63%, respectively belonged to serovars 1/2a, 1/2b, and 4b, but this distribution can vary according to place, and time of isolation. Only 58% of strains were phage-typ
