A Multi-Omic Huntington's Disease Transgenic Sheep-Model Database for Investigating Disease Pathogenesis.

BACKGROUND: The pathological mechanism of cellular dysfunction and death in Huntington's disease (HD) is not well defined. Our transgenic HD sheep model (OVT73) was generated to investigate these mechanisms and for therapeutic testing. One particular cohort of animals has undergone focused investigation resulting in a large interrelated multi-omic dataset, with statistically significant changes observed comparing OVT73 and control 'omic' profiles and reported in literature. OBJECTIVE: Here we make this dataset publicly available for the advancement of HD pathogenic mechanism discovery. METHO

Antibiotic Treatment Response of Chronic Lung Diseases of Adult Sheep in the United Kingdom Based upon Ultrasonographic Findings.

Examination of the lungs of adult sheep with chronic respiratory diseases was readily achieved using both 5 MHz linear and sector scanners. Superficial lung abscesses in eight sheep appeared as anechoic areas containing multiple hyperechoic dots bordered distally by a broad hyperechoic capsule. Unilateral fibrinous pleurisy (2 sheep) appeared as an anechoic area containing a hyperechoic latticework. Ovine pulmonary adenocarcinoma (OPA) lesions appeared as sharply demarcated hypoechoic areas in the lung parenchyma initially in the cranioventral lung lobes (21 sheep) with lesions also present in

Performance deficits in SRLV-infected sheep quantify progressive disease severity.

Managing Maedi-Visna in sheep flocks infected by Small Ruminant Lentivirus (SRLV) by limiting production losses and reducing transmission could be a more cost-effective approach than eliminating the disease. We quantified the progressive physiological costs of SRLV infection by analysing performance measures as readily accessible criteria of disease severity in data from a five-year study of a Polish flock with bi-annual tests of serostatus. SRLV infection had no effect on ewes' pre-natal reproductive traits but caused increased mortality and reduced growth rate in their lambs, and reduced ewe

Somatic CAG Repeat Stability in a Transgenic Sheep Model of Huntington's Disease.

Somatic instability of the huntingtin (HTT) CAG repeat mutation modifies age-at-onset of Huntington's disease (HD). Understanding the mechanism and pathogenic consequences of instability may reveal therapeutic targets. Using small-pool PCR we analyzed CAG instability in the OVT73 sheep model which expresses a full-length human cDNA HTT transgene. Analyses of five- and ten-year old sheep revealed the transgene (CAG)69 repeat was remarkably stable in liver, striatum, and other brain tissues. As OVT73 sheep at ten years old have minimal cell death and behavioral changes, our findings support inst

Hypoglycin A absorption in sheep without concurrent clinical or biochemical evidence of disease.

BACKGROUND: Hypoglycin A (HGA) intoxication after ingestion of Acer spp. tree material has never been confirmed in domesticated ruminants despite their similar grazing habitats. OBJECTIVES: To investigate whether sheep have low HGA bioavailability caused by rumen HGA breakdown. ANIMALS: Stomach and rumen fluid samples from 5 adult horses and 5 adult sheep respectively. Residual serum samples from 30 ewes and lambs. METHODS: Experimental and retrospective cohort study. Hypoglycin A concentration was quantified in horse gastric and sheep ruminal samples after in vitro incubation with Acer pse

Metabolic profiling of presymptomatic Huntington's disease sheep reveals novel biomarkers.

The pronounced cachexia (unexplained wasting) seen in Huntington's disease (HD) patients suggests that metabolic dysregulation plays a role in HD pathogenesis, although evidence of metabolic abnormalities in HD patients is inconsistent. We performed metabolic profiling of plasma from presymptomatic HD transgenic and control sheep. Metabolites were quantified in sequential plasma samples taken over a 25 h period using a targeted LC/MS metabolomics approach. Significant changes with respect to genotype were observed in 89/130 identified metabolites, including sphingolipids, biogenic amines, amin

Disease phenotype in sheep after infection with cloned murine scrapie strains.

OBJECTIVES: Haematopoietic stem cell transplantation (HSCT) is a treatment option for patients with severe systemic sclerosis (SSc), but the efficacy of the procedure in remodelling the nailfold microvascular array is largely unknown. Therefore, this study aimed to evaluate the effect of HSCT on microangiopathy assessed through nailfold capillaroscopy (NC) and to compare the results with findings in patients receiving conventional immunosuppression. METHODS: We included SSc patients with severe SSc and whose pre- and post-treatment NC images were available. Findings in patients treated with H

An immunohistochemical study of the distribution of border disease virus in persistently infected sheep.

Three seronegative sheep persistently infected with Border disease virus and six seropositive, non-viraemic sheep were examined for the cellular distribution of the agent. These animals originated from a closed flock which had been kept in an isolation facility for 5 years. They were killed and immediately necropsied. There were no gross abnormalities other than reduced body weight of the persistently infected sheep. Two samples of each major organ were collected. The first sample was fixed by immersion in formalin and processed for histological examination, which showed no lesions unequivocal

Astylus atromaculatus (pollen beetle) gastrointestinal disease experimentally reproduced in sheep.

Intoxication of sheep and cattle by Astylus atromaculatus recently occurred in Uruguay and Argentina in association with severe drought. Although the disease was experimentally reproduced in sheep in the 1970s, there is limited information on clinical and pathologic findings of sheep experimentally intoxicated by this beetle. Here, we described the clinical, gross, and microscopic findings in 3 sheep orally dosed with A. atromaculatus (treatment group, TG) and in 2 control sheep (control group, CG) dosed with distilled water. Anorexia, lethargy, ruminal stasis, reluctance to move, prolonged re

Moraxella nasovis sp. nov., isolated from a sheep with respiratory disease.

A novel Gram-stain-negative, aerobic, coccus-shaped bacteria, designated ZY201115T, was isolated from the nasal cavity of a sheep with respiratory disease in Yunnan Province, south-west China, and its taxonomic affiliation was studied by applying a polyphasic approach. The strain grew at 18-41 °C (optimum, 37 °C), at pH 6.0-9.0 (optimum, pH 8.0) and in 0.5-3.0% (w/v) NaCl (optimum, 1.0 % NaCl). Phylogenetic analysis based on 16S rRNA gene sequences showed that the strain is affiliated to the genus Moraxella with highest similarity to Moraxella bovis ATCC 10900T (96.6 %). Phylogenomic analysis

A missense mutation in AGTPBP1 was identified in sheep with a lower motor neuron disease.

A type of lower motor neuron (LMN) disease inherited as autosomal recessive in Romney sheep was characterized with normal appearance at birth, but with progressive weakness and tetraparesis after the first week of life. Here, we carried out genome-wide homozygosity mapping using Illumina Ovine SNP50 BeadChips on lambs descended from one carrier ram, including 19 sheep diagnosed as affected and 11 of their parents that were therefore known carriers. A homozygous region of 136 consecutive single-nucleotide polymorphism (SNP) loci on chromosome 2 was common to all affected sheep and it was the ba

Chondrodysplasia of Texel sheep - a new disease of suspected genetic aetiology (abstract).

A chondrodysplasia characterised by dwarfism and varus deformity of the forelimbs has been recognised over four seasons in Texel and Texel-cross lambs on three related properties. Affected lambs appear normal at birth, but by 2 weeks of age have reduced growth rate, shortened neck and legs, varus forelimb deformities and a progressive reluctance to walk. Most die within the first 4 months of life. Some affected lambs have had normal co-twins. Post mortem examination of two severely affected 3-month-old lambs revealed extensive loss of articular cartilage and exposure of subchondral bone on the

[Some forms of disease associated with impaired locomotion in sheep (author's transl)].

A number of flock diseases in sheep associated with impaired locomotion, which are of importance or may assume importance in the Netherlands, are reviewed. Attention is paid to the following metabolic disorders and deficiencies: acetonaemia, hypocalcaemia, hypomagnesaemia, some forms of osteopathy, enzootic ataxia and cerebrocortical necrosis. The following forms of infectious diseases are described: listeric encephalitis, purulent meningitis following injury to the throat, scrapie, visna, Aujeszky's disease and border disease. Finally, a number of viral forms of encephalitis are discussed, wh

The influence of eroding Djallonke genotype on the incidence of diseases and mortalities in Sahelian x Djallonke cross-bred sheep.

Despite cross-breeding of Djallonke and Sahelian sheep due to the higher growth rate and final body weight of the Sahelian breeds over time, little scientific evidence exists on the ability of the crosses to withstand diseases compared to their Djallonke parents. A study involving 700 Djallonke and two crosses of the Djallonke and Sahelian breeds (50% Sahel × 50% Djallonke, 75% Sahel × 25% Djallonke) was conducted, and documented incidence of diseases, parasites and mortalities in these animals. Enteritis, Ehrlichia ruminantium and orf were more prevalent in 25% Djallonke than both 100% and 50
